Abstract

Tanoak (Notholithocarpus densiflorus) is an evergreen tree in the Fagaceae family found in California and southern Oregon. Historically, tanoak acorns were an important food source for Native American tribes, and the bark was used extensively in the leather tanning process. Long considered a disjunct relictual element of the Asian stone oaks (Lithocarpus spp.), phylogenetic analysis has determined that the tanoak is an example of convergent evolution. Tanoaks are deeply divergent from oaks (Quercus) of the Pacific Northwest and comprise a new genus with a single species. These trees are highly susceptible to sudden oak death (SOD), a plant pathogen (Phytophthora ramorum) that has caused widespread deaths of tanoaks. In this study, we set out to assemble the genome and perform comparative studies among a number of individuals that demonstrated varying levels of susceptibility to SOD. First, we sequenced and de novo assembled a draft reference genome of N. densiflorus using cobarcoded library processing methods and an MGI DNBSEQ-G400 sequencer. To increase the contiguity of the final assembly, we also sequenced Oxford Nanopore long reads to 30× coverage. To our knowledge, the draft genome reported here is one of the more contiguous and complete genomes of a tree species published to date, with a contig N50 of ∼1.2 Mb, a scaffold N50 of ∼2.1 Mb, and a complete gene score of 95.5% through BUSCO analysis. In addition, we sequenced 11 genetically distinct individuals and mapped these onto the draft reference genome, enabling the discovery of almost 25 million single nucleotide polymorphisms and ∼4.4 million small insertions and deletions. Finally, using cobarcoded data, we were able to generate a complete haplotype coverage of all 11 genomes.

21. Application of diversified teaching in the clinical teaching of Endocrinology Department for Eight-year Medical Program students

Abstract

Objective To explore the influence of different teaching strategies on the clinical teaching effect of Eight-year Medical Program students in Endocrinology Department. Methods This study conducted an online questionnaire for evaluation of the teaching quality of training for attending doctors, residents and the effect of small lectures of medical students. Participants were students from 6th-8th grades of Eight-year Program of clinical medicine in Peking Union Medical Colleges Hospital, rotating in the Department of Endocrinology from December 2021 to April 2022. Results Questionnaires were distributed to 48 medical students who had completed the rotation of Endocrinology Department. A total of 40 valid questionnaires were recovered with a recovery rate of 83.3%. The attending doctors and residents in the ward performed well in the daily teaching activities. The busy clinical work was a main factor affecting the teaching quality. Most medical students prepared lectures within 6 hours. In addition to mastering relevant knowledge, lectures also improved the speech ability, literature retrieval ability and slide production ability of students. Conclusions Adopting diverse teaching strategies and organizing various forms of traning activity are conducive to arouse the learning enthusiasm of medical students and comprehensively improve their post competencies.

31. Assessment for preliminary results of ‘PUMCH model’ for urolithiasis metabolic evaluation

Abstract

Objective To review the experience of metabolic evaluation of urinary stones in Peking Union Medical College Hospital, and summarize the ‘PUMCH model’ of metabolic evaluation. Methods With descriptive evaluation method, this study reviewed and analyzed 534 metabolic evaluation cases in the urology department of Peking Union Medical College Hospital from Jan. 2016 to Dec. 2021. The distribution differences of stone patients in gender, medical history, stone shape, stone composition, renal function, systemic metabolism and 24-hour urine metabolism were observed. Results This study found that males were more than females in terms of gender differences in stone patients; The average course of stone disease was about 10 years, and the first symptom was mainly renal colic; Calcium oxalate was the most common stone component; Kidney stone was more than urethral stone; Hyperlipidemia was the most common in patients with stones; The low concentration of urinary citrate was the most significant. Conclusions As a new method of stone diagnosis and treatment, the ‘PUMCH model’ of metabolic evaluation can comprehensively evaluate the patients' stone incidence and systemic metabolic abnormalities, which has a strong guiding role in stone treatment and prevention.

Abstract

Striving for nanometer-sized solid-state single-photon sources, we investigate atom-like quantum emitters based on single germanium vacancy (GeV) centers isolated in crystalline nanodiamonds (NDs). Cryogenic characterization indicated symmetry-protected and bright (> 10^6 counts/s with off-resonance excitation) zero-phonon optical transitions with up to 6-fold enhancement in energy splitting of their ground states as compared to that found for GeV centers in bulk diamonds (i.e., up to 870 GHz in highly strained NDs vs 150 GHz in bulk). Utilizing lithographic alignment techniques, we demonstrate an integrated nanophotonic platform for deterministic interfacing plasmonic waveguides with isolated GeV centers in NDs that enables 10-fold enhancement of single-photon decay rates along with the emission direction control by judiciously designing and positioning a Bragg reflector. This approach allows one to realize the unidirectional emission from single-photon dipolar sources introducing a novel method that is alternative to the propagation-direction-dependent techniques based on chiral interactions or topological protection. The developed plasmon-based nanophotonic platform opens thereby new perspectives for quantum nanophotonics in general and for realizing entanglement between single photons and spin qubits, in particular.

Abstract

We report on the isolation of single SiV$^-$ centers in nanodiamonds. We observe the fine-structure of single SiV$^-$ center with improved inhomogeneous ensemble linewidth below the excited state splitting, stable optical transitions, good polarization contrast and excellent spectral stability under resonant excitation. Based on our experimental results we elaborate an analytical strain model where we extract the ratio between strain coefficients of excited and ground states as well the intrinsic zero-strain spin-orbit splittings. The observed strain values are as low as best values in low-strain bulk diamond. We achieve our results by means of H-plasma treatment of the diamond surface and in combination with resonant and off-resonant excitation. Our work paves the way for indistinguishable, single photon emission. Furthermore, we demonstrate controlled nano-manipulation via atomic force microscope cantilever of 1D- and 2D-alignments with a so-far unreached accuracy of about 10nm, as well as new tools including dipole rotation and cluster decomposition. Combined, our results show the potential to utilize SiV$^-$ centers in nanodiamonds for the controlled interfacing via optical coupling of individually well-isolated atoms for bottom-up assemblies of complex quantum systems.
