A decision support system, SBN-Watch, was developed to demonstrate the influence of crop rotation and the choice of sugar beets (Beta vulgaris) varieties on the sugar beet cyst nematode Heterodera schachtii Schmidt (SBN) population and sugar beets yield. The database in SBN-Watch consists of a varietal unit with five sugar beet varieties representing the three categories “Standard,” “Tolerant” and “semi-tolerant.” Data of minimal yield (m), tolerance limit (T), and population dynamic parameters were obtained from published commercial field trials conducted in Sweden and Denmark in 2011. Additionally, a sanitation intercrop unit with different resistant classes of white mustard (Sinapsis arvensis) and oil seed radish (Raphanus sativus) was included. The relationship between initial population (Pi) and sugar yield as well as SBN final population in soil (Pf) was calculated by two Seinhorst equations. Few data inputs are required to be entered by the user in SBN-Watch, mainly the initial population (Pi), expected sugar price and exchange rate of € to SEK. The calculated reproduction factor (Rf) values using SBN-Watch corresponded well with varietal characteristics, where the standard variety Mixer had the highest (Rf) values. The influence of the initial SBN population on the calculated sugar yield (tonnes ha−1) was generally small at Pi < 2.

Oilseed radish (OR; Raphanus sativus var. oleiferus) is grown as a cover crop and develops a unique taproot, absorbing nitrogen left by the previous crop. The aim of this project was to investigate the resistance of OR cultivars (cvs.) to Plasmodiophora brassicae, the causal agent of clubroot disease. Twelve market cvs. were compared with cvs. of clubroot-resistant (CR) winter oilseed rape (OSR; Brassica napus) and other selected species of the Brassicaceae family. The study was performed as a replicated bioassay in a growth chamber using a specially composed mixture of field soils holding the natural inoculum of P. brassicae. The results show that the OR cultivars were infected, which implies that OR multiplies the pathogen. The susceptibility of the OR cultivars was not significantly different from that of the CR OSR cultivars Alister and Archimedes, but it was significantly different from that of the OSR cv. Mendel. The disease severity index (DSI) for OR cultivars ranged from 2.3 to 9.3, and disease incidence was 3–17%. The best performance was shown by black radish (Raphanus sativus var. niger) with a DSI of 0.3. For sustainable brassica crop production, we suggest avoiding OR as a cover crop in crop rotations, including OSR or other brassica crops, since there is a risk of increasing inoculum in the soil.

Abstract Incidence of tick-borne encephalitis (TBE) has increased during the last years in Scandinavia, but the underlying mechanism is not understood. TBE human case data reported between 2010 and 2021 were aggregated into postal codes within Örebro County, south-central Sweden, along with tick abundance and environmental data to analyse spatial patterns and identify drivers of TBE. We identified a substantial and continuing increase of TBE incidence in Örebro County during the study period. Spatial cluster analyses showed significant hotspots (higher number of cases than expected) in the southern and northern parts of Örebro County, whereas a cold spot (lower number of cases than expected) was found in the central part comprising Örebro municipality. Generalised linear models showed that the risk of acquiring TBE increased by 12.5% and 72.3% for every percent increase in relative humidity and proportion of wetland forest, respectively, whereas the risk decreased by 52.8% for every degree Celsius increase in annual temperature range. Oilseed rape (OSR; Brassica napus) is a globally important crop which is increasingly under pressure from pests, pathogens and weeds. We investigated the potential of achieving multifunctional crop protection benefits by intercropping oilseed rape with legumes. A field experiment was conducted in which winter oilseed rape was intercropped with the annual frost sensitive legumes berseem clover (Trifolium alexandrinum) or spring faba bean (Vicia faba), or with the winter grain legumes winter faba bean or winter peas (Pisum sativum). We tracked damage to winter oilseed rape by autumn and spring pests (slugs and insects), pathogens, weed biomass, as well as oilseed rape and intercrop yield in each treatment. Intercropping treatments resulted in pest damage that was equivalent or lower than in oilseed rape alone. Follow up field and lab assessments for the frost sensitive legume intercrops provided evidence for a reduction in autumn pest damage to OSR. Each legume intercrop had its own benefits and drawbacks in relation to pest, pathogen and weed suppression, suggesting that the plant species selected for intercropping with oilseed rape should be based on the pests, pathogens and weeds of greatest concern locally to achieve relevant multifunctional benefits. Our study provides a framework for further experiments in which the multifunctional effects of intercropping on pests, pathogens and weeds can be quantified.

