SummaryPromoters of developmental genes in embryonic stem cells (ESCs) are marked by histone H3 lysine 4 trimethylation (H3K4me3) and H3K27me3 in an asymmetric nucleosomal conformation, with each sister histone H3 carrying only one mark. These bivalent domains are thought to poise genes for timely activation upon differentiation. Here we show that asymmetric bivalent nucleosomes recruit repressive H3K27me3 binders but fail to enrich activating H3K4me3 binders, despite presence of H3K4me3, thereby promoting a poised state. Strikingly, the bivalent mark combination further attracts chromatin proteins that are not recruited by each mark individually, including the histone acetyltransferase complex KAT6B (MORF). Knockout of KAT6B blocks neuronal differentiation, demonstrating that bivalency-specific readers are critical for proper ESC differentiation. These findings reveal how histone mark bivalency directly promotes establishment of a poised state at developmental genes, while highlighting how nucleosomal asymmetry is critical for histone mark readout and function.

The hydrological responses of a catchment are sensitive to, and strongly coupled to, land use and climate, and changes thereof. The hydrological responses to the impacts of changing land use and climate will be the result of complex interactions, where the change in one may moderate or exacerbate the effects of the other. Further difficulties in assessing these interactions are that dominant drivers of the hydrological system may vary at different spatial and temporal scales. To assess these interactions, a process-based hydrological model, sensitive to land use and climate, and changes thereof, needs to be used. For this purpose the daily time step ACRU model was selected. However, to be able to use a hydrological model such as ACRU with confidence its representation of reality must be confirmed by comparing simulated output against observations across a range of climatic conditions. Comparison of simulated against observed streamflow was undertaken in three climatically diverse South African catchments, ranging from the semi-arid, sub-tropical Luvuvhu catchment, to the winter rainfall Upper Breede catchment and the sub-humid Mgeni catchment. Not only do the climates of the catchments differ, but their primary land uses also vary. In the upper areas of the Mgeni catchment commercial plantation forestry is dominant, while in the middle reaches there are significant areas of commercial plantation sugarcane and urban areas, while the lower reaches are dominated by urban areas. The Luvuvhu catchment has a large proportion of subsistence agriculture and informal residential areas. In the Upper Breede catchment in the Western Cape, commercial orchards and vineyards are the primary land uses. Overall the ACRU model was able to represent the high, low and total flows, with satisfactory Nash-Sutcliffe efficiency indexes obtained for the selected catchments. The study concluded that the ACRU model can be used with confidence to simulate the streamflows of the three selected catchments and was able to represent the hydrological responses from the range of climates and diversity of land uses present within the catchments.

The International Maize and Wheat Improvement Center (CIMMYT) acts as a catalyst and leader in a global maize and wheat innovation network that serves the poor in the developing world. Drawing on strong science and effective partnerships, CIMMYT researchers create, share, and use knowledge and technology to increase food security, improve the productivity and profitability of farming systems and sustain natural resources. This people-centered mission does not ignore the fact that CIMMYT’s unique niche is as a genetic resources enhancement center for the developing world, as shown by this review article focusing on wheat. CIMMYT’s value proposition resides therefore in its use of crop genetic diversity: conserving it, studying it, adding value to it, and sharing it in enhanced form with clients worldwide. The main undertakings include: long-term safe conservation of world heritage of both crop resources for future generations, in line with formal agreements under the 2004 International Treaty on Plant Genetic Resources for Food and Agriculture, understanding the rich genetic diversity of two of the most important staples worldwide, exploiting the untapped value of crop genetic resources through discovery of specific, strategically-important traits required for current and future generations of target beneficiaries, and development of strategic germplasm through innovative genetic enhancement. Finally, the Center needs to ensure that its main products reach end-users and improve their livelihoods. In this regard, CIMMYT is the main international, public source of wheat seed-embedded technology to reduce vulnerability and alleviate poverty, helping farmers move from subsistence to income-generating production systems. Beyond a focus on higher grain yields and value-added germplasm, CIMMYT plays an “integrative” role in crop and natural resource management research, promoting the efficient use of water and other inputs, lower production costs, better management of biotic stresses, and enhanced system diversity and resilience.

Physically-based animation techniques enable more realistic and accurate animation to be created. We present a fully physically-based approach for efficiently producing realistic-looking animations of facial movement, including animation of expressive wrinkles. This involves simulation of detailed voxel-based models using a graphics processing unit-based total Lagrangian explicit dynamic finite element solver with an anatomical muscle contraction model, and advanced boundary conditions that can model the sliding of soft tissue over the skull. The flexibility of our approach enables detailed animations of gross and fine-scale soft-tissue movement to be easily produced with different muscle structures and material parameters, for example, to animate different aged skins. Although we focus on the forehead, our approach can be used to animate any multi-layered soft body.

Physically-based animation techniques enable more realistic and accurate animation to be created. Such approaches require the creation of a complex simulation model that, for computer graphics applications, can efficiently produce realistic-looking animations. We present a process to automatically create animatable nonconforming hexahedral finite element (FE) simulation models of facial soft tissue, including automatic computation of skin layers and element material properties, muscle properties and boundary conditions, making them immediately ready for simulation. Using the GPU, the detailed models can simulate complex gross and fine-scale behaviour, such as wrinkling. Our process can also be used to create a multi-layered FE model of any object (not just soft tissue).
