The aim of the study was to investigate the effects of chronic nandrolone decanoate treatment and/or swimming training on immunohistomorphometric parameters on rat pituitary gonadotropic cells. Male Wistar albino rats, 10 weeks old, were classified into four groups: control (T−N−), nandrolone (T−N+), swimming training (T+N−), and swimming training with nandrolone (T+N+). The T+ groups swam for 4 weeks, 1 h/day, 5 days/week. The N+ groups received nandrolone decanoate (20 mg/kg) once per week for 4 weeks. Pituitary tissue sections were processed and stained for immunohistochemical analysis and immunofluorescence. The volume density of luteinizing hormone (LH) cells was decreased by 48% in T−N+ and for 35% in the T+N+ group. The volume density of follicle-stimulating hormone (FSH) cells was decreased by 39% in T−N+ and for 30% in T+N+ compared to the control. Nandrolone alone, or combined with swimming training, decreased the number of LH/FSH cells compared to the control. The levels of the immunofluorescent signal of LH/FSH cells were increased in all experimental groups. Nandrolone alone decreased the serum level of LH by 17%, whereas swimming training alone increased FSH levels by 11% compared to the control. Serum levels of testosterone were increased in all experimental groups. Nandrolone alone, or combined with swimming training, decreased immunohistomorphometric parameters of gonadotropic cells, whereas the levels of immunofluorescent signal were increased.

Introduction/Objective. In this study, we have pointed out the immunohistomorphometric characteristics of somatotropic (GH) and folliculostellate (FS) cells of the male pituitary gland during ageing. Methods. On histological sections of the pituitary gland of 14 male cadavers of different ages, the GH and FS cells were immunohistochemically labeled with corresponding antibodies, monoclonal anti-GH antibody, and polyclonal anti-S100 antibody, respectively. Immunopositive GH- and FS-cells were further morphometrically analyzed using ImageJ software. Results. The obtained results of morphometric analysis showed that the surface area of GH cells increased significantly with age. In these cells, the nuclear?cytoplasmic ratio gradually decreased and became significantly higher after the age of 70 years. The volume density of GH cells has not changed during ageing, while in FS cells this parameter significantly increased in the cases older than 70 years. The nuclear?cytoplasmic ratio of GH cells is negatively correlated with the volume density of FS cells. Conclusion. Based on the obtained results, we concluded that hypertrophy of GH and FS cells occurs in men with ageing and that correlation between the morphometric parameters of these two cell types indicates their mutual interaction.

Anabolic androgenic steroids (AASs) are synthetic analogs of testosterone often used by athletes to increase the skeletal muscle mass. Our goal was to examine the effects of physical activity and physical activity combined with supraphysiological doses of nandrolone on functional morphology of the quadriceps muscle. The study included 32 peripubertal Wistar rats, divided into 4 groups: control (T-N-), nandrolone (T-N+), physical activity (T+N-) and physical activity plus nandrolone (T+N+) groups. The T+N- and T+N+ group swam for 4 weeks, 1 h/day, 5 days/week. The T-N+ and T+N+ groups received nandolone decanoate (20 mg/kg b.w.) once per week, subcutaneously. Subsequently, the rats were sacrificed and muscle specimens were prepared for the processing. Tissue sections were histochemically and immunohistochemically stained, while the image analysis was used for quantification. Longitudinal diameter of quadriceps muscle cells was increased for 21% in T-N+, for 57% in T+N- and for 64% in T+N+ group while cross section muscle cell area was increased in T-N+ for 19%, in T+N- for 47% and in T+N+ group for 59%, compared to the control. Collagen fibers covered area was increased in T-N+ group for 36%, in T+N- for 109% and in T+N+ group for 159%, compared to the control. Erythrocyte depots were decreased in T-N+ group and increased in T+N- and T+N+ group, in comparison with T-N-. VEGF depots were increased in all treated groups. Chronic administration of supraphysiological doses of AASs alone or in combination with physical activity induces hypertrophy and significant changes in the quadriceps muscle tissue structure.

