Abstract Laser weeding may contribute to less dependency on herbicides and soil tillage. Several research and commercial projects are underway to develop robots equipped with lasers to control weeds. Artificial intelligence can be used to locate and identify weed plants, and mirrors can be used to direct a laser beam towards the target to kill it with heat. Unlike chemical and mechanical weed control, laser weeding only exposes a tiny part of the field for treatment. Laser weeding leaves behind only ashes from the burned plants and does not disturb the soil. Therefore, it is an eco-friendly method to control weed seedlings. However, perennial weeds regrow from the belowground parts after the laser destroys the aerial shoots. Depletion of the belowground parts for resources might be possible if the laser continuously kills new shoots, but it may require many laser treatments. We studied how laser could be used to destroy the widespread and aggressive perennial weed Elymus repens after the rhizomes were cut into fragments. Plants were killed with even small dosages of laser energy and stopped regrowing. Generally, the highest efficacy was achieved when the plants from small rhizomes were treated at the 3-leaf stage.

Abstract Introduction EXO-CD24 are exosomes genetically manipulated to over-express Cluster of Differentiation (CD) 24. It consists of two breakthrough technologies: CD24, the drug, as a novel immunomodulator that is smarter than steroids without any side effects, and exosomes as the ideal natural drug carrier. Methods A randomized, single blind, dose-finding phase IIb trial in hospitalized patients with mild to moderate Coronavirus disease 2019 (COVID-19) related Acute Respiratory Distress Syndrome (ARDS) was carried out in two medical centers in Athens. Patients received either 109 or 1010 exosome particles of EXO-CD24, daily, for five consecutive days and monitored for 28 days. Efficacy was assessed at day 7 among 91 patients who underwent randomization. The outcome was also compared in a post-hoc analysis with an income control group (n = 202) that fit the inclusion and exclusion criteria. The massive use of herbicides since the 1950s has resulted in increasing problems with herbicideresistant weeds and pollution of the environment, including food, feed, and water. These side effects have resulted in political pressures to reduce herbicide application. The European Commission aims to reduce the use and risk of chemicals and more hazardous pesticides in the EU. Therefore, new weed control methods are in demand. Laser weeding might be an alternative to replace or supplement herbicides and other weed control methods in an Integrated Weed Management (IPM) strategy. This work aimed to investigate how increasing laser energy affected common weeds when the apical meristem was exposed to irradiation at the early stages of development. A 50 W thulium-doped fibre laser with a diameter of 2 mm and a wavelength of 2 µm was used. The highest efficacy of laser irradiation was achieved when the grass weed (Alopecurus myosuroides) had one leaf and the dicot species were at the cotyledon stage. There was a large difference between the species’ susceptibility to irradiation probably caused by differences in morphology and growth habit. At the 4-leaf stage, most of the species regrew after irradiation. Laser weeding may be a solution to replace or supplement other weed control methods in some crops, but in general the weeds must be irradiated when they are at the cotyledon to 2-leaf stage to avoid regrowth.

Background: Workplace stress and burnout in the Greek healthcare system had been considered severe even before the high pressure of the COVID-19 pandemic. We aimed to investigate occupational quality of life and burnout effects on workplace errors among the administrative staff in the Greek healthcare system. Methods: We enrolled 120 administrative healthcare employee participants between April and May 2019. Occupational burnout was assessed using the Maslach Burnout Inventory—Human Services Survey and the Hospital Anxiety and Depression Scale. Findings: Inadequate staffing, a low sense of well-being, exhaustion, and low family income were associated with workplace errors. Increased workload and staff shortages were associated with occupation related quality of life. Conclusions: Targeted interventions supporting healthcare staff mental health are warranted. Application to Practice: Wellness and professional burnout can affect professional efficiency and are associated with workplace errors in the healthcare sector. Targeted interventions are warranted to support the mental health of healthcare staff during work and to prevent incidents of post-traumatic stress. Shortages of staffing may lead to an increase in the cost of the provided services.

