Background The nuclear progesterone receptor (Pgr) is a ligand-dependent transcription factor primarily responsible for mediating progesterone actions relevant for reproduction across vertebrates. Information on the cellular localization of Pgr expression in the reproductive system is required for developing a comprehensive approach to elucidate the role of Pgr in reproduction. Results We generated transgenic zebrafish Tg(pgr:eGFP) that express enhanced green fluorescent protein (eGFP) driven by promoter sequence of pgr gene. The tissue distribution pattern of egfp mRNA is consistent with the pgr mRNA expression in Tg(pgr:eGFP). In the pituitary, GFP signals are found in the proximal pars distalis. In order to better discern the cellular localization of GFP signals in gonads, Tg(pgr:eGFP) line was crossed with Tg(gsdf:nfsB-mCherry) line, specifically expressing nitroreductase-mCherry fusion protein in granulosa and Sertoli cells in ovary and testis, respectively. Imaging of testis tissue showed that GFP expression was confined to Leydig cells. In the ovary, GFP expression colocalized with the mCherry signal in granulosa cells. Intriguingly, we also identified some non-granulosa cells close to where blood vessels branched, expressing stronger GFP signals than granulosa cells. Conclusions Analyzing Tg(pgr:eGFP) expression in zebrafish provided leads toward new routes to study the role of Pgr in reproduction.

In the context of rapid urbanization, the detailed planning of urban design lacks an integrated consideration of old and new spaces and humanized design, revealing a number of aspects that need to be improved. The living street environment, which is most intimately connected to people’s daily activities, typically lacks convenience and comfort, a humanized design, street dynamics, and a natural and humanistic appearance. In order to better understand the direction of improving the living street space in the context of urbanization, this paper discusses the research background, the concepts related to street space, analyzes the research methods, specific cases, and current problems, and through the literature research and quantitative research, puts forward ideas on the optimization strategies of living street space, and attempts to summarize the generally applicable direction of street space optimization.

The large yellow croaker (Larimichthys crocea) is one of the most valuable economic mariculture fish in China, and its yellowness in skin is considered as the most important traits that determine the market value. The fish skin colors present naturally silvery white during the daytime and golden yellow during the night-time. In the present study, we found that the xanthosomes of xanthophores aggregated during the daytime and dispersed during the night-time, while the melanosomes of melanophores always dispersed all day long in vivo. We further cloned three proopiomelanocortin (POMC) genes of the large yellow croaker. All these POMC genes were expressed in the pituitary, but only POMC-C mRNA was expressed in the skin. The results of single-cell RT-PCR showed that the POMC-C mRNA was expressed in isolated xanthophores but not in melanophores. The xanthophores, but not melanophores, were found in the ventral skin, where only one subtype of melanocortin receptor (MCR), i.e. MC5R, was expressed. MC1R, MC4R and MC5R mRNAs were detected in the dorsal skin, where there existed both the xanthophores and melanophores. The results of single-cell RT-PCR showed that only MC5R mRNA was expressed in the xanthophores, while only MC1R mRNA was detected in the melanophores. Thirty min after in vivo and in vitro treatment with melanocyte-stimulating hormone (MSH) peptides (α-MSH, Des-Ac-α-MSH, α-MSH-C and β-MSH-C), dispersion degrees of the xanthosomes and melanosomes significantly increased, even under light exposure. Interestingly, when we immediately observed the dorsal scales that have been maintained under in vitro complete dark condition without MSH peptides treatment for at least 30 min, the melanosomes aggregated as expected, but the xanthosomes dispersed. Both α-MSH and α-MSH-C exhibited similar, but stronger effects than Des-Ac-α-MSH on dispersion of both the xanthosomes and melanosomes. Although the two MSH peptides (α-MSH-C and β-MSH-C) are generated from POMC-C, the former showed stronger effect on xanthosome dispersion than the latter. Forskolin, an Adenylyl cyclase activator, dispersed pigment granules in both the xanthophores and melanophores. H 89 2HCl, a specific inhibitor of protein kinase A blocked α-MSH-induced pigment granule dispersion of xanthophores and melanophores. Taken together, our results indicated that the dispersion of melanosomes is stimulated by MSH, possibly secreted from pituitary, through endocrine pathway. The yellowness of the large yellow croaker skin was mainly due to the dispersion of xanthosomes. The dispersion of xanthosomes was regulated by MSH that is possibly released both from the pituitary through endocrine pathway and from the xanthophores through autocrine pathway.

It is well documented that androgens modify olfactory processing in vertebrates. In fish, several lines of evidence indicate that androgens increase olfactory sensitivity to prostaglandin pheromone, but the molecular mechanism is still unclear. Our previous studies showed that prostaglandin E2 (PGE2) is a sex pheromone in the burrowing-living fish Chinese black sleeper (Bostrychus sinensis) and that the PGE2 receptor 1 (Ep1) in the olfactory rosette is a candidate receptor for sensing sex pheromone PGE2. In the present study, we found that testosterone (T) and 11-ketotestosterone (11-KT) exhibited stimulatory effects on the expression of ep1 in the olfactory rosette in vivo and ex vivo. Moreover, the androgen receptor (Ar) agonist R1881 had similar effects to 11-KT on the expression of ep1 ex vivo, suggesting the up-regulatory effect is mediated by Ar. The amount of arα transcripts (˜1500 copies/100 ng total RNA) was greater than that of arβ (˜300 copies/100 ng total RNA) in the olfactory rosette, and the expression levels of arα increased with spermatogenesis and peaked at late meiosis stage. Moreover, activated Arα but not Arβ transactivated a 2k bp ep1 promoter in HEK293T cell, and some OSNs exhibited co-localization of arα mRNA and Ep1 protein signals. Taken together, our results suggest that Arα, but not Arβ, plays a crucial role in mediating the androgen-induced up-regulation of ep1 expression in B. sinensis. The present study is the first to shed light on the molecular mechanisms whereby androgens enhance responsiveness to prostaglandin sex pheromones in teleosts.

