Abstract

Japonica rice variety Kanto 194 as the donor of resistance gene Stv-bi and low-amylose content gene Wx-mq was used to improve the resistance and eating quality of Wuyujing 3 by the breeding strategy of backcross. In continuous backcross and selfcross generations, the related molecular markers with Stv-bi and Wx-mq genes were utilized for genotypic detection by associated with resistance identification of rice stripe disease and agronomic traits selection. Finally, 10 improved lines with homozygous genotype Stv-biStv-biWx-mqWx-mq were obtained from BC3F4 generation. The results of comparative analysis indicated that most characters of these lines were consistent with those of recipient parent Wuyujing 3, except for the improved resistance, appearance and eating quality. By evaluation of the comprehensive performance of them, two excellent lines K01 and K04 were selected for further experiments.

Abstract

GW2 is an important gene that regulates grain width and weight. We used cDNA clone to obtain the sequences of GW2 from large- and small-grained rice varieties, TD70 and Kasalath, respectively. Then, we developed a dCAPS (derived cleaved amplified polymorphic sequence) marker on the basis of the sequence difference between functional and nonfunctional GW2 genes to analyze the genotypes and phenotypes of recombinant inbred lines. Results showed that the sequence of GW2TD70 had a single nucleotide deletion at site 316 that generates a termination codon. This codon terminated the GW2 protein in advance. By contrast, the sequence of GW2Kasalath encoded an intact protein. A novel dCAPS marker was designed in accordance with a base A deletion at site 316 of the sequence. After the PCR product was digested by ApoI, TD70 showed 21 and 30 bp fragments, and Kasalath showed a 51 bp fragment. Up to 82 lines contained GW2TD70, and 158 lines contained GW2Kasalath. The lines that contained TD70 alleles displayed substantial increases in width and 1000-grain weight. This result suggested that GW2 played a critical role in rice breeding.

11. Photosynthetic physiological characteristics of high yield super rice variety Nanjing 5718.

Abstract

Nanjing 5718 is a rice variety with good eating quality approved by Jiangsu province in 2019 and was confirmed as super rice by the Ministry of Agriculture and Rural affairs in 2021. Studying its yield composition characteristics, photosynthetic characteristics, and physiological basis can be helpful to investigate photosynthetic traits of super rice variety, improve its photo- synthetic performance through genetic improvement, and provide the theoretical basis for high-yield breeding and cultivation of japonica rice. In this study, the parents of Nanjing 5718 and Huaidao 5, a widely popularized rice variety with the same growth period type, as the control, dry weight of aboveground of plant, chlorophyll contents, photosynthetic rates, and photochemical characteristics of photosystem II (PSII), core antenna protein expression, photosynthetic enzyme activities, and chloroplast ultrastructure were investigated in Nanjing 5718 from full expansion of flag leaves at the beginning of booting to 35 days after flowering. The results showed the pigment contents of the leaves in Nanjing 5718 were higher, which were close to male parent Yanjing 608. The net photosynthetic rates, dry weight, and PSII electron transfer activities of flag leaves at the late growth stage were significantly higher in Nanjing 5718 than those in parents and Huaidao 5. The light energy conversion performance of PSII was superior. The core antenna protein CP43 and CP47 were more stable under strong light and high temperature, and their adjustment ability of Nanjing 5718 was better than that of parents and Huaidao 5. In addition, the activities of RuBP carboxylase in flag leaves were higher, the contents of photosynthetic products were higher, the chloroplast structure was more stable, the senescence characteristics appeared later, and the chloroplast decay rates were slower in Nanjing 5718, compared with parents and Huaidao 5. The results indicated that high photosynthetic performance of flag leaves was the basis for the formation of high yield characteristics of super rice Nanjing 5718 with large panicles. The great activity and function of photosystem proteins were main factors of improving leaf photosynthesis, and the stability of chloroplast structure could provide powerful support for high photosynthetic efficiency of leaves. Abstract

Aims: Irritable bowel syndrome (IBS) is a common functional gastrointestinal disease characterized by abdominal pain. Our recent study has shown that the acid‐sensitive ion channel 1 (ASIC1) in dorsal root ganglion (DRG) is involved in stomachache of adult offspring rats subjected with prenatal maternal stress (PMS). MiR‐485 is predicted to target the expression of ASIC1. The aim of the present study was designed to determine whether miR‐485/ASIC1 signaling participates in enterodynia in the spinal dorsal horn of adult offspring rats with PMS. Methods: Enterodynia was measured by colorectal distension (CRD). Western blotting, qPCR, and in situ hybridization were performed to detect the expression of ASICs and related miRNAs. Spinal synaptic transmission was also recorded by patch clamping. Results: PMS offspring rats showed that spinal ASIC1 protein expression and synaptic transmission were significantly enhanced. Administration of ASICs antagonist amiloride suppressed the synaptic transmission and enterodynia. Besides, PMS induced a significant reduction in the expression of miR‐485. Upregulating the expression markedly attenuated enterodynia, reversed the increase in ASIC1 protein and synaptic transmission. Furthermore, ASIC1 and miR‐485 were co‐expressed in NeuN‐positive spinal dorsal horn neurons. Conclusions: Overall, these data suggested that miR‐485 participated in enterodynia in PMS offspring, which is likely mediated by the enhanced ASIC1 activities. [ABSTRACT FROM AUTHOR]

