Abstract Background The cell development atlas of transition stage from late Carnegie to fetal development (7–9 weeks) remain unclear. It can be seen that the early period of human embryos (7–9 weeks) is a critical research gap. Therefore, we employed single‑cell RNA sequencing to identify cell types and elucidate differentiation relationships. Results The single‑cell RNA sequencing analysis determines eighteen cell clusters in human embryos during the 7–9 weeks period. We uncover two distinct pathways of cellular development and differentiation. Initially, mesenchymal progenitor cells differentiated into osteoblast progenitor cells and neural stem cells, respectively. Neural stem cells further differentiated into neurons. Alternatively, multipotential stem cells differentiated into adipocyte, hematopoietic stem cells and neutrophil, respectively. Additionally, COL1A2-(ITGA1 + ITGB1) mediated the cell communication between mesenchymal progenitor cells and osteoblast progenitor cells. NCAM1-FGFR1 facilitated the cell communication between mesenchymal progenitor cells and neural stem cells. Notably, NCAM1-NCAM1 as a major contributor mediated the cell communication between neural stem cells and neurons. Moreover, CGA-FSHR simultaneously mediated the communication between multipotential stem cells, adipocyte, hematopoietic stem cells and neutrophil. Distinct cell clusters activated specific transcription factors such as HIC1, LMX1B, TWIST1, and et al., which were responsible for their specific functions. These coregulators, such as HOXB13, VSX2, PAX5, and et al., may mediate cell development and differentiation in human embryos. Conclusions We provide the cell development atlas for human embryos (7–9 weeks). Two distinct cell development and differentiation pathways are revealed.

Abstract The precise design of single-atom nanozymes (SAzymes) and understanding of their biocatalytic mechanisms hold great promise for developing ideal bio-enzyme substitutes. While considerable efforts have been directed towards mimicking partial bio-inspired structures, the integration of heterogeneous SAzymes configurations and homogeneous enzyme-like mechanism remains an enormous challenge. Here, we show a spatial engineering strategy to fabricate dual-sites SAzymes with atomic Fe active center and adjacent Cu sites. Compared to planar Fe–Cu dual-atomic sites, vertically stacked Fe–Cu geometry in FePc@2D-Cu–N–C possesses highly optimized scaffolds, favorable substrate affinity, and fast electron transfer. These characteristics of FePc@2D-Cu–N–C SAzyme induces biomimetic O2 activation through homogenous enzymatic pathway, resembling functional and mechanistic similarity to natural cytochrome c oxidase. Furthermore, it presents an appealing alternative of cytochrome P450 3A4 for drug metabolism and drug–drug interaction. These findings are expected to deepen the fundamental understanding of atomic-level design in next-generation bio-inspired nanozymes.

Abstract Objective To evaluate the value of an integrated model incorporating deep learning (DL), hand-crafted radiomics and clinical and US imaging features for diagnosing central lymph node metastasis (CLNM) in patients with papillary thyroid cancer (PTC). Methods This retrospective study reviewed 613 patients with clinicopathologically confirmed PTC from two institutions. The DL model and hand-crafted radiomics model were developed using primary lesion images and then integrated with clinical and US features selected by multivariate analysis to generate an integrated model. The performance was compared with junior and senior radiologists on the independent test set. SHapley Additive exPlanations (SHAP) plot and Gradient-weighted Class Activation Mapping (Grad-CAM) were used for the visualized explanation of the model. ABSTRACTLake distribution on the Tibetan Plateau (TP) is extensive, and lake area changes are key indicators of the TP's climate change response. Many multisource remote sensing big data for the TP, particularly optical images, are unusable due to cloud cover. Therefore, an improved isobath interpolation-based lake area extraction method is proposed and applied to obtain annual average lake areas (≥ 50 km²) on the TP from 1986 to 2020 using remote sensing big data. The lake area result accuracy was verified using existing lake area and level datasets, yielding correlation coefficients of ∼0.9. The change points and segmented trends of each lake's interannual area sequence were obtained. The relationships between lake area and climatic variables were investigated. The positive accumulation of the total precipitation minus total evaporation explains the overall lake area expansion trend after 1995. The exorheic lake interannual area is related to precipitation more than that of endorheic lakes, but endorheic lake area changes are stronger. The shrinking of lakes on the southern TP may not be climate-driven but probably attributed to lake bottom leakage. We explore detailed interannual variation characteristics of lake areas on the TP and provide reference data for studying lake responses to climate change.

Abstract Value‐added conversion of lignocellulose is a sustainable approach. Photo‐refining biomass is in line with current environmental protection strategies. However, photo‐reforming biomass suffers from poor catalyst stability and low conversion efficiency. Here, we designed fructose as a lignocellulosic model. The heterogeneous structure of Prussian blue coating was constructed with a special covalent bond structure of CoCNZn. This structure has a catalytic conversion mechanism that can accelerate electron transfer. Fructose was simultaneously converted to value‐added platform compounds (5‐HMF and formic acid) and gaseous fuels (CO, CH4) with a conversion rate of up to 92.5%, which is more than 1.7 times than that of catalysts without adding Prussian blue. Hydrogen transfer and carbon transfer on the carbon atoms of fructose facilitates the production and accelerates the spillover of CO from formic acid. This work provides new ideas for the development of Prussian blue catalysts and the conversion of pentose.

Abstract Engineering piezo/ferroelectricity in two-dimensional materials holds significant implications for advancing the manufacture of state-of-the-art multifunctional materials. The inborn nonstoichiometric propensity of two-dimensional transition metal dichalcogenides provides a spiffy ready-available solution for breaking inversion centrosymmetry, thereby conducing to circumvent size effect challenges in conventional perovskite oxide ferroelectrics. Here, we show the extendable and ubiquitous piezo/ferroelectricity within nonstoichiometric two-dimensional transition metal dichalcogenides that are predominantly centrosymmetric during standard stoichiometric cases. The emerged piezo/ferroelectric traits are aroused from the sliding of van der Waals layers and displacement of interlayer metal atoms triggered by the Frankel defects of heterogeneous interlayer native metal atom intercalation. We demonstrate two-dimensional chromium selenides nanogenerator and iron tellurides ferroelectric multilevel memristors as two representative applications. This innovative approach to engineering piezo/ferroelectricity in ultrathin transition metal dichalcogenides may provide a potential avenue to consolidate piezo/ferroelectricity with featured two-dimensional materials to fabricate multifunctional materials and distinguished multiferroic.

Abstract Background Kiwifruit bacterial canker, caused by Pseudomonas syringae pv. actinidiae (Psa), is a destructive disease worldwide. Resistance genes that respond to Psa infection urgently need to be identified for controlling this disease. Laccase is mainly involved in the synthesis of lignin in the plant cell wall and plays a prominent role in plant growth and resistance to pathogen infection. However, the role of laccase in kiwifruit has not been reported, and whether laccase is pivotal in the response to Psa infection remains unclear. Results We conducted a bioinformatics analysis to identify 55 laccase genes (AcLAC1–AcLAC55) in the kiwifruit genome. These genes were classified into five cluster groups (I–V) based on phylogenetic analysis, with cluster groups I and II having the highest number of members. Analysis of the exon–intron structure revealed that the number of exons varied from 1 to 8, with an average of 5 introns. Our evolutionary analysis indicated that fragment duplication played a key role in the expansion of kiwifruit laccase genes. Furthermore, evolutionary pressure analysis suggested that AcLAC genes were under purifying selection. We also performed a cis-acting element analysis and found that AcLAC genes contained multiple hormone (337) and stress signal (36) elements in their promoter regions. Additionally, we investigated the expression pattern of laccase genes in kiwifruit stems and leaves infected with Psa. Our findings revealed that laccase gene expression levels in the stems were higher than those in the leaves 5 days after inoculation with Psa. Objective To retrospectively compare the differences in the surgical efficacy and prognosis of laparoscopic pyeloplasty for hydronephrosis caused by symptomatic versus asymptomatic ureteropelvic junction obstruction (UPJO) in children and determine whether clinical symptoms affect the surgical outcome and prognosis. Methods Children who underwent laparoscopic pyeloplasty in our hospital from January 2018 to December 2022 were retrospectively analyzed. The children were divided into symptomatic and asymptomatic groups according to their main symptoms. The primary outcomes were the surgical success rate, change in renal parenchymal thickness, and change in renal pelvis anteroposterior diameter. The secondary outcomes were postoperative complications, reoperation rate, operative duration, intraoperative blood loss, and drainage tube indwelling time. Results In total, 224 children with UPJO were enrolled; 148 (66.1%) were symptomatic and 76 (33.9%) were asymptomatic. The symptomatic group showed a significantly greater mean change in renal parenchymal thickness, significantly higher surgical success rate, and significantly lower postoperative complication rate. Conclusions In the present study, asymptomatic children had a lower surgical success rate, less postoperative imaging improvement, and more postoperative complications than symptomatic children. The presence or absence of clinical symptoms may affect the surgical outcome and prognosis.

