Zucchini (Cucurbita pepo) is a common vegetable crop worldwide. It belongs to Family Cucurbitaceae which is one of the largest plant families and has a high economic impact worldwide. Zucchini can be infected by several plant viral pathogens which cause severe losses in cucurbits production. This study aims to identify certain viruses which infect zucchini in Assiut governorate, Egypt using Reverse Transcription Polymerase Chain Reaction (RT-PCR). The response of four zucchini cultivars (Hytech, Safa, Malat, and 03) to viral infection was evaluated under both greenhouse and field conditions. The assessment was conducted through measuring some parameters including disease severity (D.S), yield, total chlorophyll, and total carbohydrates contents. In general, zucchini cultivars differed significantly in their susceptibility to the infection with Watermelon mosaic virus (WMV) and Papaya ringspot virus (PRSV). Cultivars 03 and Malat were the most tolerant cultivars while Hytech followed by Safa were the most susceptible ones. The obtained data showed that zucchini cultivars differ significantly in their total carbohydrate and chlorophyll contents post the infection with WMV and PRSV. The highest chlorophyll and carbohydrates contents were obtained in 03 and Malat cultivars while the lowest chlorophyll and carbohydrate contents were in case of Hytech followed by Safa under both greenhouse or field conditions. Viruses employ a series of diverse translational strategies to expand their coding capacity, which produces viral proteins with common domains and entangles virus-host interactions. P3N-PIPO, which is a transcriptional slippage product from the P3 cistron, is a potyviral protein dedicated to intercellular movement. Here, we show that P3N-PIPO from watermelon mosaic virus (WMV) triggers cell death when transiently expressed in Cucumis melo accession PI 414723 carrying the Wmr resistance gene. Surprisingly, expression of the P3N domain, shared by both P3N-PIPO and P3, can alone induce cell death, whereas expression of P3 fails to activate cell death in PI 414723. Confocal microscopy analysis revealed that P3N-PIPO targets plasmodesmata (PD) and P3N associates with PD, while P3 localizes in endoplasmic reticulum in melon cells. We also found that mutations in residues L35, L38, P41, and I43 of the P3N domain individually disrupt the cell death induced by P3N-PIPO, but do not affect the PD localization of P3N-PIPO. Furthermore, WMV mutants with L35A or I43A can systemically infect PI 414723 plants. These key residues guide us to discover some WMV isolates potentially breaking the Wmr resistance. Through searching the NCBI database, we discovered some WMV isolates with variations in these key sites, and one naturally occurring I43V variation enables WMV to systemically infect PI 414723 plants. Taken together, these results demonstrate that P3N-PIPO, but not P3, is the avirulence determinant recognized by Wmr, although the shared N terminal P3N domain can alone trigger cell death. This work reveals a novel viral avirulence (Avr) gene recognized by a resistance (R) gene. This novel viral Avr gene is special because it is a transcriptional slippage product from another virus gene, which means that their encoding proteins share the common N-terminal domain but have distinct C-terminal domains. Amazingly, we found that it is the common N-terminal domain that determines the Avr-R recognition, but only one of the viral proteins can be recognized by the R protein to induce cell death. Watermelon mosaic virus (WMV) is one of the most important viruses of cucurbit plants. In this research, the reaction of 10 cantaloupe and eight watermelon Ajili (Jabani) inbred lines was evaluated under greenhouse conditions against a WMV isolate. Results indicated that only the inbred line of cantaloupe selfing 2 showed a delay in symptoms development which attributed to low disease severity index and average absorbance in ELISA test. Therefore, it was considered as resistant inbred line to the virus. While the inbred lines 3, 5, 6 and 8 were moderately susceptible and 4, 7, 10 and 11 were susceptible to the virus. Among watermelon inbred lines, 2 and 5 were found to be susceptible, whereas other accessions did not show any disease symptoms and also their severity index were recorded to be zero. Furthermore, results of enzyme-linked immunosorbent assay (ELISA) and RT-PCR tests showed that the later accessions are resistant to WMV. To assess the virus's impact on plant biomass, cantaloupe and watermelon inbred lines were mechanically inoculated with the WMV. Results indicated that the susceptible cantaloupes and watermelons against WMV were found to have significantly lower fresh leaves, dry stems and biomass than control plants (non-inoculated). Collectively, the cantaloupe inbred lines 1 and 2 and watermelon inbred lines 1, 3, 6, 7, 8 and 11 used in this study are probably resistant against WMV and can be recommended in breeding programs to establish more resistant crops. ELIZA test using antiserum to Watermelon mosaic virus (WMV) positive reaction with extract from squash plants showing mosaic, blistering and malformation leaves which indicate that the virus under study is WMV, the identification was confirmed by biological assay using quantitative indicator plant Chenopodium amaranticolor which respect chlorotic local lesions. The antibiotic Mitomycin, Para aminophenol, Chenopodium, alcoholic and aquatic extracts of thuja and chenopodium, and alcoholic extract of Eucalyptus at a high conc. showed a high inhibition on WMV in vitro when mixed with disease plant juice directly whereas the lowest inhibition effect resulted when used Bavastin at conc.0.5ml/L (3.8 spot / leaf). The alcoholic extract of Eucalyptus and Para aminophenol showed a high inhibitor effect on WMV in vivo, followed by alcoholic extract of Thuja and aquatic extract of Chenopodium, Bavastin and aquatic extract of Eucalyptus showed restricted effects on virus at 0.5ml /L and 12.5mg / ml respectively,. The higher inhibition of all treatments particularly at a high concentration were compatible with ELISA test results at 405nm.

