Objective To screen for the key genes involved in propofol-induced neurotoxicity based on metabolomics and transcriptomics. Methods HT22 cells were cultured with propofol at concentrations of 0, 25, 50, 75, and 100 mg/L for 24 h and were established as Ctrl, P25, P50, P75, and P100 groups, respectively, and CCK-9 assay was used to measure cell viability in each group. HT22 cells were collected after 24 h of treatment to extract RNA and metabolites for metabolite detection and transcriptome analysis. The extracted HT22 cell metabolites were analyzed by liquid chromatography-mass spectrometry (LC-MS), differentially expressed metabolites, and trend analysis to obtain the key metabolites. The RNA of HT22 cells was analyzed by sequence alignment and differentially expressed genes to obtain the differentially expressed genes. Weighted gene co-expression network analysis (WGCNA) and protein-protein interaction (PPI) analysis were performed for the above key metabolites and differentially expressed genes to obtain the key genes involved in propofol-induced neurotoxicity. Background: Depression and anxiety are among the most prevalent psychiatric disorders worldwide, affecting individuals of all ages. The co‐occurrence of these disorders often exacerbates their negative health impacts, underscoring the necessity of understanding their comorbid mechanisms. Methods: This study employed cross‐lagged panel networks (CLPNs) to explore the longitudinal associations between depression and anxiety symptoms across three age groups and to compare the respective symptom networks. CLPNs were constructed through cross‐temporal associations between different symptoms, reflecting both the pattern of interaction and the significance of specific symptoms in comorbidity. The sample consisted of 1258 adolescents (aged 13–19 years, M = 15.98), 1118 college students (aged 17–24 years, M = 19.94), and 548 older adults (aged 60–101 years, M = 85.19) from China. Depression and anxiety symptoms were assessed using the subscales of the Depression, Anxiety, and Stress Scale Short Version (DASS‐21) at two time points over a 6‐month period during 2020–2021. Results: The findings revealed that the prevalence of depression and anxiety in adolescents, college students, and older adults was 25.9%/46.6%, 53.7%/61.5%, and 7.2%/22.5%, respectively. The network structure varied across age groups: adolescents and college students exhibiting a tight interconnection between depression and anxiety symptoms, while older adults showed stronger small‐world network characteristics. A key finding across all age groups was the central role of irrational fear. In addition, somatic anxiety symptoms frequently emerged as outcomes of other psychological symptoms. Conclusion: Depression and anxiety are more pronounced in college students compared to adolescents and older adults. Comparisons of the overall network structure provide insights into the lifelong trajectories of depression and anxiety symptom networks. The centrality of irrational fears and somatization symptoms is emphasized. These results offer guidance for more targeted clinical interventions. [ABSTRACT FROM AUTHOR]

Background: To evaluate the effect of perioperative esketamine administration on postpartum depression in pregnant women undergoing cesarean section., Methods: Data sources was PubMed, Embase, Web of Science, and Cochrane Library from inception to February 1, 2024. Randomized controlled trials in pregnant women undergoing cesarean section were selected and compared to the use of esketamine in the perioperative period. The primary outcome measure was the incidence of postpartum maternal depression. Preferred reporting items for systematic reviews and meta-analyses were used. Data pooled by random-effects models are presented as risk ratios (RR) (95% confidence intervals, 95% CI) or mean differences (95% CI). This review was registered in PROSPERO (ID: CRD42023431197)., Results: We included 8 studies with a total of 1655 participants. The quality of the studies was rated high or unclear. Seven studies involving 1485 participants reported the incidence of postpartum depression. Compared with pregnant women undergoing cesarean section without the use of esketamine, those using esketamine in the perioperative period showed a 48% decreased risk of developing postpartum depression (RR: 0.52, 95% CI: 0.35-0.79) and a 1.43-point reduction in EPDS (Edinburgh Postnatal Depression Scale) (mean difference: -1.43, 95% CI: -2.32 to -0.54). For immediate intraoperative adverse reactions, the application of esketamine caused maternal nausea and vomiting (RR: 2.16, 95% CI: 1.22-3.81), dizziness (RR: 6.11, 95% CI: 1.49-24.98), and hallucinations (RR: 6.83, 95% CI: 1.57-29.68) compared to no esketamine use., Conclusions: Perioperative use of esketamine in pregnant women undergoing cesarean section may reduce postpartum