3. Accumulation of Mn(II) in Deinococcus radiodurans facilitates gamma-radiation resistance

Abstract

Deinococcus radiodurans is extremely resistant to ionizing radiation. How this bacterium can grow under chronic y radiation [50 grays (Gy) per hour] or recover from acute doses greater than 10 kGy is unknown. We show that D. radiodurans accumulates very high intracellular manganese and low iron levels compared with radiation-sensitive bacteria and that resistance exhibits a concentration-dependent response to manganous chloride [Mn(II)]. Among the most radiation-resistant bacterial groups reported, Deinococcus, Enterococcus, Lactobacillus, and cyanobacteria accumulate Mn(II). In contrast, Shewanella oneidensis and Pseudomonas putida have high iron but low intracellular manganese concentrations and are very sensitive. Abstract

Naphthalene dioxygenase catalyzes the enantiospecific monooxygenation of indan to (+)-(1S)-indanol and desaturation of indan to indane, serving as a substrate for the formation (+)-(1R,2S)-indandiol and (+)-(1S)-indenol. Indene is recognized as an intermediate in indan oxidation. (+)-(1S)-indanol and (+)-(1RS,2S)-indandiol are the important products formed from indene by purified naphthalene dioxygenase.

29. Haloacetonitriles Are Low KIInhibitors of Bacterial Dichloromethane Dehalogenases

Abstract

Distinct dichloromethane dehalogenases from Methylobacteriumsp. strain DM4 and MethylophilusDM11 were inhibited by low concentrations of haloacetonitriles. Chloroacetonitrile (ClCH2CN) showed maximal inhibition at a stoichiometry of 1 mol inhibitor: 1 mol holoenzyme for both enzymes. This stoichiometry is suggestive of one active site per holoenzyme or extreme negative cooperativity amongst the subunits. Radiolahcllcd ClCH2CN dissociated completely or partially from the two dehalogenases, respectively, during chromatography. This suggested ClCH2CN was bound non-covalently.