Cold acclimation has a tremendous impact on the survivability and the freshness of turbot when presented to consumers for sale. In the present study, to better understand the response of turbot to cold acclimation, physiological parameters determination, two-dimensional-based proteomic analysis and the expression of target genes by quantitative real-time PCR were analyzed. The results of physiological parameters indicated that the levels of glucose, ATP and cortisol, and the activities of lysozyme, catalase (CAT) and superoxide dismutase (SOD) were elevated. Moreover, the liver damage may be determined due to the increase of glutamic-oxalacetic transaminase (GOT) and glutamic-pyruvic transaminase (GPT) in serum. Furthermore, a total of sixteen differentially accumulated proteins were identified, which are mainly involved in carbohydrate and energy metabolism. Based on these data, we concluded that turbot accelerate the glycogen hydrolysis in liver and muscle to increase the glucose level in serum and promote the energy metabolism, enhance innate immune and heighten the antioxidant capacity to respond to cold acclimation. Meanwhile, the cold acclimation can also cause adverse effect to the innate immune system and liver of turbot. These results are helpful in understanding the response of turbot to cold acclimation and in providing the theoretical base for applying fish cold acclimation in transport.

The mudskipper Boleophthalmus pectinirostris inhabits intertidal mudflats, exhibiting semilunar reproductive rhythms. To investigate whether melanopsin is possibly involved in the synchronization of the semilunar spawning rhythm in the female mudskipper, we first cloned all four melanopsin subtypes (opn4m1, opn4m3, opn4x1, opn4x2) in B. pectinirostris. Results from RTq-PCR showed that significantly higher transcription levels of all four melanopsin subtypes were observed in the eyes rather than other tissues. In brain, all four melanopsin subtypes were also detectable in different regions, including the telencephalon, in which the expression of melanopsin has not been reported in other teleosts. The transcription levels of opn4m3 and opn4x1 in the telencephalon exhibited a daily fluctuation pattern. When females entered the spawning season, opn4m1 and opn4x1 transcript levels increased significantly in the telencephalon. During the spawning season, the transcript levels of opn4m3 and opn4x1 in the telencephalon appeared to have a cyclic pattern associated with semilunar periodicity, exhibiting two cycles with a peak around the first or the last lunar quarters. Results from ISH showed that, opn4x1 mRNA was localized in the medial of dorsal telencephalic area, dorsal nucleus of ventral telencephalic area (Vd), ventral nucleus of ventral telencephalic area (Vv), anterior part of parvocellular preoptic nucleus, magnocellular part of the magnocellular preoptic nucleus (PMmc), habenular and ventral zone of hypothalamus. Intriguingly, gnrh3 mRNA was also located in Vd, Vv and PMmc. Taken together, our results suggested that melanopsins, e.g. opn4x1, expressed in the telencephalon might mediate semilunar spawning activity in the female mudskipper.

The Chinese black sleeper ( Bostrychus sinensis ) is a commercially important fish in southeastern China, but the effects of salinity on the growth of B. sinensis fry remain unclear. In the present study, the three month-old B. sinensis fry were reared for 5 weeks at 5, 15, 25 and 35 parts per thousand (ppt) salinities to evaluate the effects of salinity on the survival rate, specific growth rate, condition factor, feed intake (FI), food conversion efficiency (FCE), plasma osmolality, plasma Cl − concentration and the gill Na + -K + -ATPase (NAK) activities. In addition, since the insulin-like growth factor (IGF) system plays important roles in osmoregulation and promoting growth in teleosts, the complete B. sinensis insulin-like growth factor-1 receptor ( igf-1r ) gene was cloned and igf-1r mRNA levels in the muscle and gill were measured at the end of this experiment. Our results showed that the fry reared at lower salinities (5 and 15 ppt) grew significantly faster than those at higher salinities (25 and 35 ppt), and the fry reared at 15 ppt exhibited lower FI and higher FCE than those reared at 5 ppt. A typical ‘U-shaped’ pattern of gill NAK activity levels with the lowest one at 15 ppt was observed, suggesting that a lower energy expenditure on osmoregulation at this level of salinity. The length of the complete igf-1r cDNA sequence was 6864 bp, and it had a wide range of tissue expression including muscle and gill. After 5 weeks of rearing, muscle igf-1r mRNA levels were similar among the four salinity groups, while the gill igf-1r mRNA level at 5 ppt was significantly higher than that at 15 ppt. Taken together, the results from the present study indicated that the optimal salinity for rearing B. sinensis fry was 15 ppt, and that IGF-1 might serve as a hyperosmoregulatory hormone in long-term low salinity acclimation in B. sinensis .

Public concern regarding silver nanoparticles (AgNPs) in the environment has been increasing since they can cause adverse effects in some aquatic species. However, few data are actually available on the effects of AgNPs on the germ cells. In the present study, we used the zebrafish ovarian follicle as a model to assess the potentially adverse effects of AgNPs on oocyte maturation (germinal vesicle breakdown, GVBD) in vitro. Similar to the maturation inducing hormone (17α, 20β-dihydroxy-4-pregnen-3-one), AgNPs induced GVBD, and reduced the total cyclic adenosine monophosphate (cAMP) concentration in zebrafish ovarian follicles. The results from transmission electron microscope observation and Hoechst 33342 staining clearly indicated that AgNPs induced apoptosis in ovarian follicle cells surrounding the oocyte. Similar to AgNPs, AgNO3 also induced GVBD, decreased cAMP concentration and induced apoptosis of ovarian follicle cells. However, the results from gene expression analysis showed that transcript levels of oxidative stress related genes were more sensitive to AgNPs than AgNO3. Further more, H2O2 has an ability to induce zebrafish oocytes maturation by induction of apoptosis in ovarian follicle cells. Taken together, the results from our study indicated that oxidative stress appeared to be one of important mechanisms in AgNP induced apoptosis in ovarian follicle cells, which further triggered the GVBD.

