Climate change scenarios predict more frequent and intense drought periods for 2071–2100 in the most important and intensively used agricultural region of Austria, the Marchfeld. Current and predicted lower precipitation scenarios were simulated at a lysimeter station for 9 years. Plant biomass, nitrogen (N) and carbon (C) content, and δ13C and δ15N values of plant compartments were monitored in years 7–9. Aboveground biomass of cereals and grain yield decreased under the predicted scenario, while the quality of grain (% N) was unaffected. Weed and catch crops grown in winter were not affected or were even positively affected, possibly due to the accumulation of nutrients in the soil following the lower plant uptake in summer. Accordingly, low plant δ15N values were mainly attributed to the presence of higher proportion of mineral fertilizer in the predicted precipitation scenario. As expected, water stress significantly increased δ13C values in plants grown over summer, while this was not seen for plants growing over winter. Fertile agricultural soil might ameliorate but cannot outbalance the negative impact of more frequent and intense drought periods.

Abstract The Pasterze is Austria's largest glacier, and it is experiencing rapid downwasting and retreat. A mosaic of complex sedimentary deposits has been produced in recent years which have not hitherto been studied, yet provide excellent lessons into the facies distribution expected from a dying valley glacier. In this paper, a new glaciological–geomorphological–geological map is presented for the glacier in July 2021. Freshly exposed (since 2018) tills and flutes constitute a subglacial sediment–landform assemblage. An ice‐marginal sediment–landform assemblage comprises meltwater streams, a delta system and proglacial lake terrace deposits. The supraglacial assemblage, meanwhile, includes fossil englacial channel deposits revealed by ablation, together with debris bands, rockfall deposits and supraglacial channel deposits. Collectively, these sediment–landform assemblages constitute the building blocks of a dying glacier landsystem.

Receptors that distinguish the multitude of microbes surrounding plants in the environment enable dynamic responses to the biotic and abiotic conditions encountered. In this study, we identify and characterise a glycan receptor kinase, EPR3a, closely related to the exopolysaccharide receptor EPR3. Epr3a is up-regulated in roots colonised by arbuscular mycorrhizal (AM) fungi and is able to bind glucans with a branching pattern characteristic of surface-exposed fungal glucans. Expression studies with cellular resolution show localised activation of the Epr3a promoter in cortical root cells containing arbuscules. Fungal infection and intracellular arbuscule formation are reduced in epr3a mutants. In vitro, the EPR3a ectodomain binds cell wall glucans in affinity gel electrophoresis assays. In microscale thermophoresis (MST) assays, rhizobial exopolysaccharide binding is detected with affinities comparable to those observed for EPR3, and both EPR3a and EPR3 bind a well-defined β-1,3/β-1,6 decasaccharide derived from exopolysaccharides of endophytic and pathogenic fungi. Both EPR3a and EPR3 function in the intracellular accommodation of microbes. However, contrasting expression patterns and divergent ligand affinities result in distinct functions in AM colonisation and rhizobial infection in Lotus japonicus. The presence of Epr3a and Epr3 genes in both eudicot and monocot plant genomes suggest a conserved function of these receptor kinases in glycan perception.

The properties of particle ensembles are defined by a complex multidimensional parameter space, namely particle size, shape, surface, structure, composition and their distributions. Macroscopic product properties are a direct result of these disperse particle properties. Therefore, the comprehensive multidimensional characterization of particle ensembles is a key task in any product design. However, the determination of complex property distributions is major challenge. We provide a broad overview of the current tools for multidimensional particle characterization. First, the mathematical handling of multidimensional (nD) property distribution is outlined as a necessary framework for the correct handling of nD particle size distributions (PSDs). Then, well-established techniques as well as recent developments with the potential to extract nD property distributions are reviewed. Ex situ imaging techniques like electron tomography or Raman spectroscopy with AFM co-localization, for instance, provide a resolution on the level of single particles but are limited in terms of sample statistics. A particular focus lies therefore on methods in the gas and the liquid phase, which provide multidimensional particle properties either directly or by a combination of one-dimensional techniques.

