1. Effects of Iron on oxidative stress of Cecropia hololeuca and Carica papaya plants

Abstract

Abstract Iron is essential to plant development. However, its excess can provoke an increase in reactive oxygen species and oxidative stress in plants. The objective of this work was to verify the effects of high concentrations of iron on the oxidative stress of seeds and young plants of Cecropia hololeuca and Carica papaya. The species were submitted to concentrations of 0.045, 4 and 8mM of iron in the form of ferrous sulfate and FeEDTA. The experiments of germination and initial growth took place in a growth chamber, with temperature of 25ºC and 12h photoperiod. We performed the lipid peroxidation test by extraction and quantification of malonaldehyde and hydrogen peroxide. The application of iron did not cause a significant elevation in the contents of malonaldehyde and hydrogen peroxide in the germination of C. hololeuca and C. papaya. In the young plants, the hydrogen peroxide did not change in any of the treatments. However, it was possible to observe an expressive increase in malonaldehyde concentration in both species when exposed to FeEDTA 4 to 8mM. The results indicate a sensibility of C. hololeuca and C. papaya to high iron levels, amplifying the oxidative stress process that can harm their growth and initial development.

2. Terapia de rescate con micofenolato mofetil en pacientes con nefritis lúpica

Abstract

La terapia a largo plazo con ciclofosfarnida combinada con esteroides mejora la sobrevida renal en pacientes con Nefritis Lúpica Proliferativa, pero con efectos tóxicos considerables. En años recientes, el Micofenolato Mofetil (MM?) un inmunosupresor usado en trasplante, parece ser efectivo en casos selectos de Nefritis Lúpica. Métodos Describiremos 6 pacientes con Nefritis Lúpica clase IV y y según OMS que por distintas razones debieron ser tratados con MM? como droga de rescate. El mismo estabilizó la función renal, controló la actividad del LES y permitió la disminución o suspensión del corticoide. Resultados En tres casos se logró remisión completa, dos presentaron remisión parcial y tuvimos un fracaso por plaquetopenia y leucopenia severa con sepsis grave a punto de partida de una neumopatía; en este paciente se suspendió el tratamiento. Uno de ellos presentó un herpes zóster que se trató con Aciclovir y la suspensión transitoria de MM?.. Conclusión EL MM? fue efectivo para conseguir remisiones en la NL y mantener inactivo el LES, por lo que se lo debiera considerar como terapia de rescate, o bien para tratamiento de mantenimiento luego de la inducción con ciclofosfamida, o directamente como terapia de inducción en aquellos pacientes donde la fertilidad es un factor importante. Seguimientos a largo plazo de las pacientes son necesarios para evaluar su efectividad en la sobreviva renal, como ya ha sido demostrado con la Ciclofosfamida.

Abstract

This article provides information on the characteristics of diverse phages of lactic acid bacteria and highlights the incidence of their presence in different dairy fermentations. As it is known, thermal treatments on raw milk and use of sanitizers in the disinfection of surfaces and equipment are strategies usually applied in dairy to prevent bacteriophage infections. In this sense, this review mainly focuses on the existing data about the resistance against thermal treatments and sanitizers usually used in the dairy industry worldwide, and the differences found among bacteriophages of diverse genera are remarked upon. Also, we provide information concerning the problems that have arisen as a consequence of the potential presence of bacteriophages in cheese whey powder and derivatives when they are added in fermented dairy product manufacturing. Finally, some important conclusions on each topic are marked and checkpoints to be considered are suggested.

5. Malignant melanoma in Chile: different site distribution between private and state patients

BACKGROUND: The body site location of primary Malignant Melanoma (MM) has been correlated with prognosis and survival. Ethnic, genetics, sun exposure factors are related to the anatomical distribution of MM. Low and high socioeconomic strata in Chile differ in ethnic, genetic and cultural conditions. The purpose of this study was to analyze the anatomical MM distribution in the Chilean population in both strata searching for differences due to their ethno-genetic-cultural differences. Records of 1148 MM, 575cases from state hospitals (Low Socioeconomic Strata, LSS) and 573 cases from private clinics (High Socioeconomic Strata, HSS) were analyzed by body site. RESULTS: Females from LSS showed a higher number of MM in soles, cheeks, and around the eye area. Females from the HSS showed a higher number of MM in dorsal feet and dorsal hands. Males from LSS showed a higher number of MM in soles, around the eye area, and cheeks. However, males from HSS showed a higher number of MM in the trunk, and in the arms. Acral MM was significantly higher in LSS than in the HSS in both sexes. The Chilean population from the HSS and LSS showed differences in the distribution of MM by site. Furthermore, gender differences in the proportion of MM analyzed by anatomical site are observed in both strata. CONCLUSIONS: Results show evidence that differential genetics factors, sun exposure, or other environmental or cultural factors of both strata may account for these differences.