The mudskipper Boleophthalmus pectinirostris is a burrow-dwelling fish inhabiting intertidal mudflats. During the spawning season, in a spawning chamber located at the center of their burrow, a pair of male and female fish mate and fertilized eggs adheres onto the inner walls and ceiling with filamentous attachments. During 5 days of incubation, the fertilized eggs are kept clean and hatch with a very high hatching rate under the natural conditions filled with microorganisms. This suggests that the male and/or female reproductive tract may synthesize antimicrobial substances to offer protection against microorganisms that may be deleterious to fertility. To study the antimicrobial strategy of this fish in the spawning season, we first cloned the two hepcidin isoforms from B. pectinirostris, and designated them as Hepcidin-1 and Hepcidin-2 based on phylogenetic analyses. Both of these hepcidin isoforms were highly expressed in the liver, but only Hepcidin-1 showed significant change in response to iron overload. Interestingly, these two hepcidin isoforms were expressed in male reproductive tracts, i.e. the testes and seminal vesicles. The monthly expression pattern indicated that Hepcidin-1 transcript levels showed a peak point only in March (before spawning) in the seminal vesicle, while Hepcidin-2 transcript levels were correlated with male reproductive status and reached their highest level in May (the peak spawning period). Under experimental conditions, the expression of these two hepcidin isoforms showed no response to iron overload in the male gonad. However, after lipopolysaccharide injection, the Hepcidin-1 transcript level was significantly up-regulated in the testes and seminal vesicle 6 h post injection, while Hepcidin-2 transcript levels exhibited a clear time-course dependent upregulation pattern and reached the highest levels 24 h post injection. More interestingly, after injection with LHRH-A3, the expression of Hepcidin-2 was significantly up-regulated in both testes and seminal vesicle. Results from in situ hybridization showed that Hepcidin-2 was expressed in the Leydig cells of the testes and in the epithelium of the seminal vesicle. Taken together, the results from our study indicated that these two hepcidin isoforms in the mudskipper may have different functions: Hepcidin-1 may play a dual role in both iron metabolism regulation in the liver and a short antimicrobial response in male reproductive tracts, while Hepcidin-2 is more specialized in reproductive immunity in male reproductive tracts.

AMP-activated protein kinase (AMPK) is a highly conserved and multi-functional protein kinase that plays important roles in both intracellular energy balance and cellular stress response. In the present study, molecular characterization, tissue distribution and gene expression levels of the AMPK α1 and α2 genes from turbot (Scophthalmus maximus) under salinity stress are described. The complete coding regions of the AMPK α1 and α2 genes were isolated from turbot through degenerate primers in combination with RACE using muscle cDNA. The complete coding regions of AMPK α1 (1722 bp) and α2 (1674 bp) encoded 573 and 557 amino acids peptides, respectively. Multiple alignments, structural analysis and phylogenetic tree construction indicated that S. maximus AMPK α1 and α2 shared a high amino acid identity with other species, especially fish. AMPK α1 and α2 genes could be detected in all tested tissues, indicating that they are constitutively expressed. Salinity challenges significantly altered the gene expression levels of AMPK α1 and α2 mRNA in a salinity- and time-dependent manners in S. maximus gill tissues, suggesting that AMPK α1 and α2 played important roles in mediating the salinity stress in S. maximus. The expression levels of AMPK α1 and α2 mRNA were a positive correlation with gill Na+, K+-ATPase activities. These findings will aid our understanding of the molecular mechanism of juvenile turbot in response to environmental salinity changes.

Accumulating evidence suggest that membrane progestin receptor α (mPRα) is the membrane receptor mediating nongenomic progestin signaling that induces oocyte maturation in teleost. However, the involvement of other members of mPR family in oocyte maturation is still unclear. In this study, we found impaired oocyte maturation in zebrafish lacking mPRα1, mPRα2, mPRβ, or mPRγ2. In contrast, no difference was observed in oocyte maturation in the single knockout of mPRγ1, mPRδ, or mPRε. To study possible redundant functions of different mPRs in oocyte maturation, we generated a zebrafish line lacking all seven kinds of mPRs (mprs(−/−)). We found oocyte maturation was further impaired in mprs(−/−). In addition, oocyte ovulation delay was observed in mprs(−/−) females, which was associated with low levels of nuclear progestin receptor (Pgr), a key regulator for ovulation. We also found reduced fertility in mprs(−/−) female zebrafish. Furthermore, eggs spawned by mprs(−/−) females were of poor quality.

The combined method with high oxygen filling under low temperature and waterless condition is suggested to be a more effective strategy to keep fish alive during turbot transportation. To investigate the biological mechanism of turbot, when turbot was subjected to waterless treatment under different oxygen conditions (water, air and oxygen), the global expression profiling of the gill tissues was performed by transcriptome analysis using Illumina HiSeq 4000 platform. In vertebrates, the aromatase coded by the cyp19a1a gene can catalyze the conversion from androgens to estrogens. Thus, the regulatory mechanisms of cyp19a1a gene expression are a critical research field in reproductive endocrinology. In this study, we use zebrafish as a model to study the dynamic methylation levels of the cyp19a1a gene core promoter during zebrafish ovarian folliculogenesis. The results show that there is an apparent fluctuation of the methylation levels of zebrafish cyp19a1a core promoter. Moreover, the methylation levels are inversely correlated with the expression levels of cyp19a1a transcripts when the ovarian follicles develop from PV into the MV stage. Also, the CpG dinucleotides which are close to the transcriptional starting site may have provided a significant blocking effect on inhibiting the transcriptional function of RNA polymerase II. Taken together, the results from the present study strongly suggest that DNA methylation was one of mechanisms that are involved in the regulation of cyp19a1a gene expression during folliculogenesis. This methylation mechanism modifying transcriptional process accompanied with zebrafish ovarian folliculogenesis might also shed new light on the regulation of cyp19a1a expression during the ovarian developmental stage in other vertebrates.

