2. Genetic and Pharmacologic Inhibition of Complement Impairs Endothelial Cell Function and Ablates Ovarian Cancer Neovascularization

Complement activation plays a critical role in controlling inflammatory responses. To assess the role of complement during ovarian cancer progression, we crossed two strains of mice with genetic complement deficiencies with transgenic mice that develop epithelial ovarian cancer (TgMISIIR-TAg). TgMISIIR-TAg mice fully or partially deficient for complement factor 3 (C3) (Tg+C3KO and Tg+C3HET, respectively) or fully deficient for complement factor C5a receptor (C5aR) (Tg+C5aRKO) develop either no ovarian tumors or tumors that were small and poorly vascularized compared to wild-type littermates (Tg+C3WT, Tg+C5aRWT). The percentage of tumor infiltrating immune cells in Tg+C3HET tumors compared to Tg+C3WT controls was either similar (macrophages, B cells, myeloid-derived suppressor cells), elevated (effector T cells), or decreased (regulatory T cells). Regardless of these ratios, cytokine production by immune cells taken from Tg+C3HET tumors was reduced on stimulation compared to Tg+C3WT controls. Interestingly, CD31+ endothelial cell (EC) function in angiogenesis was significantly impaired in both C3KO and C5aRKO mice. Further, using the C5aR antagonist PMX53, tube formation of ECs was shown to be C5a-dependent, possibly through interactions with the VEGF165 but not VEGF121 isoform. Finally, the mouse VEGF164 transcript was underexpressed in C3KO livers compare to C3WT livers. Thus, we conclude that complement inhibition blocks tumor outgrowth by altering EC function and VEGF165 expression.

El objetivo del presente trabajo fue evaluar tres métodos estadísticos para el análisis de variables discretas. La información empleada proviene de un experimento desarrollado en la Empresa Genética Camilo Cienfuegos, de la provincia de Pinar del Río en el período 2007-2008, relacionada con la producción de CT-115. Se analizaron tres muestreos, como caso de estudio se seleccionó el muestreo dos que comprendió los meses junio-julio 2007, se empleó un diseño completamente aleatorizado con tres tratamientos y 10 repeticiones. Las variables analizadas fueron: No. tallos, No. rebrotes, No. hojas totales/tallos, No. hojas totales/rebrotes, No. hojas secas/tallos y No. hojas secas/rebrotes. Se tuvo en cuenta el Análisis de Varianza paramétrico, su homólogo no paramétrico la dócima Kruskal¿Wallis y Modelo Lineal Generalizado. Se verificó el cumplimiento de los supuestos teóricos del Análisis de Varianza, para la normalidad de los errores se utilizaron las dócimas Shapiro Wilk, Kolmogorov Smirnov y Lilliefors, la dócima de Shapiro Wilk fue la más robusta para detectar la falta de normalidad, para la homogeneidad de varianza se emplearon las dócimas de Bartlett y Levene, ambas obtuvieron resultados similares. Las variables se transformaron según raíz cuadrada, la cual no mejoró el cumplimiento de distribución Normal para la variable No. hojas secas/rebrotes. Los valores de probabilidad obtenidos mantuvieron el mismo criterio de decisión con respecto a Ho para ambas dócimas, la no paramétrica Kruskal- Wallis comparado con su homóloga paramétrica la dócima F de Fisher. Los criterios de bondad de ajuste utilizados en el Modelo Lineal Generalizado permitieron conocer los efectos de mejor ajuste, Se considero que este modelo es más flexible que el Análisis de Varianza paramétrico, pues las variables en estudio no requiere del cumplimientos de los supuestos teóricos básicos.

8. ESP Root Cause Failure Analysis in Guatiquia Field, Colombia: A Case Study

This paper shows a detailed description about an uncommon electric submersible pump (ESP) failure in Candelilla-5 well. This catastrophic failure generated fishing operations to recover the well, and additional cost for the well service. Candelilla-5 is located at Guataquia block, in the Llanos Orientales basin in Colombia. It is an oil producer well, 600 BOPD, 19.7 °API, an ESP as artificial lift system, at depth of 9482 ft TVD, with runlife of 173 days. The ESP suffered a suddenly stuck pump shaft during the operation; several start attempts were made, with no success. It was determined by the given evidence in this and the neighboring wells, that the probable cause was the presence of calcium carbonate (CaCO3). The electrical measurement of the all ESP system was in good conditions. Aiming to remove the CaCO3, two barrels of hydrochloric acid (HCl) at 15% concentration was injected into the pump, but it did not work, so a rig service was programmed. During the pulling of ESP string, only the tubing string plus pump discharge head was recovered. All the screws of pump discharge head were broke and the rest of ESP remained into the well as a fish. A Root Cause Failure Analysis (RCFA) was developed. The following methodology was used: (1) fractography analysis that showed a fracture plane between head and screw's body, (2) elemental chemical analysis using optical emission technique with spark source to find the screw's composition, (3) macro-attack testing for detecting residual stresses, (4) microstructural analysis to characterize the screw's material and fractures (5) microhardness profile to identify any screw's hardening, (6) X- ray diffraction in direction to identify incrustation and corrosion products and (7) hydrogen evolution test using HCl solution at 7,5 % and 15%, with the purpose of determining the hydrogen effects on steel properties. Once the RCFA was successfully completed, it was determined that the screws were fractured mainly because of three reasons: (1) High tension zones created due to high stresses during pump service, (2) the thermally treated material and (3) the presence of an acid solution in the environment, HCl, inducing an accumulation of hydrogen on the high tension zones, giving as a result the screw's hydrogen embrittlement, phenomenon responsible for the failure of the ESP from Candenilla-5 well. This paper aims to give details of the methodology, analysis from the root cause of the failure, feedbacks and recommendations to improve operations development on ESP.

