Abstract

We introduce a novel Dynamic Graph Neural Network (DGNN) architecture for solving conditional $m$-steps ahead forecasting problems in temporal financial networks. The proposed DGNN is validated on simulated data from a temporal financial network model capturing stylized features of Interest Rate Swaps (IRSs) transaction networks, where financial entities trade swap contracts dynamically and the network topology evolves conditionally on a reference rate. The proposed model is able to produce accurate conditional forecasts of net variation margins up to a $21$-day horizon by leveraging conditional information under pre-determined stress test scenarios. Our work shows that the network dynamics can be successfully incorporated into stress-testing practices, thus providing regulators and policymakers with a crucial tool for systemic risk monitoring.

Abstract

Imaging atmospheric Cherenkov telescopes (IACTs) are used to observe very high-energy photons from the ground. Gamma rays are indirectly detected through the Cherenkov light emitted by the air showers they induce. The new generation of experiments, in particular the Cherenkov Telescope Array Observatory (CTAO), sets ambitious goals for discoveries of new gamma-ray sources and precise measurements of the already discovered ones. To achieve these goals, both hardware and data analysis must employ cutting-edge techniques. This also applies to the LST-1, the first IACT built for the CTAO, which is currently taking data on the Canary island of La Palma. This paper introduces a new event reconstruction technique for IACT data, aiming to improve the image reconstruction quality and the discrimination between the signal and the background from misidentified hadrons and electrons. The technique models the development of the extensive air shower signal, recorded as a waveform per pixel, seen by CTAO telescopes' cameras. Model parameters are subsequently passed to random forest regressors and classifiers to extract information on the primary particle. The new reconstruction was applied to simulated data and to data from observations of the Crab Nebula performed by the LST-1. The event reconstruction method presented here shows promising performance improvements. The angular and energy resolution, and the sensitivity, are improved by 10 to 20% over most of the energy range. At low energy, improvements reach up to 22%, 47%, and 50%, respectively. 3. Structuring data analysis projects in the Open Science era with Kerblam!

Abstract

Structuring data analysis projects, that is, defining the layout of files and folders needed to analyze data using existing tools and novel code, largely follows personal preferences. In this work, we look at the structure of several data analysis project templates and find little structural overlap. We highlight the parts that are similar between them, and propose guiding principles to keep in mind when one wishes to create a new data analysis project. Finally, we present Kerblam!, a project management tool that can expedite project data management, execution of workflow managers, and sharing of the resulting workflow and analysis outputs. We hope that, by following these principles and using Kerblam!, the landscape of data analysis projects can become more transparent, understandable, and ultimately useful to the wider community.

4. Assessing the Added Value of a History-Based Activity for Students with Low Mathematics Skills

Abstract

The aim of this pilot study is to evaluate the relevance of the use of history in mathematics education. This paper presents an experiment carried out in France with sixth-grade students (n=108) in which an ancient number system is used, an approach that is commonly suggested in French sixth-grade textbooks but has previously been unassessed. Based on the data of a pretest and a post-test surrounding an activity on an ancient Chinese numeration system, a statistical analysis using Rasch modeling shows a specific added value of the history of mathematics for students with low abilities in mathematics. For these students, a significant increase in observed abilities of +0.67 logit in mean is measured with a large effect size (Cliff delta +0.52). This effect is then weighted by considering the regression to the mean (RTM) effect, leading to a value around +0.14 logit in mean and a negligible effect size (Cliff delta +0.10). So, this pilot study shows the important effect of RTM, which suggests a very strong rebalancing of students' results. In the last part of the paper, we discuss how RTM can nonetheless be positively interpreted in this specific context where students' disorientation is one of the purposes of history in mathematics education.

