Abstract

Ansamycins reduced to their dihydroquinones in the presence of common reducing agents in vitro have approximately 40-fold greater affinity than the corresponding oxidized quinones. The in vivo accumulation of the high-affinity dihydroquinone ansamycins in tumor cells contributes to the antitumor activity of these compounds and alters the understanding of the active species driving the efficacy of this class of compounds.

Abstract

Herein, we report a short, three-step synthesis of d-[1-14C]-serine (4) in high enantiomeric purity. Starting from [14C]-KCN and 2-(benzyloxy)acetaldehyde, Strecker reaction using ( R)-1-phenylethylamine as the chiral auxiliary gave two diastereomeric aminonitriles 1 and 2 in the ratio of 4:3, which were conveniently separated and purified chromatographically. Following hydrolysis and subsequent hydrogenolysis, the purified major diastereomer 1, was smoothly converted to d-[1-14C]-serine (4) in an enantiomeric excess of >99%, thus circumventing time intensive chiral HPLC enantiomeric resolution. 22. Synthesis of 2H, 13C and 14C labeled vasopressin V1aV2receptor antagonist RWJ676070 and its stable labeled Ndesbenzoyl metabolite

Abstract

Stable isotopelabeled 13C2, D3 and carbon 14labeled vasopressin receptor antagonist RWJ676070, a spirobenzazepine amide, were prepared in separate syntheses for use in drug metabolism studies. The stable isotopically labeled sample was prepared starting from 13C2, D6dimethyl sulfate and 13Ccopper I cyanide in nine steps with a 14 overall isotopic yield. The 14C label was introduced in five steps starting from 14Cpotassium cyanide to provide material having a specific activity of 58 mCimmol 2.15 GBqmmol. Selective hydrolysis of the metabolically labile amide bond in 13C2, D3RWJ676070 gave the corresponding labeled metabolite in one step. Copyright © 2008 John Wiley & Sons, Ltd.