Farmers are greatly interested in reducing weed seeds in their fields to avoid unnecessary weed infestation. Autonomous vehicles equipped with plant recognition systems and lasers can be used to control weed plants and may therefore be used to replace or supplement herbicides and mechanical weed control. However, less is known about the ability of laser to control weed seeds. In this study, seeds of weeds (Alopecurus myosuroides, Anisantha sterilis, Avena fatua, Centaurea cyanus, Silene noctiflora) and crops (wheat (Triticum aestivum), maize (Zea mays)) were exposed to increasing dosages of laser energy. The species represented dicots and monocots with different seed sizes and morphology. We used a thulium-doped 50 W fiber laser with a wavelength of 2 µm and a diameter of 2 mm developed for weed control. The seeds were exposed to laser directly on the seed surface or after been covered with soil (2.5 and 5 mm). Small doses of energy (0.4 J mm−2 and 0.8 J mm−2) affected the germination ability of small seeds (S. noctiflora and C. cyanus) when the seeds were irradiated directly on the seed surface, and they were completely burned at the high doses (8.0 J mm−2 and 15.9 J mm−2). However, there was not a clear relationship between seed size and sensitivity to laser dose. Additionally, increasing the laser dose on the seed surface resulted in increasing infection of fungi. Seeds covered with soil were not affected by the laser treatments except C. cyanus seeds. Controlling large seeds on the ground while weed seedlings are controlled with laser robots seems realistic in the future. However, dosages higher than 50 J (~15.9 J mm−2) is necessary to control large seeds.

This project aimed to study the effect of laser on some living non-target organisms. We investigated the effect of laser treatment on the mortality of two species of earthworms (Enchytraeus albidus and Enchytraeus crypticus), larvae, pupas, and beetles of yellow mealworm beetles (Tenebrio molitor) and the two-spotted lady beetle (Adalia bipunctata) for increasing dosages of laser energy. In all earthworms experiments except one, the mortality rates of the worms living in the uppermost soil layer of clay, sandy, and organic soil exposed to laser heating were not significantly different from the controls even with laser dosages up to 23.8 J mm-2. Laser doses sufficient to kill plants were lethal to the insects, and lower doses that did not kill plants, killed or harmed the insects across all life stages tested. The larger beetles survived higher doses than smaller. Laser weeding is a relatively new technology and not yet widely practiced or commercialized. Therefore, we do not discuss and compare the costs of the different weeding methods at this early stage of the development of the technology.

We review the establishment of computational biology in Greece and Cyprus from its inception to date and issue recommendations for future development. We compare output to other countries of similar geography, economy, and size—based on publication counts recorded in the literature—and predict future growth based on those counts as well as national priority areas. Our analysis may be pertinent to wider national or regional communities with challenges and opportunities emerging from the rapid expansion of the field and related industries. Our recommendations suggest a 2-fold growth margin for the 2 countries, as a realistic expectation for further expansion of the field and the development of a credible roadmap of national priorities, both in terms of research and infrastructure funding.

