A highly effective Co-doped CuO photocatalyst was fabricated using a one-pot green synthetic route for photocatalytic decomposition of methylene blue (MB). The synthesized materials were confirmed using Field emission-scanning electron microscopy (FE-SEM), X-ray diffraction (XRD), Thermogravimetric analysis (TGA) and Fourier transform infrared spectroscopy (FTIR). The optical and electrochemical properties, including the energy band gap, transient photocurrent response (TPR), and charge-transfer resistance (Rct), were assessed using chronoamperometry (CA), and electrochemical impedance spectroscopy (EIS), respectively. The results from TPR, and EIS revealed that cobalt doping effectively promotes the separation of photogenerated charge carrier’s, diminishing recombination and thereby improving photocatalytic activity. The evaluation of photocatalytic activity revealed that the fabricated Co-doped CuO nanoparticles (NPs) demonstrated excellent photocatalytic activity compared to pure CuO NPs. Under UV–vis light irradiation, Co-doped CuO NPs achieved photocatalytic degradation efficiency of about 88 % within 100 min, whereas for pristine CuO NPs it was 75 % under the same irradiation time. The highest degradation efficiency achieved by Co-doped CuO NPs was ascribed to their improved electron and hole separation. Moreover, the scavenger experiment showed that the primary active species responsible for the photocatalytic decomposition of MB were hydroxyl radicals (•OH) and holes (h+), followed by electrons (e-) and superoxide radicals (•O2–). The developed Co-doped CuO photocatalyst was highly stable even after 5th cycle. Additionally, the results demonstrated that the Co-doped CuO nanocomposite displayed a significantly higher water-splitting activity. This study offers a valuable reference for metal doping in photocatalyst semiconductors for effective photocatalytic removal of organic contaminants and hydrogen production.

Summary: Caspase-8-dependent pyroptosis has been shown to mediate host protection from Yersinia infection. For this mode of cell death, the kinase activity of receptor-interacting protein kinase 1 (RIPK1) is required, but the autophosphorylation sites required to drive caspase-8 activation have not been determined. Here, we show that non-canonical autophosphorylation of RIPK1 at threonine 169 (T169) is necessary for caspase-8-mediated pyroptosis. Mice with alanine in the T169 position are highly susceptible to Yersinia dissemination. Mechanistically, the delayed formation of a complex containing RIPK1, ZBP1, Fas-associated protein with death domain (FADD), and caspase-8 abrogates caspase-8 maturation in T169A mice and leads to the eventual activation of RIPK3-dependent necroptosis in vivo; however, this is insufficient to protect the host, suggesting that timely pyroptosis during early response is specifically required to control infection. These results position RIPK1 T169 phosphorylation as a driver of pyroptotic cell death critical for host defense.

Abstract Genetic selection has remarkably helped U.S. dairy farms to decrease their carbon footprint by more than doubling milk production per cow over time. Despite the environmental and economic benefits of improved feed and milk production efficiency, there is a critical need to explore phenotypical variance for feed utilization to advance the long-term sustainability of dairy farms. Feed is a major expense in dairy operations, and their enteric fermentation is a major source of greenhouse gases in agriculture. The challenges to expanding the phenotypic database, especially for feed efficiency predictions, and the lack of understanding of its drivers limit its utilization. Herein, we leveraged an artificial intelligence approach with feature engineering and ensemble methods to explore the predictive power of the rumen microbiome for feed and milk production efficiency traits, as rumen microbes play a central role in physiological responses in dairy cows. The novel ensemble method allowed to further identify key microbes linked to the efficiency measures. We used a population of 454 genotyped Holstein cows in the U.S. and Canada with individually measured feed and milk production efficiency phenotypes. The study underscored that the rumen microbiome is a major driver of residual feed intake (RFI), the most robust feed efficiency measure evaluated in the study, accounting for 36% of its variation. Further analyses showed that several alpha-diversity metrics were lower in more feed-efficient cows. For RFI, [Ruminococcus] gauvreauii group was the only genus positively associated with an improved feed efficiency status while seven other taxa were associated with inefficiency. The study also highlights that the rumen microbiome is pivotal for the unexplained variance in milk fat and protein production efficiency. Estimation of the carbon footprint of these cows shows that selection for better RFI could reduce up to 5 kg of diet consumed per cow daily, potentially reducing up to 37.5% of CH4. These findings shed light that the integration of artificial intelligence approaches, microbiology, and ruminant nutrition can be a path to further advance our understanding of the rumen microbiome on nutrient requirements and lactation performance of dairy cows to support the long-term sustainability of the dairy community.

Non-opioid and non-pharmacologic treatments are recommended when clinically appropriate, but research on the mechanisms underlying benefits of these treatments is lacking. Here, we examined the role of sleep in the effects of three non-pharmacologic pain treatments in Veterans. Specifically, we investigated whether treatment effects on sleep predicted treatment effects on pain occurring later, or vice versa.Methods: Veterans enrolled in a randomized controlled trial were invited to participate in this supplementary sleep study. A total of 174 Veterans were randomized to one of three 8-session, in-person, group-based pain treatments: hypnosis, mindfulness meditation, or education control. Measurements included self-reported sleep disturbance, pain intensity, and pain catastrophizing; sleep duration was assessed with actigraphy. Sleep and pain measurements were obtained at baseline, posttreatment, and 3-month posttreatment follow-up.Results: At baseline, average pain intensity was moderate (mean ± SD: 5.7 ± 1.7 on the 0– 10 Numeric Rating Scale), pain catastrophizing was just below the clinically relevant threshold (mean ± SD: 28.6 ± 12.2 on the Pain Catastrophizing Scale), and subjective sleep disturbance exceeded the US population average (mean ± SD: 58.5 ± 8.1 on the Patient Reported Outcomes Measurement Information System Sleep Disturbance – Short Form). By contrast, objective sleep duration was consistent with the recommended daily sleep amount of 7– 8 h for adults (mean ± SD: 8.3 ± 1.4 h). Across treatment conditions, pain intensity, pain catastrophizing, and subjective sleep disturbance were significantly less at posttreatment and 3-month follow-up than at baseline (p < 0.001). Actigraphic sleep duration did not differ significantly as a function of time. There was a high degree of covariation among the measures of pain intensity, pain catastrophizing, and sleep disturbance (p < 0.05). However, self-reported sleep disturbance was not significantly correlated with actigraphic sleep duration (|r| 0.05). Sleep and pain variables observed at prior assessments predicted these same variables at subsequent assessments. There was no significant evidence that changes in pain preceded changes in sleep or that changes in sleep preceded changes in pain (all p > 0.05).Discussion: For this study’s Veterans, treatment-related changes in sleep and pain appeared to occur in parallel. The concomitant changes in sleep and pain suggest that therapies improving pain in Veterans may yield attendant benefits for the treatment of sleep, and possibly vice versa.Keywords: chronic pain, complementary and integrative treatments, sleep, Veterans

