Abstract

We hypothesized that small molecule transcriptional perturbation could be harnessed to target a cellular dependency involving protein arginine methyltransferase 5 (PRMT5) in the context of methylthioadenosine phosphorylase (MTAP) deletion, seen frequently in malignant pleural mesothelioma (MPM). Here we show, that MTAP deletion is negatively prognostic in MPM. In vitro, the off-patent antibiotic Quinacrine efficiently suppressed PRMT5 transcription, causing chromatin remodelling with reduced global histone H4 symmetrical demethylation. Quinacrine phenocopied PRMT5 RNA interference and small molecule PRMT5 inhibition, reducing clonogenicity in an MTAP-dependent manner. This activity required a functional PRMT5 methyltransferase as MTAP negative cells were rescued by exogenous wild type PRMT5, but not a PRMT5E444Q methyltransferase-dead mutant. We identified c-jun as an essential PRMT5 transcription factor and a probable target for Quinacrine. Our results therefore suggest that small molecule-based transcriptional perturbation of PRMT5 can leverage a mutation-selective vulnerability, that is therapeutically tractable, and has relevance to 9p21 deleted cancers including MPM.

8. Organizational Factors and Engineering Student Persistence

Abstract

Persistence and graduation rates continue to be important student success outcomes for engineering programs. In part, these outcomes reflect the effectiveness of the educational experience that has been delivered to the students. This educational experience is shaped by three main factors: 1) the organizational context, 2) the peer environment, and 3) the individual student experience. Prior research on student persistence in engineering has primarily focused on peer interactions and individual student experiences, while the organizational context has not received as much attention. Yet, engineering colleges and departments have a large degree of control over their organizational contexts which can be strategically managed to promote student success. This work investigated the relationship between organizational features of engineering programs and two student success outcomes: 1) program persistence after one academic year and, 2) engineering graduation. Student data included sociodemographics and academic records for the 2011 and 2012 engineering cohorts at a single institution. Organizational features that were evaluated included compositional diversity of students and instructors, gateway courses, program size and scale, and the curriculum. To operationalize the organizational features of the curriculum, the curricular complexity framework was used (Heileman et al., 2018). To enhance this framework, a novel method to measure the instructional complexity of a curriculum was proposed. This method determined the difficulty of a course using a performance metric called grade anomaly rather than the traditional DFW rate (the percentage of students who earn grades of D, F or withdraw). A student's grade anomaly in a specific course is calculated relative to their performance in their other courses. A course grade anomaly is the mean grade anomaly of students in a course, and a curricular grade anomaly is the mean course grade anomaly across a curriculum. Results showed that course grade anomaly measured a different aspect of the curriculum than DFW rate and was more robust. Multilevel models were used to determine which organizational variables influenced a student's likelihood of program persistence and engineering graduation after accounting for individual-level factors. Program size was positively associated with both outcomes, but class size had no effect. Gender and racial diversity in the student population had positive effects as well. More gateway courses in the curriculum had a negative impact on both outcomes, and a larger curricular DFW rate had a negative impact on engineering graduation only. The structural complexity of a curriculum had no influence on either outcome. Lastly, curricular grade anomaly had a negative effect on both outcomes, meaning students in more difficult programs were more likely to persist in the program and graduate from engineering. These results indicate that organizational features can affect student success, and engineering faculty should consider these features when evaluating their students' outcomes. Gender and racial diversity should be prioritized because it is beneficial for all students. The number of gateway courses should be minimized, and students should be given proper academic support to pass their difficult courses. The finding that students in more difficult programs had higher likelihoods of program persistence and engineering graduation was unexpected, and it raised questions about the properties of curricular grade anomaly and the cultural influences of engineering education. 9. Understanding Learners' Opinion about Participation Certificates in Online Courses Using Topic Modeling

Abstract

This study proposes a formal multi-step methodology for qualitative assessment of topic modeling results in the context of online learner motivation to purchase Statements of Participation (SoP). We developed Latent Dirichlet Allocation (LDA) based topic models on open-ended responses of three post-course survey questions from 280 open courses offered on the FutureLearn learning platform. For qualitative assessment, we first determined the theme of the topic based on the words that constituted the topic and responses that were most strongly associated with the topic. Then, we verified the theme by comparing the topics assigned by LDA model on a test set with manual annotation. We also performed sentiment analysis to check for alignment with human judgment. Learner motivations in each theme were interpreted with the Expectancy-Value-Cost framework. Our analyses indicated that, primarily, learners were motivated to purchase the SoP based on perceptions of the utility value and financial cost of the certificate. We found that human judgment agreed with the topic model more frequently when LDA topic weights were larger. [For the full proceedings, see ED593090.]

