Traditional electric vehicle （EV） load forecasting methods often fail to fully utilize the spatial correlation among EV loads， resulting in low forecasting accuracy. To address this issue， a load forecasting method using graph convolution network-long short-term memory （GCN-LSTM） is proposed. Firstly， graph data is constructed to describe the distribution of charging stations in the region， and the spatial dependency information between the charging station under study and neighboring charging stations is extracted using a GCN. Secondly， the information extracted by the GCN at different time periods is formed into a time series and input into the LSTM to forecast the EV charging loads. Finally， the proposed algorithm is validated by using load data from charging stations in an urban area in China as an example. The results show that the proposed method can effectively improve forecasting accuracy.

In badminton, power production can be enhanced through the fundamental practice of a dynamic warm-up with resistance conditioning activity to induce a post-activation performance enhancement (PAPE) effect. The use of heavy resistance exercise in the form of heavy weights to induce PAPE during competition is not logistically practical in the badminton arena. Thus, there is a need to investigate the use of easily available alternative preconditioning stimuli to induce a similar potentiating effect in badminton-specific performance. This study adopted a repeated-measures design of three warm-up conditions: control (CON), weighted wearable resistance (WWR), and resistance band variable resistance (BVR). Fourteen badminton players from the national training squad (11 males, 3 females, age 18 ± 1 y) completed the experimental sessions in random order. Change of direction speed (CODS) and smash velocity (SV) tests were performed at five timepoints—baseline test after the warm-up and at the end of each of the four exercise blocks of a simulated match play protocol. CODS was significantly faster under the two resistance warm-up conditions (WWR and BVR) compared to the CON condition at baseline (−0.2 s ± 0.39 and −0.2 s ± 0.46, p = 0.001 and 0.03, g = 0.47 and 0.40, respectively), but there were no differences at the other timepoints (all p > 0.05). SV was significantly faster for all the four exercise blocks than at baseline under all three warm-up conditions (p = 0.02), but there were no differences in SV between the three warm-up conditions across all the five measured timepoints (p = 0.15). In conclusion, implementing resistance (~10% body weight) in sport-specific plyometric exercises using WWR or BVR during warm-up routines may induce PAPE effects on the change of direction speed but not smash velocity, in well-trained badminton players, as compared with the same warm-up exercises using bodyweight (i.e., CON condition). The positive effects of CODS were, however, observed only at the start of the match and possibly lasted for up to between 5 and 10 min of match play.

Seaweeds have been studied extensively for their nutritional values but their potential nutraceutical application remained underutilized due to uncharacterized bioactive compounds. Here we demonstrated that water extracts of Kappaphycus, Halimeda, Padina and Sargassum were able to improve insulin resistance, reduced hyperglycemia and protect liver and pancreatic tissue from HFD-induced damage in mice, with both Padina and Sargasssum displayed more significant results than the other two seaweeds. A list of potential bioactive compounds was then composed by virtual screening of 276 compounds detected by LC-MS on selected Padina fractions using molecular docking by Surflex-Dock. Further analysis determined punicate as the most potent bioactive compound that inhibits both glucosidase and dipeptidyl-peptidase-4 enzymes. In conclusion, we discovered novel in vivo hypoglycemic activity in Halimeda and several potential α-glucosidase and DPP-4 inhibitors in Padina via virtual screening, demonstrating the efficacy of molecular docking to facilitate discovery of novel bioactive compounds.

