13. ASS1 gene mutation in a neonate with citrullinemia type Ⅰ

Abstract

Objective To detect blood acylcarnitine and genes in a newborn with suspected citrullinemia, and to detect gene mutations in their parents, so as to clarify their diagnosis and follow-up investigations. Methods Genetic metabolic diseases were screened in the newborn baby using tandem mass spectrometry. Peripheral blood samples were collected from the newborn patient with suspected citrullinemia to detect the products of genetic metabolic diseases. Peripheral blood samples were collected from newborns and their parents for high-throughput sequencing and Sanger sequencing. The blood samples of the newborns and their parents were sequenced and verified, and then the genetic abnormalities were analyzed. Results Tandem mass spectrometry showed that the level of citrulline increased(1 043 μmol/L), the level of citrulline increased to 2 072 μmol/L after recall immediately. Double heterozygous mutations of ASS1 gene in the proband were identified:c.256C＞T and c.577G＞A,which were confirmed in the parents.The case was diagnosed with citrullinemia type Ⅰ. Conclusions Neonatal tandem mass spectrometry, in combination with a pedigree analysis of the gene sequencing, may support the diagnosis of citrullinemia.

21. Research progress of seed dormancy and germination regulation.

Abstract

Dormancy enables plant seeds to time germination until environmental conditions become favorable for seedling survival and growth. The dormancy characteristics of seeds are of important ecological adaptive significance and notable agricultural value. Phytohormone abscisic acid (ABA) and gibberellin (GA) are the key factors for seed dormancy and germination. Mature seeds in dormancy state contain high levels of ABA and low levels of GA. ABA induces and maintains seed dormancy, while GA antagonizes ABA and promotes seed germination. DELAY OF GERMINATION-1 (DOG1) is a major regulator of seed dormancy and had a synergistic effect with ABA to delay germination. DOG1 enhances ABA signal transduction by combining with PP2C ABA hypersensitive germination (AHG1/AHG3), and inhibits the action of AHG1 to increase ABA sensitivity and impose seed dormancy. Imprinted genes are regulated by epigenetic mechanisms before and after fertilization, and are closely related to the establishment and release of seed dormancy. In recent years, remarkable progress has been made in the regulation of seed dormancy. In the present paper, we reviewed the effects of phytohormones ABA and GA on seed dormancy and germination, the action mechanism regulating seed dormancy by DOG1, and the epigenetic regulation of seed dormancy and germination. In addition, we also propose some scientific issues that need to be further investigated in this field to provide some information for understanding the molecular mechanism of seed dormancy and germination, breeding in anti-preharvest sprouting in crop plants, and promoting the germination of dormant seeds. 26. Application of Specific Surface Area Nitrogen Adsorption Method to Characterize the Alkaline Dissolution of Montmorillonite

Abstract

Montmorillonite is characterized by small particle size, large specific surface area and strong water sensitivity, which results in blocking the pore and throat of the reservoir and in turn affecting its quality. Dissolution characteristics of montmorillonite under alkaline conditions are common concerns in the development of oil fields. Mass contrast, X-ray Diffraction, solution concentration analysis and value analysis of pH methods are commonly used to characterize dissolution of montmorillonite. However, the quality comparison method is complex. Calculation of diffraction pattern by the X-ray Diffraction involves large manual operation errors. The ion concentration analysis method shows a large detection error for infusible elements. Value analysis of the pH method is influenced by temperature changes. The specific surface area, pore volume, pore diameter change, adsorption-desorption curve measured by nitrogen adsorption method was used to characterize the dissolution of montmorillonite under alkaline conditions and is reported in this paper. The experimental conditions were optimized, including the concentration of NaOH solution and filling time (reaction time). The best concentration of NaOH solution was 0.05 mol/L and the best reaction time was 3 h. Results show that the dissolved results of montmorillonite by the nitrogen adsorption method characterized by specific surface area, pore volume, pore diameter and adsorption desorption curve are consistent with the results by the change of the pH value and the concentration of the solution (such as Si4+). Therefore, the method is feasible and can be used to characterize the dissolution of minerals with multiple pores and large specific surface area.

Abstract

Proton exchange membrane fuel cells (PEMFC) are a type of the increasingly developed sustainable energy systems which have been turned into one of the most popular of them in the last decade. The performance of these types of fuel cells is the best among the others; however, they can be better by an authentic control to maintain in the maximum efficiency in the operative points during the current ripples. To do this and also to improve the value of the output voltage, the two-phase interleaved boost converter is utilized. Since boost converter has uncertain parameters and disturbances in their essence, using the ordinary methods for the control of them can ignore some of the principal parameters. In this study, a new technique is utilized for optimal control of the proton exchange membrane fuel cells with interval uncertain parameters. Here, we consider the uncertainties in the converter inherent. Because of considering the uncertainties in the system, controllability of the system is first testified based on interval arithmetic. Then, an extended version of the linear quadratic regulator strategy using interval analysis is employed for achieving a reliable and optimal solution. Chebyshev inclusion method is utilized for solving the Pontryagins problem of the LQR problem. Eventually, by solving the interval version of the Riccati equations, the robust range for optimal control of the PEMFC is obtained. The results of the proposed system are finally compared with Monte Carlo method for showing the efficiency of the presented technique.