Autophagy plays a vital role in cell homeostasis by eliminating nonfunctional components and promoting cell survival. Here, we examined the levels of autophagy signaling proteins after 7 days of overload hypertrophy in the extensor digitorum longus (EDL) and soleus muscles of control and diabetic rats. We compared control and 3‐day streptozotocin‐induced diabetic rats, an experimental model for type 1 diabetes mellitus (T1DM). EDL muscles showed increased levels of basal autophagy signaling proteins. The diabetic state did not affect the extent of overload‐induced hypertrophy or the levels of autophagy signaling proteins (p‐ULK1, Beclin‐1, Atg5, Atg12‐5, Atg7, Atg3, LC3‐I and II, and p62) in either muscle. The p‐ULK‐1, Beclin‐1, and p62 protein expression levels were higher in the EDL muscle than in the soleus before the hypertrophic stimulus. On the contrary, the soleus muscle exhibited increased autophagic signaling after overload‐induced hypertrophy, with increases in Beclin‐1, Atg5, Atg12‐5, Atg7, Atg3, and LC3‐I expression in the control and diabetic groups, in addition to p‐ULK‐1 in the control groups. After hypertrophy, Beclin‐1 and Atg5 levels increased in the EDL muscle of both groups, while p‐ULK1 and LC3‐I increased in the control group. In conclusion, the baseline EDL muscle exhibited higher autophagy than the soleus muscle. Although TDM1 promotes skeletal muscle mass loss and strength reduction, it did not significantly alter the extent of overload‐induced hypertrophy and autophagy signaling proteins in EDL and soleus muscles, with the two groups exhibiting different patterns of autophagy activation.

The global expansion of aquaculture has driven significant technological advancements, including raceways, Integrated Multitrophic Aquaculture (IMTA) and marine offshore cages. However, unregulated aquaculture escapees pose a severe threat to aquatic biodiversity, acting as a potential time bomb for the entire ecosystem. Addressing this issue requires a comprehensive understanding of the impact of unintentional escapees on aquatic ecosystems, particularly in India and riparian East African countries (Kenya, Uganda, and Tanzania). Using an exploratory research design drawing from various peer-reviewed sources, this study outlines the dynamic growth of aquaculture in these countries, identifies high-impact escape incidents, and correlates risks and remedies with global cases, especially in regions such as Norway, Scotland, Ireland, Colombia, and the United States, where serious incidents of aquaculture escapees have been reported. The research categorizes aquaculture development trends, discusses mechanisms of escapee impact, proposes remedies, assesses methods and inferential strength, and highlights gaps in the existing literature. The study revealed complex ecological shifts caused by aquaculture escapees from invasive non-native fish species, affecting predation, competition, and genetic diversity. Escaped fish from aquaculture facilities pose a significant threat to aquatic biodiversity, especially in the study regions. The escalating risk of unintentional escapes was highlighted in India and three East African countries. To mitigate this, the study proposes integrating escapee management into national fisheries systems, amending fisheries laws, holding fish farmers accountable for aquaculture system failure, and developing comprehensive regulations for non-native species in aquaculture within the study regions. It is recommended to standardize the planning for aquaculture facilities and implement emergency plans, training, local mobilization and further research on the impact thresholds of aquaculture escapees in the study regions. Ecological education in aquaculture communities and the recognition of the role of translational scientists are crucial for the dissemination of knowledge. Urgent government action is needed to address unreported aquaculture escapes, preventing further ecosystem degradation and ensuring global aquaculture sustainability.

Live fish are in high demand all over the world, especially in China, where quality standards align with consumer preferences. In Kenya, discussions on live fish, particularly in aquaculture-rich regions, are focal points. However, the lack of a systematic approach to transporting live fish poses challenges for stakeholders. This research explores attitudes and perceptions of stakeholders in Kenya regarding the adoption and transportation of live fish, shedding light on transportation procedures and sector challenges. Using a descriptive research design, primary data were collected from diverse aquaculture stakeholders through a questionnaire, addressing their knowledge, perceptions, and attitudes toward live fish transportation in the country. The study unveils varied patterns of live fish transportation in Kenya, influenced by the purpose and distance. Private cars are prominent at 26.3%, with Nyanza and Mombasa leading at 35%. Modified vans are prevalent in Central-Nairobi and Western regions (25% and 22.5%, respectively), while public transport is widespread, especially in Central and Nairobi (25%). Purpose-built live fish vehicles dominate in Western and Nyanza (20% and 17.5%, respectively). Crustaceans like lobsters and crabs are primarily exported from the Coastal region (27.5%). Participants generally assessed their knowledge of live fish transportation as moderate, with a basic understanding of fish welfare. Feedback indicates common practices in live fish transport in Kenya as sealed tanks, plastic bags, and purpose-built vehicles. Temperature and oxygen fluctuations poses a significant challenge during transportation across all regions, particularly in Mombasa and Kisumu. The use of anaesthetics, especially for food fish, is infrequently reported. The study revealed favourable perceptions of stakeholders to live fish transportation and welfare, indicating early adoption. Further research is recommended on fish welfare, best management practices, technological advancements, and interdisciplinary studies to enhance the sustainability of live fish transportation sector and fish welfare in Kenya.