During January and February 2007 a European consumer test of eight new apple varieties and three standard ones was carried out in different European countries. The most common apple varieties were compared to new ones, focusing on consumer acceptance and the potential for increasing apple consumption. We sought to test the hypothesis that the introduction of new improved varieties would help to increase fruit consumption. Data from a sensory test involving 4290 consumers in seven different countries were analysed for the effects of variety and test country, age, gender and apple consumption habit of the consumer on eating quality acceptance. The results obtained showed a strong interaction between country and variety, although a group of four varieties (‘Gold Pink’, ‘CIVni’, ‘Cripps Pink’ and ‘Fuji’) formed part of a group of the varieties that were most accepted in all of the countries except Germany. Significant interactions between variety and age and variety and gender were observed, but their magnitudes were not considered to be of any practical significance. No statistically significant interaction was found between variety and apple consumption habit on eating quality acceptance.

Al analizar datos provenientes de poblaciones de plagas agricolas, regularmente se detecta que no cumplen los requerimientos teoricos para la aplicacion del ANDEVA clasico. El uso de transformaciones Box-Cox y de metodos estadisticos no parametricos resulta la alternativa mas utilizada para resolver este inconveniente. En el presente trabajo se exponen los resultados de la aplicacion de estas tecnicas a datos provenientes de Thrips palmi Karny muestreadas en plantaciones de papa (Solanum tuberosum L.) en el periodo de incidencia de la plaga. Se utilizo la docima X2 para la bondad de ajuste a la distribucion binomial negativa y de independencia para investigar la relacion entre los estratos de las plantas y los estados del insecto, se aplicaron siete transformaciones a los datos para satisfacer el cumplimiento de los supuestos basicos del ANDEVA, con las cuales no se logro eliminar la relacion entre la media y la varianza, por lo que se aplico el ANDEVA no parametrico de Kruskal-Wallis para comparar las densidades poblacionales del insecto. Los resultados permitieron seleccionar el estado larval del insecto y el estrato medio de la planta como elementos claves para disenar planes de muestreo para esta plaga.

Analyzing data from agricultural pest populations regularly detects that they do not fulfill the theoretical requirements to implement classical ANOVA. Box-Cox transformations and nonparametric statistical methods are commonly used as alternatives to solve this problem. In this paper, we describe the results of applying these techniques to data from Thrips palmi Karny sampled in potato (Solanum tuberosum L.) plantations. The X² test was used for the goodness-of-fit of negative binomial distribution and as a test of independence to investigate the relationship between plant strata and insect stages. Seven data transformations were also applied to meet the requirements of classical ANOVA, which failed to eliminate the relationship between mean and variance. Given this negative result, comparisons between insect population densities were made using the nonparametric Kruskal-Wallis ANOVA test. Results from this analysis allowed selecting the insect larval stage and plant middle stratum as keys to design pest sampling plans.Al analizar datos provenientes de poblaciones de plagas agrícolas, regularmente se detecta que no cumplen los requerimientos teóricos para la aplicación del ANDEVA clásico. Mature apples of 'Fuji' and 'Fuji' sports sourced from around the world contain unusual clumps of multicellular branched callus hairs between the parenchyma cells of the outer cortex. The hairs are similar morphologically to the white tufts found in the seed locules of some cultivars and to the aerial cells of apple callus grown on solid culture medium. Callus hairs were found to be particularly well developed in 'Fuji Suprema' from Brazil, and least developed in 'Fuji' from China. Hairs are also found in fruit of close relatives of 'Fuji' and some unrelated cultivars. Callus hairs proliferate in intercellular air spaces and also in larger cavities or lacunae which may have poor connectivity with other air spaces. In these locations they have the potential to restrict or modify the flow of oxygen and carbon dioxide in the outer part of apples, and thus may be one of the factors contributing to internal browning, a post-harvest disorder to which large or late-picked 'Fuji' in modified atmosphere storage are known to be vulnerable. The callus hairs are found within 17 mm of the skin and frequently contain starch even when starch has been metabolised from the surrounding parenchyma during ripening. The vacuoles of callus hairs are rich in autofluorescent compounds and may accumulate red pigment, particularly when growing close to the skin of deep red 'Fuji Suprema'. Initial observations with immunogold labelling have detected low levels of the allergen Mal d 3 associated with the plasmalemma of callus hairs. In corking disorders, callus hairs can invade necrotic cells and proliferate inside them.

16. Prevalencia Del Virus Del Papiloma Humano (VPH) y Otros Factores De Riesgo Para El Desarrollo De Cáncer Cervical En Guatemala

El cáncer cervical es la malignidad más común entre las mujeres del tercer mundo y en Guatemala, es responsable del 40% de todos los cánceres en ambos sexos y del 60% de todos los cánceres femeninos, La presencia de Virus del Papiloma Humano (VPH) se considera entre otras, como el factor de riesgo más importante para el desarrollo del cáncer cervical. El presente estudio fue llevado a cabo con el propósito de determinar la prevalencia de los tipos de VPH en un grupo de pacientes de Guatemala con cáncer cervical así como para evaluar los factores de riesgo asociados al desarrollo de la enfermedad. Con ese propósito se llevó a cabo un estudio de casos y controles en 112 pacientes con cáncer cervical invasivo (casos) y 102 mujeres sanas (controles). Se obtuvieron biopsias cervicales y se investigó la presencia de, VPH usando dos métodos moleculares, captura de híbridos 2 (HC-2™. Digene Co.) y la reacción en cadena de la polimerasa (PCR). Se utilizó un cuestionario epidemiológico para evaluar otros factores de riesgo agrupados en factores de comportamiento sexual, factores reproductivos y factores sociodemográficos. La información fue recolectada durante una entrevista personal con los sujetos de estudio. Un total de 89% de las pacientes y tin 18% de los controles fueron positivos para ADN del Virus del Papiloma Humano y los tipos detectados con mayor frecuencia fueron el VPH 16, 18 y 45. La evaluación de factores de riesgo demostró valores de p de tendencia significativos (mayor riesgo de desarrollar cáncer cervical) para factores como: edad temprana de inicio de relaciones sexuales, número elevado de compañeros sexuales durante toda la vida y compañeros estables, elevado número de embarazos y partos vaginales, baja escolaridad y un origen étnico categorizado como indígena, al contrario de otros como el control prenatal durante el último embarazo, el cual es un factor que reduce el riesgo.

