Although name, image, and likeness policy officially changed on July 1, 2021, actions leading up to this policy modification provide insight into the desires and perspectives of the National Collegiate Athletic Association (NCAA). Feeling pressure from individual states and federal legislators, the NCAA engaged in discussion regarding name, image, and likeness in Fall of 2019. In response to newly introduced name, image, and likeness policy changes, the NCAA listed their official statements on the Taking Actions: Name, Image and Likeness webpage. These statements (n = 10) were analyzed using critical discourse analysis methodology underpinned with a Foucaultian perspective. Using critical discourse analysis, we extrapolated three overarching themes related to power dynamics: (a) Establishing Control While Undercutting Oppositional Power, (b) Power Shifts Away from NCAA, and (c) Power Reinforcement. This study examined the way college football players used social media to resist, highlight, and address inequity in college football. Employing a critical discourse analysis guided by poststructuralism as a theoretical framework, three public statements were analyzed to explore how the language used in the statements resisted multiple discourses that shape college football players' experiences. The ways that college football players used discourse to mobilize as activists and exert control over their college athlete experience were considered. This study investigates the distribution of southern North American glacial refugial migrants that have invaded Yukon (Canada) since the Last Glacial Maximum (LGM), which began about 9500 years ago. The analysis was based on indigenous vascular plant occurrences that were stratified according to 0.33° latitudinal bands. Among 90,397 occurrence records, about 37% were postglacial migrants as opposed to former members of the Eastern Beringian flora. Most migrants probably moved northward from LGM refugia in the western U.S. region (Montana-Idaho-Wyoming) and, to a lesser extent, from the midwestern United States, but several were potentially from a Pacific coast refugium in westernmost Washington-Oregon-northern California. Gradients in taxa composition and richness occurred, with Beringian taxa increasing and migrant taxa decreasing at greater latitudes (+3 vs -0.5 taxa per degree, respectively) based on statistically significant regression trends. An interpretation of ordination results based on taxa presence and absence suggests south-to-north migrating plants are strongly influenced by climate. Only half of all Yukon migrant taxa have geographical ranges that extend beyond the latitudinal centre of the territory. Possible constraints on greater dispersal include competition from pre-existing vegetation, colder northward climates, and the presence of the Ogilvie and Wernecke Mountains across the latitudinal centre of the territory. Just over half (24 of 44) of the taxa that occurred in only one latitudinal band were located immediately north of the southern Yukon border, which suggests glacial refugial migrants are still invading Yukon. It is clear that the societal role of meat is being challenged with ideological and simplified logic without substantiation from robust data-driven science. With this background, the international summit titled 'The societal role of meat – what the science says' was held in Dublin, Ireland, during October 2022, to provide evidence-based evaluations and the Dublin Declaration was signed by over 1000 scientists. In this paper, we provide a synopsis of the summit and then give context for evaluating the societal role of meat in Australia. The key themes of the summit were the essential roles of meat in (1) diet and health, (2) a sustainable environment and (3) society, economics and culture. Evidence clearly showed the role of meat as a nutrient-dense source of high-quality protein and micronutrients that can be safely consumed by humans. Further, the complementary role of livestock in agricultural systems was highlighted with both plant- and animal-based agriculture reliant on each other to maximise the efficient production of food. Thus, from both an Australian and world perspective, very little food considered to be human-edible is fed to livestock. The role of livestock in rural societies across the world was emphasised to underpin regional and national economies, with particular importance in those countries with developing economies to facilitate growing wealth to 'step out' of poverty and provide gender equality. Meat production, particularly from ruminants, is a critical part of Australian primary production and it is concluded that the Dublin Declaration is highly relevant to Australia. Finally, concern regarding future funding and organisation of research and extension is discussed. There is a need to continue funding highly collaborative programs that bring a broad range of disciplines together, in conjunction with undergraduate and postgraduate teaching to underpin the social license to operate for meat and livestock production. Guarantee games are among the most important revenue streams for historically Black college and university (HBCU) intercollegiate athletics departments. Some scholars, however, have critiqued these games and even questioned their legality. This study examined the contracts of guarantee games in men's basketball to assess whether HBCUs and non-HBCUs are equally compensated for their participation in these games. The findings point to the existence of a basketball "Black tax" where HBCUs are not given equal compensation for their participation in men's college basketball guarantee games. Onychomycosis is a common nail infection. Terbinafine-resistant dermatophyte infections pose an emerging global public health concern, but few cases have been described in the United States. We retrospectively reviewed and characterized clinical, histopathological, and mycological features of patients with mycologically confirmed onychomycosis who failed oral terbinafine treatment for onychomycosis at a U.S. academic nail referral center and ascertained for terbinafine-resistant isolates. During 1 June 2022–31 January 2023 at Weill Cornell Medicine in New York City, USA, 96 patients with mycologically confirmed onychomycosis were treated with oral terbinafine. Among 64 patients with adequate follow-up, 36 had clinical or complete cure. Of 28 patients who failed treatment, 17 underwent terbinafine resistance testing. Trichophyton rubrum with terbinafine resistance-conferring mutations was isolated from two patients. Overall, terbinafine failures for onychomycosis were relatively common, with some cases associated with terbinafine-resistant T. rubrum infections. These findings underscore the need for a clinical awareness of this emerging problem and public health efforts to monitor and prevent spread. We highlight the