Evapotranspiration (ET) is one of the most critical components in hydrological processes and is of great importance to water resource management. Data-oriented deep learning (DL) has been increasingly utilized to forecast hydrological variables over recent years. In this study, the ability of the widely-used DL methods, including long short-term memory (LSTM), bi-directional LSTM (BiLSTM), deep neural network (DNN), and deep belief network (DBN), on the estimation of actual daily ET over the heterogeneous surfaces, was investigated using four groups of experiments. Firstly, the influence of the different input variables on the above DL-based ET model over the various land cover types was explored and analyzed. The results showed that the DL-based ET model can accurately estimate the actual daily ET over the heterogeneous surfaces using a few key conventional observations, i.e., net radiation (Rn), relative humidity (RH), air temperature (Ta), and wind speed (u), as well as soil water content (SWC). However, the performance of the DL-based ET model varied from the different combinations of input variables. SWC was crucial to the estimation of ET, with RMSD decreased from 0.94 to 0.76 mm d−1 when SWC was added to the DL model. Then, the comparison of the four DL-based ET model performances was done. The minor difference in ET estimates was caused by the algorithm differences between LSTM, BiLSTM, DNN, and DBN. Based on the above work, a unified DL-based ET model over the heterogeneous surfaces was developed. The unified DL-based model improved the applicability of DL in ET estimation although it underperformed the separate DL-based model. Finally, the comparison of the performance of the TSEB model and DL method was done. Evaluated with the results of heterogeneous surface, the DL-based model had a better accuracy with a MAPE of 16 – 50%; while the TSEB model had a larger MAPE of 20 – 55%. The results of this research suggest that the DL-based ET model is a promising alternative for the simulation of the daily ET over the heterogeneous surfaces.

Lilium brownii var. viridulum is one of the main traditional edible and medicinal lily in China. This study aimed to explore the extraction technology, physicochemical properties and functional activity of polysaccharide from Lilium brownii var. viridulum, used Lilium brownii var. viridulum scales as materials, polysaccharide yield as the index, solid-liquid ratio, extraction time and extraction temperature as factors to optimize the extraction process of lily polysaccharide. Then the polysaccharide was purified through removing protein. The physicochemical properties of the polysaccharides were analyzed, and the antioxidant and lipid-lowering abilities were measured in vitro. The results showed that the optimal extraction conditions were the material to liquid ratio 1:20 g/mL, extraction time 20 min, extraction temperature 75℃, and polysaccharide yield was 11.98%. The contents of total sugar, uronic acid and protein of crude polysaccharide were 58.46%, 8.06% and 10.85%, respectively. After deproteinization, the contents of total sugar and uronic acid of polysaccharide were increased to 84.78% and 15.41%, respectively, and the content of protein was reduced to 4.73%. The scavenging rates of 10 mg/mL deproteinized polysaccharide for DPPH and ABTS+ free radicals were 8.75% and 38.59%, respectively, which were lower than those of control VC. The inhibition rate of deproteinized polysaccharide for pancreatic lipase was 85.78%, which was higher than that of the control orlistat, indicating good inhibit pancreatic lipase effect in vitro. These results would provide a basis for further study on the structure-activity relationship and development of potential functional food, and provide theoretical reference for the application of lily.

Abstract Incoherent interfaces with large mismatches usually exhibit very weak interfacial interactions so that they rarely generate intriguing interfacial properties. Here we demonstrate unexpected strong interfacial interactions at the incoherent AlN/Al2O3 (0001) interface with a large mismatch by combining transmission electron microscopy, first-principles calculations, and cathodoluminescence spectroscopy. It is revealed that strong interfacial interactions have significantly tailored the interfacial atomic structure and electronic properties. Misfit dislocation networks and stacking faults are formed at this interface, which is rarely observed at other incoherent interfaces. The band gap of the interface reduces significantly to ~ 3.9 eV due to the competition between the elongated Al-N and Al-O bonds across the interface. Thus this incoherent interface can generate a very strong interfacial ultraviolet light emission. Our findings suggest that incoherent interfaces can exhibit strong interfacial interactions and unique interfacial properties, thereby opening an avenue for the development of related heterojunction materials and devices.

The Cu-GO layered composites with excellent thermal conductivity were fabricated by ultrasonic-assisted electrodeposition and ultrasonic spraying composite processes. Moreover, the impact of the spraying frequency and spraying path of GO solution on the surface morphology, microstructure and thermal conductivity of the composites were investigated. The experimental findings indicated that the formation of the Cu–O–C covalent bonding and the interfacial product Cu2O among the Cu layers and the GO film enhanced the Cu/C interfacial bonding during the ultrasonic-assisted electrodeposition. The in-plane thermal conductivity of the composites reached 548.06 W m−1·-k−1 at the optimum process parameters, which was 76.2% higher compared to the pure Cu foil. In this work, the enhancement of the Cu/C interfacial bonding and the optimization of the microstructure of GO and Cu in the composites were responsible for the substantial enhancement of the thermal properties of the Cu-GO layered composites.

The soil dielectric constant model is essential for retrieving soil properties based on microwave remote sensing. However, the existing saline soil dielectric constant models perform poorly in simulating the dielectric constant of soil with high water content and salinity. In this study, the Wang Yueru (WYR) saline soil dielectric constant model, which was demonstrated to perform well in describing the effect of salinity and moisture on the dielectric constant, was validated based on experimental measurements of soil samples under different water content and salinity degrees. Furthermore, we adjusted the model form, refitted the empirical coefficient in the model, and finally acquired a two-stage model for simulating the soil dielectric constant. The enhanced model was validated under different soil moisture and salinity ranges using experimental measurements of soil samples. Compared to the original model, the proposed model exhibits a larger improvement in simulating the soil dielectric constant, and the RMSE of the simulated results dramatically decreased from 7.3 to 1.6, especially for soil with high salinity and water content. On this basis, a model suitable for L-band microwave was established. This model is of great significance for studying soil dielectric characteristics and retrieving soil parameters based on L-band data. Furthermore, this model can be used to retrieve soil salinity and water content using microwave remote sensing under a broadened application situation, such as in saline-alkali soils, wetlands, and salt marshes.