Alguns compostos de platina (II), como por exemplo, cisplatina, carboplatina e oxaloplatina, têm sido utilizados como agentes quimioterápicos no tratamento de vários tipos de câncer. Contudo, na tentativa de reduzir a toxicidade e ampliar o espectro de atividade da cisplatina e de seus análogos, milhares de complexos têm sido preparados variando a natureza dos ligantes. Este trabalho descreve a síntese e caracterização de novos complexos de platina, tendo iodeto e derivados do furano como ligantes. Os complexos foram caracterizados por RMN, IV e análise elementar.Platinum (II) compounds, for example, cisplatin, carboplatin and oxaloplatin, have been used as chemotherapeutic agents for the treatment of various types of cancer. However, in an attempt to reduce toxicity and widen the spectrum of activity of cisplatin and its analogous, thousands of platinum complexes have been prepared by varying the nature of the leaving groups and the carrier ligands. This work reports the synthesis and characterization of novel structural types of the platinum compounds, having iodide and furan derivatives as ligands. The compounds were characterized using NMR, IR and elemental analysis.

A consumer test carried out in 7 different European countries compared 3 standard apple varieties to 8 new ones. A total of 4290 consumers took part in the test. Data from this test was used to develop a preference map for apple. The preference map was constructed with 3 main dimensions (1 – sweetness, fruitiness, flowery attributes, 2 – acidity, firmness, 3 – juiciness and crispness). Consumers were segmented in 6 clusters according to their preferences. The 6 clusters were grouped into two main mega clusters A (68% of consumers) and B (32% of consumers). Megacluster A (Clusters 1, 2, 5 and 6) was characterized by preferring sweet apples. Clusters 2 and 5 (41% of consumers) liked sweet apples independently of their acidity and firmness and moderate positive values on dimension of juiciness and crispness. Cluster 1 (21% of consumers) had an optimal point in positive values of the sweetness dimension, moderate negative value for acidity and firmness and moderate positive value for juiciness and crispness. Cluster 6 (6% of consumers) besides preferring sweet varieties disliked acid-firm varieties. As to regard to megacluster B (Clusters 3 and 4) (32% of consumers), they preferred varieties that were acidic-firm and juiciy and crisp with values in the mid range of the sweetness dimension. In spite of the difficulties in translating preference dimensions into standard practical values for fruit quality and the fact of being a punctual measurement of consumer behaviour, this preference map could be of practical use of different actors on the fruit value chain like marketers and breeders.

This note implements an unstructured decaying product matrix via the quasi-least squares approach for estimation of the correlation parameters in the framework of generalized estimating equations. The structure we consider is fairly general without requiring the large number of parameters that are involved in a fully unstructured matrix. It is straightforward to show that the quasi-least squares estimators of the correlation parameters yield feasible values for the unstructured decaying product structure. Furthermore, subject to conditions that are easily checked, the quasi-least squares estimators are valid for longitudinal Bernoulli data. We demonstrate implementation of the structure in a longitudinal clinical trial with both a continuous and binary outcome variable.

Given the increasing travel of pregnant women from areas were Trypanosoma cruzi is endemic, the congenital transmission of the parasite has become a global public-health problem. In a recent pilot study, which ran in Chile from 2006 to 2010, three strategies for exploring and managing T. cruzi-infected mothers and their infected or uninfected neonates were investigated. Any protocols applied to the investigation of such mother-and-child pairs need to include the detection of infection in pregnant women, the detection of infection, if any, in the children born to the women, the appropriate treatment of the infected neonates, and the serological–parasitological follow-up of all of the neonates until their medical discharge.

The aim of this study was to gain information concerning apple and peach consumption frequency within different European countries in relation to age and gender. The survey was a part of a complex experiment with the aim of evaluating consumers’ preferences towards new varieties, and the data is based on the self-reported declarations of respondents, male and female, between 15 and 70 years old. 4271 consumers from 7 European countries were invited to supply information about their apple consumption habits, whereas 499 respondents from 5 countries answered questions relating to frequency of peach and nectarine consumption. In both, the apple and the peach surveys, data analysis of declared intake showed significant differences between nationalities. The highest apple consumption was in Poland, where over 55% declared a consumption of more than 5 apples per week. In comparison, Italian consumers most often indicated eating 3–5 apples per week (39.3%). The lowest apple consumption was in the Netherlands and Spain. In the case of peaches, the highest consumption was indicated in France where 48% of respondents declared a peach consumption of 3–5 per week with 40% eating more than 5 fruits per week. The lowest peach intake was declared in Germany. Irrespective of country women were shown to eat more apples that men. Furthermore, the group of older people (61–70 years) consume apples more often than the adult group (36–60), while within the youngest group of consumers (16–35) eating apples was not at all popular. As with apples females declared a higher peach consumption, and again significantly lower fruit consumption by the youngest group (16–35) was indicated. Although the availability of fruit at the market remains a prime factor in determining apple and peach consumption, our survey confirmed the trends of declining this popular fruit intake by the younger generation, as well as the persistent tendency of lower frequency of fruit consumption among men than women.