Gonadal soma-derived factor (Gsdf) is a new member of the transforming growth factor beta superfamily. As a teleost- and gonad-specific growth factor, several studies indicate that Gsdf plays an important role in early germ cell development. In the present study, for the first time, a 1700-bp long gsdf gene was cloned from a protandrous species, Acanthopagrus latus. We further analyzed the cellular localization and the expression patterns of gsdf in respective testicular and ovarian zones during the first maleness reproductive phase. The results showed that gsdf transcripts were highly expressed in the ovotestis during sex differentiation, and the somatic cells of the testicular zone expressed many more gsdf transcripts than those of the ovarian zone. At the onset of puberty, the gsdf expression levels decreased gradually during spermatogenesis. Conversely, the ovarian zone exhibited a stable increase pattern which was similar to the plasma 17β-estradiol (E2) levels. These results suggested that Gsdf may participates in early germ cell development, e.g. proliferation and differentiation of spermatogonia and oogonia in A. latus.

Sodium alginate (SA) and tea polyphenols (TP) are natural preservatives commonly used in the food industry, including the production of fish products. The effect of SA coating infused with TP on the quality of fresh Japanese sea bass (Lateolabrax japonicas) fillets was evaluated over a 20-day period at 4 °C. SA (1.5%, w/v) or TP (0.5%, w/v) treatment alone, and the SA coating infused with TP (SA-TP) all reduced microbial counts, with the SA-TP providing the greatest effect. Fish fillet samples treated with SA-TP had significantly lower levels of total volatile basic nitrogen, lipid oxidation, and protein decomposition during the storage period, relative to the remaining treatments. The samples treated with SA-TP had the highest sensory quality rating as well. Collectively, sodium alginate coating infused with tea polyphenols may represent a promising treatment for preservation of Japanese sea bass fillets during cold storage.The sodium alginate-tea polyphenols composite coating has strong potential to be used as a new biopreservative for maintaining fish fillet quality.

Previously we had identified adamts9 as a downstream target of Pgr, which is essential for ovulation in zebrafish. The primary goal of this study is to determine whether human chorionic gonadotropin (hCG, LH analog) also regulate adamts9 expression prior to ovulation. The expression of adamts9 was induced by hCG in a dose and time dependent manner in zebrafish preovulatory follicles in vitro. Interestingly, the stimulatory effect of hCG on adamts9 expression was not blocked in pgr(−/−) follicles but blocked in lhcgr(−/−). This effect of hCG was via Lhcgr and its associated cAMP and PKC signaling pathways. Reduced fecundity and reduced expression of adamts9 were also found in lhcgr(−/−) females in vivo. Therefore, we have provided the first evidence of gonadotropin (hCG) regulated adamts9 in zebrafish, which could be important for ovulation.

The aim of the present study was to investigate the protein-sparing effect of carbohydrate in diets for juvenile turbot (Scophthalmus maximus) reared at five salinities (12, 18, 24, 30, and 36). The fish were fed three isocaloric and isolipidic diets for 60 days. The results show that specific growth rate (SGR) and feed conversion efficiency (FCE) were higher in fish reared at salinities of 18 and 36, but lower at 12. Fish fed with diet C25P40 (25% carbohydrate and 40% protein) had lower SGR and FCE values compared with those fed with the C5P52 (5% carbohydrate and 52% protein) and C15P46 (15% carbohydrate and 46% protein) diets; however, there was no statistical difference between diet C5P52 and C15P46. SGR and FCE values were unaffected by diet composition in fish reared at salinity 36. Hepatic lipogenic enzyme activities were higher in fish reared at 18 and 36, but lower at 12, while glucokinase (GK) activity was higher in fish reared at 12, and lower at 18 and 36. Dietary starch enhanced GK activity while depressing lipogenic enzyme activity. However, lipogenic enzyme activity increased with increasing dietary starch in fish reared at 36. It is recommended that salinity should be maintained >12 in the farming of juvenile turbot. In addition, an increase in gelatinized starch from 5% to 15% could spare 6% dietary protein in fish reared at salinities of 18–30, while higher salinity (36) could improve dietary carbohydrate use and enhance the protein-sparing effect, which is linked with the induction of lipogenic capacities.

The genetic structure and demographic history of nine mudskipper Boleophthalmus pectinirostris populations on the Northwestern Pacific coast were investigated using the complete mitochondrial cytochrome b sequences. Among the 248 individuals examined, 118 haplotypes were detected. As a whole, high haplotype diversity and moderate nucleotide diversity were observed within the populations. The topology of the neighbor-joining tree of B. pectinirostris haplotypes was assigned into two closely related lineages (A and B) that appeared to have strong geographic genetic structure. Lineage A included the populations from Jiangsu, Zhejiang, Fujian, Taiwan, Guangdong, Guangxi and Vietnam while Lineage B included populations from Japan and Korea. The complete mitochondrial genome sequence of the serpent mudskipper Parapocryptes serperaster was first determined in this study. The circle genome was 17 243 bp long and consisted of 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes, and 1 control region. The nucleotide composition of the heavy strand of P. serperaster is 28.65% for A, 30.07% for C, 25.31% for T, and 15.97% for G, with a slight A + T bias of 53.96%. Only the NADH dehydrogenase subunit 6 (ND6) and eight tRNA genes were encoded on the light strand, all other mitochondrial genes were encoded on the heavy strand. A 178-bp tandem repeat, which started from the 827-bp site of the control region, was identified. Like most other gobies, P. serperaster also has the typical vertebrate mitochondrial gene arrangement. The phylogenetic analysis showed that the kinship between Parapocryptes and Boleophthalmus is closer than those between Parapocryptes and other selected genera.