Objective: The ability of a brain-computer interface (BCI) to classify brain activity in electroencephalograms (EEG) during motor imagery (MI) tasks is an important performance indicator. Because the cortical regions that drive the single-handed open and closed tasks overlap, it is difficult to classify the EEG signals during executing both tasks. Approach: The addition of special EEG features can improve the accuracy of classifying single-hand open and closed tasks. In this work, we designed a hybrid BCI paradigm based on error-related potentials (ErrP) and motor imagery (MI) and proposed a strategy to correct the classification results of MI by using ErrP information. The ErrP and MI features of EEG data from 11 subjects were superimposed. Main results: The corrected strategy improved the classification accuracy of single-hand open/close MI tasks from 52.3% to 73.7%, an increase of approximately 21%. Significance: Our hybrid BCI paradigm improves the classification accuracy of single-hand MI by adding ErrP information, which provides a new approach for improving the classification performance of BCI.

Abstract Background Obstructive sleep apnea syndrome (OSAS) is a potentially fatal sleep respiratory disorder, and hepcidin has been found to be related with OSAS onset and severity. This study aimed to examine the serum expression of microRNA‐149‐3p (miR‐149‐3p) and hepcidin in OSAS patients, and evaluate the predictive value of miR‐149‐3p and hepcidin for OSAS occurrence in obese population. Methods This study analyzed the data from 212 OSAS patients and 120 control individuals. OSAS severity was evaluated by apnea hypopnea index (AHI) from polysomnography. Serum miR‐149‐3p was examined using reverse transcription quantitative PCR, and hepcidin and inflammatory cytokines were measured using ELISA kits. Logistic regression analysis was used to evaluate the predictive value of miR‐142‐3p and hepcidin for OSAS in obese population, and ROC curve was plotted to assess the predictive accuracy. Results Serum miR‐149‐3p and hepcidin were increased in OSAS patients, especially in the severe cases, and had diagnostic potential to distinguish OSAS. High miR‐149‐3p and hepcidin were positively correlated with OSAS patients' inflammatory cytokines. Obese OSAS patients had the highest miR‐149‐3p and hepcidin levels, and the two molecules had predictive value of OSAS present in obese population, and the combination of miR‐149‐3p and hepcidin showed the highest predictive accuracy. Conclusion Serum miR‐149‐3p and hepcidin levels were elevated in OSAS patients and correlated with disease severity and systemic inflammation. miR‐149‐3p and hepcidin levels have diagnostic value to distinguish OSAS, exhibited predictive value for OSAS in obese population, and the joint detection of the two molecules showed the highest predictive accuracy.

Abstract The problem of localization and encirclement control of a moving target by multi unmanned aerial vehicles (UAVs) is considered in this paper. The main objective is to guide the UAVs to form evenly spaced formations along the circumference, with the centre of the circumference tracking the movement of the target. Firstly, each UAV is assumed only to obtain the bearing angle information of the target, for which an estimator is developed to localize the target by bearing measurements. Thereafter, a distributed encirclement control algorithm based on cooperative moving path following (CMPF) is designed to drive the multi‐UAVs to circumnavigate around a moving target at a desired distance, thus developing to pursue cooperatively. Finally, the simulation results demonstrate the effectiveness of the proposed method.

Cu-graphene oxide (GO)-Cu laminated composites foils with a “brick-mud” structure were designed and fabricated by an alternating methods of direct current deposition and electrophoresis. The effects of processing parameters and laminated structure on its micro-structure and mechanical/thermal properties were investigated, and the grain nucleation and growth behavior on GO during the electro-deposition was analyzed. The results revealed that compared with pure Cu foils, the maximum tensile strength increased from 237 MPa to 308 MPa. While, the thermal conductivity of Cu-GO-Cu laminated composites with optimized condition increased 11% to 439.60 W m−1 K−1. The existence of loose GO made it easier for copper ions to pass through and form more denser network structure, so that the performance of composite foils can be optimized. The enhanced mechanical and thermal properties are attributed to favorable interfacial bonding and well-designed laminated structure of Cu-GO composites.

Brain-computer interface (BCI) based on lower-limb motor imagery (LMI) enables hemiplegic patients to stand and walk independently. However, LMI ability is usually poor for BCI-illiterate (e.g., some stroke patients), limiting BCI performance. This study proposed a novel LMI-BCI paradigm with kinesthetic illusion(KI) induced by vibratory stimulation on Achilles tendon to enhance LMI ability. Sixteen healthy subjects were recruited to carry out two research contents: (1) To verify the feasibility of induced KI by vibrating Achilles tendon and analyze the EEG features produced by KI, research 1 compared the subjective feeling and brain activity of participants during rest task with and without vibratory stimulation (V-rest, rest). (2) Research 2 compared the LMI-BCI performance with and without KI (KI-LMI, no-LMI) to explore whether KI enhances LMI ability. The analysis methods of both experiments included classification accuracy (V-rest vs. rest, no-LMI vs. rest, KI-LMI vs. rest, KI-LMI vs. V-rest), time-domain features, oral questionnaire, statistic analysis and brain functional connectivity analysis. Research 1 verified that induced KI by vibrating Achilles tendon might be feasible, and provided a theoretical basis for applying KI to LMI-BCI paradigm, evidenced by oral questionnaire (Q1) and the independent effect of vibratory stimulation during rest task. The results of research 2 that KI enhanced mesial cortex activation and induced more intensive EEG features, evidenced by ERD power, topographical distribution, oral questionnaire (Q2 and Q3), and brain functional connectivity map. Additionally, the KI increased the offline accuracy of no-LMI/rest task by 6.88 to 82.19% (p 75.31%, and average F1_score for all subjects: 76.4% > 74.3%). The LMI-BCI paradigm of this study provides a novel approach to enhance LMI ability and accelerates the practical applications of the LMI-BCI system.

This was an initial effort to treat hemophiliac hematoma by ultrasound-guided intratumoral drainage and lavage with urokinase after adequate supplementation of coagulation factors. Two patients with severe hemophilia underwent ultrasound-guided percutaneous drainage in combination with lavage using urokinase. After 5-day and 3-day treatments, respectively, intramuscular hematomas in both patients disappeared, compression symptom was relieved, and no obvious adverse reactions or serious complications were observed during the treatment or follow-up. These findings suggest that ultrasound-guided drainage combined with lavage using urokinase is an immediate, safe, effective, and minimally invasive treatment for intramuscular hematomas in hemophiliacs, avoiding potential complications by surgical resection with relatively low treatment cost.

This study evaluates inpatients’ ontological insecurity and daily epidemic prevention behavior during the pandemic and explores the factors influencing daily epidemic prevention behaviors. The outbreak of coronavirus disease (COVID-19) in December 2019 caused a global public health crisis that has affected the very structure of society and the order of daily life. Ontological security is the ability to predict the impact of changes in social environments on personal security, such as during the pandemic. A cross-sectional study was used to collect data from 1185 inpatients of a hospital in Zhejiang, China, from July 11 to August 9, 2021. Our questionnaire recorded information on demographics, ontological insecurity, and daily epidemic prevention behaviors. The Mann-Whitney U test, Kruskal-Wallis H test, Spearman’s correlation analysis, and logistic regression analysis were used to determine the influencing factors of daily epidemic prevention behavior on ontological security. ABSTRACT Phytophthora species are devastating filamentous plant pathogens that belong to oomycetes, a group of microorganisms similar to fungi in morphology but phylogenetically distinct. They are sterol auxotrophic, but nevertheless exploit exogenous sterols for growth and development. However, as for now the mechanisms underlying sterol utilization in Phytophthora are unknown. In this study, we identified four genes in Phytophthora capsici that encode proteins containing a sterol-sensing domain (SSD), a protein domain of around 180 amino acids comprising five transmembrane segments and known to feature in sterol signaling in animals. Using a modified CRISPR/Cas9 system, we successfully knocked out the four genes named PcSCP1 to PcSCP4 (for P. capsici SSD-containing protein 1 to 4), either individually or sequentially, thereby creating single, double, triple, and quadruple knockout transformants. Results showed that knocking out just one of the four PcSCPs was not sufficient to block sterol signaling. However, the quadruple “all-four” PcSCPs knockout transformants no longer responded to sterol treatment in asexual reproduction, in contrast to wild-type P. capsici that produced zoospores under sterol treatment. Apparently, the four PcSCPs play a key role in sterol signaling in P. capsici with functional redundancy. Transcriptome analysis indicated that the expression of a subset of genes is regulated by exogenous sterols via PcSCPs. Further investigations showed that sterols could stimulate zoospore differentiation via PcSCPs by controlling actin-mediated membrane trafficking. Moreover, the pathogenicity of the “all-four” PcSCPs knockout transformants was significantly decreased and many pathogenicity related genes were downregulated, implying that PcSCPs also contribute to plant-pathogen interaction. IMPORTANCE Phytophthora is an important genus of oomycetes that comprises many destructive plant pathogens. Due to the incompleteness of the sterol synthesis pathway, Phytophthora spp. do not possess the ability to produce sterols. Therefore, these sterol auxotrophic oomycetes need to recruit sterols from the environment such as host plants to support growth and development, which seems crucial during pathogen-plant interactions. However, the mechanisms underlying sterol utilization by Phytophthora spp. remain largely unknown. Here, we show that a family of sterol-sensing domain-containing proteins (SCPs) consisting of four members in P. capsici plays a key role in sterol signaling with functional redundancy. Moreover, these SCPs play a role in different biological processes, including asexual reproduction and pathogenicity. Our study overall revealed the multiple functions of PcSCPs and addressed the question of how exogenous sterols regulate the development of heterothallic Phytophthora spp. via SSD-containing proteins.