The tuning of the luminescent properties of PtII complexes for possible use in organic light-emitting diodes (OLEDs) and sensing applications is commonly achieved by altering the electronic properties of the ligands. Our group recently demonstrated that the trifluoropropynyl ligand is strongly electron-withdrawing and possibly useful for blueshifting emission. Herein, we report the synthesis of two complexes of this trifluoropropynyl ligand, namely PtLC2CF3 and PtLFC2CF3 (L = 1,3-di(2-pyridyl)benzene; LF = 4,6-difluoro-1,3-di(2-pyridyl)benzene). The PtLC2CF3 complex crystallized in the monoclinic space group P21/n with Z = 4. The PtLFC2CF3 complex crystalized in the triclinic space group P-1 with Z = 2. Changing the tridentate ligand from L to LF resulted in a change in the packing structure, with the latter showing a metallophilic interaction (Pt-Pt distance = 3.3341(3) Å). The solution photophysics of the trifluoropropynyl complexes is compared with that of the corresponding Cl complexes, PtLCl and PtLFCl. Replacement of the chloro ligand with the trifluoropropynyl ligand blueshifted the monomer emission by less than 5 nm but blueshifted the excimer emission peaks by 15–20 nm. The complexes of the trifluoropropynyl ligand also favor the excimer emission more than the complexes of the chloro ligand. The excimer emission is quenched by dissolved oxygen significantly more than the corresponding monomer emission. The excimer emission and monomer emission are well separated, and the ratio of monomer to excimer emission is strongly dependent on oxygen concentration.

The special issue on Biomarkers of Nicotine and Tobacco Dependence reviews the science for precision treatment of nicotine dependence and future opportunities for research on biomarkers for inclusion in tobacco product cessation and switching clinical trials to advance translation. This overview summarizes the articles contributed to the special issue by leading researchers in field of addiction.

Electronic devices such as CCTVs and GPS trackers are becoming more prevalent in public utility vehicles. These electronic devices produce data that can be aggregated and processed to generate insights regarding the service quality provided by public transportation systems. We describe a methodology for processing CCTV-produced videos and GPS logs, and utilize it to perform a first order, limited assessment of the Philippines’ Service Contracting Program, which ran during the COVID-19 pandemic. Our analyses uncovered vast discrepancies between data-supported figures and manually-generated estimates provided by the transport operators, especially with regards to vehicle occupancy and total number of commuters served. Our study also uncovered imperfections in the data generated by the electronic devices, which place caveats or limitations on the insights generated by the data analyses. We provide and suggest ways on how the analyses can be modified to take into account such missing data, thus enabling us to come up with educated, reasonable insights despite incomplete input datasets.

Best-corrected visual acuity (BCVA), optical coherence tomography (OCT) at baseline and final visit, and drug-related adverse events were analyzed. Eyes with recurrent macular fluid on IVBr every 8 weeks were treated with a combination therapy alternating between IVBr and aflibercept every month.Results: Among 52 eyes (40 patients) on IVBr, all had been previously treated with other anti-VEGF therapy, with 73% having persistent macular fluid. After a mean follow-up of 46.2± 27.4 weeks on IVBr, the mean treatment interval for intravitreal therapy increased to 8.8± 2.1 weeks on IVBr from a baseline of 6.1± 3.1 weeks (p< 0.001). Macular fluid decreased and BCVA was stable/improved in 61.5% of eyes on IVBr. Ten eyes with increased macular fluid on IVBr monotherapy when extended to every 8 weeks were treated with combination therapy alternating between IVBr and aflibercept every 4 weeks. In these eyes, 80% had improved macular fluid on OCT and 70% stable or improved BCVA after a median follow-up of 53 weeks on combination therapy. Mild intraocular inflammation developed in four eyes, all occurring on IVBr monotherapy, and none had associated vision loss.Conclusion: In the real world, IVBr used to treat eyes with nAMD previously treated with other anti-VEGF therapies appears to be well tolerated and associated with an improvement in macular fluid, stabilization of BCVA, and/or increase in intravitreal treatment interval. Combination therapy alternating between IVBr and aflibercept monthly appears to be well tolerated and can be considered for eyes with macular fluid on IVBr every 8 weeks.Keywords: age-related macular degeneration, aflibercept, anti-VEGF, brolucizumab, intravitreal injection; optical coherence tomography

The presence of the non-native Nile perch (Lates niloticus) in Lake Victoria and the resulting ecological transformations have garnered significant attention from the scientific community. However, details regarding the timing, origin, and purpose of the fish introduction have remained elusive. This paper reviews fish fauna changes in Lake Victoria, investigates causes, and advocates for biodiversity conservation through diversity and sanctuary sites. It examines different historical periods to understand changes and guide effective conservation strategies. The introduction of Nile perch has led to a substantial decline and even extinction of many native fish species, particularly cichlids, resulting in ecological imbalances and economic challenges for local fishing communities. To address these pressing issues, ongoing conservation efforts are being implemented. These initiatives focus on promoting sustainable fishing practices, safeguarding critical habitats, and reintroducing native fish species. Furthermore, cage fish farming is being explored as a potential solution to alleviate pressure on wild populations, as suggested by various authors. Recommendations stemming from this study include conducting virtual population analysis for stock assessments of indigenous species, intensify conservation efforts, diversify fishing practices, expand cage fish farming, increase public awareness and education, and enhance policy and governance frameworks. By implementing these recommendations, there is optimism for the recovery and resilience of indigenous fish species in Lake Victoria, leading to the conservation of biodiversity and the promotion of sustainable livelihoods for local communities.