importance of diagnostic testing and species identification for onychomycosis patients and the increasingly important role of fungal identification and susceptibility testing to guide therapy. The development of clinical congestion resulting from volume overload, either by renal fluid retention or redistribution of blood volume from venous reservoirs, is a recurrent scenario in patients with chronic heart failure (HF). As a result, the treatment of congestion, most commonly by initiating aggressive diuretic therapy, is a front-line issue in the management of patients with HF. However, the association of clinical congestion and volume overload with physical signs and symptoms, as well as other surrogates of volume assessment, has limitations in accuracy and, therefore, reliability to direct appropriate interventions. The ability to quantitate intravascular volume and identify the variability in volume profiles among patients with HF can uniquely inform individualized volume management and aid in risk stratification. This tool is provided by contemporary nuclear medicine-based BVA-100 methodology, which uses the well-established indicator-dilution principle and is a requested topic for discussion in this review. Intravascular volume is largely regulated by the kidneys but how differences in intravascular volume profiles interact with chronic kidney disease (CKD) to influence outcomes in chronic heart failure (HF) has not been explored. Our hypothesis was that a greater degree of volume expansion (VE) would moderate the impact of CKD on HF‐related clinical outcomes. Quantitative blood volume (BV) data were available in 137 patients at the time of hospital discharge using a nuclear medicine radiolabeled albumin indicator‐dilution technique. The study patients were stratified by the cohort median glomerular filtration rate (GFR, 44 ml/min/1.73 m2). An a priori cut‐point of ≥+25% above normal BV was then used to further stratify the two GFR subgroups and prospectively analyzed for 1‐year HF‐related mortality or 1st re‐hospitalization. Persistent BV expansions ≥+25% were present in 51% of the cohort. In the subgroup with GFR above the median (N = 68) greater or lesser BV expansion from +25% did not differentiate outcomes. However, in the subgroup with GFR below the median (N = 69), BV expansion‐stratified risk (log‐rank p = 0.022) with <+25% VE associated with poorer outcomes, while VE ≥ + 25% was associated with lower risk and comparable to GFR above the median. In patients with chronic HF, significant intravascular VE and CKD are common co‐existing conditions. The presence of larger VE, however, appears to be a factor mitigating the impact of declining renal function on clinical outcomes, and as an element of volume pathophysiology warrants further study. A Tn7-transposition approach was utilized for site-specific insertion of foreign genes into the genome of simian varicella virus (SVV), the causative agent of simian varicella in nonhuman primates. The severe acute respiratory syndrome coronavirus (SARS-CoV-2) nucleocapsid (N) gene and receptor binding domain (RBD) of the spike gene were inserted into the ORF 14 region of the SVV genome cloned into a bacterial artificial chromosome and then transfected into Vero cells to generate the infectious recombinant SVV (rSVV). The rSVV replicated efficiently in infected Vero cells and expressed the N and RBD antigens as indicated by immunoblot and immunofluorescence assays. Varicella zoster virus (VZV) and simian varicella virus (SVV) cause varicella (chickenpox) in children and nonhuman primates, respectively. After resolution of acute disease, the viruses establish latent infection in neural ganglia, after which they may reactivate to cause a secondary disease, such as herpes zoster. SVV infection of nonhuman primates provides a model to investigate VZV pathogenesis and antiviral strategies. The VZV and SVV genomes are similar in size and structure and share 70–75% DNA homology. SVV and VZV DNAs are co-linear in gene arrangement with the exception of the left end of the viral genomes. Viral gene expression is regulated into immediate early, early, and late transcription during in vitro and in vivo infection. During viral latency, VZV and SVV gene expression is limited to transcription of a viral latency-associated transcript (VLT). VZV and SVV are closely related alphaherpesviruses that likely arose from an ancestral varicella virus that evolved through cospeciation into species-specific viruses. [ABSTRACT FROM AUTHOR]

Extracorporeal membrane oxygenation (ECMO) can be life-saving, but suffers from thrombus formation in the circuit with associated risks of oxygenator occlusion, hemolysis and arterial embolism. The formation of thrombin is the key step to thrombus formation and two factors are needed for sustained thrombin generation, a coagulation activator to initiate the process and a procoagulant phospholipid surface for the coagulation system to assemble on. The purpose of this study was to use thrombin generation potential (TGP) and other assays to determine the specific coagulation activators and sources of procoagulant phospholipid that are present in ECMO patient plasma. Samples were collected from 60 patients on ECMO (age 1d-19y) followed evaluation of native and stimulated TGP, measurement of factor II levels and determination of procoagulant extracellular vesicle levels by flow cytometry. During ECMO, native (unstimulated) TGP was increased, followed by a decrease back towards normal after ECMO ended. The main activator of TGP in ECMO plasma was increased FXIa (100% of samples tested), while increased tissue factor activity was present in 7%. Procoagulant phospholipids were present in plasma from ECMO patients in the form of circulating platelet and red cell extracellular vesicles, which were increased 2 to 7-fold compared to normal. Procoagulant extracellular vesicle levels correlated with increased plasma native TGP activity. ECMO activates coagulation in plasma primarily through activation of the contact system and formation of activated factor XIa and generation of circulating procoagulant extracellular vesicles through platelet and red cell activation. • In pediatric patients, ECMO stimulates increased thrombin generation potential. • ECMO activates the contact system generating activated factor IXa. • ECMO increases procoagulant phospholipids through extracellular vesicle release. • Maquet circuits showed higher TGP and platelet activation than Cardiohelp circuits. RNA polymerase (RNAP) is a highly conserved macromolecular machine that contributes to the flow of genetic information from genotype to phenotype. In Bacillus subtilis, mutations in the rpoB gene encoding the b-subunit of RNAP have been shown to alter a number of global phenotypes, including growth, utilization of unusual nutrient sources, sporulation, germination, and production of secondary