The effects of estradiol-dipropionate (EDP) or human chorionic gonadotropin (hCG) on immunohistomorphometric characteristics of pituitary GH cells in infant and peripubertal female rats were investigated. The first group of females received five injections of EDP (0.25 mg/kg b.w.) during the neonatal period of life, and was further divided into two subgroups which were sacrificed at the infantile period (17th day) or at the peripubertal period (38th day). The second group received two doses of hCG (50 IU/kg b.w.) on the 15th and 16th day of life in the first subgroup, and on the 36th and 37th days of life in the second subgroup, while they were sacrificed 24 h after the last treatment, respectively. Aging is a complex process characterized by a progressive decline in cellular function, alterations of the endocrine system and the increased incidence of thyroid diseases. Citrus flavanones naringenin (NAR) and hesperetin (HES) may contribute to maintenance of health at old age however they may also affect thyroid hormone economy. The aim of this study was to test weather NAR or HES administration alter thyroid functioning. NAR or HES were administrated orally (15 mg/kg) to male 24-month-old Wistar rats during 4 weeks. Control groups received vehicle, sunflower oil. Immunohistochemical staining of thyroglobulin (Tg), thyroxine (T4) bound to Tg (T4-Tg) and sodium iodide sysmporter (NIS) were performed. Immunopositivity of Tg or T4-Tg in thyroid follicles were measured as intensity of optical density (OD) or relative intensity of fluorescence (RIF). Thyroid stimulating hormone (TSH) and total T4 concentrations were measured in serum. After citrus flavanones treatments, thyroids showed Tg and T4-Tg immunopositivity mainly in the colloid of the follicles. The obtained results showed increased (P ! 0.05) intensity of OD for Tg and T4-Tg RIF after both flavanones treatments, while NIS expression did not change. Serum TSH increased (P ! 0.05) only after NAR, although T4 remained unchanged after both treatments. In conclusion, citrus flavanones induced stronger expression of thyroid-specific proteins in gland of old-aged males. However, only NAR elevated serum TSH, being more potent than HES. This alteration is in line with preserved capacity of the thyroid to compensate the mild interference of flavanones treatments with thyroid hormone production in old-aged males.

The pituitary appears to be vulnerable to brain trauma, and its dysfunction is a common feature after traumatic brain injury. The role of pituitary growth hormone (GH) in brain repair after injury has been envisaged, but more studies must be performed to understand completely the importance of GH in these processes. Because some of the neuroprotective effects of GH are mediated directly through the GH receptor (GHR), we examined GHR expression in the rat cerebral cortex after sensorimotor cortex ablation. RT-PCR, immunohistochemistry, and double immunofluorescence had been performed to analyze the correlation between GHR expression in the injured cortex and activity of GH cells in the pituitary. Our results showed that the volume of GH-immunopositive cells was reduced at days 2 and 7 postsurgery (dps), and volume density of GH cells was significantly decreased at 14 dps, all compared with appropriate sham controls. At 30 dps all investigated parameters had returned to control level. In the injured cortex, GHR expression was transiently upregulated. Increased GHR immunoreactivity was observed in reactive astrocytes at 7 and particularly at 14 dps. In neuronal cells, an increase of GHR immunoreactivity was seen in neuronal cell bodies and well-defined primary dendrites at 14 and especially at 30 dps. The results presented here suggest that, during recovery from brain injury, changes in activity of pituitary GH cells result in upregulation of GHR that may have a role in neuronal arborization and glial proliferation in the injured cortex. © 2014 Wiley Periodicals, Inc.

