BackgroundUnderstanding people’s knowledge, attitudes, and practices (KAP) regarding a new infectious disease is crucial to the prevention and control of it. Human infection with avian influenza A (H7N9) was first identified on March 31, 2013 in China. Out of the total number of 134 cases confirmed from March to September 2013 in China, Zhejiang Province saw the greatest number (46 cases). ObjectiveThis study employed a mobile Internet survey to assess KAP regarding H7N9 among mobile phone users in Zhejiang Province. This study intended to examine KAP by region and the association between sociodemographic variables and KAP. MethodsAn anonymous questionnaire was designed by Zhejiang Provincial Center for Disease Control and Prevention (CDC). A cross-sectional survey was executed through a mobile Internet application platform of China Unicom in 5 regions in Zhejiang Province. Stratified and clustered sampling methods were applied and mobile phone users were invited to participate in the study voluntarily. ResultsA total of 9582 eligible mobile phone users participated in the survey with a response rate of 1.92% (9582/5,000,000). A total of 9105 valid responses (95.02%) were included for statistical analysis. Generally, more than three-quarters of the participants had some basic knowledge of H7N9 and held the attitude recommended by the Zhejiang CDC toward eating cooked poultry (77.55%, 7061/9105) and visiting a hospital at the occurrence of symptoms (78.51%, 7148/9105). Approximately half of the participants worried about contracting H7N9, and took preventive practices recommended by the Zhejiang CDC. But only 14.29% (1301/9105) of participants kept eating cooked poultry as usual. Although worry about H7N9 infection did not differ by region, Hangzhou saw the largest proportion of participants with knowledge of H7N9, which was probably because Hangzhou had the greatest number of H7N9 cases. KAP varied by some sociodemographic variables. Female participants were more likely to know about symptoms of H7N9 (OR 1.32, 95% CI 1.08-1.61), to worry about contracting it (OR 1.15, 95% CI 1.04-1.27), and to report their lives being influenced by it (OR 1.27, 95% CI 1.15-1.41). They were also more likely to take the recommended precautions. Male participants and younger participants were less likely to comply with advocated protective practices. ConclusionsThe results suggest that health education should be customized depending on sociodemographic variables to achieve more effective behavioral outcomes.

The development of efficient therapeutic agents with low side effects for inflammatory bowel disease management is a longstanding challenge. Recently, hydrogen molecule (H2) is identified as an emerging spectrum‐wide, effective, and biosafe anti‐inflammatory agent, but intestine‐targeted H2 delivery is still challenging. Here, an intestine‐targeted controlled hydrogen‐releasing microcapsule (MgH2@EC@ES) is developed by confining abundant MgH2 microparticles in the hydrophobic network of ethyl cellulose (EC) before being encapsulated with Eudragit S100 (ES) by a multistep microemulsion method. The pH‐responsive swelling feature of ES enables MgH2@EC@ES microcapsules to escape from the stomach after oral administration and to hydrolytically produce a high amount of H2 in the intestinal tract in a sustained way. High‐dose oral administration of MgH2@EC@ES microcapsules exhibits a high outcome of colitis prevention, which is comparable to the first‐line drug 5‐aminosalicylic acid (5‐ASA) in the changes of body/spleen weights and disease activity and even better in the recovery of colon length and the improvement of histopathological change in the colon than 5‐ASA in a colitis mouse model. Mechanically, it is innovatively revealed that H2 released from MgH2@EC@ES microcapsules protects the complexes in the mitochondrial electron transfer chain from oxidative damage to enhance the energy metabolism of intestinal cells in support of mucosal restoration in colitis. Accurately recognizing apples in complex environments is essential for automating apple picking operations, particularly under challenging natural conditions such as cloudy, snowy, foggy, and rainy weather, as well as low-light situations. To overcome the challenges of reduced apple target detection accuracy due to branch occlusion, apple overlap, and variations between near and far field scales, we propose the Rep-ViG-Apple algorithm, an advanced version of the YOLO model. The Rep-ViG-Apple algorithm features a sophisticated architecture designed to enhance apple detection performance in difficult conditions. To improve feature extraction for occluded and overlapped