Abstract Background The mitochondrial intermembrane space (IMS) is home to proteins fulfilling numerous essential cellular processes, particularly in metabolism and mitochondrial function. All IMS proteins are nuclear encoded and synthesized in the cytosol and must therefore be correctly targeted and transported to the IMS, either through mitochondrial targeting sequences or conserved cysteines and the mitochondrial disulfide relay system. The mitochondrial oxidoreductase MIA40, which catalyzes disulfide formation in the IMS, is imported by the combined action of the protein AIFM1 and MIA40 itself. Here, we characterized the function of the conserved highly negatively charged C-terminal region of human MIA40. Results We demonstrate that the C-terminal region is critical during posttranslational mitochondrial import of MIA40, but is dispensable for MIA40 redox function in vitro and in intact cells. The C-terminal negatively charged region of MIA40 slowed import into mitochondria, which occurred with a half-time as slow as 90 min. During this time, the MIA40 precursor persisted in the cytosol in an unfolded state, and the C-terminal negatively charged region served in protecting MIA40 from proteasomal degradation. This stabilizing property of the MIA40 C-terminal region could also be conferred to a different mitochondrial precursor protein, COX19. Conclusions Our data suggest that the MIA40 precursor contains the stabilizing information to allow for postranslational import of sufficient amounts of MIA40 for full functionality of the essential disulfide relay. We thereby provide for the first time mechanistic insights into the determinants controlling cytosolic surveillance of IMS precursor proteins.

The increasing complexity in particle science and technology requires the ability to deal with multidimensional property distributions. We present the theoretical background for multidimensional fractionations by transferring the concepts known from one dimensional to higher dimensional separations. Particles in fluids are separated by acting forces or velocities, which are commonly induces by external fields, e.g., gravitational, centrifugal or electro-magnetic fields. In addition, short-range force fields induced by particle interactions can be employed for fractionation. In this special case, nanoparticle chromatography is a recent example. The framework for handling and characterizing multidimensional separation processes acting on multidimensional particle size distributions is presented. Illustrative examples for technical realizations are given for shape-selective separation in a hydrocyclone and for density-selective separation in a disc separator.

Litter decomposition plays a pivotal role in the global carbon cycle, but is difficult to measure on a global scale, especially by citizen scientists. Here, citizen scientists, i.e., school students with their teachers, used the globally applied and standardized Tea Bag Index (TBI) method to collect data on litter decomposition in urban areas in Austria. They also sampled soils to investigate the linkages between litter decomposition and soil attributes. For this study, 54 sites were selected from the school experiments and assembled into a TBI dataset comprising litter decomposition rates (k), stabilization factors (S), as well as soil and environmental attributes. An extensive pre-processing procedure was applied to the dataset, including attribute selection and discretization of the decomposition rates and stabilization factors into three categories each. Data mining analyses of the TBI data helped reveal trends in litter decomposition. We generated predictive models (classification trees) that identified the soil attributes governing litter decomposition. Classification trees were developed for both of the litter decomposition parameters: decomposition rate (k) and stabilization factor (S). The main governing factor for both decomposition rate (k) and stabilization factor (S) was the sand content of the soils. The data mining models achieved an accuracy of 54.0 and 66.7% for decomposition rates and stabilization factors, respectively. The data mining results enhance our knowledge about the driving forces of litter decomposition in urban soils, which are underrepresented in soil monitoring schemes. The models are very informative for understanding and describing litter decomposition in urban settings in general. This approach may also further encourage participatory researcher-teacher-student interactions and thus help create an enabling environment for cooperation for further citizen science research in urban school settings.

Epileptiform activity alters gene expression in the central nervous system, a phenomenon that has been studied extensively in animal models. Here, we asked whether also in vitro models of seizures are in principle suitable to investigate changes in gene expression due to epileptiform activity and tested this hypothesis mainly in rodent and additionally in some human brain slices. We focused on three genes relevant for seizures and epilepsy: FOS proto-oncogene (c-Fos), inducible cAMP early repressor (Icer) and mammalian target of rapamycin (mTor). Seizure-like events (SLEs) were induced by 4-aminopyridine (4-AP) in rat entorhinal-hippocampal slices and by 4-AP/8 mM potassium in human temporal lobe slices obtained from surgical treatment of epilepsy. SLEs were monitored simultaneously by extracellular field potentials and intrinsic optical signals (IOS) for 1–4 h, mRNA expression was quantified by real time PCR. In rat slices, both duration of SLE exposure and SLE onset region were associated with increased expression of c-Fos and Icer while no such association was shown for mTor expression. Similar to rat slices, c-FOS induction in human tissue was increased in slices with epileptiform activity. Our results indicate that irrespective of limitations imposed by ex vivo conditions, in vitro models represent a suitable tool to investigate gene expression. Our finding is of relevance for the investigation of human tissue that can only be performed ex vivo. Specifically, it presents an important prerequisite for future studies on transcriptome-wide and cell-specific changes in human tissue with the goal to reveal novel candidates involved in the pathophysiology of epilepsy and possibly other CNS pathologies.