Abstract Background We examined the association of non-cigarette tobacco use on chronic obstructive pulmonary disease (COPD) risk in the Population Assessment of Tobacco and Health (PATH) Study. Methods There were 13,752 participants ≥ 40 years with Wave 1 (W1) data for prevalence analyses, including 6945 adults without COPD for incidence analyses; W1–5 (2013–2019) data were analyzed. W1 tobacco use was modeled as 12 mutually-exclusive categories of past 30-day (P30D) single and polyuse, with two reference categories (current exclusive cigarette and never tobacco). Prevalence and incidence ratios of self-reported physician-diagnosed COPD were estimated using weighted multivariable Poisson regression. Abstract Background There are currently no global recommendations on a parsimonious and robust set of indicators that can be measured routinely or periodically to monitor quality of hospital care for children and young adolescents. We describe a systematic methodology used to prioritize and define a core set of such indicators and their metadata for progress tracking, accountability, learning and improvement, at facility, (sub) national, national, and global levels. Methods We used a deductive methodology which involved the use of the World Health Organization Standards for improving the quality-of-care for children and young adolescents in health facilities as the organizing framework for indicator development. The entire process involved 9 complementary steps which included: a rapid literature review of available evidence, the application of a peer-reviewed systematic algorithm for indicator systematization and prioritization, and multiple iterative expert consultations to establish consensus on the proposed indicators and their metadata. Results We derived a robust set of 25 core indicators and their metadata, representing all 8 World Health Organization quality standards, 40 quality statements and 520 quality measures. Most of these indicators are process-related (64%) and 20% are outcome/impact indicators. A large proportion (84%) of indicators were proposed for measurement at both outpatient and inpatient levels. By virtue of being a parsimonious set and given the stringent criteria for prioritizing indicators with “quality measurement” attributes, the recommended set is not evenly distributed across the 8 quality standards. Conclusions To support ongoing global and national initiatives around paediatric quality-of-care programming at country level, the recommended indicators can be adopted using a tiered approach that considers indicator measurability in the short-, medium-, and long-terms, within the context of the country’s health information system readiness and maturity. However, there is a need for further research to assess the feasibility of implementing these indicators across contexts, and the need for their validation for global common reporting.

The present study evaluates the quality of life of 1,139 surviving victims of the Colombian armed conflict benefited from some actions of reparation, through a brief sociodemographic and victimological survey and the WHO quality of life (WHOQOL-BREF) questionnaire, which takes into account perceived physical and mental health, social relations, and environmental factors. The results show that the quality of life of the victims of the armed conflict, once peace agreements had been reached, remained low or medium-low. The number of victimizing events suffered and the time elapsed have little effect on the quality of life, with type of trauma being more relevant. Land-related experiences, such as expropriation, had fundamentally an influence on environmental factors. Kidnappings, sexual assaults, and events where minors were involved affected mental health. Victims of sexual assault were those whose quality of life was most affected and torture victims were those with a lower index of global quality of life. Simple passing of time did not mean quality of life increase with specific interventions being needed to repair damage. To repair the damage caused, special attention should be paid to the sociodemographic characteristics of each victim, the type of aggression suffered, and their needs in the areas of physical and mental health, and economic resources.

Aim of study: To evaluate which vegetable lipid source promotes better growth performance, whole-body composition and fatty acid profile for juvenile giant trahira (Hoplias lacerdae). Area of study: Fish Nutrition Laboratory of the University of Viçosa (UFV), MG, Brazil. Material and methods: A 50-day feed trial with four treatments, consisting of diets containing different vegetable lipid sources (canola, linseed, soybean or olive oil), was conducted with juveniles of 4.76 ± 0.50 cm and 1.97 ± 0.20 g. Main results: There were no effects of vegetable lipid sources on growth performance. Fish fed diets containing canola oil had higher body lipid deposition and fish fed with linseed oil had lower body lipid content (up to -19.29%) than fish from other treatments. Fish fed canola oil showed lower proportions of saturated fatty acids (up to -11.27%) in the body. Fish fed diets containing soybean oil and linseed oil showed the highest percentages of linoleic and α-linolenic fatty acids, respectively. Fish fed diets containing soybean and linseed oils also had higher total polyunsaturated fatty acids content (up to +81.14%). Fish fed diets containing linseed oil had lower content of monounsaturated fatty acids (up to -58.59%) and higher content of docosahexaenoic (up to +175%) and eicosapentaenoic (not detectable to detectable) acids. Research highlights: Juveniles of giant thraira can alter the whole-body fatty acid profile due to their ability to desaturate and elongate the n3 and n6 series fatty acids. Linseed oil was identified as lipid source for this fish species.

There is increasing awareness among researchers and health practitioners from high income countries about the potential mental health benefits of participating in gardening activities and spending substantial time in green spaces. However, this phenomenon is not well established in low- and middle-income countries. In this commentary, we discuss the evidence base surrounding the potential mental health benefits of participating in gardening activity and spending substantial time in a green space. We hope to stimulate discourse about incorporating these activities into mental health prevention in low- and middle-income countries.

