ObjectiveTo assess the implementation of insurance-based compensation for adverse reactions following immunization in Wuxi City,Jiangsu Province.MethodsData on basic insurance and supplementary insurance for adverse reactions following immunization from 2020 to 2022 in Wuxi City was collected, and a descriptive epidemiological method was used to analyze the data.ResultsA total of 2 272 cases were compensated for adverse reactions following immunization,with a total compensation amount of 5.255 9 million yuan, and an average of 2.3 thousand yuan per case. Basic insurance accounted for 0.62% of the total cases and 10.72% of the total compensation amount, while supplementary insurance accounted for 99.43% of the total cases and 89.28% of the total compensation amount. Compensation amounts for disability and third-degree damage in basic insurance accounted for 63.18% and 88.71%, respectively, while fourth-degree and lower damage in supplementary insurance accounted for 96.52%. The compensation rate for supplementary insurance was higher than that for basic insurance. The highest number of compensated cases was in the 0‒ year age group, and vaccines included in the Expanded Program on Immunization (EPI) accounted for the majority of the cases and compensation amounts, with a percentage of 70.11% and 73.84%, respectively. For basic insurance, the largest number of compensated cases involved the Bacillus Calmette-Guérin (BCG) vaccine. For supplementary insurance, the top three compensated cases for EPI vaccines were the measles-mumps-rubella (MMR) vaccine, the hepatitis B (HepB) vaccine, and the acellular diphtheria-tetanus-pertussis (DTaP) vaccine. For non-EPI vaccines, the top three compensated cases were the 13 valent pneumococcal polysaccharide conjugate (PPCV13) vaccine, the acellular DPT-inactivated poliovirus-haemophilus influenzae type b combined vaccine (DPT-IPV/Hib), and enterovirus 71 (EV71) inactivated vaccine. In basic insurance, thrombocytopenic purpura and BCG local abscess both accounted for 21.43% of the cases, while in supplementary insurance, respiratory system diseases accounted for 77.47% of the cases. SUMMARY: Cotton fiber (Gossypium hirsutum) serves as an ideal model for investigating the molecular mechanisms of plant cell elongation at the single‐cell level. Brassinosteroids (BRs) play a crucial role in regulating plant growth and development. However, the mechanism by which BR influences cotton fiber elongation remains incompletely understood. In this study, we identified EXORDIUM‐like (GhEXL3) through transcriptome analysis of fibers from BR‐deficient cotton mutant pagoda 1 (pag1) and BRI1‐EMS‐SUPPRESSOR 1 (GhBES1.4, encoding a central transcription factor of BR signaling) overexpression cotton lines. Knockout of GhEXL3 using CRISPR/Cas9 was found to impede cotton fiber elongation, while its overexpression promoted fiber elongation, suggesting a positive regulatory function for GhEXL3 in fiber elongation. Furthermore, in vitro ovule culture experiments revealed that the overexpression of GhEXL3 partially counteracted the inhibitory effects of brassinazole (BRZ) on cotton fiber elongation, providing additional evidence of GhEXL3 involvement in BR signaling pathways. Moreover, our findings demonstrate that GhBES1.4 directly binds to the E‐box (CACGTG) motif in the GhEXL3 promoter region and enhances its transcription. RNA‐seq analysis revealed that overexpression of GhEXL3 upregulated the expression of EXPs, XTHs, and other genes associated with fiber cell elongation. Overall, our study contributes to understanding the mechanism by which BR regulates the elongation of cotton fibers through the direct modulation of GhEXL3 expression by GhBES1.4. Significance Statement: Understanding the molecular mechanisms of cotton fiber elongation is crucial for improving fiber yield. This study reveals that the EXORDIUM‐like gene (GhEXL3) positively regulates fiber elongation in response to brassinosteroids (BRs). By demonstrating that GhBES1.4, a key BR signaling transcription factor, directly activates GhEXL3, we provide insights into how BR signaling pathways influence fiber growth. In order to understand the enhancement mechanism of the interaction between functionalised carbon nanotubes and polymer matrix, and to investigate the effects of different functionalised carbon nanotubes on the properties of composites at the microscopic level, the models of pure polyurethane (PUR), carbon nanotubes/polyurethane (CNTs/PUR), hydroxyl-functionalised carbon nanotubes/polyurethane (CNTOH/PUR), and carboxyl-functionalised carbon nanotubes/polyurethane (CNTCOOH/PUR) systems were constructed, and molecular dynamics methods were used to simulate the effects of different functionalised carbon nanotubes on the mechanical properties of polyurethane composite systems. The effects of different functionalised carbon nanotubes on the properties of PUR composite systems were compared by analysing the properties of PUR and its composite systems in terms of free volume fraction (FFV), radial distribution function (RDF), binding energy, and density distribution of PUR around different functionalised carbon nanotubes. The effects of intermolecular interactions on the properties of the composite systems were elucidated at the microscopic level. The final simulation results show that the CNTCOOH/PUR composite system with added carboxylated CNTs has the highest tensile yield strength, compressive properties and modulus; the smallest free volume fraction; the strongest bonding energy between CNTCOOH and PUR matrix, the largest density distribution of PUR matrix around CNTCOOH, and the best interfacial interaction. Solar-induced chlorophyll fluorescence (SIF) is highly correlated with photosynthesis and can be used for estimating gross primary productivity (GPP) and monitoring vegetation stress. The far-red band of the solar Fraunhofer lines (FLs) is close to the strongest SIF emission peak and is unaffected by chlorophyll absorption, making it suitable for SIF intensity retrieval. In this study, we propose a retrieval window