Kiwifruit canker disease, caused by Pseudomonas syringae pv. actinidiae (Psa), is the main threat to kiwifruit production worldwide. Currently, there is no safe and effective disease prevention method; therefore, biological control technologies are being explored for Psa. In this study, Bacillus velezensis WL–23 was isolated from the leaf microbial community of kiwifruit and used to control kiwifruit cankers. Indoor confrontation experiments showed that both WL–23 and its aseptic filtrate had excellent inhibitory activity against the main fungal and bacterial pathogens of kiwifruit. Changes in OD600, relative conductivity, alkaline proteinase, and nucleic acid content were recorded during Psa growth after treatment with the aseptic filtrate, showing that Psa proliferation was inhibited and the integrity of the cell membrane was destroyed; this was further verified using scanning electron microscopy and transmission electron microscopy. In vivo, WL–23 promoted plant growth, increased plant antioxidant enzyme activity, and reduced canker incidence. Therefore, WL–23 is expected to become a biological control agent due to its great potential to contribute to sustainable agriculture.

Soft rot causes significant economic losses in the kiwifruit industry. This study isolated strain CTXW 7-6-2 from healthy kiwifruit tissue; this was a gram-positive bacterium that produced the red pigment pulcherrimin. The phylogenetic tree based on 16S ribosomal RNA, gyrA, rpoB, and purH gene sequences identified CTXW 7-6-2 as a strain of Bacillus subtilis. CTXW 7-6-2 inhibited hyphal growth of pathogenic fungi that cause kiwifruit soft rot, namely, Botryosphaeria dothidea, Phomopsis sp., and Alternaria alternata, by 81.76, 69.80, and 32.03%, respectively. CTXW 7-6-2 caused the hyphal surface to become swollen and deformed. Volatile compounds (VOC) produced by the strain inhibited the growth of A. alternata and Phomopsis sp. by 65.74 and 54.78%, respectively. Whole-genome sequencing revealed that CTXW 7-6-2 possessed a single circular chromosome of 4,221,676 bp that contained 4,428 protein-coding genes, with a guanine and cytosine (GC) content of 43.41%. Gene functions were annotated using the National Center for Biotechnology Information (NCBI) non-redundant protein, Swiss-Prot, Kyoto Encyclopedia of Genes and Genomes, Clusters of Orthologous Groups of proteins, Gene Ontology, Pathogen–Host Interactions, Carbohydrate-Active enZYmes, and Rapid Annotations using Subsystem Technology databases, revealing non-ribosomal pathways associated with antifungal mechanisms, biofilm formation, chemotactic motility, VOC 3-hydroxy-2-butanone, cell wall-associated enzymes, and synthesis of various secondary metabolites. antiSMASH analysis predicted that CTXW 7-6-2 can produce the active substances bacillaene, bacillibactin, subtilosin A, bacilysin, and luminmide and has four gene clusters of unknown function. Quantitative real-time PCR (qRT-PCR) analysis verified that yvmC and cypX, key genes involved in the production of pulcherrimin, were highly expressed in CTXW 7-6-2. This study elucidates the mechanism by which B. subtilis strain CTXW 7-6-2 inhibits pathogenic fungi that cause kiwifruit soft rot, suggesting the benefit of further studying its antifungal active substances.

Abstract Background Lung cancer mainly includes non‐small cell lung cancer (NSCLC). Lung adenocarcinoma (LUAD) is the main subtype of NSCLC. Long non‐coding RNAs (LncRNAs) had been found to exert numerous functions on the progressions of cancers. MicroRNAs often exist as the target of LncRNAs to regulate a series of signaling pathways in human. We explored the effects and molecular mechanism of LncRNA SGMS1‐AS1 on the procedures of LUAD cells. Methods The ENCORI and GEPIA databases were used to analyze the differences in SGMS1, miR‐106a‐5p, and MYLIP between LUAD and normal tissue. Their expression levels were examined by RT‐PCR. CCK8, colony formation, migration, and invasion assay were conducted in LUAD cells which had silenced SGMS1‐AS1. To verify the relationship between SGMS1‐AS1, miR‐106a‐5p, and MYLIP, we overexpressed miR‐106a‐5p inhibitor or MYLIP in LUAD cells after decreasing SGMS1‐AS1 and repeated the above assays. Results SGMS1‐AS1 was downregulated in LUAD tissue as well as cells, which was related to good prognosis of patients with lung adenocarcinoma. Additionally, knockdown of SGMS1‐AS1 promoted proliferation, migration, invasion, and epithelial mesenchymal transition (EMT) progression of LUAD cells, which meant that SGMS1‐AS1 inhibited the progression of LUAD cells. Furthermore, miR‐106a‐5p was the direct target of SGMS1‐AS1 and transfecting miR‐106a‐5p inhibitor could reversed the impact induced by knockdown of SGMS1‐AS1. Subsequently, we found that MYLIP was the target of miR‐106a‐5p, which was negatively correlated with miR‐106a‐5p, but had high positive correlation with SGMS1‐AS1. Consistently, overexpression MYLIP partly eliminated the effects on A549 cells induced by silencing of SGMS1‐AS1. Conclusion LncRNA SGMS1‐AS1 inhibits the proliferation, invasion, migration and EMT progression of LUAD cells via targeting miR‐106a‐5p/MYLIP axis.

A series polysaccharide samples extracted from three edible lilies (Lilium davidii var. willmottiae, Lilium brownii var. viridulum, and Lilium lancifolium) by subcritical water and ultrasound-assisted extraction were systematically compared. The results showed that extraction method was a more important factor than lily species. Subcritical water extracted lily polysaccharides (S-LP) with higher yield, molecular weight, neutral glucose and uronic acid content as well as apparent viscosity. Ultrasound-assisted extracted lily polysaccharides (U-LP) with higher reducing sugars and protein content. Moreover, due to the degradation of glycosidic bonds, ultrasonic extraction was easier to obtain lower molecular weight polysaccharides. In addition, the extraction method significantly affected the monosaccharide proportion of polysaccharides, but had no effect on type. Glucose was the main component in S-LP, and glucose and mannose were the main components in U-LP. The micromorphology of different polysaccharide samples was similar, and the scanning electron microscope (SEM) images showed regular/irregular particle clusters with different particle sizes. Overall, the relationships between extraction methods, lily species and polysaccharide properties were preliminarily elucidated, providing a reference for the targeted extraction of specific lily polysaccharides (LP).