6. An analysis of multiplex-PCR in the detection of BCR-ABL transcripts in hematological disorders

Abstract

In this work, we describe the advantages of multiplex-PCR in the specific detection of BCR-ABL transcripts in different hematological disorders and its sensitivity compared to nested PCR. Fifty-three patients were studied for the presence of BCR-ABL transcripts: 24 patients with chronic myeloid leukemia (CML), 20 with acute leukemia (AL), and 9 patients with other hematological disorders. A variant rearrangement (b3a3) was found in a single case of CML (4.2%). Four out of the 20 patients with AL (20.0%) (14 adults, 6 children) were bcr-abl(+), and in this group three cases were classified as B-acute lymphoblastic leukemia (B-ALL), and one as acute myeloblastic leukemia (AML). Two of the three patients with B-ALL were positive for b2a2 and the other one for e1a2, while in the BCR-ABL(+)AML patients a b3a2 rearrangement was observed. In conclusion, multiplex-PCR allows rapid, specific and simultaneous detection of different types of BCR-ABL transcripts in CML and ABL-BCR(+)AL. A full correlation was observed when multiplex-PCR was compared with nested PCR.

Abstract

Several edible agro-industrial co-products have been studied as unconventional ingredients in ruminant diets to reduce the environmental impact of food production chains. When the chemical profile of co-products is characterised by the presence of bioactive molecules, they represent a promising tool to modulate rumen microbiota activity. In the Mediterranean area, the olive oil production chain generates olive oil pomace and olive tree leaves post-milling that are animal edible bio-waste. Two in vitro trials were carried out to investigate the effect of olive oil pomace and olive tree leaves as dietary ingredients on rumen fermentation and microbiome ecology. Two experimental diets, respectively containing olive oil pomace or olive tree leaves, and the related control diets, formulated to be isoproteic and isoenergetic, were fermented and then collected after 6h and 24h. Olive oil pomace increased the content of C18:1 c9 and C18:3 c9c12c15. Considering the microbial communities, the genera Butyrivibrio, Fibrobacter, and Pseudobutyrivibrio were less abundant, while Christensenellaceae_R-7_group, Manheimia, Uruburuella were more abundant in rumen liquor fermented with olive oil pomace. Similarly, the diet containing olive tree leaves increased the content of C18:1 c9 and C18:3 c9c12c15 and decreased the abundance of Pseudobutyrivibrio and Rikenellaceae_RC9_gut_group. Data reported in this study showed that the two by-products deriving from the olive oil production chain are effective in modulating microbial community in a selective manner.

Abstract

In natural ecosystems, many plants engage in symbiotic interactions with fungal endophytes which can have impacts on host plant communities. In South American grasslands, several endophytes have been isolated from different Paspalumspecies. The aim of this work was to evaluate the antagonistic effect of endophytes Trichoderma amazonicumand Acremonium strictumagainst Claviceps paspalithrough in vitro and in vivo assays. In addition, they were evaluated as biocontrol agents against fungal pathogens which produce leaf spots such as Alternaria alternataand Bipolarissp. through in vitro experiments. The in vitro assessment was conducted by dual culture between the antagonists and the three pathogens. For the in vivo assay, plants of Paspalum notatumand Paspalum dilatatumwere planted in the field in a completely randomized block design with six replications. Three treatments were applied: T1 = endophyte inoculum, T2 = fungicide, and T3 = control. Seed yield, seed set, healthy seed set, and weight of healthy seed set per kilogram of seeds were evaluated for three timepoints. Trichoderma amazonicumexhibited a high growth inhibition, while A. strictuma moderate to low effect in vitro. In the in vivo assay, significant differences were observed among treatments in both species. T1 increased the kilogram of healthy seed set per 100 kg of seed by increasing the seed set and weight of healthy seed set in at least one of the evaluated dates in P. notatumand P. dilatatum. Based on these results, T. amazonicumand A. strictumhave antagonistic effects on the pathogens assessed. Fungal endophyte Trichoderma amazonicumcould be used to increase seed yield in Paspalum notatum.Fungal endophyte Acremonium strictumcould be used to increase seed yield in Paspalum dilatatum.Trichoderma amazonicumand Acremonium strictumhave the potential to be used as biocontrol agents for frequent grass diseases.