Abstract

Context: There are currently three pulsars firmly detected by imaging atmospheric Cherenkov telescopes (IACTs), two of them reaching TeV energies, challenging models of very-high-energy (VHE) emission in pulsars. More precise observations are needed to better characterize pulsar emission at these energies. The LST-1 is the prototype of the Large-Sized Telescope, that will be part of the Cherenkov Telescope Array Observatory (CTAO). Its improved performance over previous IACTs makes it well suited for studying pulsars. Aims: To study the Crab pulsar emission with the LST-1, improving and complementing the results from other telescopes. These observations can also be used to characterize the potential of the LST-1 to study other pulsars and detect new ones. Methods: We analyzed a total of $\sim$103 hours of gamma-ray observations of the Crab pulsar conducted with the LST-1 in the period from September 2020 to January 2023. The observations were carried out at zenith angles less than 50 degrees. A new analysis of the Fermi-LAT data was also performed, including $\sim$14 years of observations. Results: The Crab pulsar phaseogram, long-term light-curve, and phase-resolved spectra are reconstructed with the LST-1 from 20 GeV to 450 GeV for P1 and up to 700 GeV for P2. The pulsed emission is detected with a significance of 15.2$\sigma$. The two characteristic emission peaks of the Crab pulsar are clearly detected (>10$\sigma$), as well as the so-called bridge emission (5.7$\sigma$). We find that both peaks are well described by power laws, with spectral indices of $\sim$3.44 and $\sim$3.03 respectively. The joint analysis of Fermi-LAT and LST-1 data shows a good agreement between both instruments in the overlapping energy range. Abstract

Efficiently integrating renewable resources into electricity markets is vital for addressing the challenges of matching real-time supply and demand while reducing the significant energy wastage resulting from curtailments. To address this challenge effectively, the incorporation of storage devices can enhance the reliability and efficiency of the grid, improving market liquidity and reducing price volatility. In short-term electricity markets, participants navigate numerous options, each presenting unique challenges and opportunities, underscoring the critical role of the trading strategy in maximizing profits. This study delves into the optimization of day-ahead and balancing market trading, leveraging quantile-based forecasts. Employing three trading approaches with practical constraints, our research enhances forecast assessment, increases trading frequency, and employs flexible timestamp orders. Our findings underscore the profit potential of simultaneous participation in both day-ahead and balancing markets, especially with larger battery storage systems; despite increased costs and narrower profit margins associated with higher-volume trading, the implementation of high-frequency strategies plays a significant role in maximizing profits and addressing market challenges. Finally, we modelled four commercial battery storage systems and evaluated their economic viability through a scenario analysis, with larger batteries showing a shorter return on investment.

Abstract

7009 Background: The phase 2 CAPTIVATE study evaluated first-line ibrutinib (Ibr) + venetoclax (Ven) for CLL/SLL in 2 cohorts: minimal residual disease (MRD)-guided randomized discontinuation (MRD cohort) and Fixed Duration (FD cohort). Ibr±Ven retreatment was allowed in patients (pts) who had progressive disease (PD). Here, we report outcomes for pts with high-risk genomic features from the FD cohort and retreatment outcomes in pts from the FD cohort and MRD cohort placebo arm. Methods: Pts aged ≤70 y with previously untreated CLL/SLL without restriction on genomic risk factors received 3 cycles of Ibr, then 12 cycles of Ibr+Ven (Ibr, 420 mg/d orally; Ven, 5-wk ramp up to 400 mg/d orally). On-study retreatment included single-agent Ibr (FD cohort or MRD cohort placebo arm); pts with PD >2 y after end of treatment (EOT) could be retreated with FD Ibr+Ven (FD cohort). Results: In the FD cohort (n=159) with a median follow-up of 61.2 mo (range, 0.8–66.3), 5-y PFS and OS rates (95% CI) were 67% (59–74) and 96% (91–98), respectively. 5-y PFS rates were higher in pts with undetectable MRD at 3 mo after EOT in peripheral blood (83%) or bone marrow (84%) vs those without (48% and 50%, respectively). 5-y PFS rates (95% CI) in pts with genomic risk factors were: del(17p)/mutated TP53 41% (21–59), complex karyotype 57% (37–72), del(11q) 64% (30–85), and unmutated IGHV 68% (50–80) (Table). In total, 18 second malignancies occurred in 13 pts (10 events in 8 pts during FD Ibr+Ven, 6 events in 4 pts after EOT and before retreatment, and 2 events in 2 pts during retreatment). Of 202 pts who completed Ibr+Ven (FD cohort, n=159; MRD cohort placebo arm, n=43), 63 have had PD to date; PD occurred >2 y after EOT in 43/63 pts (68%). 32/63 (51%) pts initiated retreatment with Ibr (n=25) or Ibr+Ven (n=7). With a median time on Ibr retreatment of 21.9 mo (range, 0.03–50.4), ORR was 86% in 22 evaluable pts (best response: 1 CR; 1 nodular PR; 17 PR; 2 SD; 1 PD [Richter transformation]). With a median time on Ibr+Ven retreatment of 13.8 mo (range, 3.7–15.1), ORR was 71% in 7 evaluable pts (best response: 1 CR; 4 PR; 1 PR with lymphocytosis; 1 SD). Conclusions: With up to 5.5 y of follow-up, FD Ibr+Ven continues to provide clinically meaningful PFS in pts with high-risk genomic features, as well as in the overall population. Ibr-based retreatment provides promising responses in pts needing subsequent therapy after the all-oral FD regimen of Ibr+Ven. Clinical trial information: NCT02910583 . [Table: see text]