Fertilization changes nitrogen (N)-cycling processes and associated microbial communities in agricultural ecosystems. However, the long-term responses of N-cycling potential and microbial functional genes to different fertilization sources remain unclear. Soil samples were collected to a depth of 20 cm under winter wheat applied annually with N fertilizer (NF), organic manure (OM), N fertilizer plus organic manure (NM), and a control without fertilization (CK) for 35 yr, and they were analyzed for microbial functional genes involved in soil N cycling using metagenome sequencing in the Loess Plateau of China. Soil N fractions were greater with OM and NM than NF and CK. The total abundances of N-cycling genes were 9.3% (p < 0.05) greater with NM than CK, and 8.2% (p < 0.05) and 12.2% (p < 0.01) higher with OM and NM than NF, respectively. Compared to CK, OM and NM increased the abundance of genes associated with nitrification, denitrification, dissimilatory nitrate reduction, and assimilatory nitrate reduction, but decreased the abundance of genes related to organic N metabolism. However, NF increased the abundance of genes involved in nitrification. Both OM and NM also enhanced the relative abundance of Proteobacteria carrying N-cycling genes but reduced those of Firmicutes and Cyanobacteria. Soil organic carbon, total N, and potential carbon mineralization were the dominant factors affecting the abundances of N-cycling genes. Long-term application of OM and NM can promote N cycling by enhancing gene abundance due to increased soil organic matter and microbial biomass compared to NF and CK.

In this paper, rice husk ash (RHA) with different average pore diameters and specific surface areas was used to replace 10% slag in the preparation of alkali-activated slag (AAS) pastes. The effect of RHA addition on the shrinkage, hydration, and strength of AAS pastes was studied. The results show that RHA with a porous structure will pre-absorb part of the mixing water during paste preparation, resulting in a decrease in the fluidity of AAS pastes by 5–20 mm. RHA has a significant inhibitory effect on the shrinkage of AAS pastes. The autogenous shrinkage of AAS pastes decreases by 18–55% at 7 days, and the drying shrinkage decreases by 7–18% at 28 days. This shrinkage reduction effect weakens with the decrease in RHA particle size. RHA has no obvious effect on the type of hydration products of AAS pastes, whereas RHA after proper grinding treatment can significantly improve the hydration degree. Therefore, more hydration products are generated and fills the internal pores of the pastes, which significantly improves the mechanical properties of the AAS pastes. The 28 day compressive strength of sample R10M30 (the content of RHA is 10%, RHA milling time is 30 min) is 13 MPa higher than that of blank sample.

Abstract Flumorph is a carboxylic acid amide (CAA) fungicide with high activity against oomycetes. However, evolution to CAAs from low resistance to high resistance has never been reported. This study investigated the basis of resistance evolution of flumorph in Phytophthora sojae. Total of 120 P. sojae isolates were collected and their sensitivity to flumorph was evaluated. Although no spontaneous resistance was found among the field isolates, adaptation on flumorph‐amended media resulted in the selection of five stable mutant types exhibiting varying degrees of resistance to CAAs. Type I, which exhibited the lowest resistance level, was obtained when the wild‐type isolate was exposed to a low concentration of flumorph, but no resistant mutants were obtained by direct exposure to higher concentrations. However, the more resistant types (Type II, III, IV and V) were obtained when Type I were exposed to higher concentrations of flumorph. Similar results were obtained when the entire screening process was repeated, which implied that evolution of resistance to flumorph in P. sojae could be a two‐step process, where high resistance phenotypes could develop gradually from low resistance ones. Further investigation into molecular mechanism strongly confirmed that evolution of isolates highly resistant to flumorph occurs in a stepwise process with Type I as intermediary, through accumulation of mutations in their target protein of CAAs (CesA3). Together, our findings indicate that application of low rates of flumorph in field could result in selection of low resistance Type I isolates, but that raising dosage to maintain comparable levels of control could elicit rapid evolution of more resistant Type II, III, IV and V isolates with stepwise accumulation of mutations in CesA3, which would render flumorph ineffective as a control method. Precautionary resistance management strategy should be implemented. The phenomenon described in the study could have broader biological significance.

Although sterols play an important role in most eukaryotes, some oomycetes, including Phytophthora spp., have lost the sterol synthesis pathway. Nevertheless, the ERG3 gene encoding C-5 sterol desaturase in the sterol synthesis pathway is still present in the genomes of Phytophthora spp. Phytophthora capsici, a destructive pathogen with a broad range of plant hosts, poses a significant threat to the production of agriculture. This study focused on the ERG3 gene in P. capsici (PcERG3) and explored its function in this pathogen. It showed that the PcERG3 gene could be expressed in all tested developmental stages of P. capsici, with sporangium and mycelium displaying higher expression levels. A potential substrate of Erg3 (stellasterol) was used to treat the P. capsici wild-type strain and a PcERG3Δ transformant, and their sterol profiles were determined by GC-MS. The wild-type strain could convert stellasterol into the down-stream product while the transformant could not, indicating that PcErg3 retains the C-5 sterol desaturase activity. By comparing the biological characteristics of different strains, it was found that PcERG3 is not important for the development of P. capsici. The pathogenicity of the PcERG3Δ transformants and the wild-type strain was comparable, suggesting that PcERG3 is not necessary for the interaction between P. capsici and its hosts. Further investigations revealed that the PcERG3Δ transformants and the wild-type strain displayed a similar level of tolerance to external adversities such as unsuitable temperatures, high osmotic pressures, and intemperate pH, signifying that PcERG3 is not essential for P. capsici to cope with these environmental stresses.

BackgroundThe COVID-19 pandemic has changed the social environment of most individuals around the world and has profoundly impacted people's lives, ontological security, and behavior. Among them, the patients are one of the groups most influenced by the pandemic.ObjectiveThe present research aimed to study the relationship of COVID-19 pandemic-induced disruption to patients' daily lives, ontological security, and patients' responses to prevent the spread of COVID-19, and explore the role of ontological security.MethodsThis article was based on an online structured questionnaire study conducted among hospitalized patients in Taizhou Hospital of Zhejiang Province, Taizhou, China, from 8 July to 11 August 2021. We analyzed the data using the multivariate regression model and mediation analysis method.ResultsThe results showed that the higher the pandemic-induced disruption to inpatients' lives, the better behavior would be taken by hospitalized patients to prevent the spread of COVID-19, and the perceived scarcity of ontological security played a mediating role in this process. Higher pandemic-induced disruption to patients' lives increased the ontological insecurity which further, in turn, reduced patients' good practice toward measures to prevent the novel coronavirus.ConclusionThese findings provided direct evidence for the relationship between pandemic-induced disruption, scarcity of ontological security, and patients' prevention behavior. It suggested that there was a need to emphasize patients' ontological security. Overall, these findings suggested that it is important to emphasize the mental health among patients during the COVID-19 pandemic, and implement strategies to offer psychological support when needed.

The de novo biosynthesis of sterols is critical for the majority of eukaryotes; however, some organisms lack this pathway, including most oomycetes. Phytophthora spp. are sterol auxotrophic but, remarkably, have retained a few genes encoding enzymes in the sterol biosynthesis pathway. Here, we show that PcDHCR7, a gene in Phytophthora capsici predicted to encode Δ7-sterol reductase, displays multiple functions. When expressed in Saccharomyces cerevisiae, PcDHCR7 showed the Δ7-sterol reductase activity. Knocking out PcDHCR7 in P. capsici resulted in loss of the capacity to transform ergosterol into brassicasterol, which means PcDHCR7 has the Δ7-sterol reductase activity in P. capsici itself. This enables P. capsici to transform sterols recruited from the environment for better use. The biological characteristics of ΔPcDHCR7 transformants were compared with those of the wild-type strain and a PcDHCR7 complemented transformant, and the results showed that PcDHCR7 plays a key role in mycelium development and pathogenicity of zoospores. Further analysis of the transcriptome indicated that the expression of many genes changed in the ΔPcDHCR7 transformant, which involve in different biological processes. It is possible that P. capsici compensates for the defects caused by the loss of PcDHCR7 by remodelling its transcriptome.