metabolites. In addition, the spectrum of mutations in rpoB leading to rifampin resistance (Rifr) can change dramatically depending upon the environment to which B. subtilis cells or spores are exposed. Rifr rpoB mutations have historically been associated with slower growth and reduced fitness; however, these assessments of fitness were conducted on limited collections of mutants in rich laboratory media that poorly reflect natural environments typically inhabited by B. subtilis. Using a novel deep-sequencing approach in addition to traditional measurements of growth rate, lag time, and pairwise competitions, we demonstrated that the competitive advantages of specific rpoB alleles differ depending on the growth environment in which they are determined. IMPORTANCE Microbial resistance to antibiotics is a growing threat to public health across the world. Historically, resistance to antibiotics has been associated with reduced fitness. A growing body of evidence indicates that resistance to rifampin, a frontline antibiotic used to treat mycobacterial and biofilm-associated infections, may increase fitness given an appropriate environment even in the absence of the selective antibiotic. Here, we experimentally confirm this phenomenon by directly comparing the fitness of multiple rifampin-resistant mutants of Bacillus subtilis in rich LB medium and an asparagine minimal medium. Our research demonstrates that the fitness cost of rifampin resistance can vary greatly depending upon the environment. Background: Simian varicella virus (SVV) is a primate herpesvirus that causes a natural varicella‐like disease in Old World monkeys and may cause epizootics in facilities housing nonhuman primates. SVV infection of nonhuman primates is used as an experimental model to investigate varicella pathogenesis and to develop antiviral strategies. Methods: An indirect enzyme‐linked immunosorbent assay (ELISA) was developed to detect SVV antibodies in infected rhesus macaque monkeys. Results: An ELISA determined SVV antibody titers following experimental infection. SVV IgG was detected by day 14 post‐infection and remained elevated for at least 84 days. Conclusions: The SVV ELISA is a safe and rapid approach to confirm SVV seropositivity and to determine SVV antibody titers in naturally and experimentally SVV‐infected monkeys. In addition to being a useful diagnostic assay to rapidly confirm acute disease or past SVV infection, the SVV ELISA is a valuable epidemiological tool to determine the incidence of SVV in non‐human primate facilities. Expansion in blood volume (BV) is a well-recognized response to arterial underfilling secondary to impaired cardiac output in heart failure (HF). However, the effectiveness of this response in terms of outcomes remains inadequately understood. Prospective analysis was undertaken in 110 patients with HF hospitalized and treated for fluid overload. BVs were measured in a compensated state at the hospital discharge using the indicator-dilution methodology. Data were analyzed for composite 1-year HF-related mortality/first rehospitalization. Despite uniform standard of care, marked heterogeneity in BVs was identified across the cohort. The cohort was stratified by BV expansion greater than or equal to +25% above normal (51% of cohort), mild-moderate expansion (22%), and normal BV (27%). Kaplan-Meier (K-M) survival estimates and regression analyses revealed BV expansion (greater than or equal to +25%) to be associated with better event-free survival relative to normal BV (P = 0.038). Increased red blood cell mass (RBCm; RBC polycythemia) was identified in 43% of the overall cohort and 70% in BV expansion greater than or equal to +25%. K-M analysis demonstrated polycythemia to be associated with better outcomes compared with normal RBCm (P < 0.002). Persistent BV expansion to include RBC polycythemia is common and, importantly, associated with better clinical outcomes compared with normal total BV or normal RBCm in patients with chronic HF. However, compensatory BV expansion is not a uniform physiological response to the insult of HF with marked variability in BV profiles despite uniform standard of care diuretic therapy. Therefore, recognizing the variability in volume regulation pathophysiology has implications not only for impact on clinical outcomes and risk stratification but also potential for informing individualized volume management strategies. Tree-ring data for 1942 - 2013 were used to determine if boreal and subalpine Picea albertiana S. Brown emend. Strong & Hills radial growth rates changed after 1950 in southern Yukon, northwest Canada. The latter year represented the beginning of accelerated atmospheric greenhouse gas accumulation, which has subsequently caused global climate changes. Ring-width chronologies for both boreal and subalpine trees were constructed using 80 trees. Individual ring-width sequences were crossdated, age-detrended, and converted to tree ring-width indices (RWI). In 2013, boreal and subalpine RWI were 41% and 169% greater than in 1950, respectively. In contrast, elevational treeline chronologies in the region typically decreased 20% - 70%. During 1942 - 2013, boreal and subalpine chronologies were significantly, at best modestly (r < |0.36|), and exclusively correlated with either moisture or temperature variables, respectively. Within this same time frame, RWI-climate variable correlates differed between the early and late phases of each chronology. Average pre-1969 or early-phase boreal correlations based on a 19-year moving window reached a maximum of - 0.56. Late-phase chronology correlations typically shifted towards weaker associations with climate variables (typically +0.04 to +0.30). For boreal RWI, 25 of 68 tested climate variables shifted as much as 0.40 - 0.88 correlation coefficient units between the early and late phases of the chronologies. Subalpine RWI were correlated only with July - August precipitation during the early phase of its chronology. These changes suggest responses by spruce to climate have shifted since 1970. Results from previous investigations into spontaneous rifampin resistance (Rifr) mutations in the Bacillus subtilis rpoB gene suggested that the spectrum of mutations depends on the growth environment. However, these studies were limited by low sample numbers, allowing for the potential distortion of the data by the presence of "jackpot" mutations that may have arisen early in the growth of a population. Here, we addressed this issue by performing fluctuation analyses to assess both the rate and spectrum of Rifr mutations in two distinct media: LB, a complete laboratory medium, and SMMAsn, a minimal medium utilizing L-asparagine as the sole carbon source. We cultivated 60 separate populations under each growth condition and determined the mutation rate to Rifr to be slightly but significantly higher in LB cultures. We then sequenced the relevant regions of rpoB to map the spectrum of Rifr mutations under each growth condition. We found a distinct spectrum of mutations in each medium; LB cultures were dominated by the H482Y mutation (27/53 or 51%), whereas SMMAsn cultures were dominated by the S487L mutation (24/51 or 47%). Furthermore, we found through competition experiments that the relative fitness of the S487L mutant was significantly higher in SMMAsn than in LB medium. We therefore conclude that both the spectrum of Rifr mutations in the B. subtilis rpoB gene and the fitness of resulting mutants are influenced by the growth environment. [ABSTRACT FROM AUTHOR]

