Abstract Background This study examines seasonal and regional trends in chlorophyll-a concentrations and the dynamics of nitrogen (N) and phosphorus (P) in 32 fishponds (resulting in 150 pond-year cases) employed for fish production. Fishponds have a poor ecological state, requiring further insights for pond management. To gain those insights, monthly data on the pond environment were collected over the growing seasons from April to September (up to 14 years) across lowland to highland regions in Czechia. We used a ratio of dissolved inorganic nitrogen to total phosphorus (DIN:TP) to investigate seasonal patterns of N and P limitations. Results Fishponds in the lowland region (below 199 m above sea level, a.s.l.) were predominantly N-limited (80%), while ponds in the midland region (200–449 m a.s.l.) exhibited P limitation at the beginning of the growing season (April–May) and N limitation by the end of the season (August–September; 90% of fishponds). Highland fishponds (above 450 m a.s.l.) showed frequent P limitations, especially during the beginning of the season. Chlorophyll-a concentrations varied on both regional and seasonal scales, with the overall phytoplankton biomass peak at 31 ha of pond surface area. Chlorophyll-a remained stable at the beginning of the season regardless of the DIN:TP ratio but increased with a lower DIN:TP ratio by its end. The chlorophyll-a concentrations were lowest at the beginning and highest at the end of the season and decreased with altitude. Conclusions Seasonal and regional variations in nutrient limitations and chlorophyll-a occur in fishponds. Our study suggests that targeted seasonal nutrient input and enhanced monitoring can significantly improve fishpond management practices and ecosystem stability.

Abstract Background Picocyanobacteria from the genera Prochlorococcus, Synechococcus, and Cyanobium are the most widespread photosynthetic organisms in aquatic ecosystems. However, their freshwater populations remain poorly explored, due to uneven and insufficient sampling across diverse inland waterbodies. Results In this study, we present 170 high-quality genomes of freshwater picocyanobacteria from non-axenic cultures collected across Central Europe. In addition, we recovered 33 genomes of their potential symbiotic partners affiliated with four genera, Pseudomonas, Mesorhizobium, Acidovorax, and Hydrogenophaga. The genomic basis of symbiotic interactions involved heterotrophs benefiting from picocyanobacteria-derived nutrients while providing detoxification of ROS. The global abundance patterns of picocyanobacteria revealed ecologically significant ecotypes, associated with trophic status, temperature, and pH as key environmental factors. The adaptation of picocyanobacteria in (hyper-)eutrophic waterbodies could be attributed to their colonial lifestyles and CRISPR-Cas systems. The prevailing CRISPR-Cas subtypes in picocyanobacteria were I-G and I-E, which appear to have been acquired through horizontal gene transfer from other bacterial phyla. Conclusions Our findings provide novel insights into the population diversity, ecology, and evolutionary strategies of the most widespread photoautotrophs within freshwater ecosystems. ObjectiveLower limb peripheral arterial disease in the symptomatic stage has a significant effect on patients´ functional disability. Before an intervention, an imaging diagnostic examination is necessary to determine the extent of the disability. This study evaluates cost-effectiveness of duplex ultrasonography (DUS), digital subtraction angiography (DSA), computed tomography angiography (CTA) and magnetic resonance angiography (MRA) in the diagnostics of symptomatic patients with lower limb peripheral arterial disease indicated for endovascular or surgical intervention.MethodsDiscrete event simulation was used to capture lifetime costs and effects. Costs were calculated from the perspective of the health care payer, and the effects were calculated as quality-adjusted life year’s (QALY’s). The cost-effectiveness analysis was performed to pairwise compare CTA, MRA and DSA with DUS as the baseline diagnostic modality. A scenario analysis and probabilistic sensitivity analysis were carried out to evaluate the robustness of the results.ResultsIn the basic case, the DUS diagnostic was the least expensive modality, at a cost of EUR 10,778, compared with EUR 10,804 for CTA, EUR 11,184 for MRA, and EUR 11,460 for DSA. The effects of DUS were estimated at 5.542 QALYs compared with 5.554 QALYs for both CTA and MRA, and 5.562 QALYs for DSA. The final incremental cost-effectiveness ratio (ICER) value of all evaluated modalities was below the cost-effectiveness threshold whereas CTA has the lowest ICER of EUR 2,167 per QALY. However, the results were associated with a large degree of uncertainty, because iterations were spread across all cost-effectiveness quadrants in the probabilistic sensitivity analysis.ConclusionFor imaging diagnosis of symptomatic patients with lower limb peripheral arterial disease, CTA examination appears to be the most cost-effective strategy with the best ICER value. Baseline diagnostics of the DUS modality has the lowest costs, but also the lowest effects. DSA achieves the highest QALYs, but it is associated with the highest costs.

Abstract A combination of life history traits and environmental conditions has been highlighted as the main drivers of avian breeding success. While drivers of breeding success are well known in some species, especially birds in northern, temperate regions; species in other parts of the world have received relatively little attention. In this study, we used a long‐term dataset on breeding success of tropical plovers from south‐west Madagascar to investigate whether nest survival changed over time and whether the drivers of nest survival were similar for multiple species breeding in the same arid habitat. In the 12‐year period of 2009–2020, we monitored 2077 nests for three species with different breeding strategies: 1185 nests of Kittlitz's plovers (Anarhynchus pecuarius) with a flexible breeding strategy and uniparental care; and 565 nests of white‐fronted plovers (A. marginatus) and 327 nests of Madagascar plovers (A. thoracicus) which both have biparental care. We found that nest survival was associated with a combination of clutch‐size, age of nest and year among the three plover species. In addition, annual variation in climatic conditions associated with El Niño/La Niña events were included in the most supported survival models for Kittlitz's and white‐fronted plovers, but the effects were not significant. Overall estimates of daily nest survival were similar for all three species: Kittlitz's plover: 0.950 ± 0.002 SE, Madagascar plover: 0.919 ± 0.007 SE, and white‐fronted plover: 0.862 ± 0.047 SE. Estimates of nest success for the breeding season, based on increases in daily nest survival with the clutch age during the incubation periods (26 days for Kittlitz's plovers and 29 days for Madagascar and white‐fronted plovers), were relatively low: Kittlitz's plover: 0.161 ± 0.056 SE, Madagascar plover: 0.287 ± 0.022 SE, and white‐fronted plover: 0.228 ± 0.019 SE. All three species had a combination of factors affecting nest survival, both environmental and life history traits.

Agricultural intensification has affected wildlife across Europe, triggering steep declines and regional extinctions of farmland birds. Effective conservation activities are essential for the preservation of biodiversity in an agricultural landscape, but current efforts have not succeeded in halting these declines. Here we investigate a ground-nesting shorebird, the collared pratincole Glareola pratincola, which has shifted its habitat use in Central Europe over the last 20 years from alkaline grasslands to intensively managed agricultural fields. We show that nesting success was different between three agricultural habitat types, with the highest nesting success in fallow lands and the lowest in row crops. Nesting success was also associated with the timing of breeding and breeding density, as nests produced early in the breeding season and those in high-breeding-density areas hatched more successfully than those produced later in the season and at low density. We implemented direct conservation measures including marking nests and negotiating with farmers to avoid cultivating the field between nest markers, controlling nest predators and, most recently, creating suitable nesting sites and foraging areas for pratincoles. As a result of these conservation actions, nest survival increased from 11.2% to 83.5% and the size of the breeding population increased from 13 to 56 pairs during 2012–2021. Thus, we show that agricultural landscapes can continue to provide suitable habitats, and targeted conservation actions have the potential to reverse the declines of farmland species.

