Abstract

Objective: Side population (SP) cells may play a crucial role in tumorigenesis and the recurrence of cancer. Many kinds of cell lines and tissues have demonstrated the presence of SP cells, including several gastric cancer cell lines. This study is aimed to identify the cancer stem-like cells in the SP of gastric cancer cell line MKN-45. Methods: We used fluorescence activated cell sorting (FACS) to sort SP cells in the human gastric carcinoma cell line MKN-45 (cells labeled with Hoechst 33342) and then characterized the cancer stem-like properties of SP cells. Results: This study found that the SP cells had higher clone formation efficiency than major population (MP) cells. Five stemness-related gene expression profiles, including OCT-4, SOX-2, NANOG, CD44, and adenosine triphosphate (ATP)-binding cassette transporters gene ABCG2, were tested in SP and MP cells using quantitative real-time reverse transcription polymerase chain reaction (RT-PCR). Western blot was used to show the difference of protein expression between SP and MP cells. Both results show that there was significantly higher protein expression in SP cells than in MP cells. When inoculated into non-obese diabetic/severe combined immunodeficiency (NOD/SCID) mice, SP cells show higher tumorigenesis tendency than MP cells. Conclusions: These results indicate that SP cells possess cancer stem cell properties and prove that SP cells from MKN-45 are gastric cancer stem-like cells.

Abstract

In order to convert RF energy in the environment into DC power for low-power equipment, a dual band energy harvester is designed, which consists of a wideband coplanar waveguide antenna and a new matching network. The broadband antenna's bandwidth is 0.8-2.8 GHz, which can collect radio frequency energy in the environment at 0.915 GHz and 2.4 GHz. Compared with microstrip antennas, the size of the broadband antenna is reduced by nearly 1/3. The new matching network can be matched at both the 0.915 GHz and 2.4 GHz and the matching bandwidth reaches 30 MHz and 100 MHz respectively. The experimental results show that when the system is about 50 cm apart from the reader of 0.1 W, it can effectively collect 201 mV DC power. When the system is about 10 cm apart from the router less than 0.1 W, it can effectively collect 33 mV DC power. This shows that the dual-band rectifier system designed can effectively collect energy in the environment at the ISM frequency bands (0.915 GHz and 2.4 GHz). [ABSTRACT FROM AUTHOR]

Abstract

Thermoregulatory behaviours are of great importance for ectotherms buffering against the impact of temperature extremes. Such behaviours bring not only benefits but also organism level costs such as decreased food availability and foraging efficiency and thus lead to energetic costs and metabolic consequences. However, there remains an important gap in our knowledge to link thermoregulatory behaviour to population level ecological consequences. Aphids, as ectothermic invertebrates, can escape from thermal extremes by either dropping off the plant they are on or moving to suitable microclimates. Here we used the English grain aphid, Sitobion avenae , as a model system to test the hypothesis that aphids may behaviourally avoid heat stress while also altering their microhabitat utilization leading to demographic consequences. We found that heat stress drove the aphids to leave their host plant; this reduction in host plant residence was associated with increasing leaf temperatures indicating that aphids exhibit thermoregulatory behaviour to escape heat stress. Specifically, we found that behavioural thermoregulation made the aphids disperse and redistribute themselves within different microclimates and thus led to changes in microhabitat utilization. We also discovered that leaving the host plant during behavioural thermoregulation resulted in a considerable decrease in aphids' survival probability due to their inefficient relocation to other plants and increased risk of starvation. Finally, we found that the aphids' thermoregulatory behaviour prevented heat stress while concurrently resulting in decreased survival and reproduction. Together, these findings support our hypothesis that behavioural thermoregulation alters microhabitat utilization and demographic rates in aphids. This study highlights the importance of behavioural thermoregulation and its ecological consequences and has important implications for understanding population responses in the context of current climate change. Abstract

BackgroundThe locus for familial cortical myoclonic tremor with epilepsy (FCMTE) has long been mapped to 8q24 in linkage studies, but the causative mutations remain unclear. Recently, expansions of intronic TTTCA and TTTTA repeat motifs within SAMD12were found to be involved in the pathogenesis of FCMTE in Japanese pedigrees. We aim to identify the causative mutations of FCMTE in Chinese pedigrees.MethodsWe performed genetic linkage analysis by microsatellite markers in a five-generation Chinese pedigree with 55 members. We also used array-comparative genomic hybridisation (CGH) and next-generation sequencing (NGS) technologies (whole-exome sequencing, capture region deep sequencing and whole-genome sequencing) to identify the causative mutations in the disease locus. Recently, we used low-coverage (~10×) long-read genome sequencing (LRS) on the PacBio Sequel and Oxford Nanopore platforms to identify the causative mutations, and used repeat-primed PCR for validation of the repeat expansions.ResultsLinkage analysis mapped the disease locus to 8q23.3–24.23. Array-CGH and NGS failed to identify causative mutations in this locus. LRS identified the intronic TTTCA and TTTTA repeat expansions in SAMD12as the causative mutations, thus corroborating the recently published results in Japanese pedigrees.ConclusionsWe identified the pentanucleotide repeat expansion in SAMD12as the causative mutation in Chinese FCMTE pedigrees. Our study also suggested that LRS is an effective tool for molecular diagnosis of genetic disorders, especially for neurological diseases that cannot be positively diagnosed by conventional clinical microarray and NGS technologies.

Abstract

This paper presents the necessity of reading recognition in calibration process of sound level meter, and analysis the problems of existing display reading algorithm, then presents a novel reading recognition system for sound level meter. The system find the position of number string by image difference and connective region analysis, and pre-process the image by local thresholding based on neighborhood averaging method, then recognizes the number by pattern grid and template matching. The system has the characteristics of rapid and accurate. The experiment shows the system is well competent for calibration work of sound level meter.

Abstract

In OFDM-based UWB baseband, synchronizer plays a key role to the performance of the whole system. Therefore achieving an area and power efficient architecture at least SNR loss becomes the main challenge of synchronizer design. To meet 528 Msamples/s throughput, the parallel architecture is used in current approaches. But it leads to higher area and power consumption. In this paper, we propose an efficient synchronizer for OFDM-based UWB system, consisting of the filter-free symbol synchronizer, frequency synchronizer with improved Moose algorithm and sign-based auto-correlator. According to the results of synthesis using SMIC, 0.13 mum CMOS process, the proposed design can achieve the throughput requirement with only 24% gate count and 25% power consumption (in idle state) of the conventional four-parallelism approach.

Abstract

An optimum design of decision feedback equalizer (DFE) used in Ethernet is presented. This paper proposes two improving measures for physical implementation - the hybrid form and the coefficient updating unit sharing. According to the results of synthesis using SMIC, 0.18/spl mu/m CMOS process, the speed, area and power consumption of the improved DFE is optimized by 16%, 36% and 39% compared with the transposed form implementation.