It is well described that viral infections stimulate the emission of plant volatiles able to recruit viral vectors thereby promoting virus spread. In contrast, much less is known on the effects that emitted volatiles may have on the metabolism of healthy neighboring plants, which are potential targets for new infections through vector transmission. Watermelon mosaic virus (WMV) (genus Potyvirus, family Potyviridae) is an aphid-transmitted virus endemic in cucurbit crops worldwide. We have compared gene expression profiles of WMV-infected melon plants with those of healthy or healthy-but-cohabited-with-infected plants. Pathogenesis-related (PR) and small heat shock protein encoding genes were deregulated in cohabited plants, and PR deregulation depended on the distance to the infected plant. The signaling was short distance in the experimental conditions used, and cohabiting had a moderate effect on the plant susceptibility to WMV. Static headspace experiments showed that benzaldehyde and γ-butyrolactone were significantly over-emitted by WMV-infected plants. Altogether, our data suggest that perception of a volatile signal encoded by WMV-infected tissues triggers a response to prepare healthy tissues or/and healthy neighboring plants for the incoming infections. Cotton is the most important fiber crop worldwide. To determine the presence of viruses in cotton plants showing leaf roll and vein yellowing symptoms in Henan Province of China, a small RNA-based deep sequencing approach was performed. Analysis of the de novo-assembled contigs followed by reverse transcription PCR allowed the reconstruction of watermelon mosaic virus and an unknown virus. The genome of the unknown virus was determined to be 5870 nucleotides in length, and has a genomic organization with characteristic features of previously reported poleroviruses. Sequence analysis revealed that the virus was closely related to, but significantly different from, cotton leafroll dwarf virus, a polerovirus of the family Solemoviridae. This virus had less than 90% amino acid sequence identity in the products of both ORF0 and ORF1. According to the polerovirus species demarcation criteria set by the International Committee on Taxonomy of Viruses, this virus should be assigned to a new polerovirus species, for which we propose the name “cotton leaf roll virus”.

Watermelon mosaic virus (WMV) is one of the major viruses infecting cucurbit crops worldwide. Although WMV is very common worldwide, little is known about the biological traits of WMV isolates from China. Hence, this study aimed to characterize 11 WMV isolates infecting melon from different geographical origins in Xinjiang based on experimental hosts. Sap inoculation of the 11 WMV isolates onto a range of 13 plant species revealed some differences compared to the WMV isolates collected from other countries. Our results showed that, overall, there were no obvious correlations of host responses to inoculation with WMV isolates from different geographical origins. However, isolate JS-1 caused mild mosaic on Cucurbita moschata, whereas the remaining 10 isolates were asymptomatic on this plant species. Moreover, in Datura stramonium, isolate TYG-1 induced mosaic, whereas the remaining 10 isolates did not infect this species. All isolates infected systemically Cucurbita pepo and Cucumis melo plants, causing severe symptoms. All isolates did not induce any symptoms on Cucumis sativus, but the virus could be detected using RT-PCR. Additionally, all isolates infected systemically Nicotiana tabacum plants, causing mild mosaics. Chenopodium amaranticolor and Chenopodium quinoa reacted to all isolates by chlorotic local lesions in the inoculated leaves, and the virus was detected in the inoculated leaves using RT-PCR. In addition, the attempts to transmit the isolates to Luffa cylindrica, Vicia faba, Phaseolus vulgaris, Vigna unguiculata or Pisum sativum failed as confirmed by negative RT-PCR. Our results would be useful for understanding the biological variability of WMV.