The erratic supply of electricity in Nigeria has renewed interest in the utilisation of coal for energy self-sufficiency and diversification of the national energy mix. In this study, selected coal samples from three regions of the Benue Trough Basin were examined through pore water chemistry, X-ray diffraction (XRD), and thermosgravimetric analysis (TGA). The low pH observed for the Lower Benue Trough (LBT) coals indicates the release of sulphur and acids during oxidation. Mineralogical analyses reveal quartz, kaolinite, montmorillonite, albite, pyrite, gypsum and sodium chlorate in the coal samples. The genetic kaolinite species identified in Imeagha (IMG) coal are detrital kaolinite and the highly crystalline neomorphic kaolinite. Gypsum and jarosite minerals occurred in minor quantities in Garin Maiganga (GMG), whereas significant amount of pyrite was detected in Enugu (ENG) coal. Oxidative thermal degradation resulted in the residual mass (RM) ranging between 4.35% and 56.96%) and mass losses (ML) between 43.04% and 95.65%) due to the drying, devolatilization, and coke formation in the samples. The most reactive coal examined was GMG coal, whereas the least reactive was IMG coal. Lamja (LMZ), GMG, and OKB coals are suited for electricity generation, whereas ENG and IMG coals can be utilised for cement production.

Background: The role of splenectomy to diagnose and treat hematologic disease continues to evolve. In this single-center retrospective review, we describe modern morbidity, mortality, and long-term outcomes associated with splenectomy for benign and malignant hematologic disorders. Methods: We analyzed all nontrauma splenectomies performed for benign or malignant hematologic disorders from January 2009 to September 2018. Variables collected included demographics, preexisting comorbidities, laboratory results, intra- and postoperative features, and long-term follow-up. Outcomes of interest included postoperative complications, 30-day mortality, and overall mortality. Results: We identified 161 patients who underwent splenectomy for hematologic disorders. Median age was 54 years (range 19–94), and 83 (52%) were female. Splenectomy was performed for 95 (59%) patients with benign hematologic disorders and for 66 (41%) with malignant conditions. Most splenectomies were laparoscopic (76%), followed by laparoscopic hand assisted (11%), open (8%), and laparoscopic converted to open (6%). Median follow-up was 761 days (interquartile range: 179–2025 days). Major complications occurred in 21 (13%) patients. Three (2%) patients died within 30 days; 16 (9%) died more than 30 days after operation, none from surgical complications, with median time to death of 438 days (interquartile range: 231–1497 days). Among malignant cases, only preoperative thrombocytopenia predicted death (odds ratio = 5.8, 95% confidence interval = 1.1–31.8, P = .04). For benign cases, increasing age was associated with inferior survival (odds ratio = 2.3, 95% confidence interval = 1.0–5.1, P = .05). Conclusion: Splenectomy remains an important diagnostic and therapeutic option for patients with benign and malignant hematologic disorders and can be performed with a low complication rate. Despite considerable burden of comorbid disease in these patients, early postoperative mortality was uncommon.

Abstract Background Endogenous phospholipases A2 (PLA2) play a fundamental role in inflammation, neurodegenerative diseases, apoptosis and cellular senescence. Neurotoxins with PLA2 activity are found in snake venoms from the Elapidae and Viperidae families. The mechanism of action of these neurotoxins have been studied using hippocampal and cerebellar neuronal cultures showing [Ca2+]i increase, mitochondrial depolarization and cell death. Astrocytes are rarely used as a model, despite being modulators at the synapses and responsible for homeostasis and defense in the central nervous system. Preserving the cell division ability, they can be utilized to study the cell proliferation process. In the present work cultured astrocytes and glioblastoma cells were employed to characterize the action of β-micrustoxin (previously named Mlx-9), a PLA2 isolated from Micrurus lemniscatus snake venom. The β-micrustoxin structure was determined and the cell proliferation, cell cycle phases and the regulatory proteins p53, p21 and p27 were investigated. Methods β-micrustoxin was characterized biochemically by a proteomic approach. Astrocytes were obtained by dissociation of pineal glands from Wistar rats; glioblastoma tumor cells were purchased from ATCC and Sigma and cultured in DMEM medium. Cell viability was evaluated by MTT assay; cell proliferation and cell cycle phases were analyzed by flow cytometry; p53, p21 and p27 proteins were studied by western blotting and immunocytochemistry. Results Proteomic analysis revealed fragments on β-micrustoxin that aligned with a PLA2 from Micrurus lemniscatus lemniscatus previously identified as transcript ID DN112835_C3_g9_i1/m.9019. β-micrustoxin impaired the viability of astrocytes and glioblastoma tumor cells. There was a reduction in cell proliferation, an increase in G2/M phase and activation of p53, p21 and p27 proteins in astrocytes. Conclusion These findings indicate that β-micrustoxin from Micrurus lemniscatus venom could inhibit cell proliferation through p53, p21 and p27 activation thus imposing cell cycle arrest at the checkpoint G2/M.

Declining soil fertility is one of the major causes of food insecurity and high levels of poverty, both of which tend to hamper economic development in sub-Saharan Africa (SSA). To improve soil fertility, the implementation of soil organic carbon (SOC) enhancement technologies has become crucial to slowing land degradation, through increasing SOC, which is the basis of soil fertility. Using data from 381 households from Azuga-Suba and Yesir watersheds in Ethiopia, this study explores the extent of the adoption of technologies that enhance SOC. Soil organic carbon enhancing technologies include the use of manure, fertilizer, and crop residue management. The Probit model was used to assess what constrains the adoption of these technologies. The results indicate that fertilizer is the most adopted technology having over 90% adoption in both watersheds. Manure at 28% and 56% adoption while crop residue management at 37% and 26% adoption in Azuga-Suba and Yesir respectively. Technology adoption is highly constrained by lack of education, access to extension services, and access to credit services. Institutions and local farmer groups influence these constraints through training, provision of information, offering incentives, and credit services. Large plots hinder the use of manure and fertilizer due to the bulky nature of manure and the high costs of fertilizers. Insecurity in land tenure limits the adoption of manure and residue management. Perception of soil erosion and soil fertility tends to constrain the adoption of SOC technologies, as farmers are afraid that all improvements through soil amendment will be diminished through soil erosion. At the same time, farmers do not perceive the importance of SOC enhancing technologies in plots that were fertile. These results imply that strengthening institutions that enhance farmers’ knowledge and provide credit as well as strengthening social protection schemes and farmer groups is crucial in promoting the adoption of these technologies.

