Abstract

[Objective] This study aimed to investigate the chemical constituents of walnut leaves. [Methods] The volatile components of walnut leaves were analyzed by head-space solid micro-extraction and coupled with GC-MS. The chemical composition of ethanol extract was identified using ultra-high performance liquid chromatography coupled with quadrupole Orbitrap high resolution mass spectrometry (UPLC-Q-Exactive). [Results] 88 kinds of volatile compounds were identified in walnut leaves, mainly terpenoids, of which sesquiterpenoids account for 78.35%. 30 kinds of active components were identified from ethanol extracts, including twenty kinds of polyphenols (twelve kinds of flavonoids), four kinds of phenylpropanoids, three kinds of pentacyclic triterpenoids, two kinds of essential amino acids and 1 tricarboxylic acid compounds in the ethanol extract in walnut leaves. [Conclusion] The combined use of SPME-GC-MS and UPLC-Q Exactive technology can determine and analyze the chemical composition of walnut leaves systematically, accurately and rapidly.

23. TARBP2 promotes human breast cancer cell metastasis by degrading AKAP12 transcript

Abstract

Objective To study the role and mechanism of trans-activation response RNA-binding protein 2 (TARBP2) in post-transcriptional regulation of the expression of AKAP12, a metastasis suppressor gene, in breast cancer cells. Methods The expression of TARBP2 in breast tumor tissues and potential relationship between TARBP2 expression and tumor metastasis were analyzed by bioinformatics methods. Bioinformatics methods were used to analyze the correlation between TARBP2 and AKAP12 expression in different breast cancer datasets. MDA-MB-231 cells were transfected with GFP-tagged TARBP2 plasmids by Lipofectamine 2000 and screening with G418. Lung metastasis in tumor-bearing nude mice was detected by HE staining. RT-qPCR was used to test the expression of AKAP12 and its half-life. Luciferase assay was carried out to determine whether the transcript decay was mediated through targeting the 3′UTR. EMSA assay was performed to detect the physically binding of TARBP2 with stem-loop structure of AKAP12. Results The high expression of TARBP2 in human breast tumor tissue was closely related to lymph node metastasis. TARBP2 over-expression in MDA-MB-231 cells significantly promoted tumor metastasis in vitro(P＜0.05). TARBP2 inhibited the expression of AKAP12(P＜0.01) and reduced its mRNA half-life(P＜0.05). TARBP2 significantly suppressed luciferase activity of AKAP12 3′UTR reporter(P＜0.05) and bound to the stem-loop structure. There was a significant negative correlation between TARBP2 and AKAP12 expression in human breast cancer samples(P＜0.001). Conclusions TARBP2-mediated inhibition of AKAP12 gene expression might be one of the mechanisms of TARBP2 promoting breast cancer metastasis.

Abstract

The surface terminations of 122-type alkaline earth metal iron pnictides AEFe2As2 (AE = Ca, Ba) are investigated with scanning tunneling microscopy/spectroscopy (STM/STS). Cleaving these crystals at a cryogenic temperature yields a large majority of terminations with atomically resolved square-root-two (rt2) or 1*2 lattice, as well as the very rare terminations with 1*1 symmetry. By means of lattice alignment and chemical marking, we identify these terminations as rt2-AE, 1*2-As, and rt2-Fe surfaces, respectively. Layer-resolved spectroscopy on these terminating surfaces reveals a well-defined superconducting gap on the As terminations, while the gap features become weaker and absent on AE and Fe terminations respectively. The local gap features are hardly affected by the surface reconstruction on As or AE surface, whereas a suppression of them along with the in-gap states can be induced by As vacancies. The emergence of two impurity resonance peaks at +-2 meV is consistent with the sign-reversal pairing symmetry. The definite identification of surface terminations and their spectroscopic signatures shall provide a more comprehensive understanding of the high-temperature superconductivity in multilayered iron pnictides.
