Objasnění vztahu mezi mechanickým chováním a mikroskopickou strukturou laminárního spoje koňského kopyta. Cíl: Stanovit mechanické parametry a 2D délkovou hustotu profilů sekundárních lamel laminárního spoje v dermální oblasti a určit jejich možnou korelaci. Metody: Vzorky (25 celkově) laminárního spoje byly odebrány z přední, postranní a zadní části tří kopyt a vystaveny jednoosému tahovému mechanickému zatížení až do porušení tkáně. Byly stanoveny Youngovi moduli elasticity, mezní napětí a mezní deformace. Vzorky sousedící se tkání použitou na mechanické testy byly histologicky zpracovány pro získání délkové hustoty bazální membrány laminárního spoje pomocí stereologické mřížky. Výsledky: Medián (interval mezi quartily) délkové hustoty bazální membrány laminárního spoje byl 0.024 (0.020–0.027)/um. Youngův modul elasticity pro malé deformace byl 0.15 (0.11–0.35) MPa a pro velké deformace 7.58 (6.14–8.68) MPa. Mezní napětí dosahovalo hodnot 1.67 (1.41–2.67) MPa a mezni deformace 0.50 (0.38–0.70). Youngův modul elasticity v oblasti malých deformací vykazoval střední korelaci s délkovou hustotou bazální membrány laminárního spoje. Závěr: Jako většina měkkých biologických tkání laminární spoj vykazuje nelineární mechanické chování. V oblasti malých deformací, které korelují s fyziologickým zatížením laminárního spoje, větší délková hustota bazální membrány laminárního spoje odpovídá větší odolnosti laminárního spoje vůči velkému napětí přenášenému z distálního článku prstu na kopytní stěnu. Potenciální významnost: Stav laminárního spoje může být jednoduše stanovený jako délková hustota profilů sekundárních dermálních lamel. Tato kvantifikace poskytuje jednoduchý nástroj pro porovnávání náklonosti různých částí laminárního spoje k počátečním stadiím Reason for performing the study: To elucidate the relationship between mechanical behavior and microscopic morphology of the corio-epidermal junction (CEJ) of equine hooves under testing conditions. Objectives: To determine the mechanical parameters and the two-dimensional length density of profiles of secondary lamellae of the CEJ in the dermal region (LA(SL, dermal)) and to assess possible correlations. Methods: Specimens of the CEJ were taken from the front, quarter and heel parts from three equine hooves, (n=25) and exposed to a uniaxial tensile test until rupture to obtain Young’s moduli of elasticity, ultimate stress, and strain. The neighboring specimens to those used for biomechanical experiment were processed histologically to quantify the LA(SL, dermal) using stereological grids. Results: The estimated LA(SL, dermal) was 0.022±0.006 m-1 (mean±SD). Young’s modulus of elasticity in the small deformation region was 0.31±0.04 MPa, and Young’s modulus of elasticity in the linear region was 7.58±1.59 MPa. The ultimate stress was 2.09±0.96 MPa, and the ultimate strain was 0.59±0.25. The mechanical Young’s modulus of elasticity in the region of small deformations has a moderate correlation with the LA(SL, dermal). Conclusions: As with most soft biological tissues, the CEJ has a nonlinear mechanical behavior. Within the range of small deformations, which correspond to physiological loading of the CEJ, a higher LA(SL, dermal) is correlated with a higher resistance of the CEJ against high stresses transmitted from the distal phalanx to the hoof wall. Potential relevance: The condition of the CEJ apparatus may be easily quantified as the length density of the profiles of secondary dermal lamellae. Quantification of LA(SL, dermal) provides a simple tool that could be used e.g., for comparing the proneness of the various parts of the CEJ to initial stages of laminitis.