Evidence from high-income settings suggests that gardening is associated with reductions in depression, anxiety, and stress. The benefits of gardening are less well understood by mental health practitioners and researchers from low- and middle-income countries. Our study estimated the association between participation in gardening and symptoms of depression, anxiety, and stress among caregivers of people living with dementia in rural, southwestern Uganda. In a cross-sectional study, we interviewed 242 family caregivers of people with dementia to elicit their gardening activities; symptoms of depression, anxiety, and stress (Depression Anxiety Stress Scales); and caregiving burden (Zarit Burden Interview). Linear multivariable regression models estimated the association between participation in gardening and symptoms of depression, anxiety, and stress. Out of 242 participants, 131 (54%) caregivers were involved in gardening. In this study, trimetal Mg/Ce/Mn oxide-modified diatomaceous earth (DE) was synthesized at optimal conditions. Comparison of the SEM images and the results of EDX analyses of the raw and the modified DE confirmed the surface modification of the raw DE with the trimetal oxide. Groundwater fluoride removal capacity of the sorbent was evaluated by batch method at various defluoridation conditions. At a sorbent dosage of 0.6 g/100 mL (contact time: 60 min, mixing speed of 200 rpm and temperature: 297 K), the fluoride removal was >93% for solutions containing initial fluoride concentration of 10–60 mg/L. Sorbent’s optimum fluoride uptake capacity was 12.63 mg/g at the initial fluoride concentration of 100 mg/L. Fluoride removal was >91% for solutions with initial pH range of ∼4–11 (initial fluoride concentration: 9 mg/L, sorbent dosage: 0.6 g/100 mL). Appraisal of the effect of co-existing anions on fluoride removal showed that CO32− would reduce the amount of fluoride removed from solution, while other anions such as PO43−, NO3− and SO42− had no observable effect. K2SO4 solution was found to be most suitable for regeneration of spent Mg/Ce/Mn oxide-modified DE compared to Na2CO3 and NaOH. The mechanism of fluoride removal at pH > 5.45 (pHpzc = 5.45) occurred by exchange of hydroxyl groups on surface of sorbent with fluoride ions from solution. Sorption data fitted better to Langmuir isotherm and pseudo-second-order model. External diffusion was observed to be the sorption rate limiting factor. This study was to investigate the effect of dietary supplementation with xylanase and protease on growth performance, digesta viscosity, apparent ileal digestibility (AID) of nutrients, and gut health in nursery pigs. Forty-eight pigs (24 barrows and 24 gilts at 21 d of age with 7.2 ± 0.4 kg BW) were randomly allotted to 4 dietary treatments (2 × 2 factorial arrangement) in a randomized complete block design and fed in 2 phases (phase 1 for 10 d and phase 2 for 14 d). The establishment of the Morotai Island Regency as one of the 10 National Tourism Strategic Areas and Special Economic Zones is a central government strategy to accelerate the development of regencies/cities in North Maluku Province, which are still classified as underdeveloped regions in Eastern Indonesia. This study aims to analyze the influence of tourist arrival rate, price, human development index, and tourism promotion policy on economic growth by using regression of panel data of fixed-effect model (FEM) based on the feasible generalized least square (FGLS) VI method in eight regencies/cities in North Maluku Province during the period of 2012–2017. This finding shows that tourism development was closely related to economic growth, both in the short and long term. However, the development of tourism facilities and other supporting tourism facilities on the coast by coastal landfill had sacrificed the growth of coastal mangrove forests. Therefore, the local government is encouraged to issue regional regulations on the implementation of sustainable tourism businesses and educate the local community and visitors to play a role in protecting the environment of tourist destinations sustainable.

This paper presents a chemical characterization of fine particulate matter in air masses passing through Thohoyandou and further determines their sources. Fine particulate matter (PM2.5) was collected and quantified using gravimetric method. X-ray fluorescence, Smoke stain reflectometer, Optical Transmissometer and Scanning Electron Microscopy- Energy Dispersive X-Ray Spectroscopy were used to determine the chemical and morphological composition of the particulate matter. The source apportionment was done using principal component analysis while the HYSPLIT model was used to depict the long-range transport clusters. The mean of concentrations of PM2.5, soot, black carbon and UVPM were 10.9 μg/m3, 0.69x10-5 m-1, 1.22 μg/m3 and 1.40 μg/m3, respectively. A total of 24 elements were detected in the PM2.5 with Pd, Sn, Sb, Mg, Al and Si being the dominant elements. SEM-EDS have shown the presence of irregular, flat and spherical particles which is associated with crustal material and industrial emissions. Source apportionment analysis revealed six major sources of PM2.5 in Thohoyandou namely, crustal materials, industrial emissions, vehicular emissions, urban emissions, fossil fuel combustion and fugitive-Pd. Air parcels that pass-through Thohoyandou were clustered into four. The major pathways were from the SW Indian Ocean, Atlantic Ocean and inland trajectories. Clusters from the ocean are associated with low concentration, while inland clusters are associated with high concentration of PM2.5. The PM2.5 occasionally exceeds the WHO daily guideline in Thohoyandou and the sources of PM2.5 extend beyond the borders. This study recommends that further studies need to be carried out to assess the health impacts of PM2.5 in Thohoyandou.