The mudskipper Boleophthalmus pectinirostris, a burrow-dwelling fish inhabiting intertidal mudflats, spawns only once during the spawning season around either the first or last lunar quarters. To understand the molecular mechanisms regulating this semilunar spawning rhythm, we cloned all melatonin receptor subtypes (mtnr1a1.4, mtnr1a1.7, mtnr1b, and mtnr1c). Expression of three melatonin receptor subtypes (except mtnr1c) was found in the ovaries. In contrast, the expression of all receptor subtypes was found in the diencephalon and the pituitary. In the fully-grown follicles, only mtnr1a1.7 mRNA was detected in both the isolated follicle layers and denuded oocytes. Interestingly, the transcript levels of both mtnr1a1.4 in the diencephalon and mtnr1a1.7 in the ovary displayed two cycles within one lunar month, and peaked around the first and last lunar quarters. We used 17α,20β-dihydroxy-4-pregnen-3-one (DHP), a maturation-inducing hormone, as a biomarker to examine the involvement of melatonin receptors in the control of the spawning cycle. Melatonin significantly increased the plasma DHP level 1h post intraperitoneal injection. Melatonin also directly stimulated ovarian fragments in vitro to produce a significantly higher amount of DHP. Taken together, these results provided the first evidence that melatonin receptors were involved in the synchronization of the semilunar spawning rhythm in the female mudskipper by acting through the HPG axis and/or directly on ovarian tissues to stimulate the production of DHP.

The central roles of luteinizing hormone (LH), progestin and their receptors for initiating ovulation have been well established. However, signaling pathways and downstream targets such as proteases that are essential for the rupture of follicular cells are still unclear. Recently, we found anovulation in nuclear progestin receptor (Pgr) knockout (Pgr-KO) zebrafish, which offers a new model for examining genes and pathways that are important for ovulation and fertility. In this study, we examined expression of all transcripts using RNA-Seq in preovulatory follicular cells collected following the final oocyte maturation, but prior to ovulation, from wild-type (WT) or Pgr-KO fish. Differential expression analysis revealed 3,567 genes significantly differentially expressed between WT and Pgr-KO fish (fold change ≥2, p3,000) is differentially regulated in the follicular cells in zebrafish prior to ovulation, terminating programs such as growth and proliferation, and beginning processes including the inflammatory response and apoptosis. Further studies are required to establish relationships among these genes and an ovulatory circuit in the zebrafish model.

Our previous study showed that the in vivo positive effects of 17α,20β-dihydroxy-4-pregnen-3-one (DHP), the major progestin in zebrafish, on early spermatogenesis was much stronger than the ex vivo ones, which may suggest an effect of DHP on the expression of gonadotropins. In our present study, we first observed that fshb and lhb mRNA levels in the pituitary of male adult zebrafish were greatly inhibited by 3 weeks exposure to 10nM estradiol (E2). However, an additional 24h 100nM DHP exposure not only reversed the E2-induced inhibition, but also significantly increased the expression of fshb and lhb mRNA. These stimulatory effects were also observed in male adult fish without E2 pretreatment, and a time course experiment showed that it took 24h for fshb and 12h for lhb to respond significantly. Because these stimulatory activities were partially antagonized by a nuclear progesterone receptor (Pgr) antagonist mifepristone, we generated a Pgr-knockout (pgr–/–) model using the TALEN technique. With and without DHP in vivo treatment, fshb and lhb mRNA levels of pgr–/– were significantly lower than those of pgr+/+. Furthermore, ex vivo treatment of pituitary fragments of pgr–/– with DHP stimulated lhb, but not fshb mRNA expression. Results from double-colored fluorescent in situ hybridization showed that pgr mRNA was expressed only in fshb-expressing cells. Taken together, our results indicated that DHP participated in the regulation of neuroendocrine control of reproduction in male zebrafish, and exerted a Pgr-mediated direct stimulatory effect on fshb mRNA at pituitary level.

The mudskipper Boleophthalmus pectinirostris and Chinese black sleeper Bostrichthys sinensis occupy the intertidal zone. However, both species have their own unique diet. The former is an herbivore and the latter is a carnivore. In order to reveal the relationship between digestive enzyme activities and diets in the two species, the activities of protease (P), non-specific bile salt-activated lipase (BAL) and α-amylase (A) were determined in the stomach and intestine of adult mudskipper B. pectinirostris and Chinese black sleeper B. sinensis. The results showed that the activities of protease, BAL and α-amylase in the intestine of B. pectinirostris were significantly (P 0.05), while BAL and α-amylase activities of the intestine were significantly (P 0.05). Cardiac performance of the Chinese black sleeper(Bostrichthys sinensis)was examined at early stages of development (the heartbeat, blood circulation and newly hatched larval stages) under different hypoxic oxygen concentrations. Exposure of the embryos at 25°C to 0.2 mg/l dissolved oxygen (DO) indicated periodic heartbeat at both the heartbeat stage and the blood circulation stage, but it started sooner after exposure at the blood circulation stage than at the heartbeat stage. With the appearance of the gill slits, the tolerance of the newly hatched larvae to hypoxia was improved. The larvae did not display periodic heartbeat at 0.2 mg/L DO, but periodic heartbeat occurred at 0.1 mg/L DO and 0.02 mg/L DO after 23.3±6.2 minutes and 13.2±3.4 minutes of exposure. With exposure of the embryos to hypoxia, the occurrence of periodic heartbeat of excised embryos was earlier than that of whole egg embryos at both the blood circulation and heartbeat stages. These results indicated that the tolerance of embryos to hypoxia decreased with the progress of embryogenesis and suggested that the perivitelline liquid can protect the embryos by conserving oxygen. In this study, we also found that a breakdance performance (an arrhythmic heart beat) occurred in the newly hatched larvae at 0.02 mg/l DO. The breakdance performance lasted about 10 minutes, and then the heartbeat became normal.

