Abstract

Genus Pseudomonas bacteria mainly consume glucose through the Entner–Doudoroff (ED) route due to a lack of a functional Embden–Meyerhof–Parnas (EMP) pathway. In the present study, a 6-phosphogluconate dehydratase (edd) operon in the ED route was well investigated to find its structural characteristics and roles in the regulation of glucose consumption and 2-ketogluconic acid (2KGA) metabolism in the industrial 2KGA-producer P. plecoglossicida JUIM01. The edd operon contained four structural genes of edd, glk, gltR, and gtrS, encoding 6-PG dehydratase Edd, glucokinase Glk, response regulatory factor GltR, and histidine kinase GtrS, respectively. A promoter region was observed in the 5′-upstream of the edd gene, with a transcriptional start site located 129 bp upstream of the edd gene and in a pseudo-palindromic sequence of 5′-TTGTN7ACAA-3′ specifically binding to the transcription factor HexR. The knockout of the edd gene showed a remarkably negative effect on cell growth and re-growth using 2KGA as a substrate, beneficial to 2KGA production, with an increase of 8%. The deletion of glk had no significant effect on the cell growth or glucose metabolism, while showing an adverse impact on the 2KGA production, with a decrease of 5%. The outputs of the present study would provide a theoretical basis for 2KGA-producer improvement with metabolic engineering strategies and the development and optimization of P. plecoglossicida as the chassis cells. Abstract

Water and energy shortages are interdependent major worldwide issues that cannot be disregarded. In this work, graphene and BaTiO3 are used to synergistically facilitate the self‐assembly of the β‐phase that is known to induce the piezoelectric properties of the polyvinylidene fluoride (PVDF). This leads to a PVDF/graphene‐BaTiO3 nanocomposite with a unique capability of integrating Phra Phrom‐like four functions into one single asymmetric membrane: i) solar evaporation, ii) power generation, iii) piezo‐photodegradation, and iv) self‐cleaning/monitoring for environmental remediation and resources regeneration. The high heat accumulation capability and piezoelectric performance of the membrane enable it to simultaneously achieve a water production rate of 0.99 kgm−2h−1, in compliance with WHO standards, and a maximum power output of 5.73 Wm−2 in simulated natural environments. Upon subjecting the membranes to environmental cleaning, they not only show a 93% dye degradation rate due to the synergistic effect of piezoelectricity and photocatalysis but also resolve the membrane fouling issue, exhibiting ≈200% resistance change compared to the static state. The successful integration of these four functions into one membrane shows the great potential of this work toward a more sustainable and viable water and energy production approach. Abstract

Background: Tokyo, the capital of Japan, is a densely populated city of >13 million people, so the population is at high risk of epidemic severe acute respiratory coronavirus 2 (SARS-CoV-2) infection. A serologic survey of anti–SARS-CoV-2 IgG would provide valuable data for assessing the city’s SARS-CoV-2 infection status. Therefore, this cross-sectional study estimated the anti–SARS-CoV-2 IgG seroprevalence in Tokyo. Methods: Leftover serum of 23,234 hospital visitors was tested for antibodies against SARS-CoV-2 using an iFlash 3000 chemiluminescence immunoassay analyzer (Shenzhen YHLO Biotech, Shenzhen, China) with an iFlash–SARS-CoV-2 IgG kit (YHLO) and iFlash–SARS-CoV-2 IgG-S1 kit (YHLO). Serum samples with a positive result (≥10 AU/mL) in either of these assays were considered seropositive for anti–SARS-CoV-2 IgG. Participants were randomly selected from patients visiting 14 Tokyo hospitals between September 1, 2020 and March 31, 2021. No participants were diagnosed with coronavirus disease 2019 (COVID-19), and none exhibited COVID-19-related symptoms at the time of blood collection. Results: The overall anti–SARS-CoV-2 IgG seroprevalence among all participants was 1.83% (95% confidence interval [CI], 1.66–2.01%). The seroprevalence in March 2021, the most recent month of this study, was 2.70% (95% CI, 2.16–3.34%). After adjusting for population age, sex, and region, the estimated seroprevalence in Tokyo was 3.40%, indicating that 470,778 individuals had a history of SARS-CoV-2 infection. Conclusions: The estimated number of individuals in Tokyo with a history of SARS-CoV-2 infection was 3.9-fold higher than the number of confirmed cases. Our study enhances understanding of the SARS-CoV-2 epidemic in Tokyo.

Abstract

ObjectivesTo compare the temporal changes of IgM, IgG, and IgA antibodies against the SARS-CoV-2 nucleoprotein, S1 subunit, and receptor binding domain and neutralizing antibodies (NAbs) against SARS-CoV-2 in patients with COVID-19.MethodsA total of five patients in Nissan Tamagawa Hospital, Tokyo, Japan confirmed COVID-19 from August 8, 2020 to August 14, 2020 were investigated. Serum samples were acquired multiple times from 0 to 76 days after symptom onset. Using a fully automated CLIA analyzer, we measured the levels of IgG, IgA, and IgM against the SARS-CoV-2 N, S1, and RBD and NAbs against SARS-CoV-2.ResultsThe levels of IgG antibodies against SARS-CoV-2 structural proteins increased over time in all cases but IgM and IgA levels against SARS-CoV-2 showed different increasing trends among individuals in the early stage. In particular, we observed IgA antibodies increasing before IgG and IgM in 3/5 cases. The NAb levels against SARS-CoV-2 increased and kept above 10 AU/mL more than around 70 days after symptom onset in all cases. Furthermore, in the early stage, NAb levels were more than cut off value in 4/5 COVID-19 patients some of whose antibodies against RBD didn’t exceed 10 AU/mL.ConclusionsOur findings indicate that patients with COVID-19 should be examined for IgG, IgA and IgM antibodies against SARS-CoV-2 structural proteins and NAbs against SARS-CoV-2 in addition to conventional antibody testing methods for SARS-CoV-2 (IgG and IgM kits) to analyze the diversity of patients’ immune mechanisms.

49. Exploring the response mechanism of Pseudomonas plecoglossicidato high-temperature stress by transcriptomic analyses for 2-keto gluconic acid production

Abstract

High temperatures, particularly in summer, lead to decreased yields in the industrial application of Pseudomonas plecoglossicidafor 2-keto gluconic acid (2KGA) fermentation. To address this, the alterations in the transcriptomics of P. plecoglossicidain response to high-temperature stress were examined at temperatures of 32 °C, 36 °C, and 40 °C. The analysis of differential expression revealed substantial discrepancies in the number of differentially expressed genes (DEGs) at 36 °C (357) and 40 °C (1,487), primarily affecting vital biological functions. Elevated temperatures resulted in a shift in the metabolic processing of glucose, transitioning from extracellular oxidation to intracellular phosphorylation. Notable changes were observed in metabolic pathways, including the pentose phosphate pathway and tricarboxylic acid cycle. A significant observation was the decline in the activity of genes associated with extracellular glucose oxidation, accompanied by an increase in the activity of genes involved in intracellular phosphorylation pathway. This indicates a prompt and dynamic response to high-temperature stress. The investigation revealed notable alterations in genes linked to glucose metabolism, emphasizing the strain's adaptive capabilities to endure high temperatures. The reveal of adaptations are crucial for optimizing 2KGA production in challenging industrial environments.
