29. Reproductive biology of female franciscana dolphins (Pontoporia blainvillei) from Argentina.

Abstract

The ovaries of 31 franciscana dolphins (19 immature and 12 mature) by-caught in coastal waters of Argentina were examined to describe the reproductive biology. No ovarian polarity was found, both ovaries were functional and showed similar amounts of corpora. A positive trend was observed between the number of corpora and age (3-8 years old), indicating that ovulation scars are detectable for at least 4 years, and a ovulation rate of 0.39 was found for the sampled population. Age, length and weight at sexual maturity were estimated at 3.92 ± 0.09 years, 133.47 ± 11.11 cm and 32.68 ± 2.72 kg, respectively. The annual pregnancy rate was 0.36 ± 0.02 (95% CI = 0.10-0.65). The proportion of lactating and resting females found were 0.25 and 0.33 respectively. These outcomes constitute the first reproductive and life history information on franciscana dolphins from the southernmost population and are important in relation with adequate conservation management plans for this small cetacean. 36. IGF1 regulation of BOULEand CDC25Atranscripts viaa testosterone-independent pathway in spermatogenesis of adult mice

Abstract

The Deleted in AZoospermia (DAZ) gene family plays an essential role in spermatogenesis and fertility in mammals. This gene family contains two autosomal genes, BOULEand DAZL(DAZ-Like), and the DAZgene cluster in the Y chromosome. CDC25A (a cell cycle regulator) has been proposed as a putative substrate for the RNA-binding proteins of DAZ family. However, mechanisms regulating DAZgene expression have been poorly investigated. We analyzed immunohistochemical localization of DAZL, BOULE and CDC25A, as well as the involvement of testosterone (T) and insulin-like growth factor 1 (IGF1) in the modulation of mRNA expression for DAZL, BOULEand CDC25Ain the adult mouse testes. It was found that DAZL was mostly immunolocalized in spermatogonia, while BOULE and CDC25A were detected in spermatocytes and round spermatids. Three-color immunofluorescence showed that DAZL-positive cells also expressed proliferating cell nuclear antigen (PCNA). In vitroincubation of the testes showed that neither T nor IGF1 affected DAZLmRNA expression. However, either T or IGF1 increased BOULEmRNA expression. Antiandrogen flutamide abolished the T-induced increase in BOULEmRNA, but had no effect on the IGF1 induced increase in the mouse testes. Extracellular-signal-regulated kinase 1/2 (ERK1/2) inhibitor, U0126, prevented IGF1-induction of BOULEmRNA. It was found that IGF1 increased CDC25AmRNA expression and that U0126 – but not flutamide – abolished the IGF1-induced CDC25AmRNA expression. These results showed that IGF1 regulated the expression of BOULEand CDC25AmRNAs viaERK1/2 signaling and in T-independent pathway during spermatogenesis in the adult mouse testes.

39. The kinetics of oocyte activation and dynamics of polar body formation in <e1>Calomys musculinus</e1> and <e1>Calomys laucha</e1> (Rodentia–Sigmodontinae)

Abstract

The objective of this study was to determine whether Calomys laucha and Calomys musculinus super-ovulated oocytes undergo parthenogenetic activation following activation stimuli. Cumulus-intact or denuded oocytes were treated with medium containing ethanol (7%), medium containing strontium chloride, or medium alone. They were then incubated for 6–8 h to allow for activation. A group of oocytes was fixed immediately after maturation to serve as a control. The nuclear status of the oocytes was examined after staining with Hoechst 33342, to determine the timing of pronuclear progression from metaphase II to anaphase II or telophase II or to the pronuclear stage. The proportion of oocytes that underwent activation was higher for oocytes treated with ethanol or strontium chloride than in those incubated in medium alone, for the two species studied (p &lt; 0.001). There was little evidence of spontaneous activation occurring in oocytes during the treatments. Most of the activated oocytes contained a single haploid pronucleus, but it was possible to find immediate cleavage and two pronuclei. The different classes of activated oocytes were cultured for 5 days. The type of activating treatment had a marked effect on the ability of the resulting C. musculinus and C. laucha parthenogenetic embryos to develop to the preimplantation stages. Incubation with ethanol produced only 8-cell embryos while the embryos induced with strontium chloride reached the blastocyst stage. This is the first report of parthenogenesis in C. musculinus and C. laucha. The ability of strontium ions to induce matured secondary oocytes to initiate parthenogenesis and obtain further development of Calomys provides opportunities to use Calomys oocytes in vitro and, therefore, to study the genetics, cell biology and virology of development.