This paper aims at presenting the development and implementation of an approach to deal with fault location in transmission systems by Evolutionary Algorithms. The algorithm allows for the estimation of voltage sags and swells in non- monitored buses as well. An Evolutionary Strategy was applied to address the optimization problem, from the generation of a population of individuals that represent possible solutions to the problem to the evolutionary operators such as selection, mutation and crossover. Some variants of the basic approach were also considered in such a way to determine the fault location with better performance. A case study, based on the 30 bus IEEE study system, demonstrates the potential of the proposed methodology.

26. Ionic modulation of triethyllead neurotoxicity in cerebellar granule cell culture

While the molecular mechanism underlying triethyllead (TEL) neurotoxicity is unknown, we hypothesize that triethyllead mediates an accelerated Cl-/OH exchange across neuronal membranes leading to prolonged depolarization and neuronal cell injury. As a test of this hypothesis we have investigated the effect of external ion modulation on triethyllead neurotoxicity in cerebellar granule cell culture. Cultures were prepared from neonatal rats and used 10-20 days in vitro. Cytotoxicity was assessed by lactate dehydrogenase (LDH) release and trypan blue exclusion. A slow, dose-dependent (1-30 microM TEL) release of LDH occurred after a variable latent period dependent upon [TEL]. External replacement of [Cl-]e by Na isothionate dramatically shifted the dose response curve to the left reflecting an accelerated stimulation of LDH release, while replacement of extracellular [Na+]e with equimolar choline chloride had a minimal protective effect. Similarly, high [Mg2+]e or low [Ca2+]e did not protect or potentiate TEL cytotoxicity. The low [Cl-]e accelerated TEL cytotoxicity was dependent on medium pH: alkaline pH potentiated the cytotoxicity. Low [Cl-]e had no significant effect on culture ATP over 5 hrs. ATP reduction was markedly stimulated by TEL in low Cl- medium in contrast to the minimal decline in [ATP] in the control medium. The reduction of ATP in the low [Cl-]e medium occurred prior to LDH or trypan blue staining release confirming that such reduction in [ATP] was not secondary to cell damage. Substituting K sulfate or Na sulfate for the Cl(-)-free medium revealed marked loss of ATP without LDH release in control and TEL supplemented cultures. These observations provide supporting evidence for the role of an abnormal Cl- flux in mediating TEL-induced neurotoxic injury. Specifically, the membrane depolarization is proportional to the gradient imposed by Cl- efflux/OH influx, stimulated by low [Cl-]e. The rapid loss in ATP appeared early, was not a secondary reflection of neuronal damage but a result of a combination of increased ion flux at the plasma membrane, stimulation of Na+/K+ ATPase and direct TEL-induced inhibition of mitochondrial oxidative phosphorylation.

El objetivo del estudio fue evaluar el efecto del agua de arroz con zanahoria en comparación con el agua de arroz sobre la duración de la diarrea, número de deposiciones y la consistencia de las heces en el tratamiento de la diarrea aguda en niños. Se realizó en una población constituida por 22 niños entre 6 y 60 meses de edad internados en el Hospital Nacional Pedro de Bethancourt de la Antigua Guatemala, 11 niños se asignaron aleatoriamente a un grupo control y 11 al experimental. Al grupo experimental se le administró agua de arroz con zanahoria y al control solo agua de arroz. Los niños estuvieron en un período de observación mínimo de 48 y máximo de 72 horas, el volumen promedio de agua de arroz y agua de arroz con zanahoria administrado fue de 32 onzas a cada niño, los niños con lactancia materna la continuaron a libre demanda, los niños destetados o con lactancia mixta, que tomaban formula infantil se les siguió proporcionando en la forma habitual y los que recibían alimentos sólidos se les brindó la dieta del hospital. La edad promedio de los niños participantes en el estudio fue de un año con nueve meses en el grupo experimental y un año con ocho meses en el control. El 41 % de los niños del grupo experimental eran hombres y el 9.0 % mujeres, y en el control, el 27 % eran hombres y el 23 % mujeres.

Analyzing data from agricultural pest populations regularly detects that they do not fulfill the theoretical requirements to implement classical ANOVA. Box-Cox transformations and nonparametric statistical methods are commonly used as alternatives to solve this problem. In this paper, we describe the results of applying these techniques to data from Thrips palmi Karny sampled in potato (Solanum tuberosum L.) plantations. The χ⊃ test was used for the goodness-of-fit of negative binomial distribution and as a test of independence to investigate the relationship between plant strata and insect stages. Seven data transformations were also applied to meet the requirements of classical ANOVA, which failed to eliminate the relationship between mean and variance. Given this negative result, comparisons between insect population densities were made using the nonparametric Kruskal-Wallis ANOVA test. Results from this analysis allowed selecting the insect larval stage and plant middle stratum as keys to design pest sampling plans. [ABSTRACT FROM AUTHOR]