Abstract Leading digital innovation projects in smart cities requires the right human resources with the right set of competencies. Such requirement is challenging because city managers and built environment professionals are traditionally trained and work in disciplinary and professional silos. This results in a lack of knowledge, abilities, and tools to produce optimal outcomes for communities across multiple sectors. Guided by a socio‐technical and multidisciplinary approach, the DC2‐CF framework is proposed to help urban planners identify, develop and expand the competencies they need to effectively steer responsible digital innovation and ensure public value creation. The DC2‐CF proposes a digital innovation process model to facilitate the delivery of successful urban digital innovation projects based on the lessons learned from working with city digital leaders. In addition, the DC2‐CF provides a delivery structure which identifies specific tasks, competencies, and roles necessary to enable the use off the innovation process model in practice and break down the professional silos currently existing in cities. The framework is envisioned to assist city planners in fostering leadership capacity in local authorities and the private sector in digital city development, contributing to enhancing collaborative working and effective public value creation.

Abstract Homologous recombination (HR) and mismatch repair (MMR) defects are driver mutational imprints and actionable biomarkers in DNA repair-defective tumors. Although usually thought as mutually exclusive pathways, recent preclinical and clinical research provide preliminary evidence of a functional crosslink and crosstalk between HRR and MMR. Shared core proteins are identified as key players in both pathways, broadening the concept of DNA repair mechanism exclusivity in specific tumor types. These observations may result in unexplored forms of synthetic lethality or hypermutable tumor phenotypes, potentially impacting the cancer risk management, and considerably expanding in the future the therapeutic window for DNA repair-defective tumors.

Abstract The assessment of ctDNA has emerged as a minimally invasive avenue for molecular diagnosis and real-time tracking of tumor progression in NSCLC. However, the evaluation of ctDNA by amplicon-based NGS has been not endorsed by all the healthcare systems and remains to be fully integrated into clinical routine practice. To compare tissue single-gene with plasma multiplexed testing, we retrospectively evaluated 120 plasma samples from 12 consecutive patients with advanced non-squamous NSCLC who were part of a prospective study enrolling treatment-naïve patients and in which tissue samples were evaluated using a single-gene testing approach. While the plasma ctDNA detection of EGFR and BRAF mutations had an acceptable level of concordance with the archival tissue (85%), discordance was seen in all the patients in whom ALK alterations were only detected in tissue samples. Among six responders and six non-responders, early ctDNA mutant allelic frequency (MAF) reduction seemed to predict radiologic responses and longer survival, whereas increasing MAF values with the emergence of co-mutations like BRAFV600E, KRASG12V or TP53M237I seemed to be an early indicator of molecular and radiologic progression. This report using an amplicon-based NGS assay on ctDNA underscores the real-life need for plasma and tissue genotyping as complementary tools in the diagnostic and therapeutic decision-making process.

Abstract Hand visibility affects motor control, perception, and attention, as visual information is integrated into an internal model of somatomotor control. Spontaneous brain activity, i.e., at rest, in the absence of an active task, is correlated among somatomotor regions that are jointly activated during motor tasks. Recent studies suggest that spontaneous activity patterns not only replay task activation patterns but also maintain a model of the body's and environment's statistical regularities (priors), which may be used to predict upcoming behavior. Here, we test whether spontaneous activity in the human somatomotor cortex as measured using fMRI is modulated by visual stimuli that display hands vs. non-hand stimuli and by the use/action they represent. A multivariate pattern analysis was performed to examine the similarity between spontaneous activity patterns and task-evoked patterns to the presentation of natural hands, robot hands, gloves, or control stimuli (food). In the left somatomotor cortex, we observed a stronger (multivoxel) spatial correlation between resting state activity and natural hand picture patterns compared to other stimuli. No task-rest similarity was found in the visual cortex. Spontaneous activity patterns in somatomotor brain regions code for the visual representation of human hands and their use.