The genus Cucurbita comprises many popular vegetable and ornamental plants, including pumpkins, squashes, and gourds, that are highly valued in China as well as in many other countries. During a survey conducted in Zhejiang province, Southeast China in 2016, severe symptoms of viral infection were observed on Cucurbita maxima Duch. ex Lam. Diseased plants showed symptoms such as stunting, mosaicking, Shoe string, blistering, yellowing, leaf deformation, and fruit distortion. Approximately, 50% of Cucurbita crops produced in Jinhua were diseased, causing an estimated yield loss of 35%. In this study, we developed a method using all known virus genomes from the NCBI database as a reference to map small RNAs to develop a diagnostic tool that could be used to diagnose virus diseases of C. maxima. 25 leaf samples from different symptomatic plants and 25 leaf samples from non-symptomatic plants were collected from the experimental field of Jihua National Agricultural Technology Garden for pathogen identification. Small RNAs from each set of three symptomatic and non-symptomatic samples were extracted and sequenced by Illumina sequencing. Twenty-four different viruses were detected in total. However, the majority of the small RNAs were from Zucchini yellow mosaic virus (ZYMV), Watermelon mosaic virus (WMV), and Cucumber mosaic virus (CMV). Mixed infections of these three viruses were diagnosed in leaf samples from diseased plants and confirmed by reverse transcription PCR (RT-PCR) using primers specific to these three viruses. Crude sap extract from symptomatic leaf samples was mechanically inoculated back into healthy C. maxima plants growing under greenhouse conditions. Inoculated plants developed the same disease symptoms as those observed in the diseased plants and a mixed infection of ZYMV, WMV, and CMV was detected again by RT-PCR, thus fulfilling Koch’s postulates. The diagnostic method developed in this study involves fewer bioinformatics processes than other diagnostic methods, does not require complex settings for bioinformatics parameters, provides a high level of sensitivity to rapidly diagnose plant samples with symptoms of virus diseases and can be performed cheaply. This method therefore has the potential to be widely applied as a diagnostic tool for viruses that have genome information in the NCBI database.

An upgraded nicking/polymerization strategy for ultrasensitive electrochemical detection of Watermelon mosaic virus (WMV) is proposed on the basis of the exonuclease and polymerase activity of T4 DNA polymerase and Mg2+-dependent DNAzyme-assisted and hemin/G-quadruplex DNAzyme-assisted cascade amplification strategies. Briefly, the hybridized DNA of the target WMV sequence, HP1, and P1 was recognized and nicked by nicking endonuclease Nb.BbvCI, and two DNA segments (P1-25 and P1-6) were produced. P1-25 was digested in the 3′→5′ direction, and digestion was halted at the 3′-terminal G locus with the exonuclease activity of T4 DNA polymerase. When dNTP solution mix was added to the mixture, an intact enzymatic sequence of Mg2+-dependent DNAzyme was synthesized by T4 DNA polymerase, which hybridized with its substrate sequence in the loop segment of HP2 immobilized on a gold electrode and initiated the cleavage round. The caged G-quadruplex sequence was released and formed hemin/G-quadruplex-based DNAzyme, resulting in sharply increased electrochemical signals. A correlation between the differential pulse voltammetry signal and the concentration of target WMV sequence was obtained in the range from 50 fM to 1 nM, with 50 fM detection limit. Because the nicking and polymerization reactions are irreversible and share the same buffer, the cascade amplification strategy is an ultrasensitive and high-efficiency strategy, indicating potential for viral detection. Watermelon mosaic virus (WMV), a plus-strand RNA virus that belongs to the family Potyviridae, is one of the most damaging viruses that infect cucurbit crops. However, isolates of this species frequently induce mild symptoms, which makes difficult to manage the disease in the field and breeding programs. A new generation of marker genes has been recently developed to visually track plant virus infection. Virus-mediated expression of the Pantoea ananatis phytoene synthase (crtB) induces accumulation in infected tissues of pigmented carotenoids that can be readily detected by the naked eye. Here we investigated whether this visual marker may facilitate visual diagnosis of WMV infection in cucurbits. First, we cloned a mild WMV isolate (WMV-Vera) and built a recombinant clone that expresses crtB (WMV-crtB). Next, we inoculated a series of cucurbit cultivars frequently used in breeding programs. We observed that WMV-mediated expression of crtB helps to visually score infection in two susceptible and highly appreciated melon cultivars, such as Piñonet Piel de Sapo and Vedrantais, but not in susceptible Cucurbita spp. cultivars. An increase in yellow pigmentation was experimentally confirmed in susceptible melon cultivars by measuring the Hue angle using a solid colorimeter. Carotenoid analyses in infected tissues explained why the visual crtB marker performs better in susceptible melon than in Cucurbita spp. cultivars. Viruses constitute a major constraint to soybean production worldwide and are responsible for significant yield losses every year. Although varying degrees of resistance to specific viral