Plants trigger immune responses upon recognition of fungal cell wall chitin, followed by the release of various antimicrobials, including chitinase enzymes that hydrolyze chitin. In turn, many fungal pathogens secrete LysM effectors that prevent chitin recognition by the host through scavenging of chitin oligomers. We previously showed that intrachain LysM dimerization of the Cladosporium fulvum effector Ecp6 confers an ultrahigh-affinity binding groove that competitively sequesters chitin oligomers from host immune receptors. Additionally, particular LysM effectors are found to protect fungal hyphae against chitinase hydrolysis during host colonization. However, the molecular basis for the protection of fungal cell walls against hydrolysis remained unclear. Here, we determined a crystal structure of the single LysM domain-containing effector Mg1LysM of the wheat pathogen Zymoseptoria tritici and reveal that Mg1LysM is involved in the formation of two kinds of dimers; a chitin-dependent dimer as well as a chitin-independent homodimer. In this manner, Mg1LysM gains the capacity to form a supramolecular structure by chitin-induced oligomerization of chitin-independent Mg1LysM homodimers, a property that confers protection to fungal cell walls against host chitinases.

Abstract Background Cortical ischemic lesions represent the predominant pathomorphological pattern of focal lesions after aneurysmal subarachnoid hemorrhage (aSAH). Autopsy studies suggest that they occur adjacent to subarachnoid blood and are related to spasm of small cortical rather than proximal arteries. Recent clinical monitoring studies showed that cortical spreading depolarizations, which induce cortical arterial spasms, are involved in lesion development. If subarachnoid blood induces adjacent cortical lesions, it would be expected that (i) they also develop after traumatic subarachnoid hemorrhage (tSAH), and (ii) lesions after tSAH can occur in absence of angiographic vasospasm, as was found for aSAH. Case presentation An 86-year-old woman was admitted to our hospital with fluctuating consciousness after hitting her head during a fall. The initial computed tomography (CT) was significant for tSAH in cortical sulci. On day 8, the patient experienced a secondary neurological deterioration with reduced consciousness and global aphasia. Whereas the CT scan on day 9 was still unremarkable, magnetic resonance imaging (MRI) on day 10 revealed new cortical laminar infarcts adjacent to sulcal blood clots. Proximal vasospasm was ruled out using MR and CT angiography and Doppler sonography. CT on day 14 confirmed the delayed infarcts. Conclusions We describe a case of delayed cortical infarcts around sulcal blood clots after tSAH in the absence of proximal vasospasm, similar to results found previously for aSAH. As for aSAH, this case suggests that assessment of angiographic vasospasm is not sufficient to screen for risk of delayed infarcts after tSAH. Electrocorticography is suggested as a complementary method to monitor the hypothesized mechanism of spreading depolarizations.

Characterization of nanoparticles is a complex and important problem for the vast number of applications that require them. Here, the authors present a method to uncover the two-dimensional distribution of length and diameter of anisotropic nanoparticles like gold nanorods with a single measurement by combining spectroscopic and sedimentation data.