Potential Last Glacial Maximum (LGM, 26,500–15,000 bp) members of the Eastern Beringia (now Alaska-Yukon, northwest North America) vascular flora were recognized based on their contemporary co-occurrence in easternmost Siberia (formerly part of Western Beringia). Of 1,633 indigenous terrestrial Alaska-Yukon taxa, 835 also occur in easternmost Siberia, including 526 arctic/alpine and 90 subarctic taxa. Archival macrofossil and DNA evidence corroborated the occurrence of 55 LGM taxa and indicated the presence of an additional 13 non–Western Beringian taxa. The total (848) was double the number of taxa previously considered amphi-Beringian, but likely, still an underestimate. Most Eastern Beringian arctic/alpine and subarctic taxa were perennials (94%) and forbs (64%), with herbaceous plants having average median heights of ≤ 37 cm. Arctic/alpine taxa were associated with xeric to submesic environments (54%), whereas the subarctic taxa favoured moister conditions. Both arctic/alpine and subarctic taxa were potentially present in unglaciated Eastern Beringia. However, some subarctic taxa may have migrated from refugial areas peripheral to Western Beringia during the Late Wisconsinan, prior to opening of the Bering Strait (11,000 bp), rather than being migrants from mid-latitude North America because of timing and distance considerations. Currently, former LGM Eastern Beringian taxa dominate (> 80% content) the flora and plant canopy cover of Alaska-Yukon arctic vegetation, but their frequency decreases curvilinearly (~ 0.102 taxon km−1) from Alaska to Hudson Bay, and from Yukon to southern British Columbia (p < 0.05). Aims: Elevated cardiac filling pressures producing clinical congestion in heart failure (HF) patients may be secondary to intravascular volume expansion or abnormalities in cardiac diastolic properties. The objective of this study was to assess the extent to which measures of myocardial function and intravascular volume correlate with haemodynamic abnormalities in chronic HF. Methods and results: Subjects underwent invasive haemodynamic assessment, measurement of total blood volume (TBV) using radiolabel indicator‐dilution methodology, and echocardiography to evaluate cardiac structure and function. Patients were divided into those with hypervolaemia (defined as TBV > +8% above referenced normal volume) and normal volume ('euvolaemia') (TBV ≤ + 8%). Of 66 patients, 39 (59%) were hypervolaemic and 27 (41%) normal TBV. Central venous pressure (CVP, P = 0.01) and pulmonary capillary wedge pressure (PCWP, P < 0.001) were higher in hypervolaemic compared with euvolaemic patients; however, 15% of hypervolaemic patients displayed normal pressures. Of euvolaemic patients, 70% displayed elevated CVP and 63% elevated PCWP. PCWP was moderately correlated with TBV (r = 0.42), left ventricular diastolic function (e′ velocity, r = −0.44), and left atrial strain (r = −0.47). In multivariable regression TBV, left ventricular e′, and left atrial strain were independently associated with PCWP (all P < 0.05). Conclusions: While hypervolaemic patients displayed elevations in filling pressures, a substantial proportion (15%) had normal pressures, and of all subjects with elevated filling pressures nearly one third had normal TBVs. Importantly, of patients with normal volumes, a majority (>60%) display elevated filling pressures. Objective: Given the growth in national disability-associated health care expenditures (DAHE) and the changes in health insurance–specific DAHE distribution, updated estimates of state-level DAHE are needed. The objective of this study was to update state-level estimates of DAHE. Methods: We combined data from the 2013-2015 Medical Expenditure Panel Survey, 2013-2015 Behavioral Risk Factor Surveillance System, and 2014 National Health Expenditure Accounts to calculate state-level DAHE for US adults in total, per adult, and per (adult) person with disability (PWD). We adjusted expenditures to 2017 prices and assessed changes in DAHE from 2003 to 2015. Results: In 2015, DAHE were $868 billion nationally (range, $1.4 billion in Wyoming to $102.8 billion in California) accounting for 36% of total health care expenditures (range, 29%-41%). From 2003 to 2015, total DAHE increased by 65% (range, 35%-125%). In 2015, DAHE per PWD were highest in the District of Columbia ($27 839) and lowest in Alabama ($12 603). From 2003 to 2015, per-PWD DAHE increased by 13% (range, −20% to 61%) and per-capita DAHE increased by 28% (range, 7%-84%). In 2015, Medicare DAHE per PWD ranged from $10 067 in Alaska to $18 768 in New Jersey. Medicaid DAHE per PWD ranged from $9825 in Nevada to $43 365 in the District of Columbia. Nonpublic–health insurer per-PWD DAHE ranged from $7641 in Arkansas to $18 796 in Alaska. Conclusion: DAHE are substantial and vary by state. The public sector largely supports the health care costs of people with disabilities. State policy makers and other stakeholders can use these results to inform the development of public health programs that support and provide ongoing health care to people with disabilities. Wildlife-based tourism poses opportunities and challenges for species conservation. Minimizing potential negative impacts of tourism is critical to ensure business and conservation enterprises can coexist. In north-western Namibia tourism is used as a conservation tool for the Critically Endangered black rhinoceros Diceros bicornis. However, black rhinoceroses are susceptible to human disturbance and may become displaced by tourist activities, which threatens not only the security and health of the rhinoceros population but also the sustainability of the business. We examined areas avoided by black rhinoceroses to understand how they respond to the type and extent of tourism development, and to evaluate management alternatives. We used spatial data on use of water sources by rhinoceroses to create a series of a priori candidate models that described the negative influences of tourist activities on rhinoceros habitat use. A model selection approach strongly supported a cumulative zones of influence model comprised of a 6 km buffer around the airstrip combined with a 1 km buffer around roads used daily. We compared alternative management