The objective of this article was to evaluate three different statistical methods to conduct analyses of discrete variables. The information came from an experiment developed at the Camilo Cienfuegos Genetics Enterprise in the Pinar del Río province in 2007-2008 related to the CT-115 forrage production. A complete randomized design was used with three treatments and ten repetitions. The variables analysed were: number of stems, number of sprouts, total number of leaves/stem, total number of leaves/sprout, number of dried leaves/stem and number of dried leaves/sprouts. The parametric variance analysis and its homologous non-parametric, Kruskal-Wallis test and the Generalized Lineal Model were taken into account. The theoretical assumptions of the variance analyses to the test error normality were verified. The Shapiro Wilk test, Kolmogorov Smirnov and the Lilliefors test were used, Shapiro Wilk test was the most robust to detect lack of normality. For the variance homogeneity, the Bartlett and Levene test were used both with similar results. The variables were transformed with the square root transformation which did not improve the normal distribution adjustment to the variable number of dried leaves/sprout. The probability values maintained the same outcomes respect to Ho tests for the non-parametric test, compared with its homologous parametric F from Fisher test. The criteria of goodness of fit in the Generalized Linear Model permitted evaluating the best adjustment effects. It was considered that this model is more flexible than the parametric variance analyses because the variables under study did not require the theoretical assumptions fulfilment. International audience; Key message: Proof of concept of Bayesian integrated QTL analyses across pedigree-related families from breeding programs of an outbreeding species. Results include QTL confidence intervals, individuals' genotype probabilities and genomic breeding values. Abstract: Bayesian QTL linkage mapping approaches offer the flexibility to study multiple full sib families with known pedigrees simultaneously. Such a joint analysis increases the probability of detecting these quantitative trait loci (QTL) and provide insight of the magnitude of QTL across different genetic backgrounds. Here, we present an improved Bayesian multi-QTL pedigree-based approach on an outcrossing species using progenies with different (complex) genetic relationships. Different modeling assumptions were studied in the QTL analyses, i.e., the a priori expected number of QTL varied and polygenic effects were considered. The inferences include number of QTL, additive QTL effect sizes and supporting credible intervals, posterior probabilities of QTL genotypes for all individuals in the dataset, and QTL-based as well as genome-wide breeding values. All these features have been implemented in the FlexQTL (TM) software. We analyzed fruit firmness in a large apple dataset that comprised 1,347 individuals forming 27 full sib families and their known ancestral pedigrees, with genotypes for 87 SSR markers on 17 chromosomes. We report strong or positive evidence for 14 QTL for fruit firmness on eight chromosomes, validating our approach as several of these QTL were reported previously, though dispersed over a series of studies based on single mapping populations. Interpretation of linked QTL was possible via individuals' QTL genotypes. The correlation between the genomic breeding values and phenotypes was on average 90 %, but varied with the number of detected QTL in a family. The detailed posterior knowledge on QTL of potential parents is critical for the efficiency of marker-assisted breeding.

Période 2000-2012. http://prodinra.inra.fr/record/256699; International audience; Genetic parameters for apple (Malus x domestica) fruit external traits (fruit size, ground colour, proportion of over colour and attractiveness) and sensory traits (firmness, crispness, texture, juiciness, flavour, sugar, acidity and global taste) were estimated using 2,207 pedigreed genotypes from breeding programmes in six European countries. Data were scored for 3 years and four periods during storage. Analyses were performed with a restricted maximum likelihood method using VCE 5.1.2 software. Heritability estimates ranged from medium to high for instrumental traits. Genetic correlations between firmness and sugar were medium and low between firmness and acidity. Sensory traits showed low to high heritability, acidity and flavour being, respectively, the most and the less heritable. Global taste was strongly correlated with texture, juiciness, and flavour and relatively less correlated with crispness and acidity. Sensory sugar and acidity showed highly negative correlations whereas their instrumental measurements showed low and increasing positive correlations from harvest to 4 months post-harvest. Sugar exhibited a higher sensory/instrumental divergence. Conversely, instrumental and sensory firmness were highly correlated. Fruit external characteristics had medium heritability. Fruit attractiveness had highest and lowest correlations with fruit size and ground colour, respectively. Best linear unbiased predictors of breeding values were computed for all genotypes with the software PEST. The results were analysed with regard to the dynamic and the reliability of genetic parameters according to the scoring dates. Original issues of the study and the importance of the obtained results for efficient designs of further apple fruit quality breeding programmes were discussed

Copper can be opportunely complexed to modulate oncogenic pathways, being a promising strategy for cancer treatment. Herein, three new copper(II) complexes containing long-chain aliphatic hydrazides and 1,10-phenanthroline (1,10-phen), namely, [Cu(octh)(1,10-phen)(H
2 O)](NO3 )2 1, [Cu(dech)(1,10-phen)(H2 O)](NO3 )2 2 and [Cu(dodh)(1,10-phen)(H2 O)](NO3 )2 .H2 O 3 (where octh = octanoic hydrazide, dech = decanoic hydrazide, dodh = dodecanoic hydrazide) were successfully prepared and characterized by several physical-chemical methods. Furthermore, X-ray structural analysis of complex 2 indicated that the geometry around the copper(II) ion is distorted square-pyramidal, in which hydrazide and 1,10-phenanthroline act as bidentate ligands. A water molecule in the apical position completes the coordination sphere of the metal ion. All new copper(II) complexes were cytotoxic to breast cancer cell lines (MCF7, MDA-MB-453, MDA-MB-231, and MDA-MB-157) and selective when compared to the non tumor lineage MCF-10A. In particular, complex 2 showed half-maximal inhibitory concentration (IC50 ) values ranging between 2.7 and 13.4 μM in MDA-MB231 cells after 24 and 48 h of treatment, respectively. Furthermore, this complex proved to be more selective for tumor cell lines when compared to doxorubicin and docetaxel. Complex 2 inhibited the clonogenicity of MDA-MB231 cells, increasing adenosine diphosphate (ADP) hydrolysis and upregulating ecto-nucleoside triphosphate diphosphohydrolase 1 (ENTPD1) transcriptional levels. In this sense, we suggest that the inhibitory effect on cell proliferation may be related to the modulation of adenosine monophosphate (AMP) levels. 37. Advertising and Eco-Labels as Influencers of Eco-Consumer Attitudes and Awareness-Case Study of Ecuador.