Controlling the morphology and composition of semiconductor nano- and micro-structures is crucial for fundamental studies and applications. Here, Si-Ge semiconductor nanostructures were fabricated using photolithographically defined micro-crucibles on Si substrates. Interestingly, the nanostructure morphology and composition of these structures are strongly dependent on the size of the liquid–vapour interface (i.e., the opening of the micro-crucible) in the CVD deposition step of Ge. In particular, Ge crystallites nucleate in micro-crucibles with larger opening sizes (3.74–4.73 μm2), while no such crystallites are found in micro-crucibles with smaller openings of 1.15 μm2. This interface area tuning also results in the formation of unique semiconductor nanostructures: lateral nano-trees (for smaller openings) and nano-rods (for larger openings). Further TEM imaging reveals that these nanostructures have an epitaxial relationship with the underlying Si substrate. This geometrical dependence on the micro-scale vapour–liquid–solid (VLS) nucleation and growth is explained within a dedicated model, where the incubation time for the VLS Ge nucleation is inversely proportional to the opening size. The geometric effect on the VLS nucleation can be used for the fine tuning of the morphology and composition of different lateral nano- and micro-structures by simply changing the area of the liquid–vapour interface.

SYP-14288 is a fungicide as an uncoupler of oxidative phosphorylation, which is effective in controlling fungal pathogens like Rhizoctonia solani. To determine whether R. solani can develop SYP-14288 resistance and possibly multi-drug resistance (MDR), an SYP-14288-resistant mutant of R. solani X19-7 was generated from wild-type strain X19, and the mechanism of resistance was studied through metabolic and genetic assays. From metabolites of R. solani treated with SYP-14288, three compounds including M1, M2, and M3 were identified according to UPLC-MS/MS analysis, and M1 accumulated faster than M2 and M3 in X19-7. When X19-7 was treated by glutathione-S-transferase (GST) inhibitor diethyl maleate (DEM) and SYP-14288 together, or by DEM plus one of tested fungicides that have different modes of action, a synergistic activity of resistance occurred, implying that GSTs promoted metabolic resistance against SYP-14288 and therefore led to MDR. By comparing RNA sequences between X19-7 and X19, six cytochrome P450s (P450s) and two GST genes were selected as a target, which showed a higher expression in X19-7 than X19 both before and after the exposure to SYP-14288. Furthermore, heterologous expression of P450 and GST genes in yeast was conducted to confirm genes involved in metabolic resistance. In results, the P450 gene AG1IA_05136 and GST gene AG1IA_07383 were related to fungal resistance to multiple fungicides including SYP-14288, fluazinam, chlorothalonil, and difenoconazole. It was the first report that metabolic resistance of R. solani to uncouplers was associated with P450 and GST genes.

Patched (Ptc) and Patched-related (Ptr) proteins containing sterol-sensing domains (SSD) and Patched domains are highly conserved in eukaryotes for lipid transport and metabolism. Four proteins containing predicted SSD and Patched domains were simultaneously found by searching the Phytophthora sojae genome database, and one of them was identified as a Patched-like (PTL) protein. Here, we investigated the biological function of PsPTL. The expression level of PsPTL was higher during mycelial and sporulation stages, compared to zoospore (ZO), cyst, and germinated-cyst stages, without significant change during infection. However, deletion of PsPTL using CRISPR/Cas9 had no significant effect on the growth, development, or virulence of P. sojae. Further investigations showed that PsPTL is not essential for P. sojae to cope with external stresses such as temperature, pH, oxidative and osmotic pressure. In addition, this gene did not appear to play an essential role in P. sojae’s response to exogenous sterols. The transcript levels of the other three proteins containing predicted SSD and Patched domains were also not significantly upregulated in PsPTL deletion transformants. Our studies demonstrated that PsPTL is not an essential protein for P. sojae under the tested conditions, and more in-depth research is required for revealing the potential functions of PsPTL under special conditions or in other signaling pathways.

Long-term satellite observations of the water levels of lakes are crucial to our understanding of lake hydrological basin systems. The Ice, Cloud, and Land Elevation satellite (ICESat) and ICESat-2 were employed to monitor the water level of Qinghai Lake in the hydrological basin. The median of absolute deviation (MAD) method was exploited to remove the outliers. The results confirmed that the MAD range of ICESat was from 0.0525 to 0.2470 m, and the range of σ was from 0.0778 to 0.3662 m; the MAD range of ICESat-2 was from 0.0291 to 0.0490 m, and the range of σ was from 0.0431 to 0.0726 m; ICESat-2 was less than that of ICESat. The reference ellipsoid and geoid transfer equations were applied to convert the water level to the World Geodetic System (WGS84) and Earth Gravitational Model 2008 (EGM2008) geoid. The water level, as derived from laser altimeters, was validated by the Xiashe Hydrological Station; with ICESat, the coefficient of association (R) was 0.8419, the root mean square error (RMSE) was 0.1449 m, and the mean absolute error (MAE) was 0.1144 m; with ICESat-2, the R was 0.6917, the RMSE was 0.0531 m, and the MAE was 0.0647 m. The water levels from ICESat-2 are much more accurate than those from ICESat. The two combined laser altimeters showed that the R was 0.9931, the RMSE was 0.1309 m, and the MAE was 0.1035 m. The water level rise was 3.6584 m from 2004 to 2020. The rising rate was 0.2287 m/a. The collaborative use of the ICESat-2 and ICESat satellites made it easier to obtain the lake water levels.

Soil moisture is a key factor in the water and heat exchange and energy transformation of the ecological systems and is of critical importance to the accurate obtainment of the soil moisture content for supervising water resources and protecting regional and global eco environments. In this study, we selected the soil moisture monitoring networks of Naqu, Maqu, and Tianjun on the Qinghai–Tibetan Plateau as the research areas, and we established a database of surface microwave scattering with the AIEM (advanced integral equation model) and the mathematical expressions for the backscattering coefficient, soil moisture, and surface roughness of the VV and VH polarizations.We proposed the soil moisture retrieval models of empirical and machine learnings algorithms (backpropagation neural network (BPNN), support vector machine (SVM), K-nearest neighbors (KNN), and random forest (RF)) for the ascending and descending orbits using Sentinel-1 and measurement data, and we also validated the accuracies of the retrieval model in the research areas. According to the results, there is a substantial logarithmic correlation among the backscattering coefficient, soil moisture, and combined roughness. Generally, we can use empirical models to estimate the soil moisture content, with an R² of 0.609, RMSE of 0.08, and MAE of 0.064 for the ascending orbit model and an R² of 0.554, RMSE of 0.086, and MAE of 0.071 for the descending orbit model. The soil moisture contents are underestimated when the volumetric water content is high. The soil moisture retrieval accuracy is improved with machine learning algorithms compared to the empirical model, and the performance of the RF algorithm is superior to those of the other machine learning algorithms. The RF algorithm also achieved satisfactory performances for the Maqu and Tianjun networks. The accuracies of the inversion models for the ascending orbit in the three soil moisture monitoring networks were better than those for the descending orbit.