scenarios using the best-performing model and found that an optimal road-use policy combined with airstrip relocation could minimize the total area avoided by the black rhinoceros to 7.1% and loss of high quality habitat to 20.7%. Under the worst-case scenario the area avoided and loss of high quality habitat were 153 and 85% greater, respectively, than under the scenario with optimal management. Although clinostats have long been used in space microbiology studies as ground-based analogs of spaceflight, few studies to date have systematically compared -omics data from clinostats versus spaceflight. This study compared the transcriptomic response of the Gram-positive bacterium Bacillus subtilis flown in space with corresponding transcriptomes derived from 2-D clinostat (High Aspect Ratio Vessel: HARV) experiments performed under the same conditions of bacterial strain, growth medium, temperature, and incubation time. High-quality total RNA (RNA Integrity Number >9.6) was isolated from multiple biological replicates from each treatment, transcripts were quantified by RNA-seq, and raw data was processed through a previously described standardized bioinformatics pipeline. Transcriptome data sets from spaceflight-grown and corresponding clinostat-grown cells were compared by using three different methods: (i) principal component analysis, (ii) analysis of differentially expressed genes, and (iii) gene set enrichment analysis of KEGG pathways. All three analyses found a low degree of concordance between the spaceflight and corresponding clinostat transcriptome data sets, ranging from 0.9% to 5.3% concordance. These results are in agreement with prior studies that also revealed low concordances between spaceflight and clinostat transcriptomes of the Gram-negative bacteria Rhodospirillum rubrum and Pseudomonas aeruginosa. The results are discussed from the perspective of several potential confounding factors, and suggestions are offered with the aim of achieving increased concordance between clinostat and spaceflight data. Analyses of maximum early tree ring growth within a regional curve standardization (RCS) framework were used to construct ring width chronologies in mm year−1 to estimate historical growth rates in southwest Yukon (Canada), without the necessity of correcting for age related changes. The analysis relied on the use of the 11th–50th year growth rings to standardize chronology comparisons. This age range represents the period of peak radial growth for Picea albertiana and P. glauca (white spruces), which are common trees on upland boreal sites in the study area. Of 1,460 available Picea ring series, 1,029 fit the criteria for inclusion in the study and they represented five commonly occurring RCS curve types. When converted to chronologies, all curve types had year-to-year fluctuations in average ring widths (RW11–50), but nine of the 12 RCS types (four types in three study areas) also had long-term growth rate increases of 15–132% between ad 1850 and 2000, based on regression modeling (R/r = 0.54–0.76, p < 0.001). Associated with these increases were changes through time in RCS curve type and the timing of peak growth. Among studied areas, RW11–50 values on average increased 47% from 1.59 to 2.35 mm year−1 between 1850 and 2000. The greatest increase (71% vs 35–38%) occurred in the coldest study area. RW11–50 values were significantly (p < 0.001) and strongly correlated (r > 0.94, n > 100) with conventional tree ring width index values derived from multiple RCS curves. This indicates that the two chronology construction approaches have very similar temporal patterns despite the maximum early growth analysis technique representing ring widths in mm. Individuals dually eligible for Medicare and Medicaid often receive fragmented and inefficient care. Using Minnesota fee-for-service claims, managed care encounters, and enrollment data for 2010–2012, we estimated the likely impact of Minnesota Senior Health Option (MSHO)—seen as the first statewide fully integrated Medicare-Medicaid model—on health care and long-term services and supports use, relative to Minnesota Senior Care Plus (MSC+), a Medicaid-only managed care plan with Medicare fee for service. Estimates suggest that MSHO enrollees had significantly higher use of primary care and, potentially, of community-based services, combined with lower use of hospital-based care than similar MSC+ enrollees. U.S. Air Force Distributed Common Ground Station (DCGS) intelligence (Intel) operators sustain 24/7 battlefield situational awareness and facilitate precision-strike operations. DCGS operations are global, synergistic, Total Force (TF) endeavors by active duty, Air National Guard (ANG) and Reserve units, providing combatant commanders with critical real-time intelligence and shaping operational and tactical decisions. Continual surveillance of this community's psychological health is important to its military leaders. This study re-examines the most frequently reported occupational stressors, as well as the prevalence of occupational burnout (i.e. high-emotional exhaustion and cynicism, and low professional efficacy), and psychological distress within this population. Active duty (n = 1717), ANG (n = 139), and Reserve (n = 173) Intel operators participated in a comprehensive, online, occupational health assessment. Results reveal that occupational stressors contributing to elevated rates of distress, regardless of TF status (i.e. low manning, long work hours, excessive workload, and organizational communication concerns) are consistent with previous research. The prevalence of high-emotional exhaustion (AD: 29%/ANG: 25%/RES: 14%) and psychological distress (AD: 19%/ANG: 17%/RES: 5%) are above estimates for other military communities. For many potato cultivars, tuber yield is optimal at average daytime temperatures in the range 14–22 °C. Above this range, tuber yield is reduced for most cultivars. We previously reported that moderately elevated temperature increases steady-state expression of the core circadian clock gene TIMING OF CAB EXPRESSION 1 (StTOC1) in developing tubers, whereas expression of the StSP6A tuberization signal is reduced, along with tuber yield. In this study we provide evidence that StTOC1 links environmental signalling with potato tuberization by suppressing StSP6A autoactivation in the stolons. We show that transgenic lines silenced in StTOC1 expression exhibit enhanced StSP6A transcript levels and changes in gene expression in developing tubers that are indicative of an elevated sink strength. Nodal cuttings of StTOC1 antisense lines displayed increased tuber yields at moderately elevated temperatures, whereas tuber yield and StSP6A expression were reduced in StTOC1 overexpressor lines. Here we identify a number of StTOC1 binding partners and demonstrate that suppression of StSP6A expression is independent of StTOC1 complex