Abstract

Purpose: Nucleotide-based matrix-assisted laser desorption ionization time-of-flight mass spectrometry (nucleotide MALDI-TOF MS) is an emerging molecular technology used for the diagnosis of tuberculosis (TB) caused by Mycobacterium tuberculosis (MTB)and its drug resistance. This study aimed to compare the ability of nucleotide MALDI-TOF MS to detect rifampicin (RIF) resistance in drug-resistant TB (DR-TB) patients with Xpert MTB/RIF and to analyze the disparate results individually. Additionally, potential factors associated with rifampicin resistance among DR-TB patients in Qingdao were investigated. Patients and Methods: A retrospective study was conducted at Qingdao Chest Hospital, and patients with DR-TB were enrolled. Corresponding frozen isolates were recovered and subjected to nucleotide MALDI-TOF MS, Xpert MTB/RIF, and phenotypic drug susceptibility testing (pDST). Sanger sequencing was performed for the discordant results of nucleotide MALDI-TOF MS and Xpert MTB/RIF. Univariate and multivariate logistic regression analyses were used to identify potential factors associated with rifampicin resistance among patients with DR-TB. Results: A total of 125 patients with DR-TB (18.8%, 125/668) were enrolled in this study from May 1 to July 31, 2023. Rifampicin-resistant (DR-TB/RR, 29) and rifampicin-sensitive (DR-TB/RS, 96) groups were divided according to the pDST results. Nucleotide MALDI-TOF MS performed better than Xpert MTB/RIF in terms of sensitivity, specificity, accuracy, and agreement with pDST. Only six cases had inconsistent results, and the sequencing results of five cases were identical to nucleotide MALDI-TOF MS. Furthermore, chest pain (aOR=12.84, 95% CI, 2.29– 91.97, p=0.005), isoniazid sensitivity (aOR=0.14, 0.02– 0.59, p=0.013), and ethambutol sensitivity (aOR=0.02, 0.00– 0.10, p=0.000) were potential factors associated with rifampicin resistance among DR-TB patients in Qingdao. Conclusion: The overall concordance between nucleotide MALDI-TOF MS and Xpert MTB/RIF was 95.2%, with the former performing better in determining rifampicin susceptibility among DR-TB cases in Qingdao. Abstract

Electric field distortion caused by space charge accumulation is one of the challenges in the development of insulating liquid immersed converter transformers and battery energy storage. This paper studies the impact of temperature and moisture content on field distortion in synthetic ester through an improved computational model incorporating the time-development permittivity of insulating liquid, and the test results indicated highly accurate simulation results of electric field distribution can be produced compared with the previous model. Charge mobility and charge density in synthetic ester were extracted from frequency dielectric spectroscopy test results, which were then used to calculate field distortion by the developed computational model. Simulation results indicated that the field distortion in synthetic ester decreases with temperature due to the enhanced permittivity caused by space charge accumulation, while it increases with moisture content due to the increased charge density from the dissociation of water. 22. High-Frequency Partial Discharge Characteristics of Solid-State Transformer Interturn Multilayer Insulation Under Repetitive Electrical Stress

Abstract

The solid-state transformer (SST) was regarded as the ideal power router in the energy Internet. However, the partial discharge (PD) in tightly packed SST is one of the main causes of interturn multilayer insulation premature failure. The effect of high-frequency (HF) electrical-thermal coupling stress on SST multilayer insulation's PD characteristics and failure mechanism is not clarified, which is investigated in this article. An HF PD measurement system is established, and a comparative study of the PD characteristics of multilayer polyimide (PI) insulation at different test frequencies and temperatures is explored. The experimental results show that when the frequency exceeds 10 kHz, the PD exhibits a rabbit-ear-like discharge pattern. However, the higher temperature will lead to lower PD inception voltage with large PD magnitudes, which may accelerate the degradation of interturn insulation. According to the phase-resolved PD, PD magnitude, PD number, and discharge phenomenon of the PI layer, the failure process of the multilayer PI system is divided into three stages: electric tree development, electric tree breakdown, and point prebreakdown. In addition, we propose the PD development model of multilayer PI insulation. The research results can provide a theoretical reference for elucidating the PD mechanism under HF stresses.