The need for a consistent marketing evaluation framework has been highlighted by destination authorities, who in collaboration with academia and marketing professionals have sought to formulate methodologies for measuring the impact of their campaigns. Although several attempts have been made, no simple solution has emerged for evaluating destination marketing activities. This study draws on This is Athens & Partners to reveal the interaction that should take place when destination authorities employ external experts to implement and evaluate their marketing campaigns. The collaboration required adopting the appropriate destination marketing evaluation methodology, which is presented. The adapted methodology, formulated through consultation, resulted in the selection of the advertising tracking survey as the data collection method and the adaptation of a measurement instrument. The research findings show that destination marketing can positively influence the funnel process potential travelers consider when deciding on a travel destination, only when digital tools are combined with effective strategic marketing planning and, more recently, with references to resilience and sustainability. Insights from this paper regarding the importance of establishing an informative evaluation methodology to mitigate potential deficiencies in planned marketing initiatives may prove of added value to destination authorities and stakeholders. TYK2 is a nonreceptor tyrosine kinase, member of the Janus kinases (JAK), with a central role in several diseases, including cancer. The JAKs' catalytic domains (KD) are highly conserved, yet the isolated TYK2‐KD exhibits unique specificities. In a previous work, using molecular dynamics (MD) simulations of a catalytically impaired TYK2‐KD variant (P1104A) we found that this amino acid change of its JAK‐characteristic insert (αFG), acts at the dynamics level. Given that structural dynamics is key to the allosteric activation of protein kinases, in this study we applied a long‐scale MD simulation and investigated an active TYK2‐KD form in the presence of adenosine 5′‐triphosphate and one magnesium ion that represents a dynamic and crucial step of the catalytic cycle, in other protein kinases. Community analysis of the MD trajectory shed light, for the first time, on the dynamic profile and dynamics‐driven allosteric communications within the TYK2‐KD during activation and revealed that αFG and amino acids P1104, P1105, and I1112 in particular, hold a pivotal role and act synergistically with a dynamically coupled communication network of amino acids serving intra‐KD signaling for allosteric regulation of TYK2 activity. Corroborating our findings, most of the identified amino acids are associated with cancer‐related missense/splice‐site mutations of the Tyk2 gene. We propose that the conformational dynamics at this step of the catalytic cycle, coordinated by αFG, underlie TYK2‐unique substrate recognition and account for its distinct specificity. In total, this work adds to knowledge towards an in‐depth understanding of TYK2 activation and may be valuable towards a rational design of allosteric TYK2‐specific inhibitors. The genus Lysobacter includes several bacterial species which show potential for being used in biological control of plant diseases. It was shown recently that several Lysobacter type strains produce volatile organic compounds (VOCs) which controlled the growth of Phytophthora infestans in vitro when the bacteria were grown on a protein rich medium. In the present study, Lysobacter capsici AZ78 (AZ78) has been tested for its potential to produce VOCs that may contribute to the bioactivity against soilborne plant pathogens. To this end, split Petri dish assays of bacterial cultures have been combined with GC-MS measurements with the aim to reveal the identity of the VOCs which inhibit the growth of Pythium ultimum Rhizoctonia solani, and Sclerotinia minor. While AZ78 completely suppressed the growth of P. ultimum and S. minor, the growth of R. solani was still reduced significantly. The GC-MS analysis revealed 22 VOCs to be produced by AZ78, the majority of which were (putatively) identified as mono- and dialkylated methoxypyrazines. Based on additional cultivation and GC-MS experiments, 2,5-dimethylpyrazine, 2-ethyl-3-methoxypyrazine and 2-isopropyl-3-methoxypyrazine were selected as presumable bioactive compounds. Further bioassays employing indirect exposure to standard solutions (1–10 mg per Petri dish) of the synthetic compounds via the gas phase, revealed that each of these pyrazines was able to suppress the growth of the pathogens under investigation. The results of this study highlight the possible future implementation of pyrazine derivatives in the control of soilborne plant diseases and further support the biocontrol potential of L. capsici AZ78.

Determining the mode of action of microbial biocontrol agents plays a key role in their development and registration as commercial biopesticides. The biocontrol rhizobacterium Lysobacter capsici AZ78 (AZ78) is able to inhibit a vast array of plant pathogenic oomycetes and Gram-positive bacteria due to the release of antimicrobial secondary metabolites. A combination of MALDI-qTOF-MSI and UHPLC-HRMS/M was applied to finely dissect the AZ78 metabolome and identify the main secondary metabolites involved in the inhibition of plant pathogenic microorganisms. Under nutritionally limited conditions, MALDI-qTOF-MSI revealed that AZ78 is able to release a relevant number of antimicrobial secondary metabolites belonging to the families of 2,5-diketopiperazines, cyclic lipodepsipeptides, macrolactones and macrolides. In vitro tests confirmed the presence of secondary metabolites toxic against Pythium ultimum and Rhodococcus fascians in AZ78 cell-free extracts. Subsequently, UHPLC-HRMS/MS was used to confirm the results achieved with MALDI-qTOF-MSI and investigate for further putative antimicrobial secondary metabolites known to be produced by Lysobacter spp. This technique confirmed the presence of several 2,5-diketopiperazines in AZ78 cell-free extracts and provided the first evidence of the production of the cyclic depsipeptide WAP-8294A2 in a member of L. capsici species. Moreover, UHPLC-HRMS/MS confirmed the presence of dihydromaltophilin/Heat Stable Antifungal Factor (HSAF) in AZ78 cell-free extracts. Due to the production of HSAF by AZ78, cell-free supernatants were effective in controlling Plasmopara viticola on grapevine leaf disks after exposure to high temperatures. Overall, our work determined the main secondary metabolites involved in the biocontrol activity of AZ78 against plant pathogenic oomycetes and Gram-positive bacteria. These results might be useful for the future development of this bacterial strain as the active ingredient of a microbial biopesticide that might contribute to a reduction in the chemical input in agriculture.
