The increase in time-integrated divergence of a 1.7-MeV, 10-MA relativistic electron beam due to transport over 10 cm in a gas cell filled with 1 and 6 Torr of nitrogen was measured. The divergence was characterized by a multiple-pinhole beam sampling technique involving an aperture plate, an expansion region, and an attenuator plate followed by nylon radiochromic film. The divergence is determined by a fit of the measured deposition profile to response functions calculated using Monte Carlo coupled electronphoton transport codes. The initial value of 6.9° after the entrance foil is observed to increase to 12°. The errors in the measurement are quantified with Monte Carlo techniques. The response function fit gives a significantly better estimate of the divergence than a Gaussian fit.

The inverse triax diode is a low‐impedance, high‐power electron beam diode. A magnetically unbalanced electron flow in the outer gap, which results in an outward pinch, distinguishes this design. Particle code simulations show that this flow has two possible patterns, depending on the presence or absence of an anode plasma. A model for the beam dynamics based on anode plasma formation and incorporating the simulation results is developed. We compare the predictions of the model to experimental results, including time‐resolved x‐ray pinhole photographs obtained with a framing x‐ray pinhole camera, and find reasonable agreement.

Operation of a new type of high‐power electron diode, the bidirectional voltage‐dividing (BVD) diode, has been demonstrated on a 1‐MV, 2.5‐MA, 20‐ns accelerator. This diode consists of a forward‐ and an inverse‐triaxial diode in series. The diode impedance and electron flow pattern agreed reasonably well with particle‐in‐cell code calculations. The radiation spectrum was measured using two different diagnostics, a differential absorption spectrometer and the time‐projection Compton spectrometer. Operating at a voltage of 850–1000 kV, the BVD diode produced a bremsstrahlung spectrum similar to that produced by a standard diode operating at 350–500 kV.

We describe a method for the induction of different polymorphic forms of actin filament paracrystals. This polymorphism is probably based on differences in the stagger and/or polarity of adjacent filaments in single-layered paracrystals and by superposition of different layers in multilayered paracrystals. The helical parameters defining the filament geometry are indistinguishable for the different polymorphic forms observed and for the four different actins used. Analysis of these paracrystals, some of which are ordered to better than 2.5 nm, should provide a reference structure suitable for alignment and orientation within the actin filament of high resolution models of the actin monomer obtained from crystal data.

Fibrinogen molecules sprayed on mica, dried in vacuo and shadowed with platinum appear as trinodular rods. We describe here the flotation technique of negative staining by which we were able to visualize individual fibrinogen molecules. The fibrinogen molecules in our negatively stained preparations have a trinodular structure identical to that of shadowed molecules. We believe that the 20 nm diameter globules seen in previous negative staining studies are aggregates of these molecules. This is the first study in which both shadowed and negatively stained preparations of fibrinogen support a single, consistent model for the structure of fibrinogen. The molecules in our preparation are 45 nm long (±2.5 nm, our overall estimate of accuracy), the two outer nodules are 6 to 7 nm in diameter and the middle nodule is 4 to 5 nm in diameter.

We have examined rotary shadowed, purified plasmic fragments of human fibrinogen with the electron microscope and have determined the relation of these fragments to the intact fibrinogen molecule. Both intact fibrinogen and its earliest cleavage product, fragment X, are trinodular. The next largest product, fragment Y, consists of two linked nodules. The two terminal products, fragments D and E, are single nodules. From measurements of simultaneously shadowed specimens of these different species, we conclude that the outer nodules of the trinodular fibrinogen molecule are the fragment D-containing regions and the central nodule is the fragment E-containing region.

Using electron microscopy, we have visualized the substructure of human von Willebrand factor (vWf) purified by two different approaches. vWf multimers, which appear as flexible strands varying in length up to 2 micron, consist of dimeric units (protomers) polymerized linearly in an end-to-end fashion through disulfide bonds. Examination of small multimers (e.g., one-mers, two-mers, and three-mers) suggests that each protomer consists of two large globular end domains (22 X 6.5 nm) connected to a small central node (6.4 X 3.4 nm) by two flexible rod domains each approximately 34 nm long and approximately 2 nm in diameter. The protomer is 120 nm in length when fully extended. These same structural features are seen both in vWf molecules that were rapidly purified from fresh plasma by a new two-step procedure and in those purified from lyophilized intermediate-purity Factor VIII/vWf concentrates. The 240,000-mol wt subunit observed by gel electrophoresis upon complete reduction of vWf apparently contains both a rod domain and a globular domain and corresponds to one half of the protomer. Two subunits are disulfide-linked, probably near their carboxyl termini, to form the protomer; disulfide bonds in the amino-terminal globular ends link promoters to form vWf multimers. The vWf multimer strands have at least two morphologically distinct types of ends, which may result from proteolytic cleavage in the globular domains after formation of large linear polymers. In addition to releasing fragments that were similar in size and shape to the repeating protomeric unit, plasmic degradation of either preparation of vWf reduced the size of multimers, but had no detectable effect on the substructure of internal protomers.

