Abstract

BackgroundIt is necessary to differentiate high-risk human papillomavirus (HR-HPV) infection and cervical lesions during pregnancy using an effective measure, so as to guide personalized diagnosis and treatment and to reduce unnecessary invasive examinations during pregnancy.ObjectiveTo assess the value of HR-HPV E6/E7 mRNA detection in differentiating HR-HPV infection and cervical lesions during pregnancy by comparing efficacy between it and HR-HPV DNA testing.MethodsParticipants were healthy women with singleton pregnancy (20-45-years old) selected from Department of Obstetrics and Gynecology of Capital Medical University Xuanwu Hospital during January 2016 to January 2019. All had file creation and underwent regular prenatal examination, and those with abnormal liquid-based cytology and HR-HPV DNA testing results further underwent colposcopy with biopsy taken for pathological examination (serving as a gold standard) , and performed HR-HPV E6/E7 mRNA detection in cervical exfoliated cells collected as samples. Pathologically detected CIN Ⅱand Ⅲ were defined as high-grade cervical lesions.ResultsOf the 1 058 participants, 118 had cytological abnormalities and/or HPV 16 and 18 infections, and 84 of them consented to perform colposcopy with biopsy pathological results successfully obtained. The prevalence of HR-HPV E6/E7 mRNA positivity was lower than that of HR-HPV DNA positivity in women with CINⅠ, normal cervical epithelium or cervicitis detected by pathological examination (P0.05) . In predicting CINⅡ and Ⅲ, the HR-HPV DNA testing had a sensitivity of 89.7% (26/29) , a specificity of 21.8% (12/55) , a positive predictive value of 37.1% (26/69) , and a negative predictive value of 75.0% (12/29) , and the HR-HPV E6/E7 mRNA detection had a sensitivity of 65.5% (19/29) , a specificity of 54.5% (25/55) , a positive predictive value of 43.0% (19/44) , and a negative predictive value 75.0% (25/40) . McNemar's test revealed that HR-HPV E6/E7 mRNA detection had a lower sensitivity but a higher specificity than HR-HPV DNA testing in diagnosing CINⅡ and Ⅲ (P

20. Experimental Investigation on the Turn-Off Current Redistribution Characteristics of IGCT Based on an Isolated Anode Design

Abstract

The integrated gate commutated thyristor (IGCT) is promising in dc grids nowadays. However, to meet the increasing demands for handling high fault currents, the turn-off capability of IGCT still needs to be optimized, which is affected by transient turn-off current redistribution significantly. This article proposes an experiment method based on an isolated anode design for detecting the turn-off current redistribution characteristics of IGCT. Influences of ON-state current density and commutation impedance distribution on IGCT’s turn-off current redistribution characteristics are explored in detail. The results show that when IGCT’s turn-off process satisfies the “hard” gate drive condition, the anode current redistribution gets changed mainly in the current commutation period and the anode voltage rising period of the turn-off process, before IGCT’s dynamic avalanche phenomenon happens. Though the anode current distribution experiences large fluctuations during the commutation period, it will achieve a new balancing state at the end of this period, which almost does not affect the following periods. When IGCT goes into the anode voltage rising period, the ON-state current density distribution will affect the turn-off current redistribution. Under the effects of carrier extraction speed differences, the current density distribution throughout the chip tends to reduce its initial difference, which leads to the phenomenon of “current self-balancing.” Meanwhile, the region with the highest ON-state current density dominates the anode voltage’s building-up speed. To increase IGCT’s turn-off capability, improving the ON-state anode current density uniformity utilizing an optimized layout or cell structure is one of the effective methods.

Abstract

To solve the gas pre-drainage problem of high gas low permeability coal seam, based on LS-DYNA3D nonlinear dynamic finite element analysis software, the process of forming jet by perforating charge crushing shaped charge liner and penetrating coal seam is simulated. The formation of metal jet and the action range of penetrating coal seam are analyzed, and the penetrating depth is 1.4 m. The comparison is made with non-shaped charge liner penetrating coal seam. The results showed that the penetration depth increased by 250% and the active area increased by 2 592%.

Abstract

Esophageal squamous cell carcinoma (ESCC) is a deadly malignancy with notable metabolic reprogramming, yet the pivotal metabolic feature driving ESCC progression remains elusive. Here, we show that methionine cycle exhibits robust activation in ESCC and is reversely associated with patient survival. ESCC cells readily harness exogenous methionine to generate S-adenosyl-methionine (SAM), thus promoting cell proliferation. Mechanistically, methionine augments METTL3-mediated RNA m6A methylation through SAM and revises gene expression. Integrative omics analysis highlights the potent influence of methionine/SAM on NR4A2 expression in a tumor-specific manner, mediated by the IGF2BP2-dependent stabilization of methylated NR4A2mRNA. We demonstrate that NR4A2 facilitates ESCC growth and negatively impacts patient survival. We further identify celecoxib as an effective inhibitor of NR4A2, offering promise as a new anti-ESCC agent. In summary, our findings underscore the active methionine cycle as a critical metabolic characteristic in ESCC, and pinpoint NR4A2as a novel methionine-responsive oncogene, thereby presenting a compelling target potentially superior to methionine restriction.

Abstract

For problem of inconvenient energy supplement of node in ZigBee wireless sensor network of underground coal mine, a ZigBee routing algorithm adapted to underground coal mine was proposed. In the algorithm, neighbor table is adopted to reduce data transmission hops among nodes, and residual energy of node in the network is sufficiently utilized by use of dynamic control of node residual energy threshold. The simulation results show that the algorithm reduces energy consumption of node in routing process and extends running time of the network.