Variation in the production of the plasma steroid hormones E 2 , 17α-OHP and T in females and T and 11-KT in males, was investigated in the mudskipper Boleophthalmus pectinirostris during the spawning season. Females with oocytes at the vitellogenic stage (GSI 5.97–6.86%) and mature males with GSI of 0.255–0.288% were collected at intervals of 3–4 days within the two complete semilunar cycles from May 31 to June 30, 2006. The results showed that variations in the levels of plasma steroid hormones were synchronized obviously with semilunar periodicity in both females and males. Each steroid hormone level exhibited two cycles, each cycle with a peak. In females, the first peaks in plasma E 2 , 17α-OHP and T levels were observed 3 days after the first lunar quarter, and the second ones, 4 days after the last lunar quarter. In males, the first peaks of plasma T and 11-KT levels occurred 3 days after the first lunar quarter, and the second ones, at the last lunar quarter. The fact that, in the present study, changes in the levels of plasma steroid hormones were synchronized with semilunar periodicity, although the fish were at the same stages of gonadal development, suggests that variation of plasma steroid hormones is basically regulated by biological rhythms (Zeitgebers), and that tidal movement (with its semilunar periodicity) is the major environmental factor stimulating steroid hormone production in B. pectinirostris.

The mudskipper Boleophthalmus pectinirostris maintained territories in farming ponds during the cold season between November and February. There were two types of territory, one was surrounded by mud-walls, and the other was without mud-walls. Both types of territory were entirely covered by shallow water. The area of walled territories was significantly bigger than that of non-walled ones. The nearest neighbour distance of walled territories was significantly shorter than that of non-walled territories. In January, fish surface activity occurred between 1200 and 1500 hours at which time the territorial water temperature reached a maximum, being significantly higher than that of the air, mud surface or deep burrow. The territorial sediments exhibited a significantly greater benthic microalgal biomass (chlorophyll-a and total chlorophyll-a) as compared to the non-territorial sediments. The present study indicates that B. pectinirostris forms a territory during the cold season to keep a pool of water that encourages diatom growth and enables the fish to engage in surface activity.

The complete mitochondrial genome sequence of the barred mudskipper Periophthalmus argentilineatus was first determined in this study. The circle genome was 16 509 bp long and consisted of 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes, and 1 control region. The nucleotide composition of the heavy strand of P. argentilineatus is 28.28% for A, 27.83% for C, 16.01% for T, and 27.88% for G, with a slight G + C bias of 55.71%. Only the NADH dehydrogenase subunit 6 (ND6) and eight tRNA genes were encoded on the light strand, other mitochondrial genes were all encoded on the heavy strand. The mitochondrial gene arrangement of P. argentilineatus is similar to those of most other gobies. The phylogenetic analysis using the neighbor-joining method showed that the kinship between Periophthalmus and Boleophthalmus is closer than those between Periophthalmus and other selected genera. Periophthalmus argentilineatus was clustered into one branch with other four species from the same genus Periophthalmus.

In this paper, the complete mitochondrial genome sequence of Scartelaos gigas was firstly determined. The circular genome (16 717 bp) comprises 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and 1 control region. The overall base composition of S. gigas is 28.9% for C, 28.3% for A, 26.4% for T, 16.4% for G, with a slight A + T bias of 54.7%. In the control region, the termination-associated sequence and conserved sequence block domains were found, but the tandem repeat structure was not found. It has the typical vertebrate mitochondrial gene arrangement. The phylogenic analysis using the Neighbor-Joining method showed that the fishes belonging to Gobiidae, Odontoburidae, and Eleotridae formed three branches grouped with other fishes into one clade which separated from the mammals. We hope that the results from the present study will provide useful molecular information for the further studies on genetic structure and demographic history of S. gigas.

Our previous studies suggested that 17α,20β-dihydroxy-4-pregnen-3-one (DHP), an oocyte maturation inducing progestin, also acts as a sex pheromone in Chinese black sleeper Bostrichthys sinensis, a fish species that inhabits intertidal zones and mates and spawns inside a muddy burrow. The electro-olfactogram response to DHP increased during the breeding season. In the present study, we cloned the cDNAs of the nine progestin receptors (pgr, paqr5, 6, 7(a, b), 8, 9, pgrmc1, 2) from B. sinensis, analyzed their tissue distribution, and determined the expression in the olfactory rosette during the reproductive cycle in female and male fish. The deduced amino acid sequences of the nine progestin receptors share high sequence identities with those of other fish species and relatively lower homology with their mammalian counterparts, and phylogenetic analyses classified the nine B. sinensis progestin receptors into their respective progestin receptor groups. Tissue distribution of B. sinensis progestin receptors showed differential expression patterns, but all these nine genes were expressed in the olfactory rosette. Interestingly, paqr5 mRNA was found in the intermediate and basal parts of the olfactory epithelium but not in the central core using in situ hybridization, and its expression level was the highest in the olfactory rosette among the tissues examined. These results suggested Paqr5 may have an important role for transmitting progestin signaling in the olfactory system. The expression levels of paqr7a and paqr7b, pgr and pgrmc2 mRNA peaked around the mid meiotic stage, and that of paqr8 peaked at late meiotic stage in the olfactory rosette in males, while the olfactory expression of paqr5 decreased gradually as spermatogenesis progressed. In contrast, the expression of the progestin receptors did not change significantly during the development of the ovary in the olfactory rosette in females, except that of pgr. Interestingly, the changes of paqr8 expression in the olfactory rosette in males mirrored the changes of plasma DHP levels in females during the reproductive cycle, suggesting the Paqr8 may also be important for deciphering progestin signaling released by female. To our knowledge, this is the first time to demonstrate the presence of all known progestin receptors in a teleost olfactory rosette, and to show different expressions between the males and females during the reproductive cycle. This study provides the first evidence on changes of all purported progestin receptors during a reproductive cycle in teleost olfactory rosette, and suggests that distinct olfactory sensitivities to DHP may be due to the changes and compositions of each progestin receptor in B. sinensis.

