Toxoplasma gondