Fluid inclusions are a window into deep geological fluids, providing unique access to their nature and composition. The isotopic composition of CO2 and CH4 hosted in fluid inclusions is a powerful proxy to assess the origin and transformation of deep geological fluids, giving insights into carbon sources, fluxes, and degassing in a wide variety of geodynamic settings. Over the last 5 decades, techniques have been developed to extract fluid inclusions from their host minerals and measure their bulk composition. These techniques are often challenged by analytical artifacts including high blank levels of CO2 and CH4, fluid re-speciation, gas adsorption, and diffusion. Since these processes may alter the pristine composition of gases liberated from fluid inclusions, rigorous protocols are needed in order to evaluate the isotopic integrity of the extracted volatile species. In this study, we introduce new protocols for bulk off-line fluid inclusion extraction for the analysis of δ13C-CH4 and δ13C-CO2 using a Cavity Ring-Down Spectroscopy (CRDS) analyser (Picarro G2201-i). Two mechanical fluid extraction techniques are compared: ball milling in ZrO2 jars and sample crushing in a stainless steel sealed tube under a hydraulic press. Blanks and isotopically labelled tests with the ball milling technique suggest that rotation speed, grinding stock filling degree and filling type alter the CH4 and CO2 concentrations and isotopic compositions measured by the CRDS analyser. In contrast, the crushing technique does not generate measurable quantities of blank CH4 and CO2. The protocols presented in this study allow to extract, detect, and analyse δ13C of CH4 and CO2 for concentrations above 10 and 1,000 ppm respectively. Interlaboratory experiments allowed to replicate previously measured δ13C-CH4 values in natural fluid inclusions within 1‰ with both extraction techniques. This study highlights the potential of combining simple bulk off-line fluid inclusion extraction techniques with a CRDS analyser for δ13C analysis of CO2 and CH4 without gas separation being required.

Metropolitan areas represent nowadays the main drivers of European development. This puts traditional spatial governance models into crisis, with existing territorial units that are challenged by phenomena hardly manageable within their fixed administrative boundaries. A growing number of governance experimentations have emerged in Europe aiming to address the metropolitan dimension. In the Italian context, this process led to the introduction of 14 Città Metropolitane, that substituted the respective provinces in particularly complex territorial contexts. Due to its nature, however, the reform raised a number of challenges in relation to the actual role that small and medium towns should play within the new governance arrangement, and how their cooperation with the large urban core they gravitate around can bring reciprocal benefits and added value. This paper sheds some light on the matter, discussing the state of the art of metropolitan governance and planning in the country, to then use the cases of Torino and Bari to highlight how different levels of territorial fragmentation contribute to raise different challenges for metropolitan governance and planning. In doing so, the authors reflect on the advantages and disadvantages that small and medium towns encounter when participating to metropolitan governance.

Abstract During the subduction of an oceanic plate, fluids are released from metabasaltic crust, metasediment, and serpentinite under high‐pressure/low‐temperature conditions. Although some fluids may eventually leave the slab, some participate in metamorphic reactions within the slab during subduction and exhumation. To identify fluid sources and other controls influencing mineral composition, we report the in situ‐measured δ18O of lawsonite and garnet in blueschist‐ to eclogite‐facies rocks from 10 subduction zones that represent various field settings, including mélanges, structurally coherent terranes, and an eclogite xenolith derived from a subducted plate. Lawsonite records distinct δ18O depending on the host rock type and other rock types that were fluid sources during lawsonite growth. In general, lawsonite in metabasalt (7.6 ± 0.2–14.8 ± 1.1‰) is isotopically lighter than in metasediment (20.6 ± 1.4–24.1 ± 1.3‰) but heavier than in metagabbro (4.0 ± 0.4–7.9 ± 0.3‰). The extent of δ18O fractionation was evaluated for lawsonite–fluid and lawsonite–garnet pairs as a function of temperature (T). Results demonstrate that variations of >1.7‰ in lawsonite and >0.9‰ in garnet are not related to changing T. More likely, the relative contributions of fluids derived from isotopically heavier lithologies (e.g., sediments) versus lighter lithologies (e.g., ultramafic rocks) are the major control. Monte Carlo simulations were performed to investigate the sources of metasomatic fluids and the water/rock ratio that formed lawsonite‐bearing metasomatite. Results indicate that δ18OLws and δ18OGrt record interactions with fluids sourced from diverse lithologies (sediment, serpentinite), further supporting that δ18OLws is a useful indicator of subduction fluid‐rock interactions.