strains has been identified in some soybean genetic sources, the high rate of mutation in viral genomes and mixed infections of different viruses or strains under field conditions usually hinder the effective control of viral diseases. In the present study, we generated transgenic soybean lines constitutively expressing the double-strand RNA specific ribonuclease gene PAC1 from Schizosaccharomyces pombe to evaluate their resistance responses to multiple soybean-infecting virus strains and isolates. Resistance evaluation over three consecutive years showed that the transgenic lines displayed significantly lower levels of disease severity in field conditions when challenged with soybean mosaic virus (SMV) SC3, a prevalent SMV strain in soybean-growing regions of China, compared to the non-transformed (NT) plants. After inoculation with four additional SMV strains (SC7, SC15, SC18, and SMV-R), and three isolates of bean common mosaic virus (BCMV), watermelon mosaic virus (WMV), and bean pod mottle virus (BPMV), the transgenic plants exhibited less severe symptoms and enhanced resistance to virus infections relative to NT plants. Consistent with these results, the accumulation of each virus isolate was significantly inhibited in transgenic plants as confirmed by quantitative real-time PCR and double antibody sandwich enzyme-linked immunosorbent assays. Collectively, our results showed that overexpression of PAC1 can increase multiple virus resistance in transgenic soybean, and thus provide an efficient control strategy against RNA viruses such as SMV, BCMV, WMV, and BPMV. Insect endosymbionts (hereafter, symbionts) can modify plant virus epidemiology by changing the physiology or behavior of vectors, but their role in nonpersistent virus pathosystems remains uninvestigated. Unlike propagative and circulative viruses, nonpersistent plant virus transmission occurs via transient contamination of mouthparts, making direct interaction between symbiont and virus unlikely. Nonpersistent virus transmission occurs during exploratory intracellular punctures with styletiform mouthparts when vectors assess potential host-plant quality prior to phloem feeding. Therefore, we used an electrical penetration graph (EPG) to evaluate plant probing of the cowpea aphid, Aphis craccivora Koch, an important vector of cucurbit viruses, in the presence and absence of two facultative, intracellular symbionts. We tested four isolines of A. craccivora: two isolines were from a clone from black locust (Robinia pseudoacacia L.), one infected with Arsenophonus sp. and one cured, and two derived from a clone from alfalfa (Medicago sativa L.), one infected with Hamiltonella defensa and one cured. We quantified exploratory intracellular punctures, indicated by a waveform potential drop recorded by the EPG, initiation speed and frequency within the initial 15 min on healthy and watermelon mosaic virus-infected pumpkins. Symbiont associations differentially modified exploratory intracellular puncture frequency by aphids, with H. defensa-infected aphids exhibiting depressed probing, and Arsenophonus-infected aphids an increased frequency of probing. Further, there was greater overall aphid probing on virus-infected plants, suggesting that viruses manipulate their vectors to enhance acquisition-transmission rates, independent of symbiont infection. Watermelon mosaic virus (WMV) is widespread in cucurbit crops, most commonly occuring in temperate and Mediterranean regions. In Serbia WMV has been detected in single and mixed infections with Zucchini yellow mosaic virus and Cucumber mosaic virus in field-grown pumpkin and squash crops. Among pumpkin-affecting viruses WMV is the most frequent one, both by the number of localities and its incidence at each location. During the growing season of 2009, samples from 583 plants of Cucurbita pepo cvs. Olinka, Belgrade zucchini and Tosca (Zucchini group), as well as from C. maxima and C. moschata showing symptoms of virus infection were collected from 12 commercial fields at eight localities and analyzed by DAS-ELISA using polyclonal antisera specific to six most important cucurbit viruses. Interestingly, WMV was detected at fewer sites and had lower ncidence rate than in two previous years. In single infections, WMV was found in 11% of tested plants in three fields; in mixed infections with ZYMV, it was recorded in 9.9% of plants in five fields and with CMV in only 0.2% in one field. The partial coat protein gene and 3’ non-translated region from two representativeisolates of WMV originating from different localities and host plant species were amplified by RT-PCR, sequenced, and compared with the sequences available in GenBank database. The PCR-amplified fragment of predicted size of approximately 1017 bp was obtained. The sequences of isolates 137-08 (Acc. No. GQ259958) and 159-08 (GU144020) proved to be 94-99% identical at the nucleotide level with those from other parts of the world. The sequences of these two isolates differed from each other only at two nucleotide positions, without any amino acid substitution. Phylogenetic analysis of 57 isolates based on 750 bp sequences of the coat protein gene showed no correlation between isolates and their geographic origin, and italso indicated that these isolates fell into three molecular groups of strains. Serbian WMV isolates belong to group I, the most widespread group of isolates of this virus in the world.

