ObjectiveTo explore the possible mechanism of aerobic treadmill exercise in improving renal function in hyperuricemia (HUA) rats.MethodsA total of 24 male SD rats were randomly divided into the Control group, the Model group and the Exercise group according to the random number table method, with 8 rats in each group. The Control group was given 2 g/(kg·d) distilled water intragastric administration, the Model group and the Exercise group were given 2 g/(kg·d) oxonic aid potassium salt (OAPS) intragastric administration, continuous intragastric administration for 10 days. From the 11th day, the Control group was given distilled water intragastric administration every other day, and the Model group was given OAPS intragastric administration every other day to maintain high uric acid status, with a dose of 2 g/(kg·d). In both groups, diet and water were not restricted, exercise freely, without other intervention, lasted for three weeks. The Exercise group was given intragastric administration as same as the Model group, and was given aerobic treadmill training simultaneously, the speed was 12 m/min, 20 minutes a time, once a day, continuous training for six days and rest for one day, continuous training for three weeks. Hematoxylin-eosin (HE) was used to stain the renal histopathological section, and the microscope (×400) was used to observe the structural changes of renal tubules and glomerulus; the peroxidase method was used to detect the renal function indexes such as uric acid (UA), creatinine (Cre) and urea nitrogen (BUN); enzyme-linked immunosorbent assay (ELISA) method was used to determine the contents of interleukin-1β (IL-1β) in serum and kidney tissues; Western blot was used to detect the expressions of Caspase-1, IL-1β and glucose transporter 9 (GLUT-9) in the kidney tissues.Results① Pathological examination results: there was not obvious pathological changes were observed in the kidney tissues of the Control group; in the Model group, the renal tissue capsule was incomplete, the glomerulus was deformed and atrophic, and the renal tubules were disordered; in the Exercise group, the number of inflammatory cell infiltration was significantly decreased, the renal tubular structure was relatively intact, and the pathological changes such as cytoplasmic vacuolation of renal tubular epithelial cells were significantly decreased. ② UA, Cre and BUN content of serum: compared with the Control group, UA, BUN and Cre content in serum of the Model group and Exercise group was significantly increased (PPPPPP

Allergic asthma is characterized by the polarization of Th2 cells and impaired immune regulation. Macrophages occupy the largest proportion of airway immune cells. This study aims to discover the mechanism that hinders the immune regulatory functions of airway macrophages. In this study, macrophages were isolated from cells in bronchoalveolar lavage fluids (BALF) collected from asthma patients and normal control (NC) subjects. The results indicated that macrophages occupied the largest portion of the cellular components in BALF. The frequency of IL‐10+ macrophage was significantly lower in asthma patients than in NC subjects. The expression of IL‐10 in macrophages of BALF was associated with the levels of asthma‐related parameters. The immune‐suppressive functions of BALF M0 cells were defective in asthma patients. The inducibility of IL‐10 expression was impaired in BALF macrophages of asthma patients, which could be restored by exposing to CpG. In conclusion, the induction of IL‐10 in macrophages of BALF in asthma patients was impaired, and it could be restored by exposure to CpG. [ABSTRACT FROM AUTHOR]

Fruit ripening is associated with the degreening process (loss of chlorophyll) that occurs in most fruit species. Kiwifruit is one of the special species whose fruits may maintain green flesh by accumulating a large amount of chlorophyll even after ripening. However, little is known about the genetic variations related to the fruit degreening process. Here, a graph‐based kiwifruit pangenome by analyzing 14 chromosome‐scale haplotype‐resolved genome assemblies from seven representative cultivars or lines in Actinidia chinensis is built. A total of 49,770 non‐redundant gene families are identified, with core genes constituting 46.6%, and dispensable genes constituting 53.4%. A total of 84,591 non‐redundant structural variations (SVs) are identified. The pangenome graph integrating both reference genome sequences and variant information facilitates the identification of SVs related to fruit color. The SV in the promoter of the AcBCM gene determines its high expression in the late developmental stage of fruits, which causes chlorophyll accumulation in the green‐flesh fruits by post‐translationally regulating AcSGR2, a key enzyme of chlorophyll catabolism. Taken together, a high‐quality pangenome is constructed, unraveled numerous genetic variations, and identified a novel SV mediating fruit coloration and fruit quality, providing valuable information for further investigating genome evolution and domestication, QTL genes function, and genomics‐assisted breeding. Due to the rapid development of modern industry, oily wastewater, which contained poisonous chemicals, would cause damage to marine fishery resources, and finally threaten human health. To address this issue, we herein report a fibrous membrane for oil-water separation. Firstly, the silicone polymer was synthesized by the in situ polymerization of aminopropyl trimethoxysilane and 3-glycidyloxypropyl trimethoxysilane in a polycaprolactone (PCL) solution. Then, the silicone decorated PCL (S-PCL) fibrous membranes were then prepared via electrospinning the synthesized solution. The decorated silicone offers a good porous structure for fibrous membranes. Specifically, the decorated silicone endows the PCL fibrous membrane with a good lipophilic/hydrophobic performance with a water contact angle of 169° under oil. More importantly, the obtained fibrous membrane owned a good oil-water selectivity. The oil droplets could be spread and absorbed by the fibrous membrane in water within 5 s. Such efficient separation for oil and water mixtures, endowed by silicone, will make the PCL fibrous membrane an environmental-friendly candidate for treating oily wastewater. [ABSTRACT FROM AUTHOR]

Postoperative treatment of osteosarcoma is one of the major challenging clinical issues since both elimination of residual tumors and acceleration of bone regeneration should be considered. Photothermal therapy has been widely studied due to its advantages of small side-effect, low-toxicity, high local selectivity and noninversion, and bone tissue engineering is an inevitable trend in postoperative treatment of osteosarcoma. In this study, we combined the tissue engineering and photothermal therapy together, and developed a kind of multifunctional nanofibrous 3D matrixes for postoperative treatment of osteosarcoma. The flexible bioactive glass nanofibers (BGNFs) prepared by sol–gel electrospinning and calcination acted as the basic blocks, and the genipin-crosslinked gelatin (GNP-Gel) acted as the cement to bond the BGNFs forming a stable 3D structure. The stable porous 3D scaffolds were obtained through ice crystal templating method and freeze-drying technology. The obtained GNP-Gel/BGNF 3D matrixes showed a nanofibrous structure that highly biomimetics the extracellular matrix. The excellent compression recovery performance in water of these matrixes made them suitable for minimally invasive surgery. In addition, these 3D matrixes were not only biocompatible in vitro , but also benefit for the formation of mineralized bone in vivo. Furthermore, the dark blue GNP-Gel also acted as the photothermal agent, which endowed the GNP-Gel/BGNF 3D matrixes with efficient photothermal antitumor and photothermal antibacterial performance without addition of other toxic photothermal agents. Therefore, this study provides an ingenious avenue to prepare multifunctional nanofibrous 3D matrixes with photothermal therapy for postoperative treatment of osteosarcoma. [ABSTRACT FROM AUTHOR]