Abstract

Para dilucidar distintos aspectos del proceso de fecundación y desarrollo temprano, en lapresente tesis se analizaron oocitos humanos ‘no fecundados' y ‘fecundados anormalmente'luego de Fecundación In Vitro (FIV) e Inyección Intracitoplásmica de Espermatozoides (ICSI),haciendo hincapié en la arquitectura del citoesqueleto, estado de la cromatina, organizacióndel áster y presencia de activaciones abortivas. Se estudiaron un total de 815 oocitos clasificados como ‘no fecundados’ y 153 oocitosclasificados como ‘fecundados anormalmente' luego FIV e ICSI. El material se procesó parainmunofluorescencia mediante la utilización de anticuerpos monoclonales para la detección de B tubulinas y a tubulinas acetiladas. El material genético se estudió con Hoechst 33258 y seanalizó por microscopía óptica (UV). El análisis citogenético se realizó en 189 oocitosactivados luego de FIV e ICSI según la técnica de Tarkowski (1966). Durante las fallas de fecundación, la principal causa luego de FIV, fue la ausencia depenetración espermática (59,2%), el 10,5% mostró una falla de activación oocitaria y el 15,2% presentó alteraciones en la nucleación o migración de pronúcleos. Luego de ICSI, laprincipal causa de falla de fecundación fue la alteración de la activación oocitaria (36,5%). Un 18,5% de los oocitos detuvieron su desarrollo en la primera placa metafásica. El estudio cromosómico, permitió identificar la presencia de activaciones abortivas; metafases III (MIII), núcleos reticulares (NR) y núcleos telofásicos (NT). En la fecundación anormal, el patrón más frecuentemente detectado luego de FIV fue lapresencia de 3 ó 4 pronúcleos (PNs), (77,4%). Por el contrario, luego de ICSI resultó ser lapresencia de 1PN (65,2%). Se observaron leves diferencias en el desarrollo de subnúcleosentre ambos grupos. In order to dilucidate different aspects during fertilzation and early development, we analyzed “non-fertilized" and “abnormally fertilized" human oocytes after In Vitro Fertilization (IVF) and Intracytoplasmic Sperm Injection (ICSI). We focused on the cytoskeletal architecture,chromatin configuration, aster organization and the presence of abortive activations. We analyzed 815 “non-fertilized"and 153 “abnormally fertilized" oocytes after IVF and ICSI. The material was processed for immunofluorescence to detect B tubulins and a acetylatedtubulins. DNA was studied using Hoescht. Chromosomal spreads were performed by Tarkowski's air-drying method in 189 IVFand ICSI oocytes. During fertilization failure, the main reason after IVF was no sperm penetration (59.2%). Theremaining oocytes showed different abnormal patterns: oocyte activation failure (10.5%) anddefects in pronuclei apposition (15.2%). On the other hand, fertilization failure after ICSI wasmainly associated to incomplete oocyte activation (36.5%). An 18.5% of the studied oocytesstopped their development at the metaphase of the first mitosis of the pre-embryo. The Chromosomal spreads allowed identifying abortive activations, including metaphase III andothers. Immunofluorescence analysis of abnormal fertilization showed that the most frequentabnormal fertilization pattern found after IVF was the presence of 3 or 4 PNs (77.4%). Onthe other hand, the presence of 1 PN (65.2%) was the main pattern found after ICSI. Nodifferences between both groups were seen in terms of subnuclei development. Fil: Rawe, Vanesa Y.. Abstract