apple targets, we developed the inverted residual multi-scale structural reparameterized feature extraction block (RepIRD Block) within the backbone network. We also integrated the sparse graph attention mechanism (SVGA) to capture global feature information, concentrate attention on apples, and reduce interference from complex environmental features. Moreover, we designed a feature extraction network with a CNN-GCN architecture, termed Rep-Vision-GCN. This network combines the local multi-scale feature extraction capabilities of a convolutional neural network (CNN) with the global modeling strengths of a graph convolutional network (GCN), enhancing the extraction of apple features. The RepConvsBlock module, embedded in the neck network, forms the Rep-FPN-PAN feature fusion network, which improves the recognition of apple targets across various scales, both near and far. Furthermore, we implemented a channel pruning algorithm based on LAMP scores to balance computational efficiency with model accuracy. Experimental results demonstrate that the Rep-ViG-Apple algorithm achieves precision, recall, and average accuracy of 92.5%, 85.0%, and 93.3%, respectively, marking improvements of 1.5%, 1.5%, and 2.0% over YOLOv8n. Additionally, the Rep-ViG-Apple model benefits from a 22% reduction in size, enhancing its efficiency and suitability for deployment in resource-constrained environments while maintaining high accuracy. The functional integrity of [CD8.sup.+] T cells is tightly coupled to metabolic reprogramming, but how oxidative stress directs [CD8.sup.+] T cell metabolic fitness in the tumor microenvironment (TME) remains elusive. Here, we report that SUMO-specific protease 7 (SENP7) senses oxidative stress to maintain the [CD8.sup.+] T cell metabolic state and antitumor functions. SENP7-deficient [CD8.sup.+] T cells exhibited decreased glycolysis and oxidative phosphorylation, resulting in attenuated proliferation in vitro and dampened antitumor functions in vivo. Mechanistically, [CD8.sup.+] T cell-derived ROS triggered cytosolic SENP7-mediated PTEN deSUMOylation, thereby promoting PTEN degradation and preventing PTEN-dependent metabolic defects. Importantly, lowering T cell-intrinsic ROS restricted SENP7 cytosolic translocation and repressed [CD8.sup.+] T cell metabolic and functional activity in human colorectal cancer samples. Proper metabolic activities facilitate T cell expansion and antitumor function; however, the mechanisms underlying disruption of the T cell metabolic program and function in the tumor microenvironment (TME) remain elusive. Here, we show a zinc finger protein 91-governed (ZFP91-governed) mechanism that disrupts the metabolic pathway and antitumor activity of tumor-infiltrating T cells. Single-cell RNA-Seq revealed that impairments in T cell proliferation and activation correlated with ZFP91 in tissue samples from patients with colorectal cancer. T cell-specific deletion of Zfp91 in mice led to enhanced T cell proliferation and potentiated T cell antitumor function. Loss of ZFP91 increased mammalian target of rapamycin complex 1 (mTORC1) activity to drive T cell glycolysis. Mechanistically, T cell antigen receptor-dependent (TCR-dependent) ZFP91 cytosolic translocation promoted protein phosphatase 2A (PP2A) complex assembly, thereby restricting mTORC1-mediated metabolic reprogramming. Background: Inflammatory bowel diseases (IBDs) are characterized by sustained inflammation and/or ulcers along the lower digestive tract, and have complications such as colorectal cancer and inflammation in other organs. The current treatments for IBDs, which affect 0.3% of the global population, mainly target immune cells and inflammatory cytokines with a success rate of less than 40%. Results: Here we show that berberine, a natural plant product, is more effective than the frontline drug sulfasalazine in treating DSS (dextran sulfate sodium)-induced colitis in mice, and that berberine not only suppresses macrophage and granulocyte activation but also promotes epithelial restitution by activating Lgr5+ intestinal stem cells (ISCs). Mechanistically, berberine increases the expression of Wnt genes in resident mesenchymal stromal cells, an ISC niche, and inhibiting Wnt secretion diminishes the therapeutic effects of berberine. We further show that berberine controls the expression of many circadian rhythm genes in stromal cells, which in turn regulate the expression of Wnt molecules. Conclusions: Our findings suggest that berberine acts on the resident stromal cells and ISCs to promote epithelial repair in experimental colitis and that Wnt-β-Catenin signaling may be a potential target for colitis treatment.
