Abstract

Anti-HIV-active polysaccharides and polyphenols were isolated from the brown seaweed Fucus vesiculosus by hot H2O extraction of both the intact and the homogenized algae. This was followed by XAD2 chromatography and by sequential precipitation of the non-adsorbed compounds with glacial HOAc and thereafter with EtOH. The precipitate was solubilized, dialyzed against distilled H2O, and chromatographed on SP-Sephadex C25 and on QAE-Sephadex A25. This was followed by gel filtration on Sephadex G50 and Sephadex G100 and finally by hplc on a Shodex Ionpak S-804 column. For comparison, the commercial product fucoidan, a sulfated algal polysaccharide, was also further purified by the chromatographic techniques mentioned above. The isolated freeze-dried fractions obtained by these procedures were tested for inhibition of both HIV-induced syncytium formation and HIV reverse transcriptase enzyme activity. Some of these fractions inhibited both of these activities at concentrations that were not cytotoxic.

26. Toxic chlorobiphenyls in adipose tissue and whole blood of an occupationally/acidentally exposed man and the general population

Abstract

Individual polychlorinated biphenyl (CB) congeners in adipose and whole blood tissues of a capacitor manufacture worker (occupational/accidental exposure) and of the general population were determined. Nondestructive sample cleanup and multidimensional gas chromatography-electron capture detection (MDGC-ECD) techniques were applied. Special attention was given to the toxicologically relevant congeners CB-77, CB-126, CB-169, CB-105, CB-114, CB-118, CB-156, CB-167, and CB-189 (IUPAC). Lipid-based tissue contents of sigma CBs were higher in the occupationally/accidentally exposed worker (adipose = 7,000 ng/g; whole blood = 11 ng/g) than in the general population (adipose = 300 ng/g; whole blood = 1.9 ng/g). Among the non-ortho Cl CBs, CB-77 was below detection limit (< 1.0 pg/g). The contents of CB-126 for the capacitor manufacture worker were 2,000 and 3 pg/g, and in the general population were 100 and 2 pg/g for adipose and whole blood, respectively. The corresponding data for CB-169 were 3,500 and 4 for adipose and whole blood, respectively, in the exposed worker, and 40 and 2 for adipose and whole blood, respectively, in the general population. Congeners with highest contents were CB-153, CB-138, CB-180, CB-170, and CB-187. These congeners possess chlorine substitution patterns, making them resistant to metabolism. X/153 ratios suggest that both PB and 3-MC type cytochrome P-450 enzymes were induced in the exposed worker, as well as in the general population.

Abstract

In three species chronic treatment with the anorectic drug chlorphentermine causes a profound alteration of the phospholipid/lipid metabolism in the organism, resulting in an increase of the fractions of phospholipids and lipids, e.g. in lungs, livers and adrenals. The results are interpreted as drug-induced generalized phospholipidosis, which is caused by amphiphilic drugs, like chlorphentermine and others. Abstract

Chronic treatment of rats with chlorphentermine (44 mg/kg, per day, i.p. for 8 weeks) alters the lipid metabolism both of lung tissue and of alveolar macrophages (AM). Calculated for one lung three times more lipid could be extracted from lung tissue and ten times more from AM, compared with controls. The molar ratio lipids: chlorphentermine is 10∶1 in the lung tissue and 7∶1 in AM. It reflects the remarkable binding of this drug to lipids, which is supposed to be the reason for the reduced breakdown mainly of phospholipids. [ABSTRACT FROM AUTHOR]

Abstract

In order to obtain information on the behaviour of the urinary bladder wall towards pharmacologically interesting compounds, the uptake of two considerably different substances (i.e. 14C-labelled prilocaine and 14C-urea) by isolated bladder pieces (guinea pig) has been investigated. The uptake was measured as a function of the incubation time, and of the concentration and pH-value of the bath fluid. The free base (pH-value of the medium 9.0) of prilocaine (pKa=7.7 at 36° C) penetrates more rapidly into the bladder tissue than the protonated form (pH-value of the medium 6.4).