The in vitro evaluation of 3D scaffolds for bone tissue engineering in mono-cultures is a common practice; however, it does not represent the native complex nature of bone tissue. Co-cultures of osteoblasts and osteoclasts, without the addition of stimulating agents for monitoring cellular cross-talk, remains a challenge. In this study, a growth factor-free co-culture of human bone marrow-derived mesenchymal stem cells (hBM-MSCs) and human peripheral blood mononuclear cells (hPBMCs) has been established and used for the evaluation of 3D-printed scaffolds for bone tissue engineering. The scaffolds were produced from PLLA/PCL/PHBV polymeric blends, with two composite materials produced through the addition of 2.5% w/v nanohydroxyapatite (nHA) or strontium-substituted nanohydroxyapatite (Sr-nHA). Cell morphology data showed that hPBMCs remained undifferentiated in co-culture, while no obvious differences were observed in the mono- and co-cultures of hBM-MSCs. A significantly increased alkaline phosphatase (ALP) activity and osteogenic gene expression was observed in co-culture on Sr-nHA-containing scaffolds. Tartrate-resistant acid phosphatase (TRAP) activity and osteoclastogenic gene expression displayed significantly suppressed levels in co-culture on Sr-nHA-containing scaffolds. Interestingly, mono-cultures of hPBMCs on Sr-nHA-containing scaffolds indicated a delay in osteoclasts formation, as evidenced from TRAP activity and gene expression, demonstrating that strontium acts as an osteoclastogenesis inhibitor. This co-culture study presents an effective 3D model to evaluate the regenerative capacity of scaffolds for bone tissue engineering, thus minimizing time-consuming and costly in vivo experiments.

Remote rural areas are often rich in natural and landscape assets, which are in turn used as the main focus of tourism development strategies aiming at reverting their decline. However, mono-functional strategies hardly manage to achieve this goal, as in order to restore those structural conditions that are essential to liveability and local development it is necessary to engage in a more comprehensive approach. Acknowledging this challenge, the paper reflects on the possibility to include tourism within multi-level development strategies aimed at tackling marginalisation, drawing on the case of the Italian National Strategy for Inner Areas. More in detail, the authors analyse how the latter enables the integration of tourism-related actions into more comprehensive, place-based development strategies that act upon the peculiarities of the territories they focus on through a mix of top-down and bottom-up logics.

The paper highlights the realization of 3D-printed parts with complex geometries, such as chess-like pieces, using polyamide 12 (PA12) as polymeric powder via selective laser sintering (SLS). The research activity focuses on the study of the powder printability, the optimization of the printing parameters, and the tomographic evaluation of the printed objects. Morphological analyses were carried out to study the PA12 powder microstructure considering that SLS required specific particle size distribution and shape, able to guarantee a good flowability necessary to take part in a sintering process. DSC and TG analyses were performed to determine the sintering window and the crystallinity degree, and to evaluate the thermal stability of the PA12 powder due to the importance of the powder processability for the SLS process. The novelty lies in the realization of chess-like pieces very challenging to print via SLS due to their different and highly detailed structures, and the in-depth analysis of the dimensional accuracy evaluated by micro-tomography. The 3D-printed samples obtained show high printing quality and dimensional stability. The μ-CT analysis also confirms the key role of the object shape and section changes on the final porosity of the chess-like pieces. [ABSTRACT FROM AUTHOR]

The carbon isotopic signature of CO2 released from marine sediments subducted beneath volcanic arcs does not reflect their organic/inorganic fraction, but instead the fluid-rock ratios and the redox conditions in force at the top of the slab.

Using diamond anvil cell and high temperature experiments, this work proves that the interaction between deep hydrogen rich fluids and reduced carbon minerals may be an efficient mechanism for producing abiotic hydrocarbons at the upper mantle’s pressures and temperatures.