This article aims to describe, in depth, the experiences of migrant workers from China in maintaining their livelihoods in Timor-Leste through entrepreneurial activities before and during the COVID-19 pandemic. Specifically, this study discusses sustainable livelihoods with the knowledge of migration, adaptation, and entrepreneurship in Timor-Leste. To fulfill these aims, a qualitative methodology with a phenomenological approach was adopted by interviewing eight Chinese migrants who run businesses in Timor-Leste. Furthermore, the framework for the concept of sustainable livelihoods of the Department for International Development (DFID) was adapted to explain, in depth, the phenomena. The finding reveals that economic reasons encourage and attract Chinese migrant workers and entrepreneurs to Timor-Leste. The adaptation process of migrants in Timor-Leste relies on social capital to synergize with local communities in terms of culture, further affecting the comfort and security of entrepreneurship. From the perspective of sustainable livelihoods, migrant workers take advantage of human capital in the process of accumulation of financial capital to enhance their business activities. Meanwhile, migrant entrepreneurship expands their financial capital and human capital into social capital, physical capital, and natural capital. The findings also explain that the mobility and access to financial capital of Chinese migrants are supported through institutional structures, entrepreneurship activities, and policies. Furthermore, to ensure livelihood sustainability and business, Chinese migrants in Timor-Leste can carry out business intensification, extensification, diversification, and transformation strategies.

Abstract Background We have recently demonstrated that palmitoleic acid (16:1n7) increases lipolysis, glucose uptake and glucose utilization for energy production in white adipose cells. In the present study, we tested the hypothesis that palmitoleic acid modulates bioenergetic activity in white adipocytes. Methods For this, 3 T3-L1 pre-adipocytes were differentiated into mature adipocytes in the presence (or absence) of palmitic (16:0) or palmitoleic (16:1n7) acid at 100 or 200 μM. The following parameters were evaluated: lipolysis, lipogenesis, fatty acid (FA) oxidation, ATP content, oxygen consumption, mitochondrial mass, citrate synthase activity and protein content of mitochondrial oxidative phosphorylation (OXPHOS) complexes. Results Treatment with 16:1n7 during 9 days raised basal and isoproterenol-stimulated lipolysis, FA incorporation into triacylglycerol (TAG), FA oxidation, oxygen consumption, protein expression of subunits representing OXPHOS complex II, III, and V and intracellular ATP content. These effects were not observed in adipocytes treated with 16:0. Conclusions Palmitoleic acid, by concerted action on lipolysis, FA esterification, mitochondrial FA oxidation, oxygen consumption and ATP content, does enhance white adipocyte energy expenditure and may act as local hormone.

Aims: The aim is to assess for a difference in the incidence of metastasis (IM) and prostate-specific antigen (PSA) levels at diagnosis in patients with Gleason score (GS) 3+4 versus 4+3 prostate cancer using a large veterans affairs database. Subjects and Methods: A retrospective review of 1402 medical records from 5 VA hospitals was conducted. The study period was from 2009 to 2014. Primary endpoints were IM and PSA levels at diagnosis. A secondary endpoint was overall survival. Statistical Analysis Used: Chi-square tests for categorical variables, Student's t-test for continuous, normally distributed variables, and rank sum tests for continuous nonnormally distributed variables. Results: There were 1050 patients with GS3+4 and 352 with GS4+3. There were no differences in sociodemographic and clinical characteristics of the study population. PSA at the time of diagnosis was significantly higher in the GS4+3 patients compared to GS3+4 (18.0 vs. 11.4, respectively; P < 0.001). The IM at diagnosis was higher in the GS4+3 patients (10/352) compared to GS3+4 (9/1041) (2.8% vs. 0.9%; P = 0.005). In an adjusted model, GS4+3 was associated with higher PSA, higher IM at diagnosis. There was no difference in overall survival between the 2 groups though a 23% reduction in overall survival in the GS4+3 was noted (P = 0.53). Conclusions: Our results indicate that patients with GS4+3 prostate cancers have higher PSA levels at diagnosis. GS4+3 is associated with 3-fold increased risk of IM at diagnosis than GS3+4 though the overall incidence is low. Further research is needed to assess whether GS4+3 patients need routine staging imaging investigations at the time of diagnosis similar to patients with higher Gleason scores (GS ≥8).

ABSTRACT Introduction: The detection of autoantibodies in HEp-2 cells represents a relevant tool for the diagnosis of autoimmune diseases, especially rheumatic autoimmune diseases. As a result of the methodological advances, the technique gradually increased the sensitivity, as well as the need for standardization. Objective: To evaluate the implementation of the Brazilian Consensus recommendations for autoantibody determination in HEp-2 cells. Methods: A structured form in a virtual platform was filled in by experts in clinical laboratories that carry out the methodology across the country. The questionnaire addressed the adoption of the Brazilian Consensus guidelines, detailing the technical aspects, quality control, the strategy for reading slides and the release of reports. Results: The study included 53 laboratories responsible for more than 300,000 antinuclear antibody (ANA) tests/month; more than half (58.5%) reported fully adopting the recommendations of the Brazilian Consensus. The majority (83.1%) used the 1:80 for screening dilution, and 75.5% of laboratories, perform education and quality control programs. Only 39.6% reported using more than one kit brand to perform the test, and 32.1% did not report observing all phases of the cell cycle during slide reading. The study also detected some heterogeneity among participants in the identification of patterns. Conclusion: The results confirm the adoption of the Brazilian Consensus recommendations by most of participating laboratories, although with variable extent. There is need for improvement in some aspects, especially those related to the quality control.