Fungal infections in marine animals, particularly pinnipeds, have seen a notable increase, often linked to compromised immune systems in captive environments. Trichophyton species, while common in terrestrial mammals, have sporadically caused dermatophytosis in pinnipeds. A South American sea lion (Otaria byronia) presented with Trichophyton benhamiae infection, marking the first such case in this species. Effective treatment combined oral terbinafine with topical ozonized oil, supported by silymarin for liver protection. Accurate fungal identification and sensitivity testing were key to the successful management and recovery of the patient.

Abstract

The Argentinean Society of Hypertension, in agreement with the May Measurement Month (MMM) initiative of the International Society of Hypertension, implemented for the third consecutive year a hypertension screening campaign. A volunteer cross-sectional survey was carried out in public spaces and health centres during the month of May 2019 across 33 cities in Argentina. Hypertension was defined as systolic blood pressure (BP) ≥140 mmHg and/or diastolic BP ≥90 mmHg based on the mean of the second and third BP measurements, or in those on treatment for high BP. A total of 94 523 individuals (53.9 ± 17.8 years old, 55 231women and 39 292 men), were evaluated. The age and sex standardized mean BP was 124.7/77.2 mmHg. Among participants, 34.7% were overweight (25-29.9 m/kg2) and 28.7% had obesity (≥30 m/kg2). Individuals identified as being overweight had BP 3/2 mmHg higher and individuals with obesity 6/4 mmHg higher than those with normal weight. The prevalence of hypertension was 52.5%. Although 81.1% were aware and 77.7% were on antihypertensive treatment, only 46.0% of all individuals with hypertension had their BP controlled. Moreover, 19.8% of those not on any antihypertensive medication were found with raised BP. The low level of control of hypertension generates the critical need for the development of community-based prevention strategies reinforcing strategies to increase the awareness and control of hypertension.

Background: The benefits of a dedicated orthopaedic trauma room (DOTR) for patients with isolated femur fractures have not been reported from a pediatric community hospital. Methods: Twenty-three years of skeletally immature patients treated for an isolated diaphyseal femur fracture with two flexible intramedullary nails, Pre-DOTR: 2000–2015 and Post-DOTR: 2016–2022, were reviewed for comparison of patient demographics and injury patterns, timing and durations of surgery, fellow presence, duration and cost of hospitalization, and complication rates. Results: One hundred fifty-three patients were identified. One hundred twenty-six patients underwent surgery pre-DOTR and 27 post-DOTR. Demographics were similar between groups, except the post-DOTR patients were younger (mean age 7.4 years vs 9.0 years, P ​= ​0.002). There was no significant difference in the percentage of patients undergoing surgery within 18 ​h of admission pre- and post-DOTR (78% vs 93%, respectively, P ​= ​0.53). Iron is essential to plant development. However, its excess can provoke an increase in reactive oxygen species and oxidative stress in plants. The objective of this work was to verify the effects of high concentrations of iron on the oxidative stress of seeds and young plants of Cecropia hololeuca and Carica papaya. The species were submitted to concentrations of 0.045, 4 and 8mM of iron in the form of ferrous sulfate and FeEDTA. The experiments of germination and initial growth took place in a growth chamber, with temperature of 25ºC and 12h photoperiod. We performed the lipid peroxidation test by extraction and quantification of malonaldehyde and hydrogen peroxide. The application of iron did not cause a significant elevation in the contents of malonaldehyde and hydrogen peroxide in the germination of C. hololeuca and C. papaya. In the young plants, the hydrogen peroxide did not change in any of the treatments. However, it was possible to observe an expressive increase in malonaldehyde concentration in both species when exposed to FeEDTA 4 to 8mM. The results indicate a sensibility of C. hololeuca and C. papaya to high iron levels, amplifying the oxidative stress process that can harm their growth and initial development.