Abstract Background Oilseed rape (OSR; Brassica napus L.) is a highly valued crop for food, feed and industrial use. It is primarily grown in temperate climates, and over recent decades, its area of production and profitability have increased. Concurrently, several diseases negatively impact OSR production. Diseases caused by soil-borne pathogens, pose a risk of substantial yield loss since crop rotation schemes have become narrow as the time lapse between OSR crops in a field has been shortened. The aims of this paper were to provide an overview of plant protection measures available for OSR production and to identify knowledge gaps and areas where more research is needed. Methods This systematic map builds on a previously published protocol and follows the ROSES reporting standard. The search strategy was developed in collaboration with stakeholders and designed to cover available scientific evidence for OSR disease management in climate zones relevant for Scandinavian crop production (Dfc, Dfb, Cfb and Cfa in the Köppen-Geiger climate classification). Five scientific databases were used to identify peer-reviewed literature, complemented by additional searches performed in grey literature. Articles were screened at three stages: the title, abstract and full text. The eligible publications included studies of OSR crops, and all measures to control crop disease in agricultural fields were considered eligible interventions. The comparator was intervention and no intervention, and the yield per unit area, disease suppression or an increase in crop quality were determined to be outcomes of interventions. A basic assessment of the experimental design of each study was performed to assess its eligibility. All articles were coded based on the following categories: the location and climate zone, disease, pathogen, intervention and management method, outcome and study design. Articles not reporting original data but judged to be relevant (i.e., review papers, books and notes of registration of cultivars) were saved in a separate category called “books, reviews and reports”. Review findings A total of 4633 articles were collected through systematic searches. After duplicates were removed, 3513 articles were included in the screening process. After screening at the title and abstract levels, 897 articles were evaluated at the full text level, and 118 articles comprised the studies that met the eligibility criteria of the systematic map. The country (Canada) and region (Europe) with the largest OSR crop production areas also contributed the highest number of articles. In total, 17 different diseases were reported, with black leg (syn. Phoma stem canker) being the most studied disease. Nineteen different intervention methods or management types were examined. Cultivar resistance and pesticide application were the most studied control measures. Conclusion We report scientific studies on plant disease protection measures for OSR based on field trials where the results are intended to be directly implemented in crop production management. The map clearly provides an overview of research progress throughout the time period chosen, and it identifies knowledge gaps regarding important diseases where only a few studies have been published, for example, diseases caused by viruses.