depression and increase intraoperative adverse reactions, but has no significant effect on postoperative adverse reactions., Competing Interests: The authors have no conflicts of interest to disclose., (Copyright © 2024 the Author(s). Objectives: Due to the population aging, there is an urgent need to focus on how to help the oldest old (people age 80 years or older) to reach successful aging and improve their life quality. In this study, a longitudinal design spanning 1 year was used to explore the relationship between trait mindfulness and the physical and mental health of the oldest old and the mediating role of attitude toward one's own aging (ATOA)., Method: A total of 437 older adults from Jiangsu, China, participated in this study and completed questionnaires about trait mindfulness and ATOA at baseline, and their death, disability, negative affect (NA), and health-related quality of life (HRQoL) were collected after 1 year., Results: The results indicated that trait mindfulness in the oldest old was negatively associated with their mortality, disability rate, and NA and positively associated with individual HRQoL. The results of structural equation modeling analyses indicated that ATOA mediated the association of trait mindfulness with mortality, disability rate, anxiety, and three HRQoL subdimensions: role limitations because of emotional problems, vitality, and mental health., Conclusions: This study confirmed the positive longitudinal relationships between trait mindfulness and ATOA on physical health (especially mortality and disability rates), mental health, and emotional aspects of HRQoL in the oldest old population. The findings highlight the potential significance of promoting trait mindfulness and ATOA as interventions to enhance successful aging among the oldest old, which is of particular relevance in the context of the global trend of population aging. Objective: Previous studies have found that college students with left-behind experience presented high levels of aggression, and childhood trauma may be one of the contributors. This study aimed to examine the relationship between childhood trauma and aggression in Chinese college students, and to explore the mediating role of self-compassion and the moderating role of left-behind experience., Method: 629 Chinese college students completed the questionnaires at two time points: childhood trauma and self-compassion were assessed at baseline, and aggression was assessed at baseline and 3-month follow-up., Results: Among these participants, 391 (62.2%) had left-behind experience. Emotional neglect of college students with left-behind experience in childhood was significantly higher than that of college students without such experience. Childhood trauma predicted aggression after 3 months among college students. Self-compassion mediated the predictive effect of childhood trauma on aggression after controlling for gender, age, only-child status, and family residential status. However, no moderating effect of left-behind experience was found., Conclusions: These findings indicated that childhood trauma is an important predictor of aggression among Chinese college students regardless of their left-behind experience. The reason for the higher aggression of left-behind college students may be that their left-behind situation increased the possibility of childhood trauma. In addition, whether in college students with left-behind experience or without such experience, childhood trauma may increase aggression by reducing the level of self-compassion. Furthermore, interventions incorporating components to improve self-compassion could be effective in decreasing aggression of college students who perceived high childhood trauma. Parkinson's disease (PD) is a progressive and degenerative disorder of the central nervous system, characterized by the loss of dopaminergic neurons and muscular rigidity. Treatment with propofol (2,6‑diisopropylphenol) has been observed to attenuate oxidative stress injury via inhibition of programmed cell death. Results from the present study indicate that propofol treatment attenuates 1‑methyl‑4‑phenylpyridinium (MPP+)‑induced oxidative stress, which was demonstrated by increased levels of reactive oxygen species, 4‑hydroxy‑2‑nonenal and protein carbonyls. Furthermore, it was demonstrated that propofol may ameliorate MPP+‑induced mitochondrial dysfunction by increasing the level of ATP and the mitochondrial membrane potential. MTT and lactate dehydrogenase assays indicated that propofol treatment reduces cell vulnerability to MPP+‑induced insult. Propofol was also observed to prevent apoptotic signals by reducing the ratio of Bcl‑2‑associated X protein to B‑cell lymphoma 2, reducing the expression level of cleaved caspase‑3 and attenuating cytochrome c release. Thus, propofol may present as a novel therapeutic strategy for the treatment of PD.