This study measured 1) the speed and spin differences between the old celluloid versus new plastic table tennis balls at pre ball-table impact and post ball-table impact when projected with topspin at 7.56 m.s-1, and investigated 2) the effect this has on the kinematic responses of 5 elite versus 5 sub-elite players’ forehand topspin in response to topspin and backspin. Plastic balls were lower in both speed and spin at pre and post ball-table impact compared with celluloid balls but the magnitude of change in speed and spin for each ball material differed. During flight before impact plastic balls lost 3.98% more speed and 1.24% more spin than celluloid balls. Post ball-table impact, plastic balls showed a greater speed increment (0.69%) and smaller spin decrement (0.19%) than celluloid balls. Differences in players’ kinematic responses to the different ball materials were found only when players returned backspin shots. Players supinated their rackets more by 2.23% at ball-racket contact and produced 3.37% less ball spin when returning plastic compared with celluloid balls; an indication of an early adaptation to the lower spin rate of plastic balls. The lack of differences in kinematic response to topspin may be due to the similar changes in speed and spin of both types of ball at ball-table impact. It is not known if a higher initial ball projection velocity would evoke differences in movement responses from the players post ball-table impact but could be explored in future studies.

The aim of this study was to compare the nonlinear properties of the four competitive swim strokes. Sixty-eight swimmers performed a set of maximal 4x25m using the four competitive swim strokes. The hip’s speed-data as a function of time was collected with a speedo-meter. The speed fluctuation (dv), approximate entropy (ApEn) and the fractal dimension by Higuchi’s method (D) were computed. Swimming data exhibited nonlinear properties that were different among the four strokes (14.048≤dv≤39.722; 0.682≤ApEn≤1.025; 1.823≤D≤1.919). Literacy builds a foundation for further learning and necessitates human capital development that creates a knowledge economy. A clear focus on literacy policies is fundamental in achieving national and international socio-economic targets. In Tanzania, adult literacy growth has been slow, inconsistent, and characterized by regional gaps. A review of policies and practices has revealed several constraints associated with budget allocation and cultural perspectives. We suggest to the governments in the whole developing world to prioritize consistent funding and formulating culture-sensitive policies in ensuring mass literacy, poverty reduction, creation of high productive personnel, and eventually achieve the UNESCO's Agenda 2030

Literacy is a fundamental aspect of development and a step towards freedom and liberation from socio-economic constraints, because it enables all citizens to realise their basic right to learning. However, the global efforts which -- according to a 2017 estimate of the United Nations Educational, Scientific and Cultural Organization (UNESCO) -- resulted in an increase in literacy from 55.7% in 1950 to 86.2% in 2015 at the world level (an average of 5% for each decade) did not significantly affect the actual number of people with low (or no) literacy skills. Due to rapid population growth, the number of people unable to read or write was higher in 2015 (745 million) than it was in 1950 (700 million), with some world regions being more affected than others. After reflecting on global historical trends in adult literacy education, this article focuses on the African continent, and on Tanzania in particular. The authors offer a few recommendations for developing countries to progress towards ensuring that all youth and a substantial proportion of adults, both men and women, achieve literacy and numeracy by 2030.

Bisected N-glycans represent a unique class of protein N-glycans that play critical roles in many biological processes. Herein, we describe the systematic synthesis of these structures. A bisected N-glycan hexasaccharide was chemically assembled with two orthogonal protecting groups attached at the C2 of the branching mannose residues, followed by sequential installation of GlcNAc and LacNAc building blocks to afford two asymmetric bisecting "cores". Subsequent enzymatic modular extension of the "cores" yielded a comprehensive library of biantennary N-glycans containing the bisecting GlcNAc and presenting 6 common glycan determinants in a combinatorial fashion. These bisected N-glycans and their non-bisected counterparts were used to construct a distinctive glycan microarray to study their recognition by a wide variety of glycan-binding proteins (GBPs), including plant lectins, animal lectins, and influenza A virus hemagglutinins. Significantly, the bisecting GlcNAc could bestow (PHA-L, rDCIR2), enhance (PHA-E), or abolish (ConA, GNL, anti-CD15s antibody, etc.) N-glycan recognition of specific GBPs, and is tolerated by many others. In summary, synthesized compounds and the unique glycan microarray provide ideal standards and tools for glycoanalysis and functional glycomic studies. The microarray data provide new information regarding the fine details of N-glycan recognition by GBPs, and in turn improve their applications.