The paper deals with Petr Rezek’s philosophy in relation to two of his central themes, matter-of-factness (Sachlichkeit) and conflict, through the prism of the problem of non-matter-of-fact (unsachlich) quoting and its manifestations. According to Rezek, these point to the defective forms of intersubjectivity. The dialectic of quoting reason begins with a critique of these forms in order to become a true dialectic of matter-of-factness, through contrasting structures and techniques of analysis, such as the so-called boundary approach or the method of repetition.

Abstract To improve the predictability of complex computational models in the experimentally-unknown domains, we propose a Bayesian statistical machine learning framework utilizing the Dirichlet distribution that combines results of several imperfect models. This framework can be viewed as an extension of Bayesian stacking. To illustrate the method, we study the ability of Bayesian model averaging and mixing techniques to mine nuclear masses. We show that the global and local mixtures of models reach excellent performance on both prediction accuracy and uncertainty quantification and are preferable to classical Bayesian model averaging. Additionally, our statistical analysis indicates that improving model predictions through mixing rather than mixing of corrected models leads to more robust extrapolations.

Abstract Heart failure (HF) is life-threatening disease due to electro-mechanical dysfunction associated with hemodynamic overload, while alterations of extracellular matrix (ECM) along with perturbed connexin-43 (Cx43) might be key factors involved. We aimed to explore a dual impact of pressure, and volume overload due to aorto-caval fistula (ACF) on Cx43 and ECM as well as effect of renin–angiotensin blockade. Hypertensive Ren-2 transgenic rats (TGR) and normotensive Hannover Sprague–Dawley rats (HSD) that underwent ACF were treated for 15-weeks with trandolapril or losartan. Blood serum and heart tissue samples of the right (RV) and left ventricles (LV) were used for analyses. ACF-HF increased RV, LV and lung mass in HSD and to lesser extent in TGR, while treatment attenuated it and normalized serum ANP, BNP-45 and TBARS. Cx43 protein and its ser368 variant along with PKCε were lower in TGR vs HSD and suppressed in both rat strains due to ACF but prevented more by trandolapril. Pro-hypertrophic PKCδ, collagen I and hydroxyproline were elevated in TGR and increased due to ACF in both rat strains. While SMAD2/3 and MMP2 levels were lower in TGR vs HSD and reduced due to ACF in both strains. Findings point out the strain-related differences in response to volume overload. Disorders of Cx43 and ECM signalling may contribute not only to HF but also to the formation of arrhythmogenic substrate. There is benefit of treatment with trandolapril and losartan indicating their pleiotropic anti-arrhythmic potential. It may provide novel input to therapy.

Abstract Detailed electrochemical study of copper(II) complexes of cyclam bearing two phosphonate (TE2P), phenolate (TE2PhOH) or carboxylate (TE2A) pendant arms (together with one cross‐bridged‐cyclam derivative cb‐TE2A) was performed using cyclic voltammetry (CV) and polarography. The electrochemical‐heterogeneous measurements were accompanied by UV‐vis spectra describing the situation in homogeneous phase. Reduction of complexes on mercury electrode in buffered aqueous solutions (pH 2–12) occurs generally as a two‐electron process of reductive decomposition. The formed Cu‐amalgam can be (e. g. during the anodic scan of CV) reoxidized to Cu(II) cation which undergoes re‐complexation resulting in the original complex. This process of a square diagram is manifold repeatable. The results also showed presence of several isomers differing in mutual orientation of substituents on cyclam nitrogen atoms. The primary “kinetic”, pentacoordinated isomers are irreversibly transformed to the stable, hexacoordinated “thermodynamic” ones upon heating. The development of isomerization process was followed electrochemically. The cross‐bridged cb‐TE2A derivative forms only one isomer which does not change even after heating. The higher stability of the complex manifests itself at pH>8 when the complex is reversibly reduced in one‐electron step, formally CuII/CuI, without decomposition.

Five different chocolate types (i.e. extra dark, dark, milk, white, and ruby) were characterized and compared for the acoustical and textural characteristics. Acoustic properties were described in terms of longitudinal and transversal wave velocities. These velocities were used for the evaluation of elastic constants namely Young modulus and Poisson ratio. The values of the Young moduli are significantly higher than those obtained using uniaxial compression and tensile tests. At the same time, the values of Poisson ratio are in reasonable agreement with results of these methods. Texture characteristics were evaluated using of the compression test at loading rates (crosshead velocity) 1, 10 and 100 mm/min. Results are presented in form of stress vs strain. The stress-strain dependences obtained from the compression test exhibited two different regions of the sensitivity of stress to the applied strain. The linear dependence stress- strain is taken as the evidence of the elastic behavior of the tested material. The extent of this region was given by the strain, which was the same for all tested chocolates. Its value increased with the loading rate. The stress-strain behavior in this region was only slightly dependent on the loading rate with exception of the ruby and dark chocolates. The stress-strain dependence in the following region was significantly dependent on the strain rate. Values of the strain rate sensitivity were evaluated. The order of the tested chocolates according to the values of stress at the given strain was identical with that obtained using of the elastic wave velocities.

Background: In recent years, there has been an increasing effort to take advantage of the potential use of low magnetic induction devices with less than 1 T, referred to as Low-Field MRI (LF MRI). LF MRI systems were used, especially in the early days of magnetic resonance technology. Over time, magnetic induction values of 1.5 and 3 T have become the standard for clinical devices, mainly because LF MRI systems were suffering from significantly lower quality of the images, e.g., signal–noise ratio. In recent years, due to advances in image processing with artificial intelligence, there has been an increasing effort to take advantage of the potential use of LF MRI with induction of less than 1 T. This overview article focuses on the analysis of the evidence concerning the diagnostic efficacy of modern LF MRI systems and the clinical comparison of LF MRI with 1.5 T systems in imaging the nervous system, musculoskeletal system, and organs of the chest, abdomen, and pelvis. Methodology: A systematic literature review of MEDLINE, PubMed, Scopus, Web of Science, and CENTRAL databases for the period 2018–2023 was performed according to the recommended PRISMA protocol. Data were analysed to identify studies comparing the accuracy, reliability and diagnostic performance of LF MRI technology compared to available 1.5 T MRI. RESULTS: A total of 1275 publications were retrieved from the selected databases. Only two articles meeting all predefined inclusion criteria were selected for detailed assessment. Conclusions: A limited number of robust studies on the accuracy and diagnostic performance of LF MRI compared with 1.5 T MRI was available. The current evidence is not sufficient to draw any definitive insights. More scientific research is needed to make informed conclusions regarding the effectiveness of LF MRI technology.