Abstract

Brain lesion segmentation plays an essential role in neurological research and diagnosis. As brain lesions can be caused by various pathological alterations, different types of brain lesions tend to manifest with different characteristics on different imaging modalities. Due to this complexity, brain lesion segmentation methods are often developed in a task-specific manner. A specific segmentation model is developed for a particular lesion type and imaging modality. However, the use of task-specific models requires predetermination of the lesion type and imaging modality, which complicates their deployment in real-world scenarios. In this work, we propose a universal foundation model for 3D brain lesion segmentation, which can automatically segment different types of brain lesions for input data of various imaging modalities. We formulate a novel Mixture of Modality Experts (MoME) framework with multiple expert networks attending to different imaging modalities. A hierarchical gating network combines the expert predictions and fosters expertise collaboration. Furthermore, we introduce a curriculum learning strategy during training to avoid the degeneration of each expert network and preserve their specialization. We evaluated the proposed method on nine brain lesion datasets, encompassing five imaging modalities and eight lesion types. Abstract

We propose a laser resonance chromatography (LRC) experiment on $^{229}$Th$^{3+}$, with the goal of detecting the ion's electronic ground $5f$ $^2$F$_{5/2}$ state and metastable $7s$ $^2$S$_{1/2}$ state by means of their ion mobilities. To this end, we first model the ion-neutral interaction potentials for the two electronic states with a relativistic Fock space coupled cluster method and complete basis-set extrapolation scheme. The interaction potentials are used to simulate the state-specific reduced ion mobilities in terms of the operating temperature and the external electric field. The ion mobilities differ by more than 7% at 300 K and moderate field strengths; thus, separation of the $^{229}$Th$^{3+}$ metastable state lies within the reach of LRC experiments targeting optical probing and monitoring of the nuclear clock transition in this isotope., Comment: 9 pages, 4 figures

Abstract

Monochromatic gamma-ray signals constitute a potential smoking gun signature for annihilating or decaying dark matter particles that could relatively easily be distinguished from astrophysical or instrumental backgrounds. We provide an updated assessment of the sensitivity of the Cherenkov Telescope Array (CTA) to such signals, based on observations of the Galactic centre region as well as of selected dwarf spheroidal galaxies. We find that current limits and detection prospects for dark matter masses above 300 GeV will be significantly improved, by up to an order of magnitude in the multi-TeV range. This demonstrates that CTA will set a new standard for gamma-ray astronomy also in this respect, as the world's largest and most sensitive high-energy gamma-ray observatory, in particular due to its exquisite energy resolution at TeV energies and the adopted observational strategy focussing on regions with large dark matter densities. Throughout our analysis, we use up-to-date instrument response functions, and we thoroughly model the effect of instrumental systematic uncertainties in our statistical treatment. Abstract

We propose a theoretically designed laser resonance chromatography (LRC) experiment on Rf$^+$ (Z = 104) drifting in He buffer gas. To this end, we first developed a four-level rate equation model that simulates the optical pumping of Rf$^+$ from its ground state, $^2$D$_{3/2}$ (7s$^2$6d$^1$), to the metastable $^4$F$_{3/2}$ (7s$^1$6d$^2$) state via laser resonant excitation of the intermediate $^4$F$_{3/2}$ (7s$^1$6d$^1$7p$^1$) state prior to electronic state chromatography. This model predicts a 93% pumping efficiency that suffices to enable efficient laser resonance chromatography of this ion. We then performed accurate relativistic Multi-Reference Configuration-Interaction (MRCI) calculations to model the interaction of Rf$^+$ with He in the ground $^2$D$_{3/2}$ (7s$^2$6d$^1$), low-lying $^2$D$_{5/2}$ (7s$^2$6d$^1$), and metastable $^4$F$_{3/2}$ (7s$^1$6d$^2$) states. These ion-atom interaction potentials were used to calculate the state-specific ion mobilities. For gas temperatures above 100 K and small applied electric fields, the reduced ion mobilities of the ground and metastable states differ significantly. 13. Electricity Price Forecasting in the Irish Balancing Market