‘Hongyang’ kiwifruit (Actinidia chinensis, cultivar ‘Hongyang’) black spot disease is caused by the fungal pathogen Didymella glomerata, and is a serious disease, causing considerable losses to the kiwifruit industry during growth of the fruit. Hence, we aimed to identify a potential biocontrol agent against D. glomerata. In this study, bacterial isolates from the rhizosphere soil of kiwifruit were tested for their potential antifungal activity against selected fungal pathogens. Based on a phylogenetic tree constructed using sequences of 16S rDNA and the gyrA gene, BQ-33 with the best antifungal activity was identified as Bacillus mojavensis. We evaluated the antagonistic activity and inhibitory mechanism of BQ-33 against D. glomerata. Confrontation experiments showed that both BQ-33 suspension and the sterile supernatant (SS) produced by BQ-33 possessed excellent broad-spectrum antifungal activity. Furthermore, the SS damaged the cell membrane and cell wall of the mycelia, resulting in the leakage of a large quantity of small ions (Na+, K+), soluble proteins and nucleic acids. Chitinase and β-1,3-glucanase activities in SS increased in correlation with incubation time and remained at a high level for several days. An in vivo control efficacy assay indicated that 400 mL L−1 of SS completely inhibited kiwifruit black spot disease caused by D. glomerata. Therefore, BQ-33 is a potential biocontrol agent against kiwifruit black spot and plant diseases caused by other fungal pathogens. To our knowledge, this is the first report of the use of a rhizosphere microorganism as a biocontrol agent against kiwifruit black spot disease caused by D. glomerata.

Soil moisture is one of the core hydrological and climate variables that crucially influences water and energy budgets. The spatial resolution of available soil moisture products is generally coarser than 25 km, which limits their hydro-meteorological and eco-hydrological applications and the management of water resources at watershed and agricultural scales. A feasible solution to overcome these limitations is to downscale coarse soil moisture products with the support of higher-resolution spatial information. Although many auxiliary variables have been used for this purpose, few studies have analyzed their applicability and effectiveness in arid regions. To this end, we comprehensively evaluated four commonly used auxiliary variables, including NDVI (Normalized Difference Vegetation Index), LST (Land Surface Temperature), TVDI (Temperature Vegetation Dryness Index), and SEE (Soil Evaporative Efficiency), against ground-based soil moisture observations during the vegetation growing season in the Heihe River Basin, China. Performance metrics indicated that SEE is most sensitive (R2 ≥ 0.67) to soil moisture because it is controlled by soil evaporation limited by the available soil moisture. The similarity of spatial patterns also showed that SEE best captures soil moisture changes, with the STD (standard deviation) of the HD (Hausdorff Distance) less than 0.058 when compared with PLMR (Polarimetric L-band Multi-beam Radiometer) soil moisture products. In addition, soil moisture was mapped by RF (Random Forests) using both single auxiliary variables and 11 types of multiple auxiliary variable combinations. SEE was found to be the best auxiliary variable for scaling and mapping soil moisture with accuracy of 0.035 cm3/cm3. Among the multiple auxiliary variables, the combination of LST, NDVI, and SEE was found to best enhance the scaling and mapping accuracy of soil moisture with 0.034 cm3/cm3.

BackgroundEsophageal squamous cell carcinoma (ESCC) is an aggressive type of cancer, associated with poor prognosis. The development of an accurate and non-invasive method to evaluate the pathologic response of patients with ESCC to chemoradiotherapy remains a critical issue. Therefore, the aim of this study was to assess the importance of vascular permeability and texture parameters in predicting the response to neoadjuvant chemoradiotherapy (NACRT) in patients with ESCC.MethodsThis prospective analysis included patients with T1–T2 stage of ESCC, without either lymphatic or metastasis, and distant metastasis. All patients underwent surgery having received two rounds of NACRT. All patients underwent dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) twice, i.e., before the first NACRT and after the second NACRT. Patients were assessed for treatment response at 30 days after the second NACRT. Patients were divided into the complete response (CR) and partial response (PR) groups based on their responses to NACRT. Vascular permeability and texture parameters were extracted from the DCE-MRI scans. After assessing the diagnostic performance of individual parameters, a combined model with vascular permeability and texture parameters was generated to predict the response to NACRT.ResultsIn this study, the CR and PR groups included 16 patients each. The volume transfer constant (Ktrans), extracellular extravascular volume fraction (ve), and entropy values, as well as changes to each of these parameters, extracted from the second DCE-MRI scans, showed significant differences between the CR and PR groups. The area under the curve (AUC) of Ktrans, ve, and entropy values showed good diagnostic ability (0.813, 0.789, and 0.707, respectively). A logistic regression model combining Ktrans, ve, and entropy had significant diagnostic ability (AUC=0.977).ConclusionsThe use of a combined model with vascular permeability and texture parameters can improve post-NACRT prognostication in patients with ESCC.

Kiwifruit (Actinidia chinensis) is an important commercial crop in China, and the occurrence of diseases may cause significant economic loss in its production. In the present study, a new pathogen that causes brown leaf spot disease on kiwifruit was reported. The fungus was isolated from an infected sample and identified as Fusarium graminearum based on morphological and molecular evaluation. Koch’s postulates were confirmed when the pathogen was re-isolated from plants with artificially induced symptoms and identified as F. graminearum. Based on the biological characteristics of the pathogen, it was determined that: its optimal growth temperature was 25 °C; optimal pH was 7; most suitable carbon source was soluble starch; most suitable nitrogen source was yeast powder; and best photoperiod was 12 h light/12 h dark. Further investigations were conducted by determining 50% effective concentrations (EC50) of several active ingredients of biological fungicides against F. graminearum. The results showed that among the studied fungicides, tetramycin and honokiol had the highest antifungal activity against this pathogen. Our findings provide a scientific basis for the prevention and treatment of brown leaf spot disease on kiwifruit.

Phytophthora, a genus of oomycetes, contains many devastating plant pathogens, which cause substantial economic losses worldwide. Recently, CRISPR/Cas9-based genome editing tool was introduced into Phytophthora to delineate the functionality of individual genes. The available selection markers for Phytophthora transformation, however, are limited, which can restrain transgenic manipulation in some cases. We hypothesized that PcMuORP1, an endogenous fungicide resistance gene from P. capsici that confers resistance to the fungicide oxathiapiprolin via an altered target site in the ORP1 protein, could be used as an alternative marker. To test this hypothesis, the gene PcMuORP1 was introduced into the CRISPR/Cas9 system and complementation of a deleted gene in P. capsici was achieved using it as a selection marker. All of the oxathiapiprolin-resistant transformants were confirmed to contain the marker gene, indicating that the positive screening rate was 100%. The novel selection marker could also be used in other representative Phytophthora species including P. sojae and P. litchii, also with 100% positive screening rate. Furthermore, comparative studies indicated that use of PcMuORP1 resulted in a much higher efficiency of screening compared to the conventional selection marker NPT II, especially in P. capsici. Successive subculture and asexual reproduction in the absence of selective pressure were found to result in the loss of the selection marker from the transformants, which indicates that the PcMuORP1 gene would have little long term influence on the fitness of transformants and could be reused as the selection marker in subsequent projects. Thus, we have created an alternative selection marker for Phytophthora transformation by using a fungicide resistance gene, which would accelerate functional studies of genes in these species.

A whole-plant chamber system equipped with a transpiration sap flow meter was developed for measuring the transpiration rate even if leaves are wetted. A preliminary experiment in which dynamics of transpiration rate and/or evaporation rate of wetted and non-wetted plants were measured and compared with each other demonstrated the validity of the measurement system. The system was then used to analyse leaf wetting effects on gas exchange of corn under slight water stress conditions of soil (a volumetric soil water content of 9.7%). Leaf wetting decreased vapour pressure in leaves by decreasing leaf temperature but it increased vapour pressure in the air; therefore, vapour pressure difference between leaves and air, as a driving force of transpiration, was significantly lower in wetted plant. As a result, transpiration rate decreased by 44% and leaf conductance as an index of stomatal aperture was increased by leaf wetting. Such increasing leaf conductance due to leaf wetting increased the photosynthetic rate by 30% and therefore it improved water use efficiency (2.4 times). These results suggest that morning leaf wetting due to night time dew formation may have an advantage in crop production in semi-arid regions.