IntroductionThe successful cryopreservation of common carp sperm is crucial for its application in aquaculture and selective breeding programs. This study investigates the efficacy of cryopreserving sperm in large containers (5 mL) with a low dilution rate (1:1) in three different cryoprotective media and thawing in different conditions.MethodsThe developed method utilizes a low-ionic (hypotonic) cryoprotective medium, freezing with a controlled cooling rate, and high-temperature sperm thawing (60°C). The investigation employs a detailed spermatozoon motility assessment.ResultsPost-thaw motility of 32.3% ± 14% and initial curvilinear velocity of 89 ± 20 μm/s across 30 males were observed. Principal component analysis of sperm kinematic characteristics revealed distinct populations of sperm cells exhibiting varying responses to cryopreservation. The developed method achieved successful fertilization comparable to that of the non-frozen control group using sperm from a single cryotube (2.5 mL, approximately 50 * 109 spermatozoa) to fertilize 200 g of eggs (1:120,000 egg:spz).DiscussionThis novel approach demonstrates an effective cryopreservation protocol for common carp sperm in large-volume cryo-containers in combination with low-ionic cryomedia and high thawing temperature, providing methods well-suited for fisheries practices and selective breeding programs. Future studies of the biological properties of different sperm subpopulations in post-thaw sperm samples can contribute to a deeper understanding of sperm biology, improve cryopreservation techniques, and enhance the success rates of assisted reproductive technologies.

This article presents selected symbols that were adopted, either through official legislation or through political and social negotiations, as symbols of the state after the fall of the Habsburg Empire and the establishment of the Czechoslovak Republic. Their main weakness was their strong association with Czech identity. The symbols of the new state came from the period of mutual national struggles and identity affirmation in the last decades of the monarchy, but they were only significant for the modern Czech nation. This was the reason why they never became a unifying element of the democratic and multinational Czechoslovak state, nor did they appeal to most of its Slovak inhabitants, whose historical traditions and symbols were virtually ignored. What is more, they were even perceived as provocative and hostile by the German, Hungarian, Ruthenian and Polish populations of the interwar Czechoslovak Republic.

ABSTRACT Novel species of coagulase-negative staphylococci, which could serve as reservoirs of virulence and antimicrobial resistance factors for opportunistic pathogens from the genus Staphylococcus, are recognized in human and animal specimens due to advances in diagnostic techniques. Here, we used whole-genome sequencing, extensive biotyping, MALDI-TOF mass spectrometry, and chemotaxonomy to characterize five coagulase-negative strains from the Staphylococcus haemolyticus phylogenetic clade obtained from human ear swabs, wounds, and bile. Based on the results of polyphasic taxonomy, we propose the species Staphylococcus brunensis sp. nov. (type strain NRL/St 16/872T = CCM 9024T = LMG 31872T = DSM 111349T). The genomic analysis revealed numerous variable genomic elements, including staphylococcal cassette chromosome (SCC), prophages, plasmids, and a unique 18.8 kb-long genomic island SbCI ccrDE integrated into the ribosomal protein L7 serine acetyltransferase gene rimL. SbCI ccrDE has a cassette chromosome recombinase (ccr) gene complex with a typical structure found in SCCs. Based on nucleotide and amino acid identity to other known ccr genes and the distinct integration site that differs from the canonical methyltransferase gene rlmH exploited by SCCs, we classified the ccr genes as novel variants, ccrDE. The comparative genomic analysis of SbCI ccrDE with related islands shows that they can accumulate virulence and antimicrobial resistance factors creating novel resistance elements, which reflects the evolution of SCC. The spread of these resistance islands into established pathogens such as Staphylococcus aureus would pose a great threat to the healthcare system. IMPORTANCE The coagulase-negative staphylococci are important opportunistic human pathogens, which cause bloodstream and foreign body infections, mainly in immunocompromised patients. The mobile elements, primarily the staphylococcal cassette chromosome mec, which confers resistance to methicillin, are the key to the successful dissemination of staphylococci into healthcare and community settings. Here, we present a novel species of the Staphylococcus genus isolated from human clinical material. The detailed analysis of its genome revealed a previously undescribed genomic island, which is closely related to the staphylococcal cassette chromosome and has the potential to accumulate and spread virulence and resistance determinants. The island harbors a set of conserved genes required for its mobilization, which we recognized as novel cassette chromosome recombinase genes ccrDE. Similar islands were revealed not only in the genomes of coagulase-negative staphylococci but also in S. aureus. The comparative genomic study contributes substantially to the understanding of the evolution and pathogenesis of staphylococci.

Honey contains a wide range of inorganic substances. Their content can be influenced, i.e., by the type of soil on which the bee pasture is located. As part of this study, the mineral profile of 32 samples of honey from hobby beekeepers from the Czech Republic wasevaluated and then compared with soil types in the vicinity of the beehive location. Pearson’s correlation coefficient was used to express the relationship between mineral substances and soil type. There was a high correlation between antroposol and Zn (R = 0.98), Pb (R = 0.96), then between ranker and Mn (0.95), then regosol and Al (R = 0.97) (p < 0.05). A high negative correlation was found between regosol and Mg (R = −0.97), Cr (R = −0.98) and between redzinas and Al (R = −0.97) (p < 0.05). Both positive and negative high correlations were confirmed for phaeozem. The CART method subsequently proved that the characteristic elements for individual soil types are B, Ca, Mg, Ni, and Mn. The soil types of cambisol, fluvisol, gleysol, anthrosol, and kastanozem had the closest relationship with the elements mentioned, and it can therefore be assumed that their occurrence indicates the presence of these soil types within the range of beehive location.

Due to the ever-increasing consumption of fossil fuels, their impact on the environment, and the volatility of the market, it is advisable to use biofuels that can be produced locally from renewable sources, which supports the local economy, agriculture, and related processing industries. This article deals with how to improve the flow properties and pour point of biofuels for diesel engines. For the experiment, biodiesels in the form of rapeseed methyl ester (RME) and methyl ester made from waste animal fats and tallow (WAFME) were used. The pour point, viscosity, and density of WAFME were improved by mixing with RME and/or by adding bio-alcohols (alcohols produced from biomass, e.g., lignocellulosic). All used biofuels were classified as 2nd generation biofuels. The flow properties of the mixtures were monitored and subsequently modelled at temperatures from -10 to 60 °C. The addition of bio-alcohol had a statistically significant effect on the decrease in the viscosity and pour point of ternary blends (p < 0.05). Mathematical models of the dependence of kinematic viscosity on the temperature of mixtures (power law, exponential, Arrhenius, and Vogel) were created.

The study is focused on how the physical structure of the feed affects the health status of broiler chickens. The aim of this study was to evaluate the influence of feed particle size in broiler diets on gastrointestinal tract morphology, digesta viscosity, and blood biochemical parameters. A total of 90 one-day-old male Ross 308 broiler chickens were randomly divided into three different experimental groups (with five replicates per pen), with 6 birds per cage. The first experimental group (Coarse) was fed with the coarsest particle size, with feed with a geometric mean diameter (GMD) of 1111.26 µm, the next group (Medium) was fed with a less coarse feed size of GMD 959.89 µm, and the last group (Fine) was fed a diet with a fine feed particle size of GMD 730.48 µm. The use of coarse feed particle size in the diet had a positive effect on the gizzard weight and small intestinal villi height and crypt depth, which increased the surface area intended for digesting nutrients. The use of finely ground particles in the feed increased the level of gamma-glutamyl transferase and at the same time, decreased the level of urea, which could indicate adverse changes in the liver.

The paper deals with the actual topic of mechanical properties of 3D prints made from the polymer material rPET-G and its changes. Using this material in additive technology has great potential in the automotive industry. The research evaluates five different post-processing modifications of 3D prints from rPET-G (recycled polyethylene terephthalate-glycol) material produced by the Fused Filament Fabrication technology. The post-processing included the chemical smoothing of the samples using dichloromethane vapors and heat treatment in an annealing furnace. An epoxy resin, a synthetic coating, and a water-based coating were also applied. The standard was represented by samples without post-processing modifications, both in the form of recycled material and in the form of virgin PET-G. The texture of the samples was evaluated according to EN ISO 4287. Furthermore, the moisture absorption of the samples was evaluated using the gravimetric method according to EN ISO 62. The mechanical testing of the samples was carried out using a tensile test (EN ISO 527-2), a three-point bending test (EN ISO 178), impact strength (EN ISO 179-2), and a cupping test (EN ISO 1520). Degradation of the coatings took place using cyclic tests, which used a combination of exposure in a salt fog environment and in a climate chamber. The results of the experiments indicate that heat treatment appears to be a universal post-processing technology, as this method statistically improves not only the mechanical properties but also significantly reduces moisture absorption.

In this study, we aimed to observe the effects of positive airway pressure (PAP) on individual levels of obstruction during drug-induced sleep endoscopy (DISE) of the upper airways (UA), to evaluate at which pressures the obstruction disappeared or worsened, and to identify cases in which PAP was ineffective. This prospective study was conducted from June 2018 to June 2022. PAP testing was performed during DISE in patients with moderate and severe OSA. The pressure was gradually increased over the range from 6.0 to 18.0 hPa. Our findings were evaluated using the VOTE classification. The examination was performed in 56 patients, with a median apnea–hypopnea index (AHI) of 26.4. Complete obstruction of the soft palate was observed in 51/56 patients (91%), oropharyngeal obstruction in 15/56 patients (27%), tongue base obstruction in 23/56 patients (41%), and epiglottic collapse in 16/56 patients (29%). PAP was most effective in cases of complete oropharyngeal obstruction, and least effective in cases of epiglottic collapse, where it was ineffective in 11/16 patients. DISE with PAP is a simple diagnostic method that can be helpful for identifying anatomic and dynamic reasons for PAP intolerance. The main indication is ineffective PAP treatment.

The aim of this study was to compare the reliability of five sleep questionnaires in detecting the occurrence of obstructive sleep apnea (OSA). The study was conducted on a group of 201 patients. The patients completed five sleep questionnaires: the Epworth Sleepiness Scale (ESS), the STOP-Bang questionnaire, the STOP questionnaire, the Berlin questionnaire (BQ) and the Pittsburgh Sleep Quality Index (PSQI). Subsequently, the patients were examined using limited polygraphy, and the sensitivity and specificity of the questionnaires were evaluated. The STOP-Bang, Berlin and STOP questionnaires had the highest sensitivity for OSA detection (81.6%, 78.7%, and 74.2%, respectively), while the sensitivities of PSQI and ESS were low (50.8% and 34.5%). The ESS, STOP-Bang, STOP and Berlin questionnaires had the highest specificity (82.6%, 75%, 61.9%, and 61.9%). In our sample, we found the STOP-Bang and Berlin questionnaires to be the most suitable for OSA screening with the highest sensitivities (81.6%, 78.7%) and satisfactory specificities (75%, 61.9%). The STOP questionnaire was also relatively reliable, especially given its time-saving nature; though short, it preserved satisfactory sensitivity (74.2%) and specificity (61.9%). The ESS and PSQI were unsuitable for OSA screening.

This paper deals with the modification of the mechanical system of the needle bar. The purpose of this work is to reduce the vibration and noise of the sewing machine for creating a decorative stitch. A special floating needle is used to sew this stitch, in which two mechanical systems of needle bars handover through the sewn material, so that a perfect imitation of a hand stitch is created. The original system, which controls the release of the needle at the handover location by abruptly stopping the needle bar control element, could be replaced by a new system that uses magnetic force to release the needle. In addition to the usual design procedure, numerical simulations of the attractive force of the electromagnet are also used in the design of a suitable electromagnet. At the same time, an electrical circuit is also designed to allow the needle to be released and gripped quickly. The advantages of the new system lie not only in reducing vibrations and the associated increase in the operation speed of the machine, but also in making it easier for the machine to switch to possible automated or semi-automated production.

The mechanisms by which myelodysplastic syndrome (MDS) cells resist the effects of hypomethylating agents (HMA) are currently the subject of intensive research. A better understanding of mechanisms by which the MDS cell becomes to tolerate HMA and progresses to acute myeloid leukemia (AML) requires the development of new cellular models. From MDS/AML cell lines we developed a model of 5-azacytidine (AZA) resistance whose stability was validated by a transplantation approach into immunocompromised mice. When investigating mRNA expression and DNA variants of the AZA resistant phenotype we observed deregulation of several cancer-related pathways including the phosphatidylinosito-3 kinase signaling. We have further shown that these pathways can be modulated by specific inhibitors that, while blocking the proliferation of AZA resistant cells, are unable to increase their sensitivity to AZA. Our data reveal a set of molecular mechanisms that can be targeted to expand therapeutic options during progression on AZA therapy.

Staphylococci from the Staphylococcus intermedius-Staphylococcus hyicus species group include numerous animal pathogens and are an important reservoir of virulence and antimicrobial resistance determinants. Due to their pathogenic potential, they are possible causative agents of zoonoses in humans; therefore, it is important to address the properties of these strains. Here we used a polyphasic taxonomic approach to characterize the coagulase-negative staphylococcal strain NRL/St 03/464T, isolated from the nostrils of a healthy laboratory rat during a microbiological screening of laboratory animals. The 16S rRNA sequence, MALDI-TOF mass spectrometry and positive urea hydrolysis and beta-glucuronidase tests clearly distinguished it from closely related Staphylococcus spp. All analyses have consistently shown that the closest relative is Staphylococcus chromogenes; however, values of digital DNA-DNA hybridization Staphylococcus species. Whole-genome sequencing and expert annotation of the genome revealed the presence of novel variable genetic elements, including two plasmids named pSR9025A and pSR9025B, prophages, genomic islands and a composite transposon that may confer selective advantages to other bacteria and enhance their survival. Based on phenotypic, phylogenetic and genomic data obtained in this study, the strain NRL/St 03/464T (= CCM 9025T = LMG 31873T = DSM 111348T) represents a novel species with the suggested name Staphylococcus ratti sp. nov.