Abstract

Short-term electricity markets are becoming more relevant due to less-predictable renewable energy sources, attracting considerable attention from the industry. The balancing market is the closest to real-time and the most volatile among them. Its price forecasting literature is limited, inconsistent and outdated, with few deep learning attempts and no public dataset. This work applies to the Irish balancing market a variety of price prediction techniques proven successful in the widely studied day-ahead market. We compare statistical, machine learning, and deep learning models using a framework that investigates the impact of different training sizes. The framework defines hyperparameters and calibration settings; the dataset and models are made public to ensure reproducibility and to be used as benchmarks for future works. An extensive numerical study shows that well-performing models in the day-ahead market do not perform well in the balancing one, highlighting that these markets are fundamentally different constructs. The best model is LEAR, a statistical approach based on LASSO, which outperforms more complex and computationally demanding approaches.

Abstract

Space exploration and establishing human presence on other planets demand advanced technology and effective collaboration between robots and astronauts. Efficient space resource utilization is also vital for extraterrestrial settlements. The Collaborative In-Situ Resources Utilisation (CISRU) project has developed a software suite comprising five key modules. The first module manages multi-agent autonomy, facilitating communication between agents and mission control. The second focuses on environment perception, employing AI algorithms for tasks like environment segmentation and object pose estimation. The third module ensures safe navigation, covering obstacle avoidance, social navigation with astronauts, and cooperation among robots. The fourth module addresses manipulation functions, including multi-tool capabilities and tool-changer design for diverse tasks in In-Situ Resources Utilization (ISRU) scenarios. Finally, the fifth module controls cooperative behaviour, incorporating astronaut commands, Mixed Reality interfaces, map fusion, task supervision, and error control. The suite was tested using an astronaut-rover interaction dataset in a planetary environment and GMV SPoT analogue environments. Results demonstrate the advantages of E4 autonomy and AI in space systems, benefiting astronaut-robot collaboration. Abstract

Aims. LST-1, the prototype of the Large-Sized Telescope for the upcoming Cherenkov Telescope Array Observatory, is concluding its commissioning in Observatorio del Roque de los Muchachos on the island of La Palma. The proximity of LST-1 (Large-Sized Telescope 1) to the two MAGIC (Major Atmospheric Gamma Imaging Cherenkov) telescopes permits observations of the same gamma-ray events with both systems. Methods. We describe the joint LST-1+MAGIC analysis pipeline and use simultaneous Crab Nebula observations and Monte Carlo simulations to assess the performance of the three-telescope system. The addition of the LST-1 telescope allows the recovery of events in which one of the MAGIC images is too dim to survive analysis quality cuts. Results. Thanks to the resulting increase in the collection area and stronger background rejection, we find a significant improvement in sensitivity, allowing the detection of 30% weaker fluxes in the energy range between 200 GeV and 3 TeV. The spectrum of the Crab Nebula, reconstructed in the energy range ~60 GeV to ~10 TeV, is in agreement with previous measurements., Comment: Accepted for publication in Astronomy & Astrophysics

Abstract

There is a paucity of information about milk composition and quality at the quarter level from complete milking of the udder, mainly because of the difficulties of sampling procedures which imply the adoption of a vacuum system connected to four independent buckets. The objective of this study was to evaluate the effect of quarter position, intramammary infection status, and somatic cells on milk yield and composition at the mammary quarter level of Simmental cows. Milk samples were collected during three consecutive days, after milking individual mammary quarters of 10 lactating cows separately, for a total of 120 quarter milk samples. Samples were analysed for composition, somatic cell count (SCC), differential SCC, and bacterial culture. Somatic cell count was log-transformed to somatic cell score (SCS) to achieve normality of distribution. Sources of variation of milk yield and quality traits were investigated using a linear mixed model which included the fixed effects of quarter position (1 to 4), pathogen presence (positive or negative), and SCS class (low, medium-low, medium-high, and high). The interaction between cow and quarter position was included as random effect. Quarter position significantly affected milk yield, with rear udder quarters yielding more milk compared to their front counterparts, but with negligible effects on milk composition. Classes of SCS influenced milk composition, in a way that quarters with high SCS had lower lactose content and higher differential SCC. This study is a contribution towards the possibility to better understand physiological processes at cow udder quarter level.