Este artículo explora la aplicación de técnicas algebraicas en la resolución de ecuaciones diferenciales de segundo orden con coeficientes constantes en el caso homogéneo f(x)=0. Se presenta un enfoque detallado sobre la factorización y el uso de la fórmula general para resolver la ecuación característica asociada. Estas técnicas permiten obtener soluciones generales que describen el comportamiento de sistemas dinámicos sin la influencia de fuerzas externas, como circuitos eléctricos, sistemas mecánicos y osciladores armónicos. La factorización es efectiva cuando la ecuación característica puede descomponerse en factores lineales, lo que ocurre en sistemas con raíces racionales. Por otro lado, la fórmula general es un método universal que permite resolver ecuaciones cuadráticas independientemente de la naturaleza de sus raíces, ya sean reales o complejas. El estudio también resalta las implicaciones físicas de las soluciones obtenidas, como el decaimiento exponencial, el crecimiento o las oscilaciones periódicas. Asimismo, se analiza la relación entre las raíces de la ecuación característica y la estabilidad del sistema, un aspecto crucial en la ingeniería y el diseño de sistemas de control. Se concluye que las técnicas algebraicas para resolver ecuaciones diferenciales homogéneas de segundo orden no solo son esenciales desde un punto de vista teórico, sino que también tienen aplicaciones prácticas significativas en el análisis, diseño y optimización de sistemas dinámicos.

El objetivo es proponer criterios de diseño que mejoren la calidad de vida de la población mayor al facilitar las actividades básicas e instrumentales de la vida diaria al interior de sus viviendas. Este artículo presenta una revisión bibliográfica sobre el desarrollo de competencias comunicacionales en la formación de docentes de lengua y literatura en la Universidad Estatal de Milagro (UNEMI). Las competencias comunicacionales, que abarcan dimensiones verbales, no verbales y digitales, son esenciales para mejorar el proceso de enseñanza-aprendizaje, ya que permiten a los docentes interactuar de manera efectiva con sus estudiantes y promover la participación activa, el pensamiento crítico y la reflexión. La revisión de la literatura subraya la importancia de integrar tecnologías de la información y la comunicación (TIC) en la formación docente, así como el desarrollo de competencias no verbales, que contribuyen a generar un ambiente de confianza y colaboración en el aula. Además, se destaca la necesidad de fomentar una enseñanza reflexiva, en la cual los docentes ajusten sus prácticas pedagógicas en función de las necesidades de sus estudiantes. También se encontró que el uso de metodologías activas, como la gamificación y el aprendizaje basado en proyectos, es eficaz para desarrollar competencias comunicacionales en los estudiantes de lengua y literatura. La implementación de estas estrategias en los programas de formación docente en la UNEMI puede mejorar significativamente la calidad de la enseñanza y preparar a los futuros docentes para enfrentar los retos de la educación actual.

Abstract

Background: Prostate cancer (PCa) is the second most common cancer in men, and radiotherapy (RT) is one of the main treatment options. Although effective, RT can cause toxic side effects. The accurate prediction of dosimetric parameters, enhanced by advanced technologies and AI-based predictive models, is crucial to optimize treatments and reduce toxicity risks. This study aims to explore current methodologies for predictive dosimetric parameters associated with RT toxicity in PCa patients, analyzing both traditional techniques and recent innovations. Methods: A systematic review was conducted using the PubMed, Scopus, and Medline databases to identify dosimetric predictive parameters for RT in prostate cancer. Studies published from 1987 to April 2024 were included, focusing on predictive models, dosimetric data, and AI techniques. Data extraction covered study details, methodology, predictive models, and results, with an emphasis on identifying trends and gaps in the research. Results: After removing duplicate manuscripts, 354 articles were identified from three databases, with 49 shortlisted for in-depth analysis. Of these, 27 met the inclusion criteria. Most studies utilized logistic regression models to analyze correlations between dosimetric parameters and toxicity, with the accuracy assessed by the area under the curve (AUC). The dosimetric parameter studies included Vdose, Dmax, and Dmean for the rectum, anal canal, bowel, and bladder. The evaluated toxicities were genitourinary, hematological, and gastrointestinal. Conclusions: Understanding dosimetric parameters, such as DVH, Dmax, and Dmean, is crucial for optimizing RT and predicting toxicity. Enhanced predictive accuracy improves treatment effectiveness and reduces side effects, ultimately improving patients’ quality of life. Emerging artificial intelligence and machine learning technologies offer the potential to further refine RT in PCa by analyzing complex data, and enabling more personalized treatment approaches.