Liquid egg products are one of the basic raw materials for the food industry. Knowledge of their rheological and flow behaviour in real technical elements is absolutely necessary for the selection of suitable technological equipment for their processing. In this article, the rheological properties of liquid egg products were determined. Eggs from six different species of poultry are used: domestic hen (Gallus gallus domesticus) hybrid Hisex Brown; Japanese quail (Coturnix japonica); German carrier goose (Anser anser f. domestica); domestic ducks (Anas platyrhynchos f. domestica); domestic guinea fowl (Numida meleagris f. domestica); and domestic turkeys (Meleagris gallopavo f. domestica). Liquid egg products showed pseudoplastic behaviour in range of shear strain rates from 0.2 up to 200 s−1 and at the temperature of 18 °C. Thus, the flow curves were constructed using the Ostwald-de Waele rheological model, with respect to the pseudoplastic behaviour of liquid egg products. According to the values of the coefficients of determination (R2), the sum of squared estimate of errors (SSE) and the root mean square error (RMSE), this model was appropriately chosen. Using the consistency coefficient K, the flow index n and the adjusted equations for the flow rate of technical and biological fluids in standard pipelines, the 3D velocity profiles of liquid egg products were successfully modelled. The values of the Reynolds number of the individual liquid egg products were calculated, and the type of flow was also determined. A turbulent flow has been detected for some liquid egg products.

The aim of this study was to use unconventional malts in beer production and observe their effect on the wort viscosity. Six malts were analysed in this study—barley, black barley, oat, wheat, rye, and corn. Firstly, the parameters of cereals were measured after the malting process in an experimental malting house and wort production. Samples were analysed in each phase of the mashing process. Carbohydrate contents and viscosities were analytically determined from the samples. The resulting values of the dynamic viscosity were significantly higher than the values obtained by other authors, ranging from 3.4 up to 35.5 mPa·s−1. This study also confirmed the hypothesis that states that the breakdown of carbohydrates leads to a decrease in viscosity. Values measured in the black barley malt sample were higher when compared with light barley malt. Unconventional malts had a higher viscosity and were thus more difficult to filter. If these types of malts are used it is recommended to add barley malts or malts with a higher enzyme activity to them.

Advanced high-strength steels protected by zinc coatings have contributed to a reduction in CO2 emissions in the automotive industry. However, the liquid metal embrittlement (LME) of the Fe/Zn couple induced by simultaneously acting stresses and high temperatures during resistance spot welding could be the cause of unexpected failure. We investigated the possible risk of LME in spot-welded martensitic steel with Zn jet vapor-deposited coating and its influence on weld strength. The weld nugget cross-sections were analyzed (optical microscopy, SEM-EDS), and their tensile shear strengths were compared with their uncoated counterparts. LME cracks were observed in all samples meeting the process window (6, 6.5, 7 kA) located at the edge of the sheet/electrode indentation area. The frequency and length of cracks increased with current, and the occurrence of Zn within cracks indicated the LME mechanism. The shear tests showed the Zn-coated sample underwent a decrease in tensile shear strength that was most evident at a welding current of 7 kA (13.2%). However, LME was excluded as a cause of lower strength. The decrease was attributed to the smaller nugget diameter and the thin slit of Zn coating remaining in the weld notch.

Chocolate masses are one of the basic raw materials for the production of confectionery. Knowledge of their rheological and flow behaviour at different temperatures is absolutely necessary for the selection of a suitable technological process in their production and subsequent processing. In this article, the rheological properties (the effect of the shear strain rate on the shear stress or viscosity) of five different chocolate masses were determined—extra dark chocolate (EDC), dark chocolate (DC), milk chocolate (MC), white chocolate (WC), and ruby chocolate (RC). These chocolate masses showed thixotropic and plastic behaviour in the selected range of shear rates from 1 to 500 s−1 and at the specified temperatures of 36, 38, 40, 42, and 44 °C. The degree of thixotropic behaviour was evaluated by the size of the hysteresis area, and flow curves were constructed using the Bingham, Herschel–Bulkley and Casson models with respect to the plastic behaviour of the chocolate masses. According to the values of the coefficients of determination R2 and the sum of the squared estimate of errors (SSE), the models were chosen appropriately. The most suitable models are the Herschel–Bulkley and Casson models, which also model the shear thinning property of the liquids (pseudoplastic with a yield stress value). Using the coefficients of the rheological models and modified equations for the flow velocity of technical and biological fluids in standard piping, the 2D and 3D velocity profiles of the chocolate masses were further successfully modelled. The obtained values of coefficients and models can be used in conventional technical practice in the design of technological equipment structures and in current trends in the food industry, such as 3D food printing.

Owing to their low cost, good performance, and high lifetime stability, activated carbons (ACs) with a large surface area rank among the most popular materials deployed in commercially available electrochemical double-layer (EDLC) capacitors. Here, we report a simple two-step synthetic procedure for the preparation of activated carbon from natural flax. Such ACs possess a very high specific surface area (1649 m2 g–1) accompanied by a microporous structure with the size of pores below 2 nm. These features are behind the extraordinary electrochemical performance of flax-derived ACs in terms of their high values of specific capacitance (500 F g–1 at a current density of 0.25 A g–1 in the three-electrode setup and 189 F g–1 at a current density of 0.5 A g–1 in two-electrode setup.), high-rate stability, and outstanding lifetime capability (85% retention after 150,000 charging/discharging cycles recorded at the high current density of 5 A g–1). These findings demonstrate that flax-based ACs have more than competitive potential compared to standard and commercially available activated carbons.

The main objective of this study was to verify the safety of mealworm meal in the feed of laying hens from 17 to 42 weeks of age. Therefore, the feed mixtures were tested in terms of microbiological stability, fungal and mycotoxin content and selected parameters of hens’ intestinal morphology and physiology were monitored. The experiment was carried out with 30 Lohmann Brown Classic hens. Hens were divided by body mass into three equal groups with 10 replicates per treatment. The two experimental groups received feed mixtures containing 2% and 5% yellow mealworm (Tenebrio molitor L.) meal. The third group was a control group which had 0% of mealworm meal in the diet. Diets with 2% and 5% of mealworm meals did not affect the length of villi and microbiome of the caecum. The highest digesta viscosity from the ileum was found in the group with 5% mealworm, which may indicate a slower passage of the digesta through the digestive tract. Based on our results, it may be concluded that the proportion of mealworm meals does not deteriorate the quality of feeds. Mealworm meal does not negatively affect microbial stability in experimental feeds. Therefore, it can be recommended the two and (or) five percent of mealworm meal inclusion in hen’s diet.