Abstract

Abstract P. aeruginosa employs specific quorum sensing (QS) mechanisms to orchestrate biofilm formation, enhancing resistance to host defences. In physiological conditions, QS molecules permeate the lung environment and cellular membrane to reach the cytoplasmic Aryl Hydrocarbon Receptor (AhR) that is pivotal for activating the immune response against infection. In pathological conditions like cystic fibrosis (CF) this interkingdom communication is altered, favouring P. aeruginosa persistence and chronic infection. Here, we aim to investigate the molecular journey of QS molecules from CF-like environments to the cytoplasm by quantifying via HPLC-MS the permeability of selected QS molecules (quinolones, lactones, and phenazines) through in vitro models of the two main biological lung barriers: CF-mucus and cellular membrane. While QS molecules not activating AhR exhibit intermediate permeability through the cellular membrane model (PAMPA) (1.0–4.0 × 10−6 cm/s), the AhR-activating molecule (pyocyanin) shows significantly higher permeability (8.6 ± 1.4 × 10−6 cm/s). Importantly, combining the CF mucus model with PAMPA induces a 50% decrease in pyocyanin permeability, indicating a strong mucus-shielding effect with pathological implications in infection eradication. This study underscores the importance of quantitatively describing the AhR-active bacterial molecules, even in vitro, to offer new perspectives for understanding P. aeruginosa virulence mechanisms and for proposing new antibacterial therapeutic approaches.

ABSTRACT: In modern breeding systems, cows are subjected to many stress factors. Animals fed a high-grain diet may have a decreased rumen pH, which would lead to subacute ruminal acidosis syndrome. The aim of this study was to investigate the evolution of microbial community composition in cows undergoing a dietary stress challenge. Twelve cows were subjected to a challenge period that consisted of a rapid change of ration, from a normal (45.4:54.6 forage:concentrate) to a high-grain content diet (24.8:75.2 forage:concentrate) to induce subacute ruminal acidosis. Individual rumen fluid content samples were collected before (T0) and during the challenge (T3, T14, T28). The DNA from rumen contents was extracted, purified, and sequenced to evaluate bacterial populations, and sequencing was performed on Illumina MiSeq. The effect of animal conditions on rumen microbial community was quantified through a linear mixed model. The acidogenic diet created 2 main clusters: ruminal hypomotility (RH) and milk fat depression (MFD). The microbial composition did not differ in T0 between the 2 groups, whereas during the challenge Ruminococcus spp., Treponema spp., Methanobrevibacter spp., and Methanosphaera spp. concentrations increased in RH cows; and Succinivibrio spp. and Butyrivibrio spp. concentrations increased in MFD cows. Prevotella spp. and Ruminococcus spp. were negatively correlated, whereas the Christenellaceae family was positively correlated with both Methanobrevibacter spp. and Methanosphaera spp. Moreover, the same diet affected cows' microbiota composition differently, underlying the impact of the host effect. Other studies are necessary to deepen the relationship between microbiota composition and host.

Abstract Animals’ ability to orient and navigate relies on selecting an appropriate motor response based on the perception and integration of the environmental information. This is the case, for instance, of the optokinetic response (OKR) in Drosophila melanogaster, where optic flow visual stimulation modulates head movements. Despite a large body of literature on the OKR, there is still a limited understanding, in flies, of the impact on OKR of concomitant, and potentially conflicting, inputs. To evaluate the impact of this multimodal integration, we combined in D. melanogaster, while flying in a tethered condition, the optic flow stimulation leading to OKR with the simultaneous presentation of olfactory cues, based on repellent or masking compounds typically used against noxious insect species. First, this approach allowed us to directly quantify the effect of several substances and of their concentration on the dynamics of the flies’ OKR in response to moving gratings by evaluating the number of saccades and the velocity of the slow phase. Subsequently, this analysis was capable of easily revealing the actual effect, i.e. masking vs. repellent, of the compound tested. In conclusion, we show that D. melanogaster, a cost-affordable species, represents a viable option for studying the effects of several compounds on the navigational abilities of insects.