Abstract Background Oat (Avena sativa L.) is an important cereal crop for livestock feed and human consumption. The largest oat-producing countries are located in the Northern Hemisphere with Sweden as the tenth largest producer. Oat production is challenged by different diseases that can lead to significant yield reductions and impaired grain quality. The use of efficient and sustainable plant protection management is of great economic and ecological importance. The systematic map in this study aims to provide a knowledge base inventory and to identify areas that need to be researched in the future in terms of plant disease management for more sustainable oat production. Methods Literature searches were conducted in both academic bibliographic databases and relevant online sources of grey literature. A time-span restriction of 40 years (1978–2018) was applied to the searches. English was used in all searches, and Swedish, Norwegian and Danish languages were used in the grey literature searches. The screening process, which followed a protocol with eligibility criteria, was conducted at three levels: title, abstract and full text. Metadata incorporating bibliographic information, study location, climatic zone, disease name, the common and scientific names of the disease-causing organism, pathogen type, intervention and management methods, diseased plant part, plant stage, and outcome were extracted from the studies and included in the systematic map. The systematic map findings are visualized in figures and tables and described. All included studies can be found in a searchable database. Review findings A total of 58 eligible articles, most (n = 51) from scientific journals published in English, were included in the systematic map. A majority of the studies were conducted in the Northern Hemisphere in temperate climatic zones, where most of the world’s oats are produced. The earliest article was published in 1980, followed by an oscillating temporal distribution of articles over the following years. By country, Canada had the highest number of articles, and by region, Europe had the highest number. Fungi were the most studied pathogen type, and a total of 16 different diseases were reported. Fusarium head blight (Fusarium spp.) and crown rust (Puccinia coronata) were the most studied diseases. In total, 17 different intervention management approaches for controlling the diseases were analyzed in the articles, with cultivar resistance and pesticide application as the most studied methods. Conclusion The map highlights the low quantity of available relevant field research on oat disease management. To our knowledge, this is the first systematic map of crop protection. This map provides a database of scientific literature that can be used to develop sustainable disease management strategies. The method used in this study has great potential and can also be used to benefit other crops. Research is often based on the availability of funding, and this map could be a useful tool for researchers and funding organizations to identify relevant research topics that need to be further studied. In addition, this systematic map offers a useful tool for field-based advisors in providing scientifically relevant crop protection strategies for farmers.

Abstract Background Tick distribution in Sweden has increased in recent years, with the prevalence of ticks predicted to spread towards the northern parts of the country, thus increasing the risk of tick-borne zoonoses in new regions. Tick-borne encephalitis (TBE) is the most significant viral tick-borne zoonotic disease in Europe. The disease is caused by TBE virus (TBEV) infection which often leads to severe encephalitis and myelitis in humans. TBEV is usually transmitted to humans via tick bites; however, the virus can also be excreted in the milk of goats, sheep and cattle and infection may then occur via consumption of unpasteurised dairy products. Virus prevalence in questing ticks is an unreliable indicator of TBE infection risk as viral RNA is rarely detected even in large sample sizes collected at TBE-endemic areas. Hence, there is a need for robust surveillance techniques to identify emerging TBEV risk areas at early stages. Methods Milk and colostrum samples were collected from sheep and goats in Örebro County, Sweden. The milk samples were analysed for the presence of TBEV antibodies by ELISA and validated by western blot in which milk samples were used to detect over-expressed TBEV E-protein in crude cell extracts. Neutralising titers were determined by focus reduction neutralisation test (FRNT). The stability of TBEV in milk and colostrum was studied at different temperatures. Results In this study we have developed a novel strategy to identify new TBEV foci. By monitoring TBEV antibodies in milk, we have identified three previously unknown foci in Örebro County which also overlap with areas of TBE infection reported during 2009–2018. In addition, our data indicates that keeping unpasteurised milk at 4 °C will preserve the infectivity of TBEV for several days. Conclusions Altogether, we report a non-invasive surveillance technique for revealing risk areas for TBE in Sweden, by detecting TBEV antibodies in sheep milk. This approach is robust and reliable and can accordingly be used to map TBEV “hotspots”. TBEV infectivity in refrigerated milk was preserved, emphasising the importance of pasteurisation (i.e. 72 °C for 15 s) prior to consumption.