Geographical and botanical origin of honeys can be characterized on the basis of physico-chemical composition, sensory properties and on the basis of melissopalynological analysis. No comprehensive description of the characteristics of Czech honey has been published so far. This study provides insights that are important for correct classification. The study analysed 317 samples of authentic honey from randomly selected localities. Due to the diversity of the landscape, the typical honey of the region is blend honey with a predominance of blossom honey. According to the pollen profile and electric conductivity, the honeys were sorted into the following: Brassica honey (BH), Floral honey (FH), Fruit tree honey (PH), Honeydew (HD), Lime tree honey (LH), Robinia pseudoacacia honey (RH), and Trifolium honey (TH). Physico-chemical properties, including higher carbohydrates, were determined for the honeys and their pollen profiles were examined. The physico-chemical properties and pollen profile are partially in compliance with the description of European monofloral honeys, except for RH and TH. Although they had the highest proportion of acacia pollen, amounting to >10% of all the Czech honeys, these RH honeys differ from the European standard, so they cannot be considered acacia honey. Further, PH honeys and FH polyfloral honeys were described. Most honeys contained a significant proportion of rapeseed pollen, which is one of the common agricultural crops grown in the Czech Republic. All the analysed honeys met the parameters defined by the legislation. Due to direct on-site sampling, honeys were characterized by a low 5-(hydroxymethyl)furfural (HMF) content (3.0 mg/kg) and high diastase activity (24.4 DN). Honeydew honeys had the highest proportion of higher carbohydrates, primarily of Melezitose (4.8 g/100 g) and Trehalose (1.3 g/100 g). The presence of higher carbohydrates was also confirmed in LH for Maltose (4.6 g/100 g) and Turanose (2.4 g/100 g).

Butanol seems to be an eligible fuel for compensating for the increasing fuel consumption. Biobutanol could be produced from local sources in the place of use. Its properties show similar results to gasoline, so biobutanol could be added as a biocomponent into fuels. Important properties, in the case of blending biobutanol into gasoline, are its fluid properties and their dependence on the temperature. Therefore, in this paper, the volumetric mass density and viscosity of the selected ratios between biobutanol and gasoline (0, 5, 10, 85, 100 vol.%) were tested over the temperature range from −10 °C up to 40 °C. Gasolines with a 95 Research Octane Number (RON 95) and with a 98 Research Octane Number (RON 98) were used. It was observed that as the temperature increased, the viscosity and volumetric mass density of the samples decreased nonlinearly. Four mathematical models were used for modelling the viscosity. The accuracy of models was evaluated and compared according to the coefficient of determination R2 and sum of squared estimate of errors (SSE). The results show that blends with 5 vol.% and 10 vol.% of biobutanol promise very similar fluid properties to pure gasoline. In contrast, a blend with 85 vol.% of biobutanol shows different fluid properties from gasoline, especially in negative temperatures, a lot. For practical applications, mathematical polynomial multivariate models were created. Using these models, three-dimensional graphs were constructed.

Recently we have shown the importance of hypergolic reactions in carbon materials synthesis. However, hypergolic reactions could be certainly expanded beyond carbon synthesis, offering a general preparative pathway towards a larger variety of materials. Cyclopentadienyls are one of the most common ligands in organometallic chemistry that react hypergolicly on contact with strong oxidizers. By also considering the plethora of cyclopentadienyl compounds existing today, herein we demonstrate the potential of such compounds in hypergolic materials synthesis in general (carbon or inorganic). In a first example, we show that cyclopentadienyllithium reacts hypergolicly with fuming nitric acid to produce carbon. In a second one, we show that ferrocene and cobaltocene also react hypergolicly with the concentrated acid to afford magnetic inorganic materials, such as γ-Fe2O3 and metallic Co, respectively. The present results further emphasize the importance and universal character of hypergolic reactions in materials science synthesis, as an interesting new alternative to other existing and well-established preparative methods.

The effect of phase composition and morphology on high-temperature strength in the compression of Fe-Al-Si-based iron aluminides manufactured by casting was investigated. The structure and high-temperature strength in the compression of three alloys—Fe28Al5Si, Fe28Al5Si2Mo, and Fe28Al5Si2Ti—were studied. Long-term (at 800 °C for 100 h) annealing was performed for the achievement of structural stability. The phase composition and grain size of alloys were primarily described by means of scanning electron microscopy equipped with energy dispersive analysis and Electron Backscatter Diffraction (EBSD). The phase composition was verified by X-ray diffraction (XRD) analysis. The effect of Mo and Ti addition as well as the effect of long-term annealing on high-temperature yield stress in compression were investigated. Both additives—Mo and Ti—affected the yield stress values positively. Long-term annealing of Fe28Al5Si-X iron aluminide alloyed with Mo and Ti deteriorates yield stress values slightly due to grain coarsening.

Hatchery-reared sterlet originating from the Danube and Volga river basins that showed population-discriminatory alleles on at least one microsatellite locus were used to produce purebred (within-population) and hybrid crosses to evaluate intraspecific hybridization with respect to the genetic polymorphism and physiological fitness of fish for commercial aquaculture and, conservation programs. Reciprocal crossing assessed the effect of parent position. The fish were reared in indoor and outdoor tanks and monitored over 504 days for growth traits. The highest final mean body weight (144.9 ± 59.5 g) was recorded in the Danube (♀) × Volga (♂) hybrid and the highest survival in the Volga (♀) × Danube (♂) hybrid. The Volga purebred exhibited the lowest mean body weight (124.8 ± 57.6 g). A set of six microsatellites was used to evaluate the heterozygosity. The mean number of alleles was highest in the Danube (♀) × Volga (♂) hybrid and lowest in the Volga purebred, suggesting an influence of the parent position in the hybridization matrix. The higher level of genetic polymorphism, as in the Danube (♀) × Volga (♂) hybrid, may confer greater fitness in a novel environment. Our analysis revealed that the intraspecific hybrids performed better than the purebred fish in the controlled and suboptimal rearing conditions.

Multivariate tail coefficients are an important tool when investigating dependencies between extreme events for different components of a random vector. Although bivariate tail coefficients are well-studied, this is, to a lesser extent, the case for multivariate tail coefficients. This paper contributes to this research area by (i) providing a thorough study of properties of existing multivariate tail coefficients in the light of a set of desirable properties; (ii) proposing some new multivariate tail measurements; (iii) dealing with estimation of the discussed coefficients and establishing asymptotic consistency; and, (iv) studying the behavior of tail measurements with increasing dimension of the random vector. A set of illustrative examples is given, and practical use of the tail measurements is demonstrated in a data analysis with a focus on dependencies between stocks that are part of the EURO STOXX 50 market index.

TGM dedicated this book to the Czech and Slovak soldiers joining the Allied armies in Russia, France, USA, Britain, Italy and other states. These soldiers demonstrated their anti-Austrian feelings and willingness to fight for the freedom and independence of our nation. The book was also dedicated to these who were forced to fight for Austria. The author explained the Czech national programme and his view on the First World War. He assessed its importance for the future of the independent Czech nation and advocated the restoration of an independent Czech and Slovak state which should be an equal partner in the political and social organisation of Europe.

This study deals with the activities of the Spanish ambassador Juan de Borja in the circumstances of Rudolph II's Court during the early era of his reign (1578–1579), when the catholic reform began to play an important role.

Sulphite is a food additive used worldwide. Globally, for concentrations above 10 mg.kg-1, sulphite compounds must be labelled as sulphur dioxide (SO2) on the packaging due to their potential health risks. This study compares spectrophotometric (S), titration (T) (modified optimised Monier-Williams, OMW), and reflectoquant (R) methods for measuring sulphur dioxide in twenty fruit and vegetable products. The samples comprise sulphited, unsulphited, and naturally sulphur-containing products (Allium genus such as garlic and onion). The article discusses the strengths and weaknesses of the method used. Allium genus samples yielded false-positive results, especially in fresh garlic samples with average SO2 concentrations of 46, 1 152, and 40 mg.kg-1 obtained by titration, spectrophotometric, and reflectoquant methods, respectively, therefore, none of the methods is suitable for testing this type of vegetables or products containing a low proportion of them. For other types of samples, the methods showed acceptable working characteristics. Recovery tests showed 89.5, 82.0, and 75.2% recovery with 2.8, 3.9, and 13.2% repeatability and the limit of quantification of 1, 10, and 25 mg.kg-1 in the spectrophotometric, titration, and reflectoquant methods. The result highlights the importance of method selection based on sample characteristics and regulatory compliance.

Abstract Objectives Human papillomavirus (HPV) infections are highly prevalent sexually transmitted infections, notably associated with various cancers. This study analyses the health and economic impacts of HPV-associated diseases in the Czech Republic and evaluates the cost-effectiveness of a catch-up vaccination program. Methods Utilizing a Markov multistate model, the study assesses the lifetime impacts and costs related to HPV infections. Cohorts of ages 15–21 were simulated to assess the impact of catch-up vaccination outside the 11-year-old age group. Results The total quality-adjusted life years (QALYs) for the female and male cohorts (together 119,362 individuals) were higher in the vaccination scenario compared to the non-vaccination scenario. The increase in QALYs was 122,246 and 200,852 respectively, when considering the actual vaccination rates. Across both cohorts, 329 cancer-related deaths were prevented. In the probabilistic sensitivity analysis for the female population, vaccination was the dominant strategy in 99.3% of iterations. In the male population, vaccination was the dominant strategy in 80.3% of iterations. The implementation of catch-up vaccination for the 15–21 age group significantly increased QALY gains and reduced life-years-lost (LYLs). In the female cohort, all analysed rates of catch-up vaccination were the dominant strategy, while in the male cohort, the incremental cost-effectiveness ratios (ICERs) remained consistently below 42,000 CZK/QALY. Conclusions The catch-up vaccination program for 15-21-year-olds is cost-effective and can prevent a significant number of HPV-related cancers in both men and women.

This study analyses diploid and hydrostatic-pressure-induced triploid brook trout (Salvelinus fontinalis) sampled from a fish farm before (360 days post-hatch, d.p.h.) and during the normal time of sexual maturation for pan-sized market fish (555 and 667 d.p.h.). Biometric indices and slaughtering traits of examples of both ploidy levels were compared and their genomic heterozygosity and gonadal development were evaluated to assess the possible link between these two parameters and growth performance. At 555 d.p.h., triploids – irrespective of sex – had greater carcass yield than their diploid conspecifics. At 667 d.p.h., all triploids (females, males and intersex fish) were significantly larger and heavier and had far lower gonadosomatic indices than their diploid conspecifics; however, in terms of carcass and fillet yields these differences were not as notable. Delayed gonadal growth in triploids was confirmed. Microsatellite analysis at five polymorphic loci suggest that triploids could have higher levels of heterozygosity than their diploid counterparts, a trend in multilocus heterozygosity that was consistent in all three age groups. Nevertheless, the link between heterozygosity and body weight was inconclusive. Suppressed gonadal development seems to be more probable explanation for the improved growth performance of triploids. Remarkable occurrence of intersex fish in triploids at both 555 and 667 d.p.h suggests that intersexes can commonly arise from artificial triploidization in brook trout. External appearance, biometric indices and slaughtering traits of triploid intersex were highly similar to that of triploid females.

The article presents the international digital humanities project called History of Everyday Life Database and its possibilities for use in humanities research. The foundations of this project, which is run by the Institute of History of the Czech Academy of Sciences, were laid in the 1990s. Its main goal is to form a digital collection of sources (mainly biographical documents) on the history of “everyday life”. The database includes hundreds of such sources in the Czech and German languages and makes them accessible to scholars, students and the public, helping them get a different perspective on the past, one that was not shaped only by “great history”. The project also aims to create an interdisciplinary platform for mutual discussion and a common research space for scholars dealing with the topics of the history of everyday life and memory studies.

Abstract Microclimates have been recognised as one of the key drivers in global change biology. Durable microclimate loggers, detailed in‐situ measurements and sophisticated modelling tools are increasingly available, but a lack of standardised workflows for microclimate data handling hinders synthesis across the studies and thus progress in the global change biology. To overcome these limitations, we developed an R package myClim for microclimate data processing, storage and analyses. The myClim package supports complete workflow for microclimate data handling, including reading raw logger data files, their preprocessing and cleaning, time‐series' aggregation, calculation of ecologically relevant microclimatic variables, data export and storage. The myClim package stores data in a size‐efficient, hierarchical structure which respects the hierarchy of field microclimate measurement (locality > loggers > sensors). For imported microclimatic data, myClim provides an informative summary and automatically detects and corrects common issues like duplicated and wrongly ordered measurements. The myClim package also provides advanced functions for microclimate data aggregation to various timescales (e.g. days, months, years or growing seasons) as well as tools for sensor calibration, data conversion and joining of multiple microclimatic time series. The myClim package provides advanced functions for standardised calculation of ecologically relevant microclimatic variables like freezing and growing degree days, snow cover period, soil volumetric water content and atmospheric vapour pressure deficit. Calculated microclimatic variables are stored efficiently in myClim data format and can be easily exported to long or wide tables for further analyses and visualisations. Adopting myClim can facilitate large‐scale syntheses, boost data sharing and increase the comparability and reproducibility of microclimatic studies. The stable version of myClim is available on CRAN (https://cran.r‐project.org/web/packages/myClim) and the development version is available on GitHub (https://github.com/ibot‐geoecology/myClim).

Abstract Purpose The goal of our study is to find an optimal approach to the preparation and preservation of corneal stromal tissue. We want to compare different methods of corneal stromal tissue creation and storage to optimize the efficacy of this process under the conditions of an eye bank. After we find the most suitable method to create a safe high quality product, we want to prove the possibility of using a single donor cornea for more than one patient. We would also like to verify the feasibility of making more corneal lenticules after the removal of a corneal endothelium for DMEK transplantation. Methods We provided morphological (histology, scanning electron microscope) and microbiological analysis in order to compare different methods of corneal lenticule and corneal stromal lamellae preparation and preservation. We also tested the surgical handling of the tissue to secure a safe manipulation of the tissue for clinical use. We compared two methods of corneal lenticule preparation: microkeratome dissection and femtosecond laser. As methods of preservation, we tested hypothermia, cryopreservation at -80 degrees Celsius in DMSO (dimethyl sulfoxide) and storage at room temperature with glycerol. Some intrastromal lenticules and lamellae in each group were previously irradiated with gamma radiation of 25 kGy (KiloGray). Results Corneal stromal lamellae prepared with a microkeratome have a smoother cut – side surface compared to lamellae prepared with a femtosecond laser. Femtosecond laser preparation caused more irregularities on the surface and we detected more conglomerates of the fibrils, while lamellae made with microkeratome had more sparse network. Using femtosecond laser, we were able to make more than five lenticules from a single donor cornea. Gamma irradiation led to damage of collagen fibrils in corneal stroma and a loss of their regular arrangement. Corneal tissue stored in glycerol showed collagen fibril aggregates and empty spaces between fibrils caused by dehydration. Cryopreserved tissue without previous gamma irradiation showed the most regular structure of the fibrils comparable to storage in hypothermia. Conclusion Our results suggest that formation of a corneal lenticule lamellae by microkeratome results in smoother corneal lenticules, while being much cheaper than formation by femtosecond laser. Gamma irradiation of 25 kGy caused damage of the collagen fibres as well as their network arrangement, which correlated with loss of transparency and stiffer structure. These changes impair possible surgical utilisation of gamma irradiated corneas. Storage in glycerol at room temperature and cryopreservation had similar outcomes and we believe that both methods are appropriate and safe for further clinical use .

Abstract Background Planktonic microbial communities have critical impacts on the pelagic food web and water quality status in freshwater ecosystems, yet no general model of bacterial community assembly linked to higher trophic levels and hydrodynamics has been assessed. In this study, we utilized a 2-year survey of planktonic communities from bacteria to zooplankton in three freshwater reservoirs to investigate their spatiotemporal dynamics. Results We observed site-specific occurrence and microdiversification of bacteria in lacustrine and riverine environments, as well as in deep hypolimnia. Moreover, we determined recurrent bacterial seasonal patterns driven by both biotic and abiotic conditions, which could be integrated into the well-known Plankton Ecology Group (PEG) model describing primarily the seasonalities of larger plankton groups. Importantly, bacteria with different ecological potentials showed finely coordinated successions affiliated with four seasonal phases, including the spring bloom dominated by fast-growing opportunists, the clear-water phase associated with oligotrophic ultramicrobacteria, the summer phase characterized by phytoplankton bloom-associated bacteria, and the fall/winter phase driven by decay-specialists. Conclusions Our findings elucidate the major principles driving the spatiotemporal microbial community distribution in freshwater ecosystems. We suggest an extension to the original PEG model by integrating new findings on recurrent bacterial seasonal trends. Video Abstract

The reduction of NaCl content in cheeses is nutritionally desirable but quite challenging due to NaCl's key role during cheese production and ripening. We focused on reformulated Dutch-type cheeses ripened for 120 days and their microbiological and sensory characteristics, including of determining organic acids (electrophoresis) and volatile substances (SPME-GC-MS analysis). Experimental batches contained 0.64, 0.90, and 1.19% NaCl or 0.77% NaCl together with 0.33% KCl. The influence of salts on lactose and citrate metabolism (the formation of lactic, acetic, and formic acid, ethanol, diacetyl, acetoin and 2,3-butanediol), proteolysis (the formation of glutamic acid), lipolysis and β-oxidation of fatty acids (the formation of 2-butanone, 2-butanol, hexanal, hexanoic and octanoic acid) was undetected. Contrarily, brining conditions affected the contamination of cheese surfaces with yeasts and halotolerant microorganisms and cheese consistency. While a typical consistency was formed only in the cheeses with 1.19% NaCl acceptable saltiness was declared in the cheeses with the content of salts 0.90% or higher. The partial replacement of NaCl with KCl caused metallic off-taste in the cheeses that ripened longer than Consistent acceptance seems to be the most limiting factor for the tested reformulation appears.

Abstract Background The phytoplankton spring bloom in freshwater habitats is a complex, recurring, and dynamic ecological spectacle that unfolds at multiple biological scales. Although enormous taxonomic shifts in microbial assemblages during and after the bloom have been reported, genomic information on the microbial community of the spring bloom remains scarce. Results We performed a high-resolution spatio-temporal sampling of the spring bloom in a freshwater reservoir and describe a multitude of previously unknown taxa using metagenome-assembled genomes of eukaryotes, prokaryotes, and viruses in combination with a broad array of methodologies. The recovered genomes reveal multiple distributional dynamics for several bacterial groups with progressively increasing stratification. Analyses of abundances of metagenome-assembled genomes in concert with CARD-FISH revealed remarkably similar in situ doubling time estimates for dominant genome-streamlined microbial lineages. Discordance between quantitations of cryptophytes arising from sequence data and microscopic identification suggested the presence of hidden, yet extremely abundant aplastidic cryptophytes that were confirmed by CARD-FISH analyses. Aplastidic cryptophytes are prevalent throughout the water column but have never been considered in prior models of plankton dynamics. We also recovered the first metagenomic-assembled genomes of freshwater protists (a diatom and a haptophyte) along with thousands of giant viral genomic contigs, some of which appeared similar to viruses infecting haptophytes but owing to lack of known representatives, most remained without any indication of their hosts. The contrasting distribution of giant viruses that are present in the entire water column to that of parasitic perkinsids residing largely in deeper waters allows us to propose giant viruses as the biological agents of top-down control and bloom collapse, likely in combination with bottom-up factors like a nutrient limitation. Conclusion We reconstructed thousands of genomes of microbes and viruses from a freshwater spring bloom and show that such large-scale genome recovery allows tracking of planktonic succession in great detail. However, integration of metagenomic information with other methodologies (e.g., microscopy, CARD-FISH) remains critical to reveal diverse phenomena (e.g., distributional patterns, in situ doubling times) and novel participants (e.g., aplastidic cryptophytes) and to further refine existing ecological models (e.g., factors affecting bloom collapse). This work provides a genomic foundation for future approaches towards a fine-scale characterization of the organisms in relation to the rapidly changing environment during the course of the freshwater spring bloom. This paper deals with a phenomenon in combat sports called the glass jaw or glass chin. It is the ability of a fighter to withstand punches to the jaw area without falling unconscious and thus receiving a knockout. The main objective of this paper is to investigate the possibility of developing a glass jaw over the course of a fighter's career, and whether certain fighters are more predisposed to experiencing knockouts. Utilizing data from the largest Mixed Martial Arts organization, the Ultimate Fighting Championship, spanning from 2010 to 2021, statistical models reveal that certain fighters may be more susceptible to knockouts compared to others. Conversely, it could not be shown that the probability of a future knockout loss, that is developing a glass jaw, increased with the number of knockouts received. However, there remains space for a more detailed investigation of this phenomenon. The historically debated resistance training of punches to the head area cannot be recommended. Conversely, it is advisable to observe the fighters' reactions to punches to the head in order to pick out those with a greater predisposition for a knockout. They should then be especially cautious when practicing combat sports.