formation with the potato homolog StPIF3. Chromogenic anti‐Xa assays for unfractionated heparin monitoring (heparin activity) are susceptible to interference from hemolysis and icterus. The purpose of this study was to better understand the effect of hemolysis and icterus on anti‐Xa heparin activity and to predict the magnitude of the error. Increasing levels of hemoglobin and unconjugated bilirubin were added to pooled normal plasma or buffer containing known levels of heparin. Increased plasma hemoglobin or bilirubin produced falsely increased residual factor Xa activity as measured by the absorbance change (OD/min) in the Stago heparin activity assay. This increased absorbance change slope resulted in falsely lower estimates of heparin activity. The falsely lower heparin activity measurement occurred even when heparin was not present, indicating it was not due to heparin neutralization. In a sample containing 0.62 ± 0.06 U/mL heparin and 228 mg/dL hemoglobin, the measured heparin activity was 0.41 ± 0.03 U/mL, underestimating heparin activity by 0.21 ± 0.07 U/mL. Interference occurred if plasma hemoglobin was above 70 mg/dL or bilirubin was above 16 mg/dL, which happened in 16%–26% of samples from pediatric patients on extracorporeal life support (ECLS). In conclusion, hemolysis and icterus were common in ECLS patients, leading to underestimates of unfractionated heparin activity and potentially higher doses of heparin than intended. The magnitude of the heparin activity measurement error could be predicted based on plasma hemoglobin and bilirubin levels until these levels exceeded the technical limits of the assay, ~230 mg/dL hemoglobin and 55 mg/dL bilirubin. Pro-environmental behaviors (PEBs) may be associated with a personal relationship with nature. We conducted a quasi-experiment with 423 residents who had or had not actively participated in a tree-planting scheme and lived in 20 neighborhoods that differ in their greenness level. We tested whether exposure to nature (specified by the amount of vegetation cover in residents' neighborhoods) and/or past PEB (specified by tree-planting participation) was associated with residents' PEB and whether key psychological constructs would mediate these relationships. Structural equation mediation models revealed that the level of neighborhood vegetation and involvement in the planting scheme explained 46% of the variance in PEB, with connection to nature, the use of nature for psychological restoration, and environmental attitudes mediating the relationships. We demonstrate the application of a modified form of the configurational-bias algorithm for the simulation of chain molecules on the second-nearest-neighbor-diamond lattice. Using polyethylene and poly(ethylene-oxide) as model systems we show that the present configurational-bias algorithm can increase the speed of the equilibration by at least a factor of 2–3 or more as compared to the previous method of using a combination of single-bead and pivot moves along with the Metropolis sampling scheme [N. Metropolis, A. W. Rosenbluth, M. N. Rosenbluth, A. H. Teller, and E. Teller, J. Chem. Phys. 21, 1087 (1953)]. The increase in the speed of the equilibration is found to be dependent on the interactions (i.e., the polymer being simulated) and the molecular weight of the chains. In addition, other factors not considered, such as the density, would also have a significant effect. The algorithm is an extension of the conventional configurational-bias method adapted to the regrowth of interior segments of chain molecules. Appropriate biasing probabilities for the trial moves as outlined by Jain and de Pablo for the configurational-bias scheme of chain ends, suitably modified for the interior segments, are utilized [T. For a model system of polyethylene of chain lengths 40 and 100 carbon atoms, we calculated the pressure at different densities and compared them with the experimental values. The simulation was conducted on the second nearest neighbor diamond lattice, and the pressure was calculated using the virtual-volume-variation method after the system was reverse mapped to its fully atomistic form in continuous space and energy minimized. In addition, the pressure was also calculated from the virial route by conducting a short molecular dynamics simulation starting from the energy minimized structure. We show that the pressure obtained from our simulations is quite reasonable in the length of simulation time (in Monte Carlo steps) normally employed in our group. These results provide additional evidence for the equilibration of our model systems, and methodology to calculate the pressure in our lattice models. © 2004 American Institute of Physics. An amorphous 50/50 atactic polypropylene (aPP)/isotactic polypropylene (iPP) mixture at 125 °C was simulated using a second nearest neighbor diamond lattice and a three states rotational isometric state model. The result suggests that at the liquidlike density that corresponds to the atmospheric pressure, aPP prefers to interact with other aPP chains rather than with iPP chains. The result is consistent with the inference of Keith and Padden [J. Appl. Phys. 35, 1286 (1964)] that aPP and iPP will tend to separate from one another in their melt at 125 °C, before the onset of crystallization of iPP. The tendency for immiscibility of the amorphous aPP/iPP blend is likely attributed to the presence of short syndiotactic sequences in the aPP chains adopting all-trans conformations. The attractive intermolecular interaction of pairs of such subchains at 125 °C promotes the separation of aPP from iPP. This interaction is weakened at higher temperature, where aPP and iPP become miscible. The result also shows that miscibility of the blend increases with increasing pressure. Recently we described a coarse-grained model of poly(ethylene oxide) and then employed that model to study the amount of spontaneous threading of cyclic molecules by linear chains in the melt [C. A. Helfer, G. Xu, W. L. Mattice, and C. Pugh, Macromolecules 36, 10071 (2003)]. Since the amount of statistical threading at equilibrium is small, there is interest in identifying physical changes in the system that will increase the threading. We now use that coarse-grained model to investigate the effect on threading of various hypothetical (but feasible) modifications to the two-component system of macrocycles and linear chains in the melt, and different confinement geometries, that can bring about correlations in the arrangement of the rings. Our work follows on the concept of an amphiphilic approach [C. Pugh, J.