Clubroot, caused by Plasmodiophora brassicae, is a crucial oilseed rape disease worldwide. Information on the virulence of P. brassicae populations is essential to apply disease control with proper clubroot-resistant cultivars. In 2016–2020, 84 isolates of P. brassicae were collected in the Czech Republic (CZ), Germany (DE), Poland (PL), and Sweden (SW). Pathotypes were designated using 17 Brassica hosts, including the European Clubroot Differentials (ECD), Somé set, and clubroot-resistant oilseed rape cv. Mendel. According to the ECD set, virulence analyses differentiated the isolates into 42 pathotypes. The most common pathotypes were 16/31/31 (in DE, PL, and SW) and 16/06/12 (in CZ, DE, and PL). Six pathotypes were found according to the Somé set, including 1–4 pathotypes per country. P1 was most prevalent in DE, PL, and SW, while P3 was abundant in CZ, DE, and PL. The current study provides clear evidence for a shift towards increased virulence in P. brassicae populations compared to previous studies. Several isolates overcame the resistance of cv. Mendel and of Brassica rapa genotypes ECD 01 to ECD 04. Considering all investigated samples, significant negative correlations were found between clubroot incidence and the frequency of oilseed rape in crop rotation, as for clubroot incidence and soil pH.

Optimal seed yield and quality requires that the sulphur (S) demand of faba bean (Vicia faba L.) is ensured. The effect of S fertilization on organic cultivated faba bean was investigated under field conditions during two growing seasons (2017–2018), in Sweden. Kieserite (MgSO4) and gypsum (CaSO4) were applied at a rate of 20 and 40 kg ha-1 to study the effect on faba bean growth, yield, crude protein (CP) content and amino acid (AA) composition. Gypsum and kieserite significantly increased S concentration of faba bean dry matter (DM) in 2017. The S concentration ranged from 0.20% to 0.23% of DM compared to 0.18% in the untreated control. In 2018, kieserite application at 40 kg ha-1 significantly increased S concentration to 0.15% compared to 0.12% in the untreated control. The faba bean plants did, however, not respond neither with increased growth nor increased seed yield. The seed quality in terms of CP and S-containing AA, was not affected by S fertilization, however, significant differences were observed between the experimental sites.

Soil analysis is crucial for estimating the risk of crop damage by the root-knot nematode Meloidogyne hapla. Here, we developed an analysis assay based on Loop-mediated Isothermal Amplification (LAMP). The LAMP primers were verified for specificity against 10 different nematode species. A manual soil DNA extraction, referred to as SKMM, was developed and compared with a FastDNA kit followed by DNA purification. DNA was extracted with both methods from artificially inoculated soils as well as from naturally infested soil collected from farm fields. The primers exclusively amplified DNA from M. hapla with both colorimetric and real-time LAMP. The detection limit was 193 gene copies and 0.0016 juveniles (12 pg µL−1) per reaction. DNA concentrations and purity (A260/A230) were significantly higher using the SKMM procedure compared with the kit. From the field samples collected in 2019, DNA was amplified from 16% of samples extracted with SKMM and from 11% of samples using the kit. Occurrence of M. hapla DNA was confirmed in soil samples from two out of six field soils in 2020 using both real-time LAMP and qPCR. In conclusion, the developed real-time LAMP is a fast and specific assay for detection and quantification of M. hapla DNA in soil.

Seed treatment with mineral nutrients in spring oilseed rape (SOR) under cold climatic conditions has the potential to improve the outcome of production. The objectives of this study were to investigate the effects of mineral nutrients applied as seed treatment on germination and growth of roots and biomass in SOR. In addition, seed yield and oil content were also measured. Seed vigour tests and in-field experiments were performed in Central Sweden in 2016 and 2017. Seven products with different composition of mineral nutrients were applied to the seed in a combination with binder, filler and fungicide (BFF), or applied directly onto the seed. The results showed that a treatment with mineral nutrients increased the plant size at stage BBCH 60 (early flowering stage) when the products were applied in combination with BFF in the field experiments. However, the increased plant size did not influence seed yield. Furthermore, when seeds were treated only with a nutrient product the addition of BFF decreased plant size by 10% on average and seed and crude fat yields by 4% on average. The use of BFF may thereby be questioned and is subject to further investigation.