Abstract

Many countries have ethnically diverse populations and marketing practitioners need to consider these diversities when undertaking research, particularly when exploring sensitive topics. In Australia, Indigenous Australians make up 3.3% of the population and are a commonly researched audience to gauge attitudes and ensure cultural offense does not occur due to unintended consequences of marketing activity. However, obtaining information from such a vulnerable group using quantitively based surveys is often inappropriate or insensitive. This paper introduces to Euro-western market researchers the concepts of flipping and yarning as a market research approach that has been used by Indigenous Australians for thousands of years. This circular market research approach demonstrates that ensuring a cultural understanding of the community can provide a foundation for a research approach that is 'considered' and respectful. 17. Analyzing Large Collections of Open-Ended Feedback From MOOC Learners Using LDA Topic Modeling and Qualitative Analysis

Abstract

There is a large variation in background and purpose of massive open online course (MOOC) learners. To improve the overall MOOC learning experience, it is important to identify which MOOC characteristics are most important for learners. For this purpose, in this article, we analyzed about 150,000 open-ended learner responses from 810 MOOCs to three postcourse survey questions about their learning experience: (Q1) What was your most favorite part and why? (Q2) What your least favorite part and why? (Q3) How could the course be improved? We used the latent Dirichlet allocation topic model to identify prominent topics present in learner responses to each question. We determined the theme of each identified topic through qualitative analysis. Our results show that the following aspects of MOOCs can significantly impact the learning experience: quality of course content, accurate description of prerequisites and required time commitment in course syllabus, quality of assessment and feedback, meaningful interaction with peers and educators, engaging instructor and videos, accessibility of learning materials, and usability of platform.

A newly discovered population of Monetaria caputdraconis has been identified on Easter Island. This new population has a high occurrence of specimens expressing morphology that appear both melanistic and rostrate, and is named herein Monetaria caputdraconis form darkini. It is suggested that the geology of the island, which comprises Roiho basalt containing olivine tholeiite with relatively high concentrations of heavy metals, may contribute to this unique morphology. We have tested this hypothesis in a retrospective analysis of the two in similar groups of patients.Methods: In a 4-year experience, we performed 177 consecutive LVR procedures: 83 patients underwent Robot Assisted Thoracoscopic (RATS) LVRS and 94 EBLVR. EBLVR was intentionally precluded by evidence of incomplete fissure integrity or intra-operative assessment of collateral ventilation. Unilateral RATS LVRS was performed in these cases together with those with unsuitable targets for EBLVR.Results: EBLVR was uncomplicated in 37 (39%) cases; complicated by post-procedure spontaneous pneumothorax (SP) in 28(30%) and required revision in 29 (31%). In the LVRS group, 7 (8%) patients were readmitted with treatment-related complications, but no revisional procedure was needed. When compared with uncomplicated EBLVR, LVRS had a significantly longer operating time: 85 (14– 82) vs 40 (15– 151) minutes (p< 0.001) and hospital stay: 7.5 (2– 80) vs 2 (1– 14) days (p< 0.01). However, LVRS had a similar total operating time to both EBLVR requiring revision: 78 (38– 292) minutes and hospital stay to EBLVR complicated by pneumothorax of 11.5 (6.5– 24.25) days. Abstract

OBJECTIVES Robotic-assisted thoracoscopic surgery (RATS) facilitates complex pulmonary segmentectomy which offers one-stage diagnostic and therapeutic management of small pulmonary nodules. We aimed to explore the potential advantages of a faster, simplified pathway and earlier diagnosis against the disadvantages of unnecessary morbidity in benign cases. METHODS In an observational study, patients with small, solitary pulmonary nodules deemed suspicious of malignancy by a multidisciplinary team were offered surgery without a pre or intraoperative biopsy. We report our initial experience with RATS complex segmentectomy (using >1 parenchymal staple line) to preserve as much functioning lung tissue as possible. RESULTS Over a 4-year period, 245 RATS complex segmentectomies were performed; 140 right: 105 left. A median of 2 (1–4) segments was removed. There was no in-hospital mortality and no requirement for postoperative ventilation. Complications were reported in 63 (25.7%) cases, of which 36 (57.1%) were hospital-acquired pneumonia. A malignant diagnosis was found in 198 (81%) patients and a benign diagnosis in 47 (19%). The malignant diagnoses included: adenocarcinoma in 136, squamous carcinoma in 31 and carcinoid tumour in 15. The most frequent benign diagnosis was granulomatous inflammation in 18 cases. CONCLUSIONS RATS complex segmentectomy offers a precise, safe and effective one-stop therapeutic biopsy in incidental and screen-detected pulmonary nodules. [ABSTRACT FROM AUTHOR]

Abstract

The most recent b-physics results from the OPAL experiment at LEP are reviewed. A measurement of semileptonic B meson decays to narrow orbitally-excited charm mesons is presented first, followed by a study of charm production in b-hadron decays.