Abstract

In human teeth, the cementoenamel junction (CEJ) can have different morphologies, known as Choquet cases or types: in type 1, the cementum overlaps the enamel; in type 2, the enamel overlaps the cementum; in type 3, the relationship between the cementum and enamel is edge-to-edge; and in type 4, there is a gap between the enamel and the cementum, with exposed dentin. Since little is known on the morphology of the CEJ in animals, the aim of this study was to characterize the structure of the CEJ of dog teeth by SEM. We studied 82 permanent teeth extracted from 4 dogs of similar size, aged from 7 months to 2 years. Teeth were grouped according to their size, sectioned and prepared for SEM. The teeth evaluated showed the 4 CEJ types. Type 3 was the most prevalent, being present, either alone or in combination, in 90.79% of the samples, followed by type 4 (present in 31.58% of the samples). Of the samples, 65.13% showed only one type, while 34.87% presented a combination of them. Types 3 and 4 were the only ones present alone (in 55.92% and 9.21% of the samples respectively), whereas the combinations between types 3 and 4 (20.40%) and types 1 and 3 (9.87%) were the most frequent. In all combinations, type 3 was predominant. The CEJ of the dogs evaluated showed a high degree of variability in the different types of teeth, within a same tooth, within the dentition of a single individual, and among individuals.

Incluye contenido parcial de los autores Abstract.Mammalian carnivores are considered a key group in maintaining ecologicalhealth and can indicate potential ecological integrity in landscapes where they occur. Carni-vores also hold high conservation value and their habitat requirements can guide managementand conservation plans. The order Carnivora has 84 species from 8 families in the Neotropicalregion: Canidae; Felidae; Mephitidae; Mustelidae; Otariidae; Phocidae; Procyonidae; andUrsidae. Herein, we include published and unpublished data on native terrestrial Neotropicalcarnivores (Canidae; Felidae; Mephitidae; Mustelidae; Procyonidae; and Ursidae). NEOTRO-PICAL CARNIVORES is a publicly available data set that includes 99,605 data entries from35,511 unique georeferenced coordinates. Detection/non-detection and quantitative data wereobtained from 1818 to 2018 by researchers, governmental agencies, non-governmental organi-zations, and private consultants. Data were collected using several methods including cameratrapping, museum collections, roadkill, line transect, and opportunistic records. Literature(peer-reviewed and grey literature) from Portuguese, Spanish and English were incorporated inthis compilation. Most of the data set consists of detection data entries (n=79,343; 79.7%) butalso includes non-detection data (n=20,262; 20.3%). Of those, 43.3% also include count data(n=43,151). The information available in NEOTROPICAL CARNIVORES will contribute tomacroecological, ecological, and conservation questions in multiple spatio-temporal perspec-tives. As carnivores play key roles in trophic interactions, a better understanding of their distri-bution and habitat requirements are essential to establish conservation management plans andsafeguard the future ecological health of Neotropical ecosystems. Our data paper, combinedwith other large-scale data sets, has great potential to clarify species distribution and relatedecological processes within the Neotropics. There are no copyright restrictions and no restric-tion for using data from this data paper, as long as the data paper is cited as the source of theinformation used. We also request that users inform us of how they intend to use the data.

Abstract

Although it may seem an impossible task to offer a panorama of contemporary Brazilian poetry, we will discuss a selection of poets who possibly epitomize the most significant current trends. We start with a brief presentation of four well-regarded authors whose works represent major poetic currents: Augusto de Campos, Francisco Alvim, Armando Freitas Filho and Adélia Prado. We then comment on excerpts from a few important younger poets, whose works embrace several different tones and styles, from indigenous symbolic mythology to crude or ironic testimonials of life in the big city, from melodious lyrical intimacy to a more social perspective. For the second part of this article, we have chosen Paulo Henriques Britto, Josely Vianna Baptista, Ricardo Aleixo, Carlito Azevedo, Eucanaã Ferraz and Ana Martins Marques. We hope this preliminary selection may awaken the interest of future readers to the enormous variety and richness of Brazilian poetry.

