Objective To investigate the correlation of Wnt5a expression and vasculogenic mimicry (VM) in prostate cancer tissues, and analyze their relationships with cancer stem cells (CSCs) characteristics and epithelial–mesenchymal transition (EMT). Methods Immunohistochemistry was conducted to detect the expression of Wnt5a in 50 prostate cancer tissues and 50 benign prostatic hyperplasia tissues. The expression levels of CD133, vimentin, and E-cadherin were detected in the prostate cancer tissues, and CD34/PAS double staining was used to detect VM structures. We analyzed the difference in Wnt5a level between prostate cancer and benign prostatic hyperplasia tissues, the clinical significance of Wnt5a and VM, the relationship of Wnt5a expression and VM, and the relationships of Wnt5a expression and VM with CD133, Vimentin, E-cadherin. Results The expression of Wnt5a was significantly higher in prostate cancer tissues than in benign prostatic hyperplasia (P < 0.05). A positive correlation was observed between Wnt5a expression and VM (P < 0.05). The expression levels of Wnt5a and VM were positively correlated with those of CD133 and vimentin (P < 0.05). Wnt5a expression and VM were positively correlated with Gleason score, vas deferens invasion and lymphatic metastasis (P < 0.05) of prostate cancer, and VM was also positively correlated with T stage of prostate cancer (P < 0.05). Conclusion The expression level of Wnt5a in prostate cancer tissues is elevated and positively related with VM formation. Wnt5a expression and VM are correlated with cancer stem cells characteristics and the expression of epithelial–mesenchymal transition marker proteins.

Prostate cancer (PCa) represents a substantial global health concern and a prominent contributor to male cancer-related mortality. The aim of this study is to explore the role of B-type endothelin receptor (EDNRB) in PCa and evaluate its therapeutic potential. The investigation employed predictive methodologies encompassing data acquisition from the GEO and TCGA databases, gene screening, enrichment analysis, in vitro experiments involving PCR, Western blotting, wound healing, and Transwell assays, as well as animal experiments. Analysis revealed a significant downregulation of EDNRB expression in PCa cells. Overexpression of EDNRB demonstrated inhibitory effects on tumor cell growth, migration, and invasion, likely mediated through activation of the cGMP-Protein Kinase G pathway. In vivo experiments further confirmed the tumor-suppressive properties of EDNRB overexpression. These findings underscore the prospect of EDNRB as a therapeutic target for PCa, offering novel avenues for PCa treatment strategies.

Objective To explore the inhibition effect and mechanism of miR-146a-5p on proliferation and invasion of prostate cancer (PCa) cell line PC-3 by targeting SMAD4. Methods RT-qPCR was used to detect the expression of miR-146a-5p in PCa tissues and cell lines. The relevance of miR-146a-5p expression with Gleason score was also analyzed. MTT, BrdU experiment, cell colony formation experiment, scratch experiment, Transwell assay and nude mouse xenograft model experiment were conducted to detect the effect of miR-146a-5p on cell proliferation, tumorigenicity, migration and invasion. The expression of SMAD4 in PCa tissues was detected by RT-qPCR, and the targeting relationship of SMAD4 and miR-146a-5p was confirmed by double luciferase reporter gene assay and rescue experiment. Western blot was used to detect the expression of SMAD2/SMAD3 complex in nucleus affected by miR-146a-5p and SMAD4. Finally, double luciferase reporter gene assay and ChIP experiment were performed to examine the targeting regulation of TIM3 by miR-146a-5p/SMAD4/SMAD2/SMAD3 signaling axis. Results miR-146a-5p was low expressed in PCa tissues and cell lines; its expression was negatively correlated to Gleason score and had the lowest expression in PC-3 cells. miR-146a-5p inhibited the proliferation and invasion of PC-3 cells by targeting SMAD4. SMAD2/SMAD3/TIM3 axis seemed to be the downstream mechanism of miR-146a-5p/SMAD4 signaling pathway. Conclusions miR-146a-5p can inhibit the proliferation and invasion of PC-3 cells by targeting SMAD4, and the downstream mechanism might be related to the SMAD2/SMAD3/TIM3 signaling pathway.