This study examined the impact of green advertising and eco-labels on the attitudes and environmental awareness of millennials purchasing eco-friendly products in shopping centers across Ecuador. The research utilized a quantitative, correlational, cross-sectional methodology with 430 millennials participating. A 20-item survey was administered face-to-face at shopping centers in Quito and Guayaquil, Ecuador. The validity of the research model was established through Confirmatory Factor Analysis (CFA) and Structural Equation Modeling (SEM), employing SPSS 20 and AMOS 24 for statistical evaluations. Findings reveal that green advertising significantly shapes environmental attitudes (β: 0.245) and awareness (β: 0.110), as well as directly influences the purchasing behavior (β: 0.154) towards green products. While eco-labels do not exert a direct effect on purchasing behavior (β: 0.128), they significantly inform attitudes (β: 0.406) and ecological awareness (β: 0.277) of millennials who purchase organic products. This paper is among the pioneering research to delineate the correlation between green advertising elements and the purchasing patterns of green products among millennials in a developing nation. It concludes that marketing strategies centered on green advertising and eco-labels do affect millennials' attitudes and environmental consciousness, but only advertising has a direct impact on purchasing behaviors, contrary to eco-labels. The research bears social significance as it affirms that millennials are attentive to environmental issues and are actively engaged in promoting sustainability.

- Abstract
Bismuth-based drugs have been used primarily to treat ulcers caused by Helicobacter pylori and other gastrointestinal ailments. Combined with antibiotics, these drugs also possess synergistic activity, making them ideal for multiple therapy regimens and overcoming bacterial resistance. Compounds based on bismuth have a low cost, are safe for human use, and some of them are also effective against tumoral cells, leishmaniasis, fungi, and viruses. However, these compounds have limited bioavailability in physiological environments. As a result, there is a growing interest in developing new bismuth compounds and approaches to overcome this challenge. Considering the beneficial properties of bismuth and the importance of discovering new drugs, this review focused on the last decade's updates involving bismuth compounds, especially those with potent activity and low toxicity, desirable characteristics for developing new drugs. In addition, bismuth-based compounds with dual activity were also highlighted, as well as their modes of action and structure-activity relationship, among other relevant discoveries. In this way, we hope this review provides a fertile ground for rationalizing new bismuth-based drugs.

This work describes the preparation, characterization and antimicrobial activity of four palladium(II) complexes, namely, [Pd(meg)(1,10-phen)] 1 , [Pd(meg)(PPh
3 )2 ] 2, [Pd(og)(1,10-phen)] 3 and [Pd(og)(PPh3 )2 ] 4 , where meg = methyl gallate, og = octyl gallate, 1,10-phen = 1,10-phenanthroline and PPh3 = triphenylphosphine. As to the chemical structures, spectral and physicochemical studies of 1 - 4 indicated that methyl or octyl gallate coordinates a palladium(II) ion through two oxygen atoms upon deprotonation. A chelating bidentate phenanthroline or two triphenylphosphine molecules complete the coordination sphere of palladium(II) ion, depending on the complex. The metal complexes were tested against the Mycobacterium tuberculosis H37Rv strain and 2 exhibited high activity (MIC = 3.28 μg/mL). As to the tests with Campylobacter jejuni , complex 1 showed a significant effect in reducing bacterial population (greater than 7 log CFU) in planktonic forms, as well as in the biomass intensity (IBF: 0.87) when compared to peracetic acid (IBF: 1.11) at a concentration of 400 μg/mL. The effect provided by these complexes has specificity according to the target microorganism and represent a promising alternative for the control of microorganisms of public health importance.- Published

As to the structure of these complexes, the X-ray structural analysis of 1 indicates that the geometry around the platinum(II) ion is distorted square-planar, where two 5-alkyl-1,3,4-oxadiazol-2-thione derivatives coordinate a platinum(II) ion through the sulfur atom. A chelating bidentate phenanthroline molecule completes the coordination sphere. We tested these complexes in two breast cancer cell lines, namely, MCF-7 (a hormone responsive cancer cell) and MDA-MB-231 (triple negative breast cancer cell). In both cells, the most lipophilic platinum compound, complex 2, was more active than cisplatin, one of the most widely used anticancer drugs nowadays. DNA binding studies indicated that such complexes are able to bind to ct-DNA with Kb values of 104 M-1 . According to data from dichroism circular and fluorescence spectroscopy, these complexes appear to bind to the DNA in a non-intercalative, probably via minor groove. Molecular docking followed by semiempirical simulations indicated that these complexes showed favorable interactions with the minor groove of the double helix of ct-DNA in an A-T rich region. This work describes the synthesis and characterization of two metal complexes of the type [M(L1)2(phen)], where M = Pt2+ (complex I) or Pd2+ (complex II), L1 = 5-amino-1,3,4-thiadiazole-2(3H)-thiolate and phen = 1,10-phenanthroline. The in vitro antibacterial activity of these complexes was investigated in isolation and synergistically with ciprofloxacin (CIP) and erythromycin (ERY) in three strains of Campylobacter jejuni (MIC = 32 mg/L for CIP and ERY), selected from a bank of 235 strains representative of three poultry exporting states of the country (A, B and C), previously analyzed for epidemiology and resistance to CIP and ERY. A total of 53/235 (22.55%) strains showed co-resistance to CIP and ERY. Isolated resistance to CIP was higher than to ERY. Epidemiological analysis showed that resistance to CIP was more evident in state B (p < 0.0001), as well as a higher susceptibility to ERY in state C (p = 0.0028). Co-resistance was expressive in state A and in the spring and fall seasons. The evaluation of I alone and in synergy with CIP and ERY found values up to 0.25 mg/L not significant for ERY. Complex II did not show an antimicrobial effect on the three strains of tested C. jejuni. The effect provided by complex I represents a promising alternative for control of resistant strains of C. jejuni.