26. Analysis of Quantitative Trait Loci for Resistance to Rice False Smut

Abstract

A recombinant inbred line (RILs) population with 157 lines derived from an inter-subspecies cross of Daguandao/IR28 by the single seed descent method was used to detect quantitative trait loci (QTLs) conferring resistance to rice false smut (caused by Ustilaginoidea virens (Cooke) Takahashi) in 2007 and 2009. The disease rate index of the two parents and 157 RILs caused by rice false smut were scored and the QTLs for rice false smut resistance were detected accordingly by QTL Cartographer software. Seven QTLs controlling false smut resistance were detected on chromosomes 1, 2, 4, 8, 10, 11, and 12, respectively, with the phenotypic variance of 9.8-22.5%. There were five and four QTLs detected in 2007 and 2009, respectively, and only two QTLs were found in both two years, the phenotypic variation was explained by individual QTL ranged from 18.0% to 19.3% for these two QTLs, and the additive effects of these two QTLs contributed to the 8.0-16.3% decrease of disease index and therefore the disease resistance increased. The direction of the additive effects at six loci qFsr1, qFsr2, qFsr8, qFsr10, qFsr11, and qFsr12 coincided with that predicted by phenotypes of the parents, and the IR28 alleles at these loci had positive effect against rice false smut while the negative effects were found in Daguandao alleles at qFsr4. Both qFsr11 and qFsr12 should be useful in rice breeding for resistance to rice false smut in marker-assisted selection (MAS) program.

Abstract

This paper proposes an oxide filled extended trench gate super junction (SJ) MOSFET structure to meet the need of higher frequency power switches application. Compared with the conventional trench gate SJ MOSFET, new structure has the smaller input and output capacitances, and the remarkable improvements in the breakdown voltage, on-resistance and switching speed. Furthermore, the SJ in the new structure can be realized by the existing trench etching and shallow angle implantation, which offers more freedom to SJ MOSFET device design and fabrication.

31. Characterization of eating quality and starch properties of two Wxalleles japonicarice cultivars under different nitrogen treatments

Abstract

To understand the effect of nitrogen (N) fertilizer on rice (Oryza sativaL.) eating and cooking quality (ECQ). Here, we investigated the ECQ attributes, physicochemical foundation of ECQ, and amylopectin fine structure of two Waxy(Wx) alleles japonicarice cultivars Nanjing 9108 (NJ9108) and Huaidao 5 (HD5) under four N rates (0, 150, 300, and 450 kg ha−1). Sensory and pasting properties of the two cultivars varied depending on N rates. Compared with the control (0 kg ha−1), the overall eating quality and sensory value were significantly decreased under the N rates of 300 and 450 kg ha−1. Further, conventional descriptive analysis showed that the stickiness and retrogradation of cooked rice were significantly decreased. These results indicated that application of N fertilizer seems to affect the texture of cooked rice, causing it to be less sticky, lowering its retrogradation, and consequently reducing its palatability. Results from rapid visco analyzer (RVA) revealed that the peak and breakdown viscosities significantly decreased, while the setback viscosity and peak time increased under the N rates of 300 and 450 kg ha−1. However, no significant difference was observed when the N rate was 150 kg ha−1, indicating that less N fertilization can maintain rice ECQ. As the N rate increasing, protein content increased, whereas apparent amylose content, starch content, and gel consistency almost unchanged. Interestingly, compared with the control, under N treatments, the percentage of short amylopectin branches in NJ9108 was decreased, but increased in HD5, as controlled by amylopectin synthesis-related genes. Notably, SSIand BEIIbwere down-regulated in NJ9108, whereas BEIIbwas up-regulated in HD5. Thus, the palatability of both rice cultivars was significantly decreased under excessive N fertilization as a consequence of reduced stickiness and retrogradation of the cooked rice, which might have resulted from an elevated protein content and altered amylopectin fine structure. In addition, amylopectin synthesis appeared to be affected by N fertilizer and the genotype of the rice cultivar.

Abstract

A novel type of p+(SiGe)–n−–n+ heterojunction switching power diode with high-speed capability is presented to overcome the drawbacks of existing power diodes. The improvement is achieved by using a p+–n+ mosaic layer as a substitute for the n+ region in the conventional p+(SiGe)–n−–n+ diode to realize an `ideal ohmic' contact for electrons and holes simultaneously. Compared with conventional p+(SiGe)–n−–n+ diodes, the ideal ohmic contact p+(SiGe)–n−–n+ diodes have about one third of the reverse recovery time and a half of peak reverse recovery current. Furthermore, the softness factor increases nearly two times and the leakage current decreases 1–2 orders of magnitude. These improvements are achieved without resorting special process step to lower the carrier lifetime and thus the devices could be easily integrated into power ICs. The Ge percentage content of p+(SiGe) layer is an important parameter for the optimal device design.

Abstract

In this study we report the results of a decade-long breeding program for japonicasuper rice made by Nanjing Branch of Chinese National Center for Rice Improvement in Jiangsu Academy of Agricultural Sciences. We concluded that selection of parents with good comprehensive traits and complementary advantages and disadvantages of both parents in the hybrid combination, and early selection of high heritability traits in earlier segregating generations could significantly improve the breeding efficiency. The use of closely-linked functional markers in pyramiding of multiple genes could greatly increase breeding efficiency, avoiding time-consuming and laborious steps that were used in traditional breeding program. It is also important to coordinate the yield components with variety characteristics such as yield stability, wide adaptability, lodging resistance, and an attractive grain appearance during late growth stage of rice.
