Plants of the genus Physalis are of economic interest because of their fleshy edible fruits with high nutritional value. Some species have high medicinal value with a long history of ethno-medicinal use to treat diverse diseases. There is therefore a need to correctly discriminate the different species of Physalis for proper utilization. Although most Physalis species have unique morphologies, their vegetative stages are identical, making it difficult to accurately identify them based on morphological characteristics. DNA barcoding has the potential to discriminate species accurately. In this study, ribulose bisphosphate carboxylase large (rbcL) and internal transcribed spacer 2 (ITS2) regions were used to discriminate Physalis species and to reveal their phylogenetic relationships and genetic diversity. Physalis plant samples were collected from seven counties in Kenya based on the availability of the germplasm. The voucher specimens were identified using the botanical taxonomy method and were deposited in the University of Nairobi herbarium. Genomic DNA was isolated from leaf samples of 64 Physalis accessions and used for PCR amplification and the sequencing of rbcL and ITS2 barcode regions. The discriminatory ability of the barcodes was based on BLASTn comparison, phylogenetic reconstruction and cluster analysis, and the determination of inter- and intra-specific distances. The nucleotide polymorphism, genetic diversity and distance of the identified Physalis species were determined using DnaSP and MEGA 11.0 software. Species discrimination was more robust using ITS2 sequences. The species identified and discriminated by ITS2 sequences were Physalis purpurea, Physalis peruviana and Physalis cordata. The rbcL sequences were only able to identify Physalis to the genus level. There was high interspecific and low intraspecific divergence within the identified Physalis species based on ITS2 sequences. The ITS2 barcode is an ideal DNA barcode for use in the discrimination of species, as well as in genetic diversity studies of Physalis accessions in Kenya. [ABSTRACT FROM AUTHOR]

Context: In Kenya, most people use traditional medicine and medicinal plants to treat many diseases including malaria. To manage malaria, new knowledge and products are needed. Traditional herbal medicine has constituted a good basis for antimalarial lead discovery and drug development. Objectives: To determine in vivo antimalarial activity and brine shrimp toxicity of five medicinal plants traditionally used to treat malaria in Msambweni district, Kenya. Materials and methods: A 0.2 ml saline solution of 100 mg/kg aqueous crude extracts from five different plant parts were administered orally once a day and evaluated for their in vivo chemosuppressive effect using Plasmodium berghei berghei-infected Swiss mice for four consecutive days. Their safety was also determined using Brine shrimp lethality test: Grewia trichocarpa Hochst ex A. Rich (Tiliaceae) root, Dicrostachys cinerea (L) Wight et Am (Mimosaceae) root, Tamarindus indica L. (Caesalpiniaceae) stem bark, Azadirachta indica (L) Burn. (Meliaceae) root bark, and Acacia seyal Del. (Mimosaceae) root. Results: Parasitaemia was as follows: A. indica, 3.1%; D. cinerea, 6.3%; T. indica, 25.1%; A. seyal, 27.8%; and G. trichocarpa, 35.8%. In terms of toxicity, A. indica root bark extract had an LC50 of 285.8 µg/ml and was considered moderately toxic. T. indica stem bark extract and G. trichocarpa root extract had an LC50 of 516.4 and 545.8 µg/ml, respectively, and were considered to be weakly toxic while A. seyal and D. cinerea root extracts had a LC50 >1000 µg/ml and were, therefore, considered to be non-toxic. Discussion and conclusion: All extracts had antimalarial activity that was not significant compared to chloroquine ( p ≥ 0.05). No extract was toxic to the arthropod invertebrate, Artemia salina L. Mycotoxin contamination of wheat kernels and diversity of fungal pathogens were monitored in a survey of 26 fields in Nakuru district, Kenya, in 2006. Sampling was performed twice; at mid-anthesis in July for leaves, stems and spikelets and at harvest in September for spikelets and kernels. Kernels were analysed for the occurrence of 32 mycotoxins using a high performance liquid chromatography – electrospray tandem mass spectrometry (LC-ESI-MS/MS) method. Fungi were isolated from surface-sterilized tissues and differentiated morphologically to the genus level. Fusarium isolates were identified based on the sequence of translation elongation factor 1-alpha gene. Eleven Fusarium-related mycotoxins were quantified with deoxynivalenol being detected at highest frequency (69%) and highest concentrations. Occurrence of enniatins in wheat (50%) is reported for the first time in Kenya. Non- Fusarium mycotoxins detected included aflatoxin G2, ochratoxin A, alternariol and alternariol monomethyl ether. Prevalence of Fusarium species on different wheat parts was 100% at both growth stages. Nineteen Fusarium species were identified with seven species – Fusarium chlamydosporum, Fusarium boothi, Fusarium poae, Fusarium scirpi, Fusarium arthrosporioides, Fusarium oxysporum and Fusarium graminearum– accounting for 80% of infections. At anthesis, spikelets and leaves were the most and least susceptible tissue, respectively. At harvest, infection of spikelets was 59% higher than that of kernels. [ABSTRACT FROM AUTHOR]