Gonadal soma-derived factor (gsdf) is a teleost- and gonad-specific growth factor involved in early germ cell development. The red spotted grouper, Epinephelus akaara, as a protogynous hermaphrodite, provides a novel model for understanding the mechanisms of sex determination and differentiation in teleosts. In the present study, a 2307-bp long gsdf gene was cloned from E. akaara and there was further analysis of its tissue distribution and gonadal patterns of gene expression during the female phase and sex change developmental stages. The cellular localization of gsdf at the late transitional developmental stage was also analyzed. In addition, the concentrations of serum sex steroid hormones (E2, 11-KT and DHP) were determined. The gsdf transcripts were exclusively localized in the gonad. During the female phase at an early developmental stage, when the ovotestis contained mainly oogonia and primary growth oocytes, the gsdf mRNA was relatively more abundant. The relative abundance of gsdf decreased, however, and the lesser amount was sustained with the advancement of oocyte development. During the transitional phase, the relative abundance of gsdf mRNA increased slightly at the early developmental stage and there were further increases in relative abundance in the late developmental stage, and the gsdf transcripts were observed in the Sertoli cells surrounding early developing spermatogonia. Among the sex steroids, 11-KT concentrations were positively correlated with amount of gsdf mRNA during sex change. These results suggest that gsdf could have roles in regulating pre-meiotic germ cell proliferation and be involved in sex change in E. akaara.

Anatomical studies showed that all collected feral fish of Chinese black sleepers (Bostrichthys sinensis L.) were gonochoristic. In contrast, an average of 12.4% of cultured fish appeared to be hermaphroditic. The shape of the genital papilla of hermaphroditic specimens resembled that of gonochoristic males. The gonadosomatic index (IG) of hermaphroditic fish varied from 0.24 to 4.48%, while that of gonochoristic conspecifics varied from 6.1 to 12.2% for females and 0.11 to 0.14% for males. Histological studies revealed that the sex organs of hermaphrodites consisted of ovarian, testicular, and seminal vesicle tissues, located in the central, peripheral, and caudal region of the gonads, respectively. During the spawning season, ovarian tissue contained poorly developed oocytes, that is, hydrated oocytes were absent and only a few degenerated vitellogenic oocytes were present. Testicular tissue, however, contained fully developed spermatozoa. The fertilization rate of these spermatozoa was significantly lower than those of gonochoristic conspecifics. It is concluded that part of the cultured stock of Chinese black sleeper became hermaphroditic during culture and that the hermaphrodites are able to function as males. The lowered fertilization rate, however, indicates that hermaphroditism in cultured fish of this species is disadvantageous to reproduction.

Ovarian, testicular and seminal vesicle extracts of mature Chinese black sleeper (Bostrichthys sinensis Lacepede), synthetic steroids 17[alpha]-progesterone (17[alpha]-P), 17[alpha],20[beta]-dihydroxy-4-pregnen-3one (17[alpha],20[beta]-P) and prostaglandins PGE₂ and PGF[subscript 2[alpha]] were used in an attempt to attract broodfish and to induce spawning. Furthermore, 17[alpha]-P, PGE₂ and PGF[subscript 2[alpha]] levels were determined in extracts, and PGE₂ and PGF[subscript 2[alpha]] levels were determined in holding and spawning waters. The results showed that ovarian extracts attracted more males than females to enter artificial nests, while testicular and seminal vesicle extracts attracted more females than males. The highest percentage of spawned nests were found when 17[alpha],20[beta]-P and PGE₂ were administered to the nests, while the largest egg numbers and the mean highest fertilization rate were observed when PGE₂ was added. The artificial breeding of marine fish in China has been developed over a period of more than 50 years. Both species diversity and fry production have greatly increased since the 1980s. By the year 2000, at least 52 species belonging to 24 families of marine fish had been successfully bred. Large quantities of cultured fry can meet the needs of both sea cage culture and pond culture for most species. Fish fry production has entered a period of sustainable development. The number of artificially produced fry of large yellow croaker (Pseudosciaena crocea) exceeded 1.3 billion in year 2000. The species for which more than 10 million fry is produced annually through aquaculture are: red drum (Sciaenops ocellatus), Japanese sea perch (Lateolabrax japonicus), redeye mullet (Liza haematocheila), cuneate drum (Nibea miichthioides), red seabream (Pagrosomus major), milkfish (Chanos chanos) and three-banded sweetlip (Plectorhynchus cinctus). Several millions of artificial fry are cultured annually for the following species: Japanese flounder (Paralichthys olivaceus), black porgy (Sparus macrocephalus), silver sea perch (Lates calcarifer), Russell's snapper (Lutjanus russelli), javelin grunt (Pomadasys hasta), miiuy croaker (Miichthys miiuy), Chinese black sleeper (Bostrichthys sinensis) and skewband grunt (Hapalogenys nitens). Fishes belonging to the family Sciaenidae are now the major species for artificial propagation.

Sox genes play important roles in various developmental processes such as sex determination, embryogenesis, oogenesis, neurogenesis, and larval development. In order to clarify the roles of Sox genes in the developmental process of large yellow croaker, the full-length cDNA of the Sox11a gene (Lc-Sox11a) was cloned for the first time. Bioinformatics analysis indicated that Lc-Sox11a contains a protein of 366 amino acids with a Ser-rich region, a C-terminal conserved region, and a high mobility group box. The expression of Lc-Sox11a in different tissues of both sexes and in different developmental embryonic stages revealed that Lc-Sox11a were expressed with tissue and gender specificity, of which the expression level in female was ovary>brain>eye>gill; in male was brain>testis>gill. The gender differences occurred in the brain and eye with the male brain>female brain, female eye>male eye. Moreover, the expression of Lc-Sox11a in the gonad and brain at different growth stages was detected. Significant up-regulated expression of Lc-Sox11a was found in the ovary and the male brain at 1000dph (days post hatching) compared with 270dph and 635dph. However, significant down-regulated expression of Lc-Sox11a occurred in the testis with growth. Besides, the expression of Lc-Sox11a in the female brain showed a trend of first rising then falling, with the highest peak in 635dph. The results of in situ hybridization displayed that Lc-Sox11a was widely distributed only in cytoplasm of oocytes at each stage in oogenesis. In early stage of oocytes, Lc-Sox11a was expressed weakly and evenly. As the appearance of vacuoles and synthesis of yolks, positive signals of Lc-Sox11a distributed intensively in the residual cytoplasm. In spermatogenesis, Lc-Sox11a was distributed in cytoplasm of all male germ cells except spermatozoon with spermatogonium>spermatocyte>spermatid. During embryogenesis, Lc-Sox11a was expressed in most embryonic stages, the highest expression occurred in the formation-of-eye-lens stage, closely followed by the closure-of-blastopore stage, then the beginning-of-heart-pulsation stage. The results of whole mount in situ hybridization showed that the expression of Lc-Sox11a began to increase beginning with the multiple-cell stage, with the major distribution of Lc-Sox11a in the brain and eye areas in the pre-hatching stage.