Abstract

The Himalayan marmot (Marmota himalayana), a natural host and transmitter of plague, is also susceptible to the hepadnavirus infection. To reveal the genetic basis of the hepadnavirus susceptibility and the immune response to plague, we systematically characterized the features of immune genes in Himalayan marmot with those of human and mouse. We found that the entire major histocompatibility complex region and the hepatitis B virus pathway genes of the Himalayan marmot were conserved with those of humans. A Trim (tripartite motif) gene cluster involved in immune response and antiviral activity displays dynamic evolution, which is reflected by the duplication of Trim5 and the absence of Trim22 and Trim34. Three key regions of Ntcp, which is critical for hepatitis B virus entry, had high identity among seven species of Marmota. Moreover, we observed a severe alveolar hemorrhage, inflammatory infiltrate in the infected lungs and livers from Himalayan marmots after infection of EV76, a live attenuated Yersinia pestis strain. Lots of immune genes were remarkably up-regulated, which several hub genes H2r[gamma], Tra29, and Nlrp7 are placed at the center of the gene network. These findings suggest that Himalayan marmot is a potential animal model for study on the hepadnavirus and plague infection. Key words: Himalayan marmot, immune system, hepadnavirus, plague. La marmotte de l'Himalaya (Marmota himalayana), un hote naturel qui peut transmettre la peste, est egalement sensible a l'infection par l'hepadnavirus. Afin de reveler l'assise genetique de la sensibilite a l'hepadnavirus et de la reponse immunitaire a la peste, les auteurs ont systematiquement caracterise les genes du systeme immunitaire chez la marmotte de l'Himalaya avec ceux chez l'humain et la souris. Les auteurs ont trouve que la region entiere du complexe majeur d'histocompatibilite et les genes du sentier du virus de l'hepatite B chez la marmotte de l'Himalaya sont conserves avec ceux presents chez l'humain. Un groupe de genes Trim (<< a motif tripartite >>) implique dans la reponse immunitaire et l'activite antivirale presentait une evolution dynamique, laquelle se reflete par la duplication de Trim5 et l'absence des genes Trim22 et Trim34. Trois regions cles de Ntcp, lequel est critique pour l'entree du virus de l'hepatite B, presentaient un fort niveau d'identite au sein de sept especes de Marmota. Abstract

The lumped element network model has been proven to be an efficient tool for the interpretation of transformer Frequency Response Analysis. However, it is challenging to obtain parameters of the model if transformer design information is unavailable. In such a case, optimisation algorithms can be used for gray‐box model parameter estimation. A methodology is developed by the authors to establish a transformer network model without winding design data, and instead, end‐to‐end open circuit Frequency Response and other terminal test results are utilised; and Genetic Algorithm is applied to approximate the unknown parameters of the model. The modelling approach developed is independent of the unit number of the network model and therefore guarantees the accuracy of optimisation. Furthermore, it can deal with transformers with a complicated winding structure such as interleaved disc type winding. Two single windings, helical and interleaved disc type, and a single‐phase 144/13 kV 60 MVA transformer are used to demonstrate the method. FRA spectra produced by the best estimated gray‐box model and the corresponding white‐box model are compared. The main features in amplitude and phase spectra are well matched, with low values of Relative Standard Deviation, for both single windings and transformer. Estimated electrical parameters show a high consistency with reference values, which are calculated based on winding design data. Abstract

At present, the development of electric vehicles (EV) is limited by the lack of charging facilities. While the construction of charging stations is slowing down because of the small ownership of EV and the high operational cost. The mobile charging vehicles (MCV) are proposed to replace the charging station. At first, this paper introduces the MCV’s operation architecture. Then the spatial charging demand is estimated based on the travel chain. At last, the allocation method of mobile charging vehicles is studied in detail with the result of estimation result. Abstract

Partial discharge (PD) techniques have been used for many years for detecting incipient insulation faults in power transformers. However, reliable electrical techniques for locating the PDs still need to be developed. This lack of development is caused by the difficulty in extracting information from PD terminal signal measurements and relating this information to PD location. The main objective of this thesis is to develop an electrical PD location technique for use in power transformers. The overall approach consists of three elements. First, a computer simulation model is developed. Simulated PD pulses are injected into the winding at all locations at which a PD might occur. A file of results is created of all the terminal responses to the injected PDs. Second, a PD is injected into the transformer winding undergoing experimentation at some specific site, and the PD signals measured at the terminals are then analysed and the characteristics relating to the location of the PD extracted. Third, algorithms are constructed for matching the file of calculated results with the experimental results for the purpose of identifying the location of the PD in the transformers. A computer simulation model was specifically developed for studying the characteristics of PD propagation in power transformer windings. The model includes a lumped-element, multi-winding transformer model and a PD waveform model. Simulation analyses are carried out to define the transfer functions from the PD locations to the measuring terminals. Features contained in the spectra of the PD terminal signals relating to PD location are extracted. The effect of different winding construction, and terminal connections, on PD propagation are studied. Experimental tests were performed on full-size transformer windings to investigate the differences between the PD terminal signals produced when a PD pulse was injected at different locations. Experimental results were obtained for a multi-winding 220kV transformer winding and for a single 110kV transformer winding. Finally, PD location algorithms are developed based on comparing the characteristics of the PD signals measured at the transformer terminals experimentally, with those obtained from simulation analysis. Feature template matching methods and artificial neural network techniques are developed. It is shown that for the 110kV transformer winding, the feature template matching PD location algorithm predicts the location of the PD source with an accuracy better than 5% of the winding length. However, for high power, high voltage, multi-winding transformers, further refinements of the feature template matching PD location algorithm are required. Likewise the artificial neural network PD location algorithm, whilst showing promise, also needs further research to be applied to such transformer windings. In brief, the work presented indicates that the overall approach used to locate PDs in power transformers is sound. Methods, techniques and algorithms have been developed which with further refinement will allow substantial advances to be made in this field.