Prostate Cancer (PCa) is the second leading cause of cancer-related deaths among men worldwide. The treatment of advanced cases is based on chemotherapy, which lacks specificity and efficacy, due to severe side effects and resistance to the traditional drugs. Copper complexes have shown antitumoral efficacy and low toxicity, being considered a promising class of metal-based drugs for the treatment of malignant neoplasms. Thus, the present study aimed to evaluate the cellular effects of a copper(II) complex with 4-fluorophenoxyacetic acid hydrazide and 1,10-phenanthroline ( 1 ) on PCa cell lines, as well as the mutagenic/recombinogenic and anticarcinogenic potential of 1 in Drosophila melanogaster. PNT-2 (non-tumorigenic), LNCaP (hormone-responsive PCa) and PC-3 (androgen-independent PCa) cells were cultured, and cytotoxicity was assessed using the MTT assay. The expression levels of the proliferation markers Ki-67 and Cyclin D1 were analyzed by flow cytometry. Furthermore, the Somatic Mutation and Recombination Test (SMART) and the Epithelial Tumor Test (ETT) were performed. Complex 1 was selective to LNCaP cells, significantly reducing Ki-67 and Cyclin D1 expression levels. Sub-toxic concentrations of complex 1 were defined by the toxicity test in D. melanogaster , and no mutagenic/recombinogenic/carcinogenic effects were observed. Anticarcinogenic potential was observed in D. melanogaster , suggesting modulating activity of the complex 1 against Doxorubicin, a drug used as control by its carcinogenic properties. Therefore, complex 1 is a possible starting point for the development of new antitumor agents for the treatment of PCa.

Salmonella spp. continues to figure prominently in world epidemiological registries as one of the leading causes of bacterial foodborne disease. We characterised 43 Brazilian lineages of Salmonella Typhimurium (ST) strains, characterized drug resistance patterns, tested copper (II) complex as control options, and proposed effective antimicrobial measures. The minimum inhibitory concentration was evaluated for seven antimicrobials, isolated and combined with the copper (II) complex [Cu(4-FH)(phen)(ClO
4 )2 ] (4-FH = 4-fluorophenoxyacetic acid hydrazide and phen = 1,10-phenanthroline), known as DRI-12, in planktonic and sessile ST. In parallel, 42 resistance genes were screened (PCR/microarray). All strains were multidrug resistant (MDR). Resistance to carbapenems and polymyxins (86 and 88%, respectively) have drawn attention to the emergence of the problem in Brazil, and resistance is observed also to CIP and CFT (42 and 67%, respectively), the drugs of choice in treatment. Resistance to beta-lactams was associated with the genes blaTEM / blaCTX-M in 39% of the strains. Lower concentrations of DRI-12 (62.7 mg/L, or 100 μM) controlled planktonic and sessile ST in relation to AMP/SUL/TET and AMP/SUL/TET/COL, respectively. The synergistic effect provided by DRI-12 was significant for COL/CFT and COL/AMP in planktonic and sessile ST, respectively, and represents promising alternatives for the control of MDR ST.- Published

Implementation of genomic tools is desirable to increase the efficiency of apple breeding. Recently, the multi-environment apple reference population (apple REFPOP) proved useful for rediscovering loci, estimating genomic predictive ability, and studying genotype by environment interactions (G × E). So far, only two phenological traits were investigated using the apple REFPOP, although the population may be valuable when dissecting genetic architecture and reporting predictive abilities for additional key traits in apple breeding. Here we show contrasting genetic architecture and genomic predictive abilities for 30 quantitative traits across up to six European locations using the apple REFPOP. A total of 59 stable and 277 location-specific associations were found using GWAS, 69.2% of which are novel when compared with 41 reviewed publications. Average genomic predictive abilities of 0.18-0.88 were estimated using main-effect univariate, main-effect multivariate, multi-environment univariate, and multi-environment multivariate models. The G × E accounted for up to 24% of the phenotypic variability. This most comprehensive genomic study in apple in terms of trait-environment combinations provided knowledge of trait biology and prediction models that can be readily applied for marker-assisted or genomic selection, thus facilitating increased breeding efficiency., (© The Author(s) 2022. II complexes emerged as an alternative to chemotherapy; however, its biological properties need to be better understood. Thus, we report in vitro the antitumor effects of two CuII complexes named [Cu(4-fh)(phen)(ClO4 )2 ] (complex 1) and [Cu(4-nh)(phen)(ClO4 )2 ]·H2 O (complex 2), in which 4-fh = 4-fluorophenoxyacetic acid hydrazide; 4-nh = 4-nitrobenzoic hydrazide and phen = 1,10-phenanthroline. Both complexes presented cytotoxic activity against tumor cells, but only complex 1 showed significant selectivity. Complex 1 also induced DNA-damage, led to G0/G1 arrest and triggered apoptosis, which was initiated by an autophagy dysfunction. The design and manufacture of highly efficient nanocatalysts for the oxygen reduction reaction (ORR) is key to achieve the massive use of proton exchange membrane fuel cells. Up to date, Pt nanocatalysts are widely used for the ORR, but they have various disadvantages such as high cost, limited activity and partial stability. Therefore, different strategies have been implemented to eliminate or reduce the use of Pt in the nanocatalysts for the ORR. Among these, Pt-free metal nanocatalysts have received considerable relevance due to their good catalytic activity and slightly lower cost with respect to Pt. Consequently, nowadays, there are outstanding advances in the design of novel Pt-free metal nanocatalysts for the ORR. In this direction, combining experimental findings and theoretical insights is a low-cost methodology-in terms of both computational cost and laboratory resources-for the design of Pt-free metal nanocatalysts for the ORR in acid media. Therefore, coupled experimental and theoretical investigations are revised and discussed in detail in this review article.