En el presente trabajo se analizaron las características biológicas de las gametasfemenina y masculina de C. musculinus y C. laucha y sus interaccionesinterespecíficas dentro y fuera del género. Brevemente, los parámetros estudiados (superovulación, activación partenogenética, dinámica de capacitación in vitro delespermatozoide, fecundaciones homólogas interespecíficas) han mostrado para estasespecies que: 1- tienen características propias inherentes a su propio género y especie (por ejemplo,ritmo endógeno de LH, activación partenogenética, capacitación in vitro delespermatozoide) por lo cual no pueden ser homologables a los modelosconvencionales de roedores de laboratorio sino que constituyen en sí mismos unmodelo nuevo para estudios de biología del desarrollo. 2- las células del cúmulo, la zona pelúcida, y/o la membrana plasmática del oocito noconstituyen una barrera efectiva a la fecundación heteróloga intragenérica. 3- la diferencia en la morfología espermática entre C. musculinus (con ganchoacrosomal) y C. laucha (ausencia total de gancho acrosómico) mostró una relaciónespecie-específica en el proceso de activación 4- las gametas de Calomys mostraron mayor afinidad en la interacción con lasgametas de hamster (Cricetidae) que con las gametas de ratón (Muridae). Estasobservaciones apoyan la hipótesis de Reig (1982) que sostiene que estas especiespertenecen a la familia Cricetidae. The present study focused on the characterization of the biological properties offemale and male gametes in C. musculinus and C. laucha and the interspecificinteractions between them and with other extra-generic rodent species. Briefly, themain parameters analyzed (superovulation, parthenogenetic activation, in vitrocapacitation of spermatozoa, interespecies fertilization) showed that: 1- C. musculinus and C. laucha had specific and inherent features related to theirgenus and species (i.e. endogenous LH rhythm, a high parthenogenetic activation,in vitro capacitation of spermatozoa). They can not be directly related to thetypical laboratory rodents. They constitute a new model for developmental biologyresearch. 2- cumulus cells, zona pellucida and plasma membrane of the oocyte are not effectivebarriers to interspecific fertilization. 3- the differences on sperm head morphology between C. musculinus (hooked head)and C. laucha (hookless head) showed a specific relation to the activation process. 4- Calomys' gametes interacted more easily with golden hamster (Cricetidae) than Mus musculus (Muridae) gametes. 43. Immunolocalization of the TGFB1 system in submandibular gland fibrosis after experimental periodontitis in rats.

Abstract

Saliva is the first barrier to entry of bacteria and viruses into the body. The submandibular glands (SMG) contribute to the maintenance of oral health and regulation of immune/ inflam matory responses. Previous studies suggest that transforming growth factor beta 1 (TGFB1) may contribute to salivary gland fibrosis but the expression of the TGFB1 system in the SMG has not been elucidated. Thus, the aim of this study was to analyze in rat SMG the immunolocalization of TGFB1 and its specific receptors ALK5 (profibrotic) and ALK1 (proproliferative) and the coreceptor endoglin (EDG) in a bilateral experimental periodontitis (EP) model (cotton thread ligature around the neck of the first lower molars) for 1 and 6 weeks. Fixed SMG were embedded in paraffin and serially cut for routine hematoxylin-eosin staining for histological analysis or immunohistochemical techniques by diaminobenzidine detection. SMG histology from animals with EP showed timedependent structural changes involving marked reduction in the height of the contoured ducts, cell destruction, loss of secretory granules, periductal congestion and excess connective tissue surrounding these ducts indicative of a fibrotic process, compared to control SMG. TGFB1, ALK5 and ALK1 receptors and the coreceptor EDG were mainly immunolocalized in ductal cells and in the fibrotic areas in EP groups. The expression of the profibrotic ALK5 receptor was increased in areas of fibrosis in SMG of animals with EP. In SMG of rats with EP, the localization of the TGFB1 specific receptors in the ducts and cells from fibrotic areas, due to the expression of TGFB1 in the surrounding areas, might indicate paracrine and autocrine actions exerted by TGFB1 via its specific receptors. The results of this study suggest that TGFB1 promotes fibrosis, inducing cell proliferation via ALK1 and EDG receptors and stimulates fibrosis relatedprocesses via ALK5 receptor, which could lead to abnormal secretor activity of the SMG during periodontal disease., (Sociedad Argentina de Investigación Odontológica.)

Abstract

Lagostomus maximus is a notable mammalian model for reproductive studies. Females have an extremely high ovulation rate, which is due to down-regulation of the follicular apoptosis pathway, which ensures a large pool of developing follicles. This large pool is supported by the convoluted anatomy of the mature ovary, whose germinal tissue is found in irregularly curved ridges throughout the cortex. Medullary tissue is restricted to a minimum. Lyso Tracker Red reconstruction under confocal laser scanning microscopy was used to recognize and measure all follicular stages from primordial to antral. Unlike most mammals in which early primordial follicles are just found in fetal life, the adult ovary shows regions packed with early primordial follicles. Follicle size ranged from 24 to 316 microm. We discuss the relationships of L. maximus follicles size with regard to other species of mammals and propose that the physiology of the adult viscacha ovary obeys to a neoteny process in the evolution of this species.