In this study, the complete mitogenome sequence of the Royal dottyback, Pictichromis paccagnellae (Perciformes: Pseudochromidae), has been sequenced by the next-generation sequencing method. The assembled mitogenome was of 16 976 bp in length, consisting of 13 protein-coding genes, 22 transfer RNAs, and two ribosomal RNAs genes. The overall base composition was 27.4% for A, 28.1% for C, 17.3% for G, and 27.2% for T and showed 76% identities to Fire-tail devil Labracinus cyclophthalmus in the same family. The complete mitogenome of P. paccagnellae provides essential and important DNA molecular data for further phylogeography and evolutionary analysis for Pseudochromidae.

In this study, the complete mitogenome sequence of the palette surgeonfish, Paracanthurus hepatus (Perciformes: Acanthuridae), has been sequenced by next-generation sequencing method. The assembled mitogenome was 16 498 bp in length, consisted of 13 protein-coding genes, 22 transfer RNAs, 2 ribosomal RNAs genes. The overall base composition of palette surgeonfish was 28.6% for A, 28.6% for C, 16.3% for G, 26.4% for T and showed 87% identities to somber surgeonfish Zebrasoma flavescens. The complete mitogenome of the palette surgeonfish provides essential and important DNA molecular data for further phylogeography and evolutionary analysis for surgeonfish's phylogeny.

Our previous studies suggested that prostaglandin E2 (PGE2) is a putative sex pheromone in Chinese black sleeper Bostrichthys sinensis, a fish species that inhabits intertidal zones and mates and spawns inside a muddy burrow. We found immunoreactivities of PGE2 receptor subtypes (Ep1-3) expressed in the olfactory sac, but only Ep1 presented higher density of immunoreactivity in mature fish than that in immature fish in both sexes. To gain a better understanding of the underlying molecular mechanism for the detection of PGE2 in the olfactory system, we cloned an ep 1 cDNA from the adult olfactory sac. The open-reading frame of the ep 1 consisted of 1,134-bp nucleotides that encoded a 378-amino acid-long protein with a seven-transmembrane domain, typical for the G protein-coupled receptors superfamily. Expression of ep 1 mRNA was observed in all tissues examined, with higher levels obtained in the olfactory sacs and testes. The expression of ep 1 mRNA in the olfactory sacs and gonads was significantly higher in both sexes of mature fish than in those of immature ones. Taken together, our results suggested that Ep1, which is highly expressed in the olfactory sacs and gonads of mature fish, is important for the control of reproduction and may be involved in PGE2-initiated spawning behavior in B. sinensis.

The Boddart's goggle-eyed mudskipper, Boleophthalmus boddarti (Perciformes, Gobiidae) is an amphibious fish, inhabiting brackish waters of estuaries and builds burrows in soft mud along the intertidal zone. In this paper, the complete mitochondrial genome sequence of B. boddarti was firstly determined. The circle genome (16,727 bp) comprises 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes and 1 control region. The overall base composition of B. boddarti is 29.1% for C, 28.9% for A, 25.9% for T, and 16.0% for G, with a slight A + T bias of 54.8%. The termination-associated sequence, conserved sequence block domains, and a 131-bp tandem repeat were found in the control region. It has the typical vertebrate mitochondrial gene arrangement.

Mudskippers are amphibious fishes that have developed morphological and physiological adaptations to match their unique lifestyles. Here we perform whole-genome sequencing of four representative mudskippers to elucidate the molecular mechanisms underlying these adaptations. We discover an expansion of innate immune system genes in the mudskippers that may provide defence against terrestrial pathogens. Several genes of the ammonia excretion pathway in the gills have experienced positive selection, suggesting their important roles in mudskippers’ tolerance to environmental ammonia. Some vision-related genes are differentially lost or mutated, illustrating genomic changes associated with aerial vision. Transcriptomic analyses of mudskippers exposed to air highlight regulatory pathways that are up- or down-regulated in response to hypoxia. The present study provides a valuable resource for understanding the molecular mechanisms underlying water-to-land transition of vertebrates., Mudskippers are amphibious fishes that have adapted to live on mudflats. Here, the authors sequence the genomes of four different mudskipper species and highlight genetic changes that may have had an evolutionary role in the water-to-land transition of vertebrates.

The changes of ultrastructures of hepatic cells of Boleophthalmus pectinirostris were investigated after the fish were exposed under benzo(a) pyrene in different concentrations under experimental condition. The results showed that the organelles in hepatic cells of B. pectinirostris were damaged to different extents after the fish was exposed under lower concentration of BaP (0.5 mg.L-1) for up to 7 d, in which, mitochondria and endoplasmic reticulum were the chief organelles affected by BaP exposure. While the fish was exposed under higher concentration of BaP (5 mg.L-1) for 2 h, almost all of the organelles including mitochondria and endoplasmic reticulum in hepatic cells of B. pectinirostr were affected by BaP exposure. The structures of liver cells were seriously damaged. It was demonstrated that BaP could produce multiorganalle lesions in hepatic cells of B. pectinirostris, and the severity extent of such lesions was dependent on the concentration level of BaP.