SummaryTo test the validity of a proposed two step fibrin assembly mechanism and directly visualize the molecular species present at various stages of fibrin formation, we have carried out an electron microscopic investigation. Assembly conditions duplicated those of a recent light scattering study and specimens were prepared at different time points with the use of a negative staining technique recently employed to visualize the trinodular structure of fibrinogen. Under near-physiological buffer conditions, protofibrils structurally similar to those postulated by Ferry have been found at early stages of fibrin assembly. In parallel with the light scattering results, a dramatic increase in fiber diameter was found in specimens prepared during the postulated lateral association stage of gelation. Light scattering and electron microscopic results both showed that high ionic strength reduces the rate and extent of fiber formation. Reptilase cleavage is shown to result in typical cross striated fibrin.

The effects of Staphylococcus aureus V8 protease (V8) on the multimeric structure of human von Willebrand factor (vWF) were studied to test and expand our model for the substructure of vWF. Electron microscopy of V8 digests of vWF revealed that the multimers were cleaved where the flexible rod (R) domains join the large elongated globular (G) domains. The resulting two major fragments, which were purified by affinity and hydrophobic interaction chromatography and by glycerol-gradient ultracentrifugation, are disulfide-linked homodimers of these domains (i.e. RR and GG) and are morphologically identical to the alternating RR and GG domains of intact vWF. The glycoprotein fragment GG (6.5 X 35 nm) has mass 343 kDa by sedimentation equilibrium and the amino-terminal sequence of intact plasma vWF. It contains the binding site for heparin within 300 residues of its amino terminus and a separate site for the platelet GPIb receptor responsible for platelet agglutination in the presence of ristocetin. With approximately 18% alpha-helix and approximately 15% beta-pleated sheet, fragment GG accounts for most of the ordered secondary structure present in whole vWF. The two thin flexible rod domains (1.8-2.0 X 30-34 nm) of fragment RR are joined at a small central nodule (approximately 5 nm diameter) and also have a small nodule at each free end. Fragment RR contains an extraordinarily high cystine content, lower than average amounts of other hydrophobic residues, and essentially no alpha-helix, as judged by circular dichroism. The amino-terminal sequence and amino acid composition of fragment RR corresponded to that of the COOH-terminal 685 residues of the intact vWF subunit (Titani, K., Kumar, S., Takio, K., Ericsson, L. H., Wade, R. D., Ashida, K., Walsh, K. A., Chopek, M. W., Sadler, J. E., and Fujikawa, K. (1986) Biochemistry 25, 3171-3184). This sequence analysis gives a mass of 180 kDa for glycosylated fragment RR, somewhat higher than the 130 kDa we obtained by sedimentation equilibrium. Our sequence analysis of a 110-kDa plasmic vWF peptide also permitted identification of a major plasmin cleavage site 705 residues from the COOH terminus and a half-cystine residue (1360) involved in maintaining the multimeric structure of plasmin-degraded vWF.(ABSTRACT TRUNCATED AT 400 WORDS)

