Abstract

The total synthesis of a novel siderophore, acinetoferrin, is described. The key transformation involves the tandem oxidation and acylation of the [N.sub.3]-amino group of [N.sub.1]-BOC propane diamine prior to coupling with the external carboxyls of citric acid. A 'one-pot-two-step' reaction converted a primary amine (RN[H.sub.2]) into a O-benzoyl hydroxamate (i.e., RN(OOCPh)COR[prime]) in good yield (68%). These studies demonstrated the utility of the O-benzoyl protecting group in the synthesis of [Alpha],[Beta]-unsaturated hydroxamic acids.

Abstract

A study that illustrates how O-benzoyl protected hydroxamates can be synthesized under biphasic conditions and using primary amines as material is presented. In this process, a mixture of an aqueous carbonate buffer and CH2Cl2resulted to a rise in the N-oxidized product at the expense of benzamide. The yields of the desired product and the byproduct was quantified through reverse-phase high performance liquid chromatography.

Abstract

A typical experiment in a high school or college organic chemistry laboratory is synthesis of soap. Based on the thinking that different fats likely would give different soaps, an experiment for students entering chemistry labs for the first time and needing to develop ability to observe changes using their senses was developed. Students were to read the experimental procedure and bring in a unique triglyceride source to saponify for a designer or exotic soap. Vietnamese garlic oil to grill sludge from a restaurant showed up for this lab, a favorite for many students.

Abstract

Prefrontal cortical (PFC) circuits provide top-down control of threat reactivity. This includes ventromedial PFC (vmPFC) circuitry, which plays a role in suppressing fear-related behavioral states. Dynorphin (Dyn) has been implicated in mediating negative affect and mal-adaptive behaviors induced by severe threats and is expressed in limbic circuits, including the vmPFC. However, there is a critical knowledge gap in our understanding of how vmPFC Dyn-expressing neurons and Dyn transmission detect threats and regulate expression of defensive behaviors. Here, we demonstrate that Dyn cells are broadly activated by threats and release Dyn locally in the vmPFC to limit passive defensive behaviors. We further demonstrate that vmPFC Dyn-mediated signaling promotes a switch of vmPFC networks to a fear-related state. In conclusion, we reveal a previously unknown role of vmPFC Dyn neurons and Dyn neuropeptidergic transmission in suppressing defensive behaviors in response to threats via state-driven changes in vmPFC networks.