22. Where Less Is More: Limited Feedback in Formative Online Multiple-Choice Tests Improves Student Self-Regulation

Abstract

Background: Formative online multiple-choice tests are ubiquitous in higher education and potentially powerful learning tools. However, commonly used feedback approaches in online multiple-choice tests can discourage meaningful engagement and enable strategies, such as trial-and-error, that circumvent intended learning outcomes. These strategies will not prepare graduates as self-regulated learners, nor for the complexities of contemporary work settings. Objectives: To investigate whether providing only a score after formative online multiple-choice test attempts (score-only feedback) increases the likelihood of students to engage in self-regulated learning compared with more directive feedback. Measurable outcomes included deeper learning, collaboration, information seeking, and satisfaction. Methods: Data in this mixed methods study were collected from nursing students through surveys, test results, focus groups, and student discussion board contributions. A quasi-experimental design was used for quantitative data, and qualitative data were analysed thematically against domains of self-regulated learning. Results and Conclusions: Students receiving score-only feedback were more cognitively engaged with the content, collaborated constructively, and sought out richer sources of information. However, it was also associated with lower satisfaction. In this study, minimal feedback created states of uncertainty, which resulted in the activation of self-regulatory actions. Implications for Practice: Providing overly directive feedback for formative online multiple-choice tests is conducive to surface-level learning strategies. By minimising feedback and allowing for extended states of uncertainty, students are more likely to regulate their learning through self-assessment and problem-solving strategies, all of which are required by graduates to meet the challenges of real-world work settings.

23. International Student Mobility and Academic Performance: Does Timing Matter?

Abstract

We examine the impact of credit mobility exchange programs' timing on students' academic performance, focusing on the moment in which students travel and the length of the period spent abroad. To provide causal evidence, we exploit unique data from more than 10,000 students from a well-known and internationalized Brazilian university from 2010 to 2020. By combing Propensity Score Matching with Difference in Differences techniques, we find that international mobility impacts groups of students differently. Students who travel closer to the end of their undergraduate courses benefit the most from the mobility experience, while negative effects are found for those who travel at the beginning of their university program. Results also show that, while student mobility impacts positively and significantly students who participate in programs lasting from one semester to 1 year, negative effects are associated with shorter periods abroad. Our findings also reveal heterogeneity across destination countries. Mobility has a positive impact on students' grades for those students traveling to English-speaking countries. Our analysis presents empirical evidence that can be used to design international student mobility programs, providing insights to policymakers engaged in maximizing their effects.

Abstract

Abstract Colistin is a polymyxin antibiotic currently experiencing renewed clinical interest due to its efficacy in the treatment of multidrug resistant (MDR) bacterial infections. The frequent onset of acute dose-dependent kidney injury, with the potential of leading to long-term renal damage, has limited its use and hampered adequate dosing regimens, increasing the risk of suboptimal plasma concentrations during treatment. The mechanism of colistin-induced renal toxicity has been postulated to stem from mitochondrial damage, yet there is no direct evidence of colistin acting as a mitochondrial toxin. The aim of this study was to evaluate whether colistin can directly induce mitochondrial toxicity and, if so, uncover the underlying molecular mechanism. We found that colistin leads to a rapid permeability transition of mitochondria isolated from mouse kidney that was fully prevented by co-incubation of the mitochondria with desensitizers of the mitochondrial transition pore cyclosporin A or L-carnitine. The protective effect of L-carnitine was confirmed in experiments in primary cultured mouse tubular cells. Consistently, the relative risk of colistin-induced kidney damage, calculated based on histological analysis as well as by the early marker of tubular kidney injury, Kim-1, was halved under co-administration with L-carnitine in vivo. Notably, L-carnitine neither affected the pharmacokinetics of colistin nor its antimicrobial activity against relevant bacterial strains. In conclusion, colistin targets the mitochondria and induces permeability transition thereof. L-carnitine prevents colistin-induced permeability transition in vitro. Moreover, L-carnitine co-administration confers partial nephroprotection in mice treated with colistin, without interfering with its pharmacokinetics and antibacterial activity.