-Y. Bae, J. R. Scott, and C. L. Wilkins, Macromolecules 30, 8139 (1997)] for increasing the statistical threading in homopolyrotaxane melts. We investigate whether introducing such correlations in the macrocycles can increase the spontaneous threading. A recently developed high coordination lattice based Monte Carlo method is applied to investigate the glass transition of free-standing atactic polypropylene thin films. In this method the short-range interactions are described by a rotational isomeric state model and the long-range interactions are obtained from a discretized form of a Lennard-Jones function. Simulations of five thin films of C[sub 150]H[sub 302] with thickness of 20-100 Å at T[sub g] are performed by a stepwise cooling from the melts to the glass states. The temperature dependence of the thickness of the thin films is obtained from the fitting of the density profile by a hyperbolic function. The glass transition temperatures of the thin films are evaluated from the change in thermal expansion coefficients at T[sub g]. The T[sub g] decreases as the films become thinner. For all but the thinnest film, 1/T[sub g], is inversely proportional to the thickness of the film. The proportionality constant is within the range cited in experimental studies of other systems. Thin films of a mixture of two kinds of short polyethylene chains, n-tricontane and n-tetracontane, are simulated both at high and low temperatures by a dynamic Monte Carlo method on a high coordination lattice, which bridges the coarse-grained and the fully atomistic simulations. Films are obtained from an equilibrated model for the melt by increasing one of the three periodic boundaries to a very large value. The melting temperatures (T[sub m]) of the two pure components in the simulation are obtained from a sharp transition of various parameters, such as the probability of trans conformation of C-C bonds, orientation order parameters, energies, etc. At high temperature, 420 K (above T[sub m] of the two pure components), the chains have a trend to separate with each other. The shorter chains are enriched on the free surface due to their larger fraction of chain ends. At low temperature (below T[sub m] of the two pure components), two situations exist, which depend on the prior history of the mixture. If the quench takes place from a homogeneous mixture the chains crystallize together with almost the same density profile except for a large fluctuation in the bulk region of the thin film. On the other hand, if the quench takes place from the equilibrated thin film at 420 K, the shorter chains remain enriched on the surface, and the segregation of the shorter chain is enhanced. The radial local dynamics in poly(1,4-trans-butadiene) melts is studied using molecular dynamics simulations. In this work, the hopping peak is observed in the Van Hove space-time correlation function G[sub s],(r,t) for the first time in polymeric systems. The hopping motion, observed only for methine hydrogen, is also identified in the mean-square displacement 〈Λr²(t)) and the intermediate scattering function F[sub s],(k,t) through the relative dynamics of the hopping methine hydrogen and the nonhopping methylene hydrogen. The hopping motion is found to cause an unusual broadening of the dispersion width in the dynamic structure factor S[sub inc](k, ω). Active free volume is proposed in terms of G[sub s],(r, t) at a short time, which offers a consistency to the relationship between free volume and local dynamics. Fast counterrotation at a pair of CH-CH[sub 2] bonds across a CH=CH bond is found responsible for the hopping peak in this polymer, and a new hopping criterion modified for polymeric liquids is proposed. A freestanding thin film of n-tetracontane chains is simulated by a Monte Carlo (MC) method on a high coordination lattice. The coarse-grained chains, represented by 20 beads each, can be reverse-mapped into the fully atomistic description, C[sub 40]H[sub 82]. The Hamiltonian includes a short-range interaction based on a rotational isomeric state model and a long-range interaction obtained from a Lennard-Jones potential energy function. When the melt is instantaneously quenched from 473 to 298 K, crystallization initiates in the surface region and propagates into the interior of the film, as was found in a prior molecular dynamics simulation of a united atom model of polyethylene [M. Ito, M. Matsumoto, and M. Doi, Fluid Phase Equilibria, 144, 395 (1998)]. Several repetitions of the MC simulation, starting from different configurations of the melt at 473 K, reveal that two distinctly different structures can be obtained. Usually the independently initiated crystals at the two surfaces of the thin film produce a disordered grain boundary when they impinge on one another as a consequence of propagation into the interior of the film. This grain boundary was also observed by Ito et al. However, if the MC simulation is repeated many times, there are a few instances in which the independently initiated crystals happen to have a similar orientation, and then crystallization propagates completely through the thin film without producing a grain boundary in the interior. A well-defined melting phenomenon is observed at about 390 K when the film without the grain boundary is heated. Annealing at 380 K of the film with the grain boundary causes growth of one crystal at the expense of the other. This growth eventually leads to a completely crystalline film, with elimination of the grain boundary. Recently Clancy et al. reported a Monte Carlo simulation that reproduces the tendency, observed by Mu¨lhaupt and co-workers, for demixing of melts of isotactic polypropylene (iPP) and syndiotactic polypropylene (sPP). The short-range intermolecular interactions in the coarse-grained chains in the simulation are controlled by a rotational isomeric state model for PP that was described by Suter et al. The intermolecular interactions of the beads are controlled by a discretized representation of a continuous Lennard-Jones (LJ) potential that describes the interaction of two molecules of propane. The work reported here reveals that the outcome is sensitive to the truncation of this LJ potential. A weak tendency for demixing of the 50:50 iPP:sPP melt is seen if the LJ potential is truncated so that only its repulsive part is used in the simulation. Inhomogeneity is enhanced if the truncation of the LJ potential is shifted to larger distances, so that the attractive branch of the LJ potential is incorporated in the simulation. This observation in the simulation leads to conclusive identification of the mechanism responsible for demixing of the 50:50 melt. Calculations of an order of magnitude greater strain hardening coefficient over compression or tensile test measurements are demonstrated for continuous indentation hardness measurements past "pop-in". Analyses were performed at small indentation strains for a macro-ball test on a NaCl crystal and at larger strains measured for rounded points of micro- and nano-tipped indenters in tests of MgO and copper crystal surfaces. The exceptional strain hardening is attributed to the smaller spacing and consequent interactions