BackgroundDespite the known health benefits of physical activity (PA), less than half and less than one-third of older adults in Germany reach the PA recommendations for endurance training and strength training, respectively, of the World Health Organization. The aim of this study is to investigate the implementation and effectiveness over the course of 9 months of two interventions (information technology [IT]-based vs print-based) for PA promotion among initially inactive older adults in a randomized, crossover trial. This study is part of a large research consortium (2015-2021) investigating different aspects of PA promotion. The IT-based intervention was previously developed and refined, while the print-based intervention was newly developed during this funding phase. ObjectiveWe aim to compare the effectiveness and examine the preferences of study participants regarding both delivery modes. MethodsOur target sample size was 390 initially inactive community-dwelling older adults aged ≥60 years at baseline (3-month follow-up [T1]: expected n=300; 9-month follow-up [T2]: expected n=240) who were randomized to one of two interventions for self-monitoring PA: IT-based (50%) or print-based (50%) intervention. In addition, 30% of the IT-based intervention group received a PA tracker. At T1, participants in both groups could choose whether they prefered to keep their assigned intervention or cross over to the other group for the following 6 months (T2). Participants’ intervention preferences at baseline were collected retrospectively to run a post hoc matched-mismatched analysis. During the initial 3-month intervention period, both intervention groups were offered weekly group sessions that were continued monthly between T1 and T2. A self-administered questionnaire and 3D accelerometers were employed to assess changes in PA between baseline, T1, and T2. Adherence to PA recommendations, attendance at group sessions, and acceptance of the interventions were also tracked. ResultsThe funding period started in February 2018 and ends in January 2021. We obtained institutional review board approval for the study from the Medical Association in Bremen on July 3, 2018. Data collection was completed on January 31, 2020, and data cleaning and analysis started in February 2020. We expect to publish the first results by the end of the funding period. ConclusionsStrategies to promote active aging are of particular relevance in Germany, as 29% of the population is projected to be ≥65 years old by 2030. Regular PA is a key contributor to healthy aging. This study will provide insights into the acceptance and effectiveness of IT-based vs print-based interventions to promote PA in initially inactive individuals aged ≥60 years. Results obtained in this study will improve the existing evidence base on the effectiveness of community-based PA interventions in Germany and will inform efforts to anchor evidence-based PA interventions in community structures and organizations via an allocation of permanent health insurance funds. Abstract Observational practice is discussed as a substitute for physical practice for motor learning and adaptation. We systematically reviewed the literature on observational practice in reaching and aiming tasks. Our objectives were to identify (i) performance differences between observational and physical practice; (ii) factors that contribute to adaptation following observational practice; and (iii) the neural correlates of observational practice. We found 18 studies, all investigated adaptation of reaching in visuomotor rotations or force-field perturbations. Results of the studies showed that observational practice led to adaptation in both, visuomotor rotation and force-field paradigms (d = −2.16 as compared to no practice). However, direct effects were considerably smaller as compared to physical practice (d = 4.38) and aftereffects were absent, suggesting that observational practice informed inverse, but not forward modes. Contrarily, neurophysiological evidence in this review showed that observational and physical practice involved similar brain regions.

Abstract Background Executive functions (EFs) and episodic memory are fundamental components of cognition that deteriorate with age and are crucial for independent living. While numerous reviews have explored the effect of exercise on these components in old age, these reviews screened and analyzed selected older adult populations, or specific exercise modes, thus providing only limited answers to the fundamental question on the effect of exercise on cognition in old age. This article describes the protocol for a systematic review and multilevel meta-analytic study aiming at evaluating the effectiveness of different types of chronic exercise in improving and/or maintaining EFs and long-term episodic memory in older adults. Methods and analysis The study protocol was written in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. Several databases will be searched. Randomized controlled trials (RCTs) conducted in older adults aged ≥ 60 years providing any kind of planned, structured, and repetitive exercise interventions, and EFs and/or episodic memory measures as outcomes, published in English in peer-reviewed journals and doctoral dissertations will be included. Two independent reviewers will screen the selected articles, while a third reviewer will resolve possible conflicts. The Cochrane risk-of-bias tool will be used to assess the quality of the studies. Finally, data will be extracted from the selected articles, and the formal method of combining individual data from the selected studies will be applied using a random effect multilevel meta-analysis. The data analysis will be conducted with the metafor package in R. Discussion and conclusion This review will synthesize the existing evidence and pinpoint gaps existing in the literature on the effects of exercise on EFs and episodic memory in healthy and unhealthy older adults. Findings from this meta-analysis will help to design effective exercise interventions for older adults to improve and/or maintain EFs and episodic memory. Its results will be useful for many researchers and professionals working with older adults and their families. Systematic review registration PROSPERO CRD42022367111.

Abstract There is a global trend of increasing antibiotic resistance to Gram‐negative bacteria and the medical research community is looking for alternative antibiotics. Antibiotics such as polymyxin B (PMB) show excellent efficacy and low resistance rates against Gram‐negative bacteria. The topical administration of PMB, however, has clinical limitations such as minimal residence time, making it difficult for its use to treat skin infections. Here, we present porous silicon microneedle (pSi MN) patches to deliver PMB to the skin. pSi MN patches are fabricated by, first, generating the projections by dry Si etching techniques, followed by an electrochemical etching to create porous layers around the MNs. After loading PMB into the porous layer, the antibacterial activity of pSi MNs is determined by measuring the inhibition zone of Gram‐negative Escherichia coli. Next, PMB is efficiently delivered into the epidermal layer of ex vivo skin models, as confirmed by chemical mapping using matrix‐assisted laser desorption/ionization mass spectrometry imaging (MALDI‐MSI). The results demonstrate that pSi MN patches are an efficient and versatile tool for the transdermal delivery of PMB with maintained bioactivity and antimicrobial efficacy, thus expanding the possibilities of current transdermal drug delivery systems.

Carbon-based nanomaterials are key to developing high-performing electrochemical sensors with improved sensitivity and selectivity. Nonetheless, limitations in their fabrication and integration into devices often constrain their practical applications. Moreover, carbon nanomaterials-based electrochemical devices still face problems such as large background currents, poor stability, and slow kinetics. To advance towards a new class of carbon nanostructured electrochemical transducers, we propose the in-situ polymerization and carbonization of furfuryl alcohol (FA) on porous silicon (pSi) to produce a tailored and highly stable transducer. The thin layer of polyfurfuryl alcohol (PFA) that conformally coats the pSi scaffold transforms into nanoporous carbon when subjected to pyrolysis above 600 °C. The morphological and chemical properties of PFA-pSi were characterized by scanning electron microscopy, and Raman and X-ray photoelectron spectroscopies. Their stability and electrochemical performance were investigated by cyclic voltammetry and electrochemical impedance spectroscopy in [Fe(CN)6]3-/4-, [Ru(NH3)6]2+/3+, and hydroquinone. PFA-pSi showed superior electrochemical performance compared to screen-printed carbon electrodes while also surpassing glassy carbon electrodes in specific aspects. Besides, PFA-pSi has the additional advantage of easy tuning of the electroactive surface area. To prove its potential for biosensing purposes, a DNA sensor based on quantifying the partial pore blockage of the pSi upon target hybridization was built on PFA-pSi. The sensor showed a limit of detection of 1.4 pM, outperforming other sensors based on the same sensing mechanism.

Abstract In order to grasp and transport an object, grip and load forces must be scaled according to the object’s properties (such as weight). To select the appropriate grip and load forces, the object weight is estimated based on experience or, in the case of robots, usually by use of image recognition. We propose a new approach that makes a robot’s weight estimation less dependent on prior learning and, thereby, allows it to successfully grasp a wider variety of objects. This study evaluates whether it is feasible to predict an object’s weight class in a replacement task based on the time series of upper body angles of the active arm or on object velocity profiles. Furthermore, we wanted to investigate how prediction accuracy is affected by (i) the length of the time series and (ii) different cross-validation (CV) procedures. To this end, we recorded and analyzed the movement kinematics of 12 participants during a replacement task. The participants’ kinematics were recorded by an optical motion tracking system while transporting an object, 80 times in total from varying starting positions to a predefined end position on a table. The object’s weight was modified (made lighter and heavier) without changing the object’s visual appearance. Throughout the experiment, the object’s weight (light/heavy) was randomly changed without the participant’s knowledge. To predict the object’s weight class, we used a discrete cosine transform to smooth and compress the time series and a support vector machine for supervised learning from the achieved discrete cosine transform parameters. Results showed good prediction accuracy (up to $$95\%$$ 95 % , depending on the CV procedure and the length of the time series). Even at the beginning of a movement (after only 300 ms), we were able to predict the object weight reliably (within a classification rate of $$88-94\%$$ 88 - 94 % ).

The investigation of gait and its neuronal correlates under more ecologically valid conditions as well as real-time feedback visualization is becoming increasingly important in neuro-motor rehabilitation research. The Gait Real-time Analysis Interactive Lab (GRAIL) offers advanced opportunities for gait and gait-related research by creating more naturalistic yet controlled environments through immersive virtual reality. Investigating the neuronal aspects of gait requires parallel recording of brain activity, such as through mobile electroencephalography (EEG) and/or mobile functional near-infrared spectroscopy (fNIRS), which must be synchronized with the kinetic and /or kinematic data recorded while walking. This proof-of-concept study outlines the required setup by use of the lab streaming layer (LSL) ecosystem for real-time, simultaneous data collection of two independently operating multi-channel EEG and fNIRS measurement devices and gait kinetics. In this context, a customized approach using a photodiode to synchronize the systems is described. This study demonstrates the achievable temporal accuracy of synchronous data acquisition of neurophysiological and kinematic and kinetic data collection in the GRAIL. By using event-related cerebral hemodynamic activity and visually evoked potentials during a start-to-go task and a checkerboard test, we were able to confirm that our measurement system can replicate known physiological phenomena with latencies in the millisecond range and relate neurophysiological and kinetic data to each other with sufficient accuracy.