Abstract

IntroductionAlthough hereditary male neoplasms are quite rare, individuals harbouring germline BRCA1/2 pathogenic variants (PVs) may have a risk of developing tumours associated with Hereditary Breast and Ovarian Cancer (HBOC) syndrome, including male breast (MBC), prostate (PCa) and pancreatic (PC) cancers, and melanoma. Women and men showed a comparable genetic architecture of cancer susceptibility, but there are some gender-specific features. Since little is known about cancer genetic susceptibility in male population, our study was aimed at investigating the frequency of BRCA1/2 PVs in men with HBOC syndrome-associated tumors, in order to understand whether differences in gender may reflect in the prevalence and spectrum of germline alterations.Patients and methodsWe retrospectively collected and analysed clinical information of 352 HBOC-associated male cancer patients genetically tested for germline BRCA1/2 PVs by Next-Generation Sequencing analysis, enrolled, from February 2018 to January 2024, at the “Regional Center for the prevention, diagnosis and treatment of rare and heredo-familial tumors of adults” of the University-Hospital Policlinico “P. Giaccone” of Palermo (Italy).ResultsOur investigation revealed that 7.4% of patients was carrier of a germline BRCA PV, with an almost total prevalence of BRCA2 alterations. In particular, 65.4% of BRCA-positive patients developed MBC, 19.2% had PC, 11.6% developed PCa, and only 3.8% had melanoma. Specifically, MBC individuals showed a BRCA-associated genetic predisposition in 17% of cases, whereas patients with PCa or PC exhibited a lower frequency of BRCA2 PVs, taking into account the current national criteria for access to germline genetic testing.DiscussionOur study showed a high heterogeneity in prevalence of germline BRCA2 PVs among men which could reflect a potential gender-specific genetic heterogeneity. Therefore, BRCA-associated male tumours could be due to BRCA2 PVs different from those usually detected in women. In the event that it is demonstrated, in future, that male cancers are genetically distinct entities from those female this could improve personalized risk evaluation and guide therapeutic choices for patients of both sexes, in order to obtain a gender equality in cancer care.

Abstract

Summary: Humans are the most versatile tool users among animals. Accordingly, our manual skills evolved alongside the shape of the hand. In the future, further evolution may take place: humans may merge with their tools, and technology may integrate into our biology in a way that blurs the line between the two. So, the question is whether humans can embody a bionic tool (i.e., experience it as part of their body) and thus if this would affect behavior. We investigated in virtual reality how the substitution of the hand with a virtual grafting of an end-effector, either non-naturalistic (a bionic tool) or naturalistic (a hand), impacts embodiment and behavior. Across four experiments, we show that the virtual grafting of a bionic tool elicits a sense of embodiment similar to or even stronger than its natural counterpart. In conclusion, the natural usage of bionic tools can rewire the evolution of human behavior.

Abstract

Background and purposeThis study aimed to investigate the factors that influence physiotherapists’ decision in choosing restorative or compensatory rehabilitation during gait training in people with neurological disorders (PwNDs) and the different treatments used in the approaches.MethodsThis cross-sectional analysis used the baseline data from an observational cohort study. We analyzed data from 83 PwNDs (65 people after stroke, 5 with multiple sclerosis, and 13 with Parkinson’s disease) who underwent at least 10 sessions of physiotherapy (PT) focusing on gait function. Performance was quantified using the modified Dynamic Gait Index (MDGI), three impairment domains of Fugl–Meyer Assessment for lower extremity (mFM-LL), Activities-specific Balance Confidence (ABC), modified Barthel Index (mBI), Mini-Mental State Examination (MMSE), and Motivational Index (MI). Forty-three physiotherapists completed a treatment report form categorizing the rehabilitation approach and specifying treatments used (e.g., resistance training and proprioceptive exercises).ResultsFifty-six subjects underwent restorative rehabilitation approach. The univariate predictors of restorative approach were being in the subacute phase with a disease onset of less than 180 days, (odds ratio [95%CI]; 3.27[1.19–9.24]), mFM-LL (1.25[1.11–1.44]), MMSE (0.85[0.67–1.00]), and number of sessions (1.03[1–1.01]). The backward stepwise analysis revealed an association between restorative and subacute phase (36.32[4.11–545.50]), mFM-LL (3.11[1.55–9.73]), mBI (1.79[1.08–3.77]), MMSE (0.46[0.25–0.71]), and the interaction between mFM-LL and mBI (0.99[0.98–1.00]). No statistically significant association between treatments used and approach was found (p = 0.46).Discussion and conclusionThe restorative approach was more commonly used to improve gait. The main variables associated with this approach were: being in the subacute phase of the disease, a low level of impairment, and a high level of functional independence at baseline. However, few differences were found between the treatments used for the restorative or compensatory approaches, as similar PT treatments were used for both.