Abstract Background The Shewanella oneidensis MR-1 genome encodes a predicted orphan DNA-binding response regulator, SO2426. Previous studies with a SO2426-deficient MR-1 strain suggested a putative functional role for SO2426 in the regulation of iron acquisition genes, in particular, the siderophore (hydroxamate) biosynthesis operon so3030-3031-3032. To further investigate the functional role of SO2426 in iron homeostasis, we employed computational strategies to identify putative gene targets of SO2426 regulation and biochemical approaches to validate the participation of SO2426 in the control of siderophore biosynthesis in S. oneidensis MR-1. Results In silico prediction analyses revealed a single 14-bp consensus motif consisting of two tandem conserved pentamers (5'-CAAAA-3') in the upstream regulatory regions of 46 genes, which were shown previously to be significantly down-regulated in a so2426 deletion mutant. These genes included so3030 and so3032, members of an annotated siderophore biosynthetic operon in MR-1. Electrophoretic mobility shift assays demonstrated that the SO2426 protein binds to its motif in the operator region of so3030. A "short" form of SO2426, beginning with a methionine at position 11 (M11) of the originally annotated coding sequence for SO2426, was also functional in binding to its consensus motif, confirming previous 5' RACE results that suggested that amino acid M11 is the actual translation start codon for SO2426. Alignment of SO2426 orthologs from all sequenced Shewanella spp. showed a high degree of sequence conservation beginning at M11, in addition to conservation of a putative aspartyl phosphorylation residue and the helix-turn-helix (HTH) DNA-binding domain. Finally, the so2426 deletion mutant was unable to synthesize siderophores at wild-type rates upon exposure to the iron chelator 2,2'-dipyridyl. Conclusions Collectively, these data support the functional characterization of SO2426 as a positive regulator of siderophore-mediated iron acquisition and provide the first insight into a coordinate program of multiple regulatory schemes controlling iron homeostasis in S. oneidensis MR-1.

Abstract Background Shewanella oneidensis MR-1 exhibits diverse metal ion-reducing capabilities and thus is of potential utility as a bioremediation agent. Knowledge of the molecular components and regulatory mechanisms dictating cellular responses to heavy metal stress, however, remains incomplete. In a previous work, the S. oneidensis so2426 gene, annotated as a DNA-binding response regulator, was demonstrated to be specifically responsive at both the transcript and protein levels to acute chromate [Cr(VI)] challenge. To delineate the cellular function of SO2426 and its contribution to metal stress response, we integrated genetic and physiological approaches with a genome-wide screen for target gene candidates comprising the SO2426 regulon. Results Inactivation of so2426 by an in-frame deletion resulted in enhanced chromate sensitivity and a reduced capacity to remove extracellular Cr(VI) relative to the parental strain. Time-resolved microarray analysis was used to compare transcriptomic profiles of wild-type and SO2426-deficient mutant S. oneidensis under conditions of chromate exposure. In total, 841 genes (18% of the arrayed genome) were up- or downregulated at least twofold in the Δso2426 mutant for at least one of six time-point conditions. Hierarchical cluster analysis of temporal transcriptional profiles identified a distinct cluster (n = 46) comprised of co-ordinately regulated genes exhibiting significant downregulated expression (p < 0.05) over time. Thirteen of these genes encoded proteins associated with transport and binding functions, particularly those involved in Fe transport and homeostasis (e.g., siderophore biosynthetic enzymes, TonB-dependent receptors, and the iron-storage protein ferritin). A conserved hypothetical operon (so1188-so1189-so1190), previously identified as a potential target of Fur-mediated repression, as well as a putative bicyclomycin resistance gene (so2280) and cation efflux family protein gene (so2045) also were repressed in the so2426 deletion mutant. Furthermore, the temporal expression profiles of four regulatory genes including a cpxR homolog were perturbed in the chromate-challenged mutant. Conclusion Our findings suggest a previously unrecognized functional role for the response regulator SO2426 in the activation of genes required for siderophore-mediated Fe acquisition, Fe storage, and other cation transport mechanisms. SO2426 regulatory function is involved at a fundamental molecular level in the linkage between Fe homeostasis and the cellular response to chromate-induced stress in S. oneidensis.