Abstract The study synthesizes a low-cost adsorbent made from Mn2+-modified bentonite clay for groundwater defluoridation. The clays were characterized using X-ray diffraction, X-ray fluorescence, scanning electron microscopy, and Fourier transform infrared techniques. The fluoride adsorption capacity of the modified clay was evaluated using batch experiments. The adsorption kinetics results showed that the optimum fluoride (F−) uptake was achieved within the 30 min’ contact time. The data fitted well to pseudo-second-order of reaction kinetics indicating that adsorption of F− occurred via chemisorption. In addition, the adsorption isotherm data fitted well to Langmuir isotherm model indicating that adsorption occurred on a mono-layered surface. Maximum F− removal of 57% was achieved from groundwater with an initial F− concentration of 5.4 mg L−1 and natural pH of 8.6 using adsorbent dosage of 1 g/100 mL. Fluoride adsorption occurred through ligands and ion exchange mechanisms. The synthesized adsorbent was successfully regenerated for up to five times. The study shows that Mn2+-intercalated bentonite clay has potential for application in defluoridation of groundwater.

Cigarette abstinence for 12+ months at W3 was ~10% in each group. Half of the cigarette abstainers in the e-cigarette group were using e-cigarettes at W3. Different methods to help quitting had statistically comparable 12+ month cigarette abstinence at W3 (e-cigarettes vs no product: Risk Difference (RD) = 0.01, 95% CI: -0.04 to 0.06; e-cigarettes vs pharmacotherapy: RD = 0.02, 95% CI:-0.04 to 0.09). Likewise, daily e-cigarette users at W2 did not show a cessation benefit over comparable no-e-cigarette users and this finding was robust to sensitivity analyses. Abstinence for 30+ days at W3 was also similar across products.LimitationsThe frequency of e-cigarette use during the LQA was not assessed, nor was it possible to assess continuous abstinence from the LQA.ConclusionAmong US daily smokers who quit cigarettes in 2014-15, use of e-cigarettes in that attempt compared to approved cessation aids or no products showed similar abstinence rates 1-2 years later.

Summary: Receptor-interacting protein kinase 1 (RIPK1) and 3 (RIPK3) are well known for their capacity to drive necroptosis via mixed-lineage kinase-like domain (MLKL). Recently, RIPK1/3 kinase activity has been shown to drive inflammation via activation of MAPK signaling. However, the regulatory mechanisms underlying this kinase-dependent cytokine production remain poorly understood. In the present study, we establish that the kinase activity of RIPK1/3 regulates cytokine translation in mouse and human macrophages. Furthermore, we show that this inflammatory response is downregulated by type I interferon (IFN) signaling, independent of type I IFN-promoted cell death. Specifically, low-level constitutive IFN signaling attenuates RIPK-driven activation of cap-dependent translation initiation pathway components AKT, mTORC1, 4E-BP and eIF4E, while promoting RIPK-dependent cell death. Altogether, these data characterize constitutive IFN signaling as a regulator of RIPK-dependent inflammation and establish cap-dependent translation as a crucial checkpoint in the regulation of cytokine production. : Balancing inflammatory responses is critical for host survival. Muendlein et al. show that constitutive type I IFN signaling inhibits translation machinery activated downstream of the kinase activity of RIPK1/3, preventing the production of a subset of inflammatory cytokines. This work identifies cap-dependent translation as a checkpoint in regulation of RIPK1/3-kinase-dependent inflammation. Abandoned gold mine sites are generally characterised by severe environmental problems and physical hazards. Because of socio-economic problems confronting communities around abandoned mine sites, historic and abandoned gold mines have become hot-spots for artisanal and small-scale miners. These mining activities at times thwart the efforts of rehabilitation at these sites. This article details how artisanal mining operations have frustrated rehabilitation efforts of abandoned mine shafts in the Sutherland goldfield. The field investigation of abandoned shafts and analysis of the nature of artisanal mining operations in the Sutherland goldfield revealed that artisanal mining involving digging around collars of sealed shafts is a major threat to the stability of the shafts and their sealing structures. In addition, artisanal mining operations have increased the safety risks of the abandoned shafts in the area. This has also been worsened by the fact that a large number of people, especially women and children, are exposed to the hazards of the abandoned mine sites. This article emphasises an urgent need for the development of holistic and cohesive strategies for dealing with the problems of abandoned gold mine shafts wherever they exist in the country as opposed to simply closing them up.

In the present study, silver nanoparticles (AgNPs) were synthesized using both the chemical and biological methods and conjugated with Pyrenacantha grandiflora extracts. These were then characterized and evaluated for antimicrobial activities against multi-drug resistant pathogens, such as methicillin-resistant Staphylococcus aureus (MRSA), Klebsiella pneumonia, and Escherichia coli. Nanoparticles were analyzed with UV-visible spectrophotometer, transmission electron microscopy (TEM), and energy dispersive X-ray analysis (EDX). Silver nanoparticles, P. grandiflora extracts, and the conjugates were also analyzed with Fourier transform infrared spectroscopy (FTIR). As a result, quasi-sphere-shaped AgNPs with sizes ranging from 5 to 33 nm and spherically shaped AgNPs with sizes ranging from 3 to 25 nm were formed from chemical and biological synthesis, respectively. A well diffusion assay showed that the activity of silver nanoparticles was most improved with acetone extract against all tested bacteria with diameters in the range of 19–24 mm. The lowest MIC value of 0.0063 mg/mL against MRSA was observed when biologically synthesized AgNPs were conjugated with acetone and water extracts. Chemically synthesized silver nanoparticles showed the lowest MIC value of 0.0063 mg/mL against E. coli when conjugated with acetone and methanol extracts. This study indicates that silver nanoparticles conjugated with P. gandiflora tubers extracts exhibit strong antibacterial activities against multi-drug resistant bacterial pathogens. Therefore, biosynthesized conjugates could be utilized as antimicrobial agents for effective disease management due to the synergistic antibacterial activity that was observed.