Multispecies cropping systems contribute to sustainable agriculture with multiple ecosystem services. Effects of intercropping of organically managed maize and faba beans to silage on acquisition of mineral nutrients in shoots of both crops and on leaf spot progression in faba beans were investigated. Three field experiments were performed with maize and faba bean intercropped or grown separately. Intercropping increased shoot concentrations of K, Ca, Mg, Na, S and B in faba bean, and shoot concentrations of Cu, Zn and Mo in maize. Thus, the ecological complementary effects enhance feed quality. Disease severity index (DSI) of leaf spots in faba beans was reduced by intercropping by 42–57%, partly due to an increased Cu acquisition at sites where the Cu availability was low. There was a significant negative linear relationship between Cu concentration in shoots and DSI of leaf spots. Total uptake of mineral nutrients per land area was greater in the intercropping system with a total LER > 1 for all mineral nutrients, except for P, Ca and Mn at one of the sites. Increased nutrient use efficiency, due to facilitative uptake from the soil, and the production of crops with higher contents of minerals compared with monocropping, are benefits of intercropped maize and faba beans.

Tripartite motif-containing (TRIM) proteins are defined by the sequential arrangement of RING, B-box and coiled-coil domains (RBCC), where the B-box domain is a unique feature of the TRIM protein family. TRIM21 is an E3 ubiquitin-protein ligase implicated in innate immune signaling by acting as an autoantigen and by modifying interferon regulatory factors. Here we report the three-dimensional solution structure of the TRIM21 B-box2 domain by nuclear magnetic resonance (NMR) spectroscopy. The structure of the B-box2 domain, comprising TRIM21 residues 86-130, consists of a short α-helical segment with an N-terminal short β-strand and two anti-parallel β-strands jointly found the core, and adopts a RING-like fold. This ββαβ core largely defines the overall fold of the TRIM21 B-box2 and the coordination of one Zn2+ ion stabilizes the tertiary structure of the protein. Using NMR titration experiments, we have identified an exposed interaction surface, a novel interaction patch where the B-box2 is likely to bind the N-terminal RING domain. Our structure together with comparisons with other TRIM B-box domains jointly reveal how its different surfaces are employed for various modular interactions, and provides extended understanding of how this domain relates to flanking domains in TRIM proteins.

Outbreaks of clubroot disease caused by the soil-borne obligate parasite Plasmodiophora brassicae are common in oilseed rape (OSR) in Sweden. A DNA-based soil testing service that identifies fields where P. brassicae poses a significant risk of clubroot infection is now commercially available. It was applied here in field surveys to monitor the prevalence of P. brassicae DNA in field soils intended for winter OSR production and winter OSR field experiments. In 2013 in Scania, prior to planting, P. brassicae DNA was detected in 60% of 45 fields on 10 of 18 farms. In 2014, P. brassicae DNA was detected in 44% of 59 fields in 14 of 36 farms, in the main winter OSR producing region in southern Sweden. P. brassicae was present indicative of a risk for >10% yield loss with susceptible cultivars (>1300 DNA copies g soil−1) in 47% and 44% of fields in 2013 and 2014 respectively. Furthermore, P. brassicae DNA was indicative of sites at risk of complete crop failure if susceptible cultivars were grown (>50 000 copies g−1 soil) in 14% and 8% of fields in 2013 and 2014, respectively. A survey of all fields at Lanna research station in western Sweden showed that P. brassicae was spread throughout the farm, as only three of the fields (20%) showed infection levels below the detection limit for P.brassicae DNA, while the level was >50,000 DNA copies g−1 soil in 20% of the fields. Soil-borne spread is of critical importance and soil scraped off footwear showed levels of up to 682 million spores g−1 soil. Soil testing is an important tool for determining the presence of P. brassicae and providing an indication of potential yield loss, e.g., in advisory work on planning for a sustainable OSR crop rotation. This soil test is gaining acceptance as a tool that increases the likelihood of success in precision agriculture and in applied research conducted in commercial oilseed fields and at research stations. The present application highlights the importance of prevention of disease spread by cleaning of farm equipment, footwear, etc.