Background: Pythium ultimum is a ubiquitous oomycete plant pathogen responsible for a variety of diseases on a broad range of crop and ornamental species.Results: The P. ultimum genome (42.8 Mb) encodes 15,290 genes and has extensive sequence similarity and synteny with related Phytophthora species, including the potato blight pathogen Phytophthora infestans. Whole transcriptome sequencing revealed expression of 86% of genes, with detectable differential expression of suites of genes under abiotic stress and in the presence of a host. The predicted proteome includes a large repertoire of proteins involved in plant pathogen interactions, although, surprisingly, the P. ultimum genome does not encode any classical RXLR effectors and relatively few Crinkler genes in comparison to related phytopathogenic oomycetes. A lower number of enzymes involved in carbohydrate metabolism were present compared to Phytophthora species, with the notable absence of cutinases, suggesting a significant difference in virulence mechanisms between P. ultimum and more host-specific oomycete species. Although we observed a high degree of orthology with Phytophthora genomes, there were novel features of the P. ultimum proteome, including an expansion of genes involved in proteolysis and genes unique to Pythium. We identified a small gene family of cadherins, proteins involved in cell adhesion, the first report of these in a genome outside the metazoans.Conclusions: Access to the P. ultimum genome has revealed not only core pathogenic mechanisms within the oomycetes but also lineage-specific genes associated with the alternative virulence and lifestyles found within the pythiaceous lineages compared to the Peronosporaceae. © 2010 Lévesque et al.; licensee BioMed Central Ltd.

The pathogenic oomycete Saprolegnia parasitica secretes effector proteins that translocate into host cells through unclear mechanisms. Here, Trusch et al. show that the uptake of effector protein SpHtp3, resulting in RNA degradation, depends on a gp96-like host receptor and a second effector protein.

Citizen science is a powerful tool for collecting data in inaccessible places and at scales that would otherwise not be possible. Studies using complex, laboratory-based technical analysis with samples derived from easy to conduct experiments could also capitalize on this approach, by including the public in the experimental undertaking. This approach offers practical communication opportunities to raise awareness about the scientific method. We used an experimental citizen science approach in order to communicate the concept of land-based carbon sequestration and the potential role of biochar (i.e. charcoal added to soil). At four gardening events (between 100 and 7,000 attendees) we encouraged participation in our pot-scale citizen science project. We aimed to assess drought resilience of pot plants in soils amended with biochar. Participants sent their mature plant samples to our laboratory for stable isotope analysis to get results and additional information on drought stress, that was otherwise not possible. We observed no significant negative or positive effects of biochar, neither on the water use efficiency, as determined by isotopic methods or on the growth of the bean plants. Our two-stage strategy (experimental citizen science and laboratory analysis) was an effective way of involving people. We identified some challenges sustaining commitment and made some improvements to the project design. Overall we successfully avoided the “learning deficit” trap by engaging the people in an experimental learning activity; demonstrating that combining experimental citizen science with lab-based analysis is a promising and inspiring approach for future studies.

Up to date, preclinical screening for new antiepileptic substances is performed by a combination of different in vivo models of acute seizures, for which large numbers of animals are necessary. So far, little attention has been paid to in vitro models, which are also able to detect antiepileptic efficacy and in principle could likewise serve for exploratory preclinical screening. One of the established in vitro models of acute seizures is the 4-aminopyridine (4-AP) model. Previous studies have shown that the 4-AP model is capable to recapitulate the antiepileptic efficacy of standard antiepileptic drugs (AEDs) such as valproate or carbamazepine. Here, we employed a dual methodological approach using electrophysiology and optical imaging to systematically test the antiepileptic efficacy of three new-generation AEDs with distinct mechanisms of action (lacosamide, zonisamide, and levetiracetam). We found that frequency of 4-AP induced seizure like events (SLE) was the most sensitive parameter to detect dose-dependent antiepileptic effects in these compounds. Specifically, levetiracetam reduced SLE frequency while lacosamide and zonisamide at higher doses completely blocked SLE incidence. Analysis of the intrinsic optical signal additionally revealed a subiculum-specific reduction of the area involved in the propagation of ictal activity when lacosamide or zonisamide were administered. Taken together, our data adds some evidence that acute seizure models in vitro are in principle capable to detect antiepileptic effects across different mechanisms of action with efficacy similar to acute models in vivo. Further studies with negative controls, e.g., penicillin as a proconvulsant, and other clinically relevant AEDs are needed to determine if this acute in vitro model might be useful as exploratory screening tool. In view of the increasing sensitivity toward animal welfare, an affective in vitro model may help to reduce the number of laboratory animals deployed in burdening in vivo experiments and to preselect substances for subsequent testing in time- and cost-laborious models of chronic epilepsy.