Abstract Background Serotypes of the Foot-and-Mouth disease viruses (FMDVs) were generally determined by biological experiments. The computational genotyping is not well studied even with the availability of whole viral genomes, due to uneven evolution among genes as well as frequent genetic recombination. Naively using sequence comparison for genotyping is only able to achieve a limited extent of success. Results We used 129 FMDV strains with known serotype as training strains to select as many as 140 most serotype-specific nucleotide strings. We then constructed a linear-kernel Support Vector Machine classifier using these 140 strings. Under the leave-one-out cross validation scheme, this classifier was able to assign correct serotype to 127 of these 129 strains, achieving 98.45% accuracy. It also assigned serotype correctly to an independent test set of 83 other FMDV strains downloaded separately from NCBI GenBank. Conclusion Computational genotyping is much faster and much cheaper than the wet-lab based biological experiments, upon the availability of the detailed molecular sequences. The high accuracy of our proposed method suggests the potential of utilizing a few signature nucleotide strings instead of whole genomes to determine the serotypes of novel FMDV strains.

Abstract Background Codon usage bias has been widely reported to correlate with GC composition. However, the quantitative relationship between codon usage bias and GC composition across species has not been reported. Results Based on an informatics method (SCUO) we developed previously using Shannon informational theory and maximum entropy theory, we investigated the quantitative relationship between codon usage bias and GC composition. The regression based on 70 bacterial and 16 archaeal genomes showed that in bacteria, SCUO = -2.06 * GC3 + 2.05*(GC3)2 + 0.65, r = 0.91, and that in archaea, SCUO = -1.79 * GC3 + 1.85*(GC3)2 + 0.56, r = 0.89. We developed an analytical model to quantify synonymous codon usage bias by GC compositions based on SCUO. The parameters within this model were inferred by inspecting the relationship between codon usage bias and GC composition across 70 bacterial and 16 archaeal genomes. We further simplified this relationship using only GC3. This simple model was supported by computational simulation. Conclusions The synonymous codon usage bias could be simply expressed as 1+ (p/2)log2(p/2) + ((1-p)/2)log2((l-p)/2), where p = GC3. Purpose: To compare the accuracy of intraocular lens (IOL) power calculation formulas in cataract patients with keratoconus (KC). Methods: This study followed the Preferred Reporting Items for Systematic Reviews and Meta-analysis statementand and was registered on PROSPERO (CRD42024568997). Pubmed, Web of Science, Cochrane Library, and EMBASE were searched for retrospective and prospective clinical studies published until October 2024. The outcome measurement was the percentage of eyes with a predicted error (PE) within ± 0.50 or ± 1.00 diopter (D). Results: The study have nine retrospective clinical trials, involving a total of 637 eyes and 18 calculation formulas. According to the ranking based on the surface under the cumulative ranking curve by Bayesian method, the top three formulas were Barrett True-K formula for keratoconus predicted posterior corneal astigmatism (Barrett KC P-PCA), EVO2.0, and Barrett True-K formula for keratoconus measured posterior corneal astigmatism (Barrett KC M-PCA) on the percentage of PE within ± 0.50 D, and the comparison between the three formulas and Barrett Universal II formula has statistical significance. In the range of ± 1.00D, the top three formulas were Barrett KC P-PCA, Barrett KC M-PCA and Kane for keratoconus formula, and the difference was significant. Thereforewe recommend using the Barrett KC P-PCA formula and the Barrett KC M-PCA formula for calculating IOL power in cataract patients with KC. Conclusion: This study revealed that the KC-specific IOL formulas, notably the Barrett KC P-PCA and Barrett KC M-PCA formulas, demonstrated superior accuracy. In clinical practice, when managing patients with different degrees of KC, surgeons should take into account the individual characteristics of each patient and adopt multiple formulas to improve the accuracy of refractive prediction.