The effects of estradiol (EDP) and human chorionic gonadotropin (HCG) on histological and stereological features of adrenocorticotropic (ACTH) cells in peripubertal female rats were examined. The first group of females received five injections of EDP (0.25 mg/kg b.w.), every second day from the 4th to 14th day after birth, and was killed at the peripubertal stage. The second group of females was given two injections of pregnyl-gonadotrophinum chorionicum (HCG; 50 IU/kg body weight) on the 36th and 37th days after birth, and it was killed 24 h after the last treatment. The controls were injected with an equivalent volume of the corresponding vehicle. ACTH cells were immunohistochemically labeled and stereologically evaluated. Stereological analysis showed that the volume of ACTH cells and their volume density in peripubertal females treated with EDP, were decreased by 15.6\% and 53.8\% (p < 0.05), respectively, compared to the controls. In HCG-treated animals, the observed parameters were increased by 39.2\% and 15.4\% (p < 0.05), respectively, in comparison with the control females. These findings suggest that the application of EDP or HCG exerted opposite effects on the stereological features of pituitary ACTH cells. Modified incubation factors affect skeletal musculature development of broiler chickens during embryonic and postnatal periods of development. These changes appear to have great influence on the proliferation of myoblasts and muscle growth after hatching. In this study, the histomorphological and -morphometric parameters of broiler chicken leg musculature, after thermal and light treatments during embryogenesis, were examined. The applied treatments included thermal manipulation (exposure of fertilized eggs to increased temperature), light manipulation (exposure of fertilized eggs to monochromatic green light) and simultaneous thermal and light manipulations. Following the specific parameters that were observed, it was determined that in the late postnatal period of development, treated groups have a larger diameter and smaller nucleocytoplasmic ratio of muscle cells compared to those of the control group, in which fertilized eggs were incubated at a constant temperature of 37.8 degrees C, without light. Diameter was increased by 4.20\%, 3.77\% and 4.55\% on day 42 after hatching in thermal, light and combined thermal and light treated groups, respectively, compared to the control. The nucleocytoplasmic ratio was increased by 37.5\% on day 42 after hatching in the control group compared to all treated groups. The volume density of the muscle connective tissue was approximately at the same level between the groups. It can be concluded that thermal and light treatments stimulate the proliferation of myoblasts and enhance development and growth of leg muscles in broiler chickens. Hyperthermia can cause significant structural and functional reorganization of tissues and organs. The proliferative and apoptotic processes of rat adrenal cortex were analyzed by light and electron microscopy after an acute exposure to high ambient temperature. Animals were divided in two groups. The soybean phytoestrogen, genistein, is increasingly consumed as an alternative therapeutic for age-related diseases. The aim of this study was to examine the morphofunctional characteristics of adrenocorticotrophic (ACTH) cells and blood concentrations of ACTH in sham-operated, orchidectomized and genistein-treated orchidectomized, 16-month-old Wistar male rats. Genistein (10 mg/kg/day) was administered subcutaneously for three weeks, while the control groups received the vehicle alone. Orchidectomy and genistein treatment decreased the volume density of ACTH cells and reduced (p < 0.05) circulating ACTH concentrations in comparison with control groups. Traumatic brain injury (TBI) represents a serious event with far reaching complications, including pituitary dysfunction. Pars distalis corticotropes (ACTH cells), that represent the active module of hypothalamo-pituitary-adrenocortical axis, seem to be affected as well. Since pituitary failure after TBI has been associated with neurobehavioral impairments the aim of this study was to evaluate the effects of TBI on recovery of motor functions, morphology and secretory activity of ACTH cells in the pituitary of adult rats. Wistar male rats, initially exposed to sensorimotor cortex ablation (SCA), were sacrificed at the 2nd, 7th, 14th and 30th days post-surgery (dps). A beam walking test was used to evaluate the recovery of motor functions. Pituitary glands and blood were collected for morphological and hormonal analyses. During the first two weeks post-injury increased recovery of locomotor function was detected, reaching almost the control value at day 30. SCA induces significant increase of pituitary weights compared to their time-matched controls. The volume of ACTH-immunopositive cells was reduced at the 7th dps, while at the 14th dps their volume was enlarged, in comparison to corresponding sham controls. Volume density of ACTH cells was increased only at 14th dps, while at day 30 this increase was insignificant. The plasma level of ACTH transiently increased after the injury. The most pronounced changes were observed at the 7th and 14th dps, and were followed by decrease toward control levels at the 30th dps. Thus, temporal changes in the hypothalamic-pituitary-adrenal axis after traumatic brain injury appear to correlate with the recovery process. (C) 2013 Elsevier Inc. All rights reserved. The effects of chronic treatments with SRIH-14 and octreotide on pituitary corticotropes (ACTH cells) and on the adrenal cortex of male Wistar rats were examined. Adult males received two daily s.c. injections of 20 ?g/100 g of body weight of either SRIH-14 or octreotide for 28 consecutive days.