Managing crop viruses is difficult due to complex interactions among vectors, reservoirs, and mediating factors such as land cover. Identifying the appropriate ecological neighborhood, or the spatial area in which the most influential interactions occur affecting virus epidemiology, would therefore be beneficial in exposing which of the many explanatory variables to target in the plant-pathogen system. We constructed partial least squares path models to find the neighborhood size for vectors of stylet-borne nonpersistent viruses infecting pumpkins, and compare the relative influence of within-field and extra-field land cover. Two economically important aphid-vectored viruses in the U.S. Midwest are included in these analyses: papaya ringspot virus (PRSV) and watermelon mosaic virus (WMV). In 2010 and 2011, we used commercial pumpkin fields to quantify virus infection, vector (aphid) alightment (i.e., landing rates), and within-field weed communities, and subsequently analyzed extra-field cover from 1 to 5 km concentric buffer zones within the surrounding landscape. Alightment rates of total noncolonizing vectors and the top three numerically dominant species ( Aphis craccivora Koch, Therioaphis trifolii (Monell), and Rhopalosiphum padi [L.]) were included in individual path models. Overall, we found that extra-field landscape composition had a far stronger influence on vector alightment than within-field weed cover; this pattern was consistent for seven of eight statistical models. In one exception, weed cover influenced alightment of the putative PRSV vector, A. craccivora, while surrounding landscape had no effect. In this case, weed coverage and vector alightment were inversely related, demonstrating that aphids were less likely to land in weed-infested fields. However, weed cover did not predict alightment of T. trifolii or total noncolonizers. The neighborhood size scales for total noncolonizers’ tended to be larger than for individual species (4–5 km), suggesting future studies of dispersal by multi-species aphid groups may benefit from an extended gradient. On balance, our results indicate that while surrounding land cover interactions are complex, they exert greater influence over vector dispersal than within-field weed cover, calling into question whether management of local weeds is an effective method of crop virus prevention in some systems. Watermelon mosaic virus (WMV) is an important disease of cucumber (Cucumis sativus L.) that is difficult to control in the field. Some resistant lines are available, and understanding the inheritance of the disease and mapping the gene or genes that confer resistance will facilitate the development of further resistant varieties. In this study, the inheritance and gene mapping for resistance to WMV in cucumber were conducted using populations derived from the cross between susceptible ‘65G’ and resistant ‘02245’ inbred lines. Genetic analysis showed that resistance to WMV in ‘02245’ is controlled by a single recessive gene designated as wmv02245 mapped to chromosome 6 (Chr.6). The region was flanked by the molecular markers SSRWMV60-23 and CAPS-W1 with genetic distances of 0.34 and 1.19 cM from the wmv02245 locus, respectively. The 134.7 kb physical distance of this region includes 21 candidate genes. Comparison of the genotypic and phenotypic results showed that the accuracy rate of the most closely linked marker SSRWMV60-23 was 94.0 %. This marker will be used for molecular marker-assisted selection to select resistant lines, and future research will be directed at identifying and cloning the resistance gene. [ABSTRACT FROM AUTHOR]