of the plastically-induced dislocations, including for MgO, the formation of nano-scale sessile dislocations accompanying the imposed three-dimensional deformation. The dislocation-based hardening is much greater than the smaller so-called "Indentation Size Effect (ISE)" of softening obtained at larger, constant strain, penetration depths with Berkovich-type indenters. The nutritional benefits of consuming potato are increasingly recognized, however improvements in nutritional quality must proceed with good consumer quality if a variety is going to achieve commercial success. There is a major challenge to provide breeders with the selection tools needed to evaluate and use quality early enough in the breeding program. Reports of flavor and texture trait dissection are starting to surface and rapid advances in "omic" technologies provide the means for assessment of these traits. Translational of this knowledge to high throughput phenotyping platforms that can be used on large numbers of selections will be an important next step with some promising technologies in development. Having identified genes and markers coupled to beneficial alleles for flavor and textural traits, new breeding technologies may provide promising avenues for variety improvement linking consumer preferences for flavor and texture to enhanced nutritional properties. Objectives: To determine the relationship between baseline variations in the partial thromboplastin time (PTT) and the discordance between the PTT and anti-Xa heparin activity (anti-Xa) during heparin therapy.Methods: The baseline PTT on heparin was determined using automated heparin neutralization with protamine (prPTT). The prPTT was used to calculate a baseline-corrected PTT on heparin to reduce discordance with anti-Xa measurements.Results: The prPTT removed up to 1 U/mL of heparin, returning baseline values for normal, factor-deficient, and lupus inhibitor plasmas. A prolonged prPTT was seen in 97 (53%) of 182 samples from heparinized patients. The heparinized PTT was discordant compared with anti-Xa in 64 (35%) of 182 samples and 43 (67%) of 64 discordant samples, and 46% of concordant samples showed a prolonged prPTT. A baseline-corrected PTT reduced discordance with anti-Xa measurements by 64%.Conclusions: PTT/anti-Xa discordance due to baseline PTT prolongation could be reduced using a baseline-corrected PTT. Context.--The College of American Pathologists (CAP) developed proficiency testing for platelet function assays by using blood collected by the participant added to challenge tubes containing either saline (normal) or tirofiban (abnormal). Objective.--To analyze platelet function proficiency testing for Platelet Function Analyzer PFA-100, platelet aggregation, PlateletWorks, and PlateletMapping. Design.--Proficiency testing data from 2012--2016 were analyzed. Results.--For PFA-100, a total of 1200 laboratories participated; the coefficient variation (CV) of cartridge closure times was 22% (saline); 44,952 of 45,616 survey responses (99%) provided an interpretation, and 42,934 of 44,952 (96%) were correct. For optical platelet aggregation, 190 laboratories participated; the CV was 17% (saline), 7444 of 7813 survey responses (95%) provided an interpretation, and 7015 of 7444 (94%) were correct. For PlateletWorks, 60 laboratories participated; the CV was 3% to 11% (saline); 2412 of 2454 survey responses (98%) provided an interpretation, and 1207 of 1276 (95%) were correct for adenosine diphosphate (ADP) and 936 of 1136 (82%) for collagen. For PlateletMapping, 200 laboratories participated. For ADP, 1128 of 2697 survey responses (42%) provided an interpretation, but only 927 of 1128 (82%) were correct. For arachidonic acid, 1139 of 2604 survey responses (44%) provided an interpretation and 964 of 1139 (85%) were correct. Conclusions.--CAP is the first to provide proficiency testing for platelet aggregation, PlateletWorks, and PlateletMapping. Platelet aggregation, PFA-100, and Platelet- Works using ADP as an agonist performed well with more than 90% of laboratories providing an interpretation and a similar number providing correct results. Results reveal that occupational stressors contributing to elevated rates of distress, regardless of TF status (i.e., low manning, long work hours, excessive workload, and organizational communication concerns) are consistent with previous research. The prevalence of high-emotional exhaustion (AD: 29%/ANG: 25%/RES: 14%) and psychological distress (AD: 19%/ANG: 17%/RES: 5%) are above estimates for other military communities. Potato tuber bud dormancy break followed by premature sprouting is a major commercial problem which results in quality losses and decreased tuber marketability. An approach to controlling premature tuber sprouting is to develop potato cultivars with a longer dormancy period and/or reduced rate of sprout growth. Our recent studies using a potato diploid population have identified several quantitative trait loci (QTLs) that are associated with tuber sprout growth. In the current study, we aim to characterize a candidate gene associated with one of the largest effect QTLs for rapid tuber sprout growth on potato chromosome 3. Underlying this QTL is a gene encoding a TERMINAL FLOWER 1/CENTRORADIALIS homologue (PGSC0003DMG400014322). Here, we use a transgenic approach to manipulate the expression level of the CEN family member in a potato tetraploid genotype (cv. Désirée). We demonstrate a clear effect of manipulation of StCEN expression, with decreased expression levels associated with an increased rate of sprout growth, and overexpressing lines showing a lower rate of sprout growth than controls. Associated with different levels of StCEN expression were different levels of abscisic acid and cytokinins, implying a role in controlling the levels of plant growth regulators in the apical meristem. Abstract Earthworms have a profound effect on the soil they inhabit, but our current understanding of the chemical factors and molecular mechanisms that attract or repel earthworms from a specific area of soil is limited. In this study, we developed a soil T-maze assay allowing for the examination of earthworm behavioral responses to repellent chemicals. This tests earthworms individually, and thus the potential confounds of worms being influenced by conspecifics signals are avoided. Canadian nightcrawlers (Lumbricus terrestris) can detect the canonical Transient Receptor Potential Ankyrin 1 (TRPA1) agonist, allyl isothiocyanate (AITC), but not the canonical Transient Receptor Potential Vanilloid 1 (TRPV1) agonist capsaicin. In the T-maze assay L. terrestris avoided TRPA1 activators AITC, cinnamaldehyde and menthol in a dosage dependent manner. The single subject T-maze assay is a useful tool to study the avoidance of earthworms to chemical compounds and provides behavioral evidence for a TRPA1 ortholog in L. terrestris.