for far-red SIF, significantly enhancing the sensitivity of data-driven methods to SIF signals near 757 nm. This window introduces a weak O2 absorption band based on the FLs window, allowing for better separation of SIF signals from satellite spectra by altering the shape of specific singular vectors. Additionally, a frequency shift correction algorithm based on standard non-shifted reference spectra is proposed to discuss and eliminate the influence of the Doppler effect. SIF intensity retrieval was achieved using data from the GOSAT satellite, and the retrieved SIF was validated using GPP, enhanced vegetation index (EVI) from the MODIS platform, and published GOSAT SIF products. The validation results indicate that the SIF products provided in this study exhibit higher fitting goodness with GPP and EVI at high spatiotemporal resolutions, with improvements ranging from 55% to 129%. To determine the genes associated with the fiber strength trait in cotton, three different cotton cultivars were selected: Sea Island cotton (Xinhai 32, with hyper-long fibers labeled as HL), and upland cotton (17–24, with long fibers labeled as L, and 62–33, with short fibers labeled as S). These cultivars were chosen to assess fiber samples with varying qualities. RNA-seq technology was used to analyze the expression profiles of cotton fibers at the secondary cell wall (SCW) thickening stage (20, 25, and 30 days post-anthesis (DPA)). The results showed that a large number of differentially expressed genes (DEGs) were obtained from the three assessed cotton cultivars at different stages of SCW development. For instance, at 20 DPA, Sea Island cotton (HL) had 6,215 and 5,364 DEGs compared to upland cotton 17–24 (L) and 62–33 (S), respectively. Meanwhile, there were 1,236 DEGs between two upland cotton cultivars, 17–24 (L) and 62–33 (S). Gene Ontology (GO) term enrichment identified 42 functions, including 20 biological processes, 11 cellular components, and 11 molecular functions. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis identified several pathways involved in SCW synthesis and thickening, such as glycolysis/gluconeogenesis, galactose metabolism, propanoate metabolism, biosynthesis of unsaturated fatty acids pathway, valine, leucine and isoleucine degradation, fatty acid elongation pathways, and plant hormone signal transduction. Through the identification of shared DEGs, 46 DEGs were found to exhibit considerable expressional differences at different fiber stages from the three cotton cultivars. These shared DEGs have functions including REDOX enzymes, binding proteins, hydrolases (such as GDSL thioesterase), transferases, metalloproteins (cytochromatin-like genes), kinases, carbohydrates, and transcription factors (MYB and WRKY). Therefore, RT-qPCR was performed to verify the expression levels of nine of the 46 identified DEGs, an approach which demonstrated the reliability of RNA-seq data. Background: The grading of adult isocitrate dehydrogenase (IDH)-mutant astrocytomas is a crucial prognostic factor. Purpose: To investigate the value of conventional magnetic resonance imaging (MRI) features and apparent diffusion coefficient (ADC) in the grading of adult IDH-mutant astrocytomas, and to analyze the correlation between ADC and the Ki-67 proliferation index. Material and Methods: The clinical and MRI data of 82 patients with adult IDH-mutant astrocytoma who underwent surgical resection and molecular genetic testing with IDH and 1p/19q were retrospectively analyzed. The conventional MRI features, ADCmin, ADCmean, and nADC of the tumors were compared using the Kruskal–Wallis single factor ANOVA and chi-square tests. Receiver operating characteristic (ROC) curves were drawn to evaluate conventional MRI and ADC accuracy in differentiating tumor grades. Pearson correlation analysis was performed to determine the correlation between ADC and the Ki-67 proliferation index. Results: The difference in enhancement, ADCmin, ADCmean, and nADC among WHO grade 2, 3, and 4 tumors was statistically significant (all P <0.05). ADCmin showed the preferable diagnostic accuracy for grading WHO grade 2 and 3 tumors (AUC=0.724, sensitivity=63.4%, specificity=80%, positive predictive value (PPV)=62.0%; negative predictive value (NPV)=82.5%), and distinguishing grade 3 from grade 4 tumors (AUC=0.764, sensitivity=70%, specificity=76.2%, PPV=75.0%, NPV=71.4%). Enhancement + ADC model showed an optimal predictive accuracy (grade 2 vs. 3: AUC = 0.759; grade 3 vs. 4: AUC = 0.799). The Ki-67 proliferation index was negatively correlated with ADCmin, ADCmean, and nADC (all P <0.05), and positively correlated with tumor grade. In cancer genomics, variant calling has advanced, but traditional mean accuracy evaluations are inadequate for biomarkers like tumor mutation burden, which vary significantly across samples, affecting immunotherapy patient selection and threshold settings. In this study, we introduce TMBstable, an innovative method that dynamically selects optimal variant calling strategies for specific genomic regions using a meta-learning framework, distinguishing it from traditional callers with uniform sample-wide strategies. The process begins with segmenting the sample into windows and extracting meta-features for clustering, followed by using a pre-trained meta-model to select suitable algorithms for each cluster, thereby addressing strategy-sample mismatches, reducing performance fluctuations and ensuring consistent performance across various samples. We evaluated TMBstable using both simulated and real non-small cell lung cancer and nasopharyngeal carcinoma samples, comparing it with advanced callers. The assessment, focusing on stability measures, such as the variance and coefficient of variation in false positive rate, false negative rate, precision and recall, involved 300 simulated and 106 real tumor samples. Benchmark results showed TMBstable's superior stability with the lowest variance and coefficient of variation across performance metrics, highlighting its effectiveness in analyzing the counting-based biomarker.