Nanohydroxyapatite (nanoHA) is the major mineral component of bone. It is highly biocompatible, osteoconductive, and forms strong bonds with native bone, making it an excellent material for bone regeneration. However, enhanced mechanical properties and biological activity for nanoHA can be achieved through enrichment with strontium ions. Here, nanoHA and nanoHA with a substitution degree of 50 and 100% of calcium with strontium ions (Sr-nanoHA_50 and Sr-nanoHA_100, respectively) were produced via wet chemical precipitation using calcium, strontium, and phosphorous salts as starting materials. The materials were evaluated for their cytotoxicity and osteogenic potential in direct contact with MC3T3-E1 pre-osteoblastic cells. All three nanoHA-based materials were cytocompatible, featured needle-shaped nanocrystals, and had enhanced osteogenic activity in vitro. The Sr-nanoHA_100 indicated a significant increase in the alkaline phosphatase activity at day 14 compared to the control. All three compositions revealed significantly higher calcium and collagen production up to 21 days in culture compared to the control. Gene expression analysis exhibited, for all three nanoHA compositions, a significant upregulation of osteonectin and osteocalcin on day 14 and of osteopontin on day 7 compared to the control. The highest osteocalcin levels were found for both Sr-substituted compounds on day 14. These results demonstrate the great osteoinductive potential of the produced compounds, which can be exploited to treat bone disease.

Geological sources of H2 and abiotic CH4 have had a critical role in the evolution of life and sustainability of the deep subsurface biosphere, yet the origins of these sources remain largely unconstrained. Here the authors show that deep serpentinization (40–80 km) during subduction generates significant amounts of H2 and abiotic CH4, potentially providing energy to the overlying subsurface biosphere.

Backcasting is a scenario-building technique that can help decision-makers face uncertain and complex dynamics. For this reason, backcasting is often mentioned as suitable to deal with the transition to autonomous driving. However, at present the applications of backcasting to this transition in real-world cases are quite few. The article presents a backcasting carried out in the city of Turin (Italy), aimed at defining a policy pathway to steer the transition to autonomous driving towards objectives of sustainability and liveability of the city and its neighbourhoods. It reflects on this exercise and highlights some critical issues for backcasting that emerged from its application to autonomous vehicles (AVs). The transition to AVs displays some issues that proved to challenge the effectiveness and the potentialities of implementation of backcasting. These are mainly related to: factors and levels of uncertainty, contextualization of the vision, involvement of relevant stakeholders, definition of the policy pathway. Nevertheless, the exercise showed that some solutions can be adopted to deal with these challenges, in terms of definition of background socioeconomic scenarios, combination of a range of participatory techniques, integration of collaborative and think-tank methodologies, reference to mid-term planning tools. These solutions can support further backcasting exercises for AVs.

The COVID-19 pandemic raises questions on the ever-growing urban concentration that characterizes our society and various experts are pointing out the need to rethink our cities. At the same time, a number of voices advocate for a rediscovery of the rural. However naive they may sound, these claims have the merit to call for a thorough reflection on the challenges and potentials that alternative modes of urbanisation could bring along. In order to provide a contribution in this direction, the article introduces the challenges that the present sanitary emergency raises for contemporary urban environments, to then reflect on the reasons behind the progressive abandonment of remote rural areas, and especially on the increasingly scarce accessibility to services they offer to their inhabitants. The authors argue that policies dedicated to counteract these trends are needed, in order to enhance the overall resilience of our societies. In this concern, the National Strategy for Inner Areas, promoted by the Italian Government to trigger place-based territorial development in the remote areas of the country, is a promising way forward, and the present crisis constitutes a window of opportunity to further strengthen and refine its approach in the view of the coming EU programming period.

The application of mechanical stimulation on bone tissue engineering constructs aims to mimic the native dynamic nature of bone. Although many attempts have been made to evaluate the effect of applied mechanical stimuli on osteogenic differentiation, the conditions that govern this process have not yet been fully explored. In this study, pre-osteoblastic cells were seeded on PLLA/PCL/PHBV (90/5/5 wt.%) polymeric blend scaffolds. The constructs were subjected every day to cyclic uniaxial compression for 40 min at a displacement of 400 μm, using three frequency values, 0.5, 1, and 1.5 Hz, for up to 21 days, and their osteogenic response was compared to that of static cultures. Finite element simulation was performed to validate the scaffold design and the loading direction, and to assure that cells inside the scaffolds would be subjected to significant levels of strain during stimulation. None of the applied loading conditions negatively affected the cell viability. The alkaline phosphatase activity data indicated significantly higher values at all dynamic conditions compared to the static ones at day 7, with the highest response being observed at 0.5 Hz. Collagen and calcium production were significantly increased compared to static controls. These results indicate that all of the examined frequencies substantially promoted the osteogenic capacity.