Ocean resource exploration expands into deep and ultra-deep waters, which has posed great challenges to the 6-DOF parallel platform that requires to finish the long-span and high-velocity wave compensation task with high precision and anti-interference ability. The control strategy employed in the asymmetric hydraulic system of large aspect ratio requires more careful considerations when operating in the harsh and severe environment. An adaptive feedback linearization control strategy was proposed by employing the radial basis function neural network (RBFNN) for identification. First, a nonlinear model of the asymmetric hydraulic system was established. Then, an adaptive controller was designed based on RBFNN and feedback linearization. Finally, simulations were performed by using MATLAB/Simulink under the five-stage wave environment at a 90° wave encounter angle and under the external interference condition. The result shows that this method has a good traceability and robustness compared to classic PID and sliding mode control methods, which is more suitable in control of the wave compensation platform in complex sea conditions. The new controller can significantly increase the compensation accuracy and anti-interference ability, and provide a workbench for the 6-DOF parallel platform operation in deep waters.

This article introduces three mixed integer programming (MIP) models to address a network design problem for mixed plastic waste (MPW) supply chains. By tracking waste compositions throughout the supply chain, the models optimize the technologies needed to process MPW. The three models adopt different approaches to preserve composition information in the supply chain. We also remark on how to improve solution times with additional constraints, and how the models can be easily modified to handle larger‐scale problems. The proposed models provide an approach for examining emerging MPW recycling technologies that may be more sensitive to input composition, as well as determining the extent to which advanced sorting is useful. Existing conventional materials are usually difficult to achieve multiple tunable stress plateau characteristics, which hinders their multifunctional applications. This study proposes a multiple stable stress plateau metamaterial. A theoretical model for predicting stress plateaus was established, and its accuracy was verified by experimental and numerical methods. The proposed material exhibits a three-stage deformation mode owing to sequential snap-through and Euler buckling at the microscopic unit cell level under compression. The effect of geometric parameters on the stress plateau was studied. The results indicated that the first plateau stress can be tuned independently by altering the angle between the inclined thin and thick beams, whereas the third plateau stress can be tuned independently by changing the thickness of the vertical thick beams. The first and second plateau stresses can be tuned by varying the thickness ratio of the inclined and vertical thin beams. Moreover, the total number of stress plateaus and the range of each stress plateau could also be tuned by changing the thickness ratio of the inclined and vertical thin beams and the vertical thick beams. Using the energy efficiency method, we obtained the experimental and numerical plateau stresses for each specimen. Notably, the experimental, numerical, and theoretical results were in good agreement. The main reducer is the main component of the whole vehicle, and its main function is to realize deceleration and torque increase. For Baha racing car, in order to improve the dynamic performance of the whole car, the main reducer is designed from the aspects of layout, transmission ratio distribution, shift mode, overall size and shell structure. Calculate the transmission ratio range of reducer according to the performance parameters of transmission parts, and verify the rationality of transmission ratio; Then determine the parameters of gear according to the transmission ratio and related parameters, and finally design the parameters of gear according to the transmission ratio and related parameters, and finally design the parameters of other parts of reducer. Based on the determined parameters, 3D modeling software UG is used to build 3D models of various parts of the reducer, and finite element analysis software ANSYS is used to simulate and analyze the parts to check whether the comprehensive mechanical properties meet the requirements. In this paper, the design of the main reducer realizes the comprehensive design of small size, light weight, reasonable transmission ratio distribution, high reliability, shifting gears during driving, and the comprehensive mechanical properties also meet the requirements.

The study discusses the characteristics of Jurassic reservoir in Shenquan Oilfield in Xinjiang. The reservoir of Qiketai Sanjianfang formation mainly develops delta front facies and shore shallow lake facies sand bodies.The lithological characteristics of the reservoir are mainly fine sandstone and siltstone, and the physical properties of the target layer are mainly medium porosity and medium permeability. The Jurassic reservoir space in Shenquan Oilfield is sandstone pore type, mainly composed of secondary pores, mainly including intergranular dissolved pores, intragranular dissolved pores and intercrystalline pores, with good connectivity between pores. According to the analysis data of well logging, core analysis, production performance and DST in Shenquan Oilfield, porosity, permeability and water saturation models are established respectively:The porosity interpretation model is established by using acoustic wave and shale content; the permeability parameter interpretation model is realized by applying graph based cluster analysis method and neural network algorithm. The oil saturation interpretation model is obtained by Archie formula. According to the logging interpretation model and interpretation results, it can meet the needs of reservoir evaluation, oil-water distribution and original geological reserve parameters. After analyzing the logging parameter model and reservoir parameter calculation method, it is considered that the Jurassic in the Shenquan Oilfield in Xinjiang has better physical properties.