Two new bismuth(III) complexes, [BiL
1 Cl2 ] (1) and [BiL2 Cl2 ] (2), in which L1 is (2-hydroxy-4-6-di-tert-butylbenzyl-2-pyridylmethyl)amine and L2 is 2,4-diiodo-6-((pyridine-2-ylmethylamino)methyl)phenol, were synthesized and characterized by elemental and conductivity analyses, atomic absorption spectrometry, infrared and1 H NMR spectroscopies. The molecular structure of 1 reveals that the NN'O ligand forms a 1:1 complex with bismuth through coordination via the nitrogen of the aliphatic amine, the nitrogen of the pyridine ring and the oxygen of the phenolate. The coordination sphere is completed with two chloride anions in a distorted square pyramidal geometry. Bismuth exhibits the same coordination mode in compound 2. The cytotoxic activity of 1 and 2 was investigated in a chronic myelogenous leukemia cell line. The complexes are approximately three times more potent than the corresponding free ligands, with the IC50 values 0.30 and 0.38 μM for complex 1 and 2, respectively. To address the cellular mechanisms underlying cell demise, apoptosis was quantified by flow cytometry analysis. From 0.1 μM, both complexes induce apoptosis and there is a remarkable concentration-dependent increase in the population of cells in apoptosis. The complexes were also evaluated against Gram-positive and Gram-negative bacteria. 48. Metabolomic Characterization of Commercial, Old, and Red-Fleshed Apple Varieties.

- Abstract
In this study, a metabolomic investigation was presented to correlate single polyphenolic compounds in apple pulp with quality characteristics such as antioxidant activity and content of phenolic compounds and anthocyanins in apple skin. Since the concentration of these compounds is influenced by environmental factors, the twenty-two apple cultivars originate from the same site. The polyphenolic compounds were analyzed by ultra-high-performance liquid chromatography coupled with triple quadrupole mass spectrometry (UHPLC-QqQ-MS/MS). The antioxidant activity, phenolic content, and anthocyanins were evaluated on the sunny and the shady sides of apple skin by spectrometric assays. In old apple varieties, the measured parameters were higher than in the commercial and red-fleshed varieties. By contrast, the profile of flavan-3-ols and anthocyanins was variable amongst commercial and red-fleshed varieties. The partial least square (PLS) method was applied to investigate the association between the skin proprieties and the metabolic profile of the pulp. The highest coefficients of determination in prediction (Q2) were obtained for compounds quantified in old cultivars. These results provided information to define the old apple varieties as a reliable group based on the pathway of the antioxidant compounds and anthocyanins content. Our results show the possibility to find cultivars with promising health features based on their content of polyphenols suitable for commercialization or breeding.

- Abstract
Breeding of apple is a long-term and costly process due to the time and space requirements for screening selection candidates. Genomics-assisted breeding utilizes genomic and phenotypic information to increase the selection efficiency in breeding programs, and measurements of phenotypes in different environments can facilitate the application of the approach under various climatic conditions. Here we present an apple reference population: the apple REFPOP, a large collection formed of 534 genotypes planted in six European countries, as a unique tool to accelerate apple breeding. The population consisted of 269 accessions and 265 progeny from 27 parental combinations, representing the diversity in cultivated apple and current European breeding material, respectively. A high-density genome-wide dataset of 303,239 SNPs was produced as a combined output of two SNP arrays of different densities using marker imputation with an imputation accuracy of 0.95. Based on the genotypic data, linkage disequilibrium was low and population structure was weak. Two well-studied phenological traits of horticultural importance were measured. We found marker-trait associations in several previously identified genomic regions and maximum predictive abilities of 0.57 and 0.75 for floral emergence and harvest date, respectively. With decreasing SNP density, the detection of significant marker-trait associations varied depending on trait architecture. Regardless of the trait, 10,000 SNPs sufficed to maximize genomic prediction ability. We confirm the suitability of the apple REFPOP design for genomics-assisted breeding, especially for breeding programs using related germplasm, and emphasize the advantages of a coordinated and multinational effort for customizing apple breeding methods in the genomics era.

- Abstract
Plants have evolved a range of adaptive mechanisms that adjust their development and physiology to variable external conditions, particularly in perennial species subjected to long-term interplay with the environment. Exploiting the allelic diversity within available germplasm and leveraging the knowledge of the mechanisms regulating genotype interaction with the environment are crucial to address climatic challenges and assist the breeding of novel cultivars with improved resilience. The development of multisite collections is of utmost importance for the conservation and utilization of genetic materials and will greatly facilitate the dissection of genotype-by-environment interaction. Such resources are still lacking for perennial trees, especially with the intrinsic difficulties of successful propagation, material exchange, and living collection maintenance. This work describes the concept, design, and realization of the first multisite peach ( Prunus persica ) reference collection ( PeachRefPop ) located across different European countries and sharing the same experimental design. Other than an invaluable tool for scientific studies in perennial species, PeachRefPop provides a milestone in an international collaborative project for the conservation and exploitation of European peach germplasm resources and, ultimately, as a true heritage for future generations., (© 2020 American Society of Plant Biologists.