Polypeptides immunologically related to erythrocyte spectrin and ankyrin have been detected in brain. The cross-reacting proteins include soluble as well as membrane-associated forms. A class of soluble cross-reacting polypeptides have been identified as high molecular mass microtubule-associated proteins (MAPS). MAP1, a group of polypeptides of molecular mass ca . 370 kDa contains a component that cross-reacts with anti-ankyrin IgG. MAP2, a polypeptide of molecular mass 300 kDa cross-reacts with anti-spectrin IgG, with the shared antigenic sites localized to the α chain of spectrin. The functional basis for structural homology between MAP1 and ankyrin may involve association with tubulin, since erythrocyte ankyrin binds to microtubules polymerized from pure brain tubulin. Spectrin did not associate with microtubules, but does have in common with MAP2 the ability to bind to actin (Brenner & Korn 1979; Sattilaro et al . 1981) and the shape of a flexible rod as visualized by rotary shadowing (Shotton et al . 1979; Voter & Erickson 1981). Immunoreactive forms of spectrin and ankyrin are also present in membrane fractions. A homologue of spectrin which constitutes 3% of the total membrane protein has been purified from low ionic strength extracts of membranes. This protein contains two non-identical polypeptide chains of molecular masses of 260 and 265 kDa, binds to F-actin, and displaces binding of erythrocyte spectrin to erythrocyte membranes. The brain protein pas been visualized by rotary shadowing as an extended rod-like molecule 195 nm in length. These studies indicate that the organization of proteins in the membrane-cytoskeleton complex of erythrocytes has direct relevance to other types of cells, and suggest the existence of families of proteins related to spectrin and ankyrin.

A moisture-condensation technique previously described was utilized in a study of the effect of polishing on fracture patterns in glass surfaces. The experiments show that the apparent controversy concerning the polishing out or “bridging over” of surface fractures may be resolved on the basis of crack dimensions. If the fracture width is less than 1/4 λ, it may be sealed over by polishing, even though the depth may be 1 mm. or more. A modified condensation chamber was devised to show in detail the effect of aging and fracture growth in polished surfaces.

Two accelerator magnets have been designed and developed to a manufacturable prototype. Most of the dipole calculations presented will be based on a four shell, five kiloampere/turn, five centimeter aperature magnet, approximately six meters long. Two designs are considered: a dipole bending magnet and the quadrupole focusing magnet. The mechanical and electrical coil parameters are listed.

Fibrin assembly in vivo is initiated by the limited proteolytic action of the enzyme thrombin on the 340,000-dalton soluble protein fibrinogen. In this communication, attention will be focused on the molecular mechanism of fibrin formation. By employing non-rate limiting concentrations of the activating enzyme, it has been possible to separate (in time) the proteolytic steps which initiate gelation from the two subsequent physical steps: polymerization and lateral association, both of which can be directly observed by light scattering. Network formation has been studied by sensitive time-dependent measurement of clot rigidity. More recently, a new negative staining technique developed to study the molecualr structure of fibrinogen in the electron microscope has been applied to follow the time-dependent structural changes characteristic of fibrin assembly. Results of this electron microscope investigation are consistent with the assembly mechanism proposed on the basis of the light scattering kinetic data.

We have identified the fibrin protofibril and studied its structure in electron micrographs of negatively stained specimens. Based on these images and considerations of symmetry we have constructed a model of the protofibril, in which two strands of the trinodular fibrin molecules are related by a two-fold screw axis between the strands and two-fold axes perpendicular to them. The molecules in a strand are joined by longitudinal contacts between outer nodules, and the two strands are held together by a single type of lateral contact between a central nodule on one strand and an outer nodule on the other. This interpretation of the structure of protofibrils is supported by images of trimer complexes, which show a single molecule of one strand attached laterally (by its central nodule) to the outer nodules of two molecules of the other strand. These lateral contacts, which are presumably the site of thrombin activation, are entirely saturated within the protofibril. We conclude that the association of protofibrils to form thicker fibers must involve a second type of lateral contact, probably between outer nodules of adjacent, inregister strands.

We describe the preparation and structural analysis of ordered tubular arrays of the actin-DNase I complex. These structures consist of helically stacked rings; each ring is 73 A thick, has a 240 A outer and a 120 A inner diameter, and has 7-fold rotational symmetry. The actin-DNase I complex forms tubes under conditions in which actin alone aggregates into crystalline sheets-i.e., in the presence of the trivalent cation gadolinium. Moreover, upon addition of an equimolar amount of DNase I, crystalline actin sheets are slowly converted to tubes. The rings making the tubes contain a radial dyad axis that may be identical to the dyad axis of the unit cell of the crystalline actin sheet. Evidence is presented for this identification, which in turn allows tentative assignment of actin- and DNase I-containing regions in three-dimensional reconstructions of the rings. The structural analysis presented here may be useful in aligning available three-dimensional molecular models of actin determined from crystals of the actin-DNase I complex and from crystalline actin sheets with each other and ultimately within the biologically important actin filament.