Investigating the energy and water vapor exchange in oasis riparian forest ecosystems is of significant importance to improve scientific understanding of land surface processes in extreme arid regions. The Heihe Watershed Allied Telemetry Experimental Research (HiWATER) provided many observations of water vapor and heat fluxes from riparian forest ecosystem by using a network of eddy-covariance (EC) systems installed over representative surfaces in the Ejina Oasis, which is located in the downstream areas of the Heihe River Basin, northwestern China. Based on EC flux measurements and meteorological data performed at five stations and covering representative surface types of Populus euphratica tree with associated Tamarix chinensis shrub, Tamarix chinensis shrubland, cantaloupe cropland, and barren-land, this study explored the spatio-temporal patterns of heat and water vapor fluxes over the Ejina Oasis riparian forest ecosystem with five different surface types over the course of a growing season in 2014. Energy balance closure of the flux data was evaluated; footprint analysis for each EC site was also performed. Results showed that energy balance closure for the flux data was reasonably good, with average energy balance ratio (EBR) of 1.03. The seasonal variations in net radiation (Rn), latent (LE), and sensible heat flux (H) over the five contrasting surfaces were similar, and a reverse seasonal change was observed in energy partitioning into LE and H. Remarkable differences in Rn, LE, and H between the five surfaces were explored preliminarily, associated closely with the soil properties and foliage phenology. Over the growing season (May–October) in 2014, the total ET ranged 622–731 mm for mixed forest of P. euphratica trees with associated T. chinensis shrubs with average daily ET of 3.6–4.2 mm; ET from T. chinensis shrubland was about 541 mm, with average daily ET of 3.6 mm. ET for barren-land was 195 mm. The total ET in irrigated cantaloupe cropland with plastic mulch was 431 mm for its four-month growing period with a total average of 3.8 mm d−1. Determination of ET over riparian forest ecosystem helps to improve reasonable use of limited water resource in the Ejina Oasis.

Purpose: To examine diagnostic performance of qualitative shear wave elastography (SWE) for evaluation of status of axillary lymph nodes (ALN) in comparison with conventional ultrasonograghy (US) and quantitative SWE parameters.Methods: A total of 118 patients were enrolled, who were all scheduled for breast cancer surgery and core needle biopsy. Conventional US and SWE were performed before biopsy. Based on qualitative evaluation of each ALN, the SWE images were classified into four color patterns: Color Pattern 1: homogeneous; Color Pattern 2: filling defect within lymph node (LN); Color Pattern 3: homogeneous within LN with a localized colored area at the margin; and Color Pattern 4: filling defect within LN with a localized colored area at the margin. The diagnostic performances of the three methods were compared.Results: There were 60 metastatic nodes and 61 benign nodes in the 121 ALNs. Benign ALNs were presented as Color Pattern 1 while metastatic ALNs usually were presented as Color Pattern 2 to 4 (p < 0.05). Research on land surface processes at the catchment scale has drawn much attention over the past few decades, and a number of watershed observatories have been established worldwide. The Heihe River Basin (HRB), which contains the second largest inland river in China, is an ideal natural field experimental area for investigation of land surface processes involving diverse landscapes and the coexistence of cold and arid regions. The Heihe Integrated Observatory Network was established in 2007. For long-term observations, a hydrometeorological observatory, ecohydrological wireless sensor network, and satellite remote sensing are now in operation. In 2012, a multiscale observation experiment on evapotranspiration over heterogeneous land surfaces was conducted in the midstream region of the HRB, which included a flux observation matrix, wireless sensor network, airborne remote sensing, and synchronized ground measurements. Under an open data policy, the datasets have been publicly released following careful data processing and quality control. The outcomes highlight the integrated research on land surface processes in the HRB and include observed trends, scaling methods, high spatiotemporal resolution remote sensing products, and model–data integration in the HRB, all of which are helpful to other endorheic basins in the “Silk Road Economic Belt.” Henceforth, the goal of the Heihe Integrated Observatory Network is to develop an intelligent monitoring system that incorporates ground-based observatory networks, unmanned aerial vehicles, and multi-source satellites through the Internet of Things technology. Furthermore, biogeochemical processes observation will be improved, and the study of integrating ground observations, remote sensing, and large-scale models will be promoted further.

Botrytis cinerea is a destructive necrotrophic pathogen that can infect many plant species. The control of gray mold mainly relies on the application of fungicides, and the fungicide fludioxonil is widely used in China. However, the field fungicide resistance of B. cinerea to this compound is largely unknown. In this study, B. cinerea isolates were collected from different districts of Shanghai province in 2015–2017, and their sensitivity to fludioxonil was determined. A total of 65 out of 187 field isolates (34.76%) were found to be resistant to fludioxonil, with 36 (19.25%) showing high resistance and 29 (15.51%) showing moderate resistance. Most of these resistant isolates also showed resistance to iprodione, and some developed resistance to fungicides of other modes of action. AtrB gene expression, an indicator of MDR1 and MDR1h phenotypes, was not dramatically increased in the tested resistant isolates. Biological characteristics and osmotic sensitivity investigations showed that the fitness of resistant isolates was lower than that of sensitive ones. To investigate the molecular resistance mechanisms of B. cinerea to fludioxonil, the Bos1 amino acid sequences were compared between resistant and sensitive isolates. Resistant isolates revealed either no amino acid variations or the mutations I365S, I365N, Q369P/N373S, and N373S.

Downward surface shortwave radiation (DSSR) plays an important role in the energy balance of the earth’s surface. Accurate estimate of DSSR is of great significance for the rational and effective use of solar energy. Some parameterization schemes were proposed to estimate DSSR using meteorological measurements given ground-based radiation observation sites are scare and uneven. With the development of remote sensing technique, remotely sensed data can be applied to obtain continuous DSSR in space. Commonly, the spatial resolution of most radiation products is relatively low and cannot meet the needs of certain fields. Moreover, some retrieval algorithms based on the radiation transfer models are complicated for non-professionals. In this study, a back-propagation (BP) neural network method with Levenberg–Marquardt (LM) algorithm (hereafter referred to as LM-BP) was applied to predict DSSR by building the relationship between measured DSSR and high-resolution remote sensing data from the Advanced Space-borne Thermal Emission Reflectance Radiometer (ASTER). The DSSR observations from the four-component radiation sensor installed at the land covered by vegetable, village, maize, orchard, Gobi, sandy desert, desert steppe, and wetland were used to validate the model estimates. The results showed that the estimates of DSSR from LM-BP agreed well with the site measurements, with the root mean square error (RMSE) and the mean bias error (MBE) values of 27.34 W/m2 and −1.59 W/m2, respectively. This indicates that by combining the LM-BP network model and ASTER images can obtain precise DSSR in heterogenous surface. The DSSR results of this study can provide accurate high-spatial resolution input data for hydrological, evapotranspiration, and crop models.

A Zr4+-doped anatase TiO2 nanotube array electrode was prepared using a process that included Ti anodizing, chemical immersion, and heat treatment. The compositions, microstructure, and electrochemical properties of the prepared electrodes were characterized. The results show that Zr4+ was successfully introduced into the TiO2 nanotube array electrodes. Because Zr4+ was doped into the crystal structure of the TiO2and replaced a part of Ti4+ to form more oxygen vacancies and Ti3+, the electrocatalytic activity of the prepared electrodes, for the reduction of L-cystine, was significantly improved.