Abstract Background Intramammary infection is the result of invasion and multiplication of microorganisms in the mammary gland and commonly leads to mastitis in dairy animals. Although much has been done to improve cows’ udder health, mastitis remains a significant and costly health issue for dairy farmers, especially if subclinical. In this study, quarter milk samples from clinically healthy cows were harvested to detect pathogens via quantitative PCR (qPCR) and evaluate changes in individual milk traits according to the number of quarters infected and the type of microorganism(s). A commercial qPCR kit was used for detection of Mycoplasma bovis, Mycoplasma spp., Staphylococcus aureus, coagulase-negative staphylococci (CNS), Streptococcus agalactiae, Streptococcus dysgalactiae, Streptococcus uberis, Prototheca spp., Escherichia coli, Klebsiella spp., Enterococcus spp. and Lactococcus lactis ssp. lactis. Quarter and pooled milk information of 383 Holstein, 132 Simmental, 129 Rendena, and 112 Jersey cows in 9 Italian single-breed herds was available. Results Among the cows with pathogen(s) present in at least 1 quarter, CNS was the most commonly detected DNA, followed by Streptococcus uberis, Mycoplasma bovis, and Streptococcus agalactiae. Cows negative to qPCR were 206 and had the lowest milk somatic cell count. Viceversa, cows with DNA isolated in ≥ 3 quarters were those with the highest somatic cell count. Moreover, when major pathogens were isolated in ≥ 3 quarters, milk had the lowest casein index and lactose content. In animals with pathogen(s) DNA isolated, the extent with whom milk yield and major solids were impaired did not significantly differ between major and minor pathogens. Conclusions The effect of the number of affected quarters on the pool milk quality traits was investigated in clinically healthy cows using a commercial kit. Results remark the important negative effect of subclinical udder inflammations on milk yield and quality, but more efforts should be made to investigate the presence of untargeted microorganisms, as they may be potentially dangerous for cows. For a smarter use of antimicrobials, analysis of milk via qPCR is advisable – especially in cows at dry off - to identify quarters at high risk of inflammation and thus apply a targeted/tailored treatment.

Abstract

Predicting future resource demand in Cloud Computing is essential for optimizing the trade-off between serving customers' requests efficiently and minimizing the provisioning cost. Modelling prediction uncertainty is also desirable to better inform the resource decision-making process, but research in this field is under-investigated. In this paper, we propose univariate and bivariate Bayesian deep learning models that provide predictions of future workload demand and its uncertainty. We run extensive experiments on Google and Alibaba clusters, where we first train our models with datasets from different cloud providers and compare them with LSTM-based baselines. Results show that modelling the uncertainty of predictions has a positive impact on performance, especially on service level metrics, because uncertainty quantification can be tailored to desired target service levels that are critical in cloud applications. Moreover, we investigate whether our models benefit transfer learning capabilities across different domains, i.e. dataset distributions. Experiments on the same workload datasets reveal that acceptable transfer learning performance can be achieved within the same provider (because distributions are more similar). Also, domain knowledge does not transfer when the source and target domains are very different (e.g. from different providers), but this performance degradation can be mitigated by increasing the training set size of the source domain.

IntroductionThe identification of baseline prognostic factors in Classical Hodgkin Lymphoma could help in tailoring a risk-based approach as the therapeutic landscape expands. Currently, the International Prognostic Score (IPS) represents the most used prediction tool in clinical practice, but other potential baseline risk predictors have been identified.MethodsWe performed a retrospective analysis in a cohort of 274 patients treated with 18FDG-PET/CT-guided ABVD to assess the prognostic significance of the IPS risk factors, and to validate the impact of the peripheral blood lymphocyte to monocyte (LMR) and neutrophil to lymphocyte (NLR) ratios on prognosis definition.ResultsAmong the considered risk factors, stage IV disease (HR 1.83), leukocytosis (HR 2.28), anemia (HR 3.23) and low LMR (HR 2.01) significantly predicted PFS, whereas male sex (HR 2.93), stage IV disease (HR 3.00) and lymphopenia (HR 7.84) significantly predicted OS. A 4 variable and a 3 variable prognostic system was subsequently proposed for PFS and OS, respectively. In both cases, a stark decrease in the survival probability was documented as the score increased. Moreover, by selecting only the significant IPS items and considering a more recently proposed prognostic factor (LMR) we were able to better identify patients at higher risk of relapsing after PET/CT-guided ABVD.DiscussionAlthough the IPS was still able to identify a subgroup of high-risk patients within our cohort of individuals treated with PET/CT-guided ABVD, not all the risk factors that it considers were found to have an impact on survival times. Moreover, by selecting only the significant IPS items and considering a more recently proposed prognostic factor (LMR) we were able to better identify patients at higher risk of relapse, in an effort to contribute to the building of a modern risk prediction tool that can help guide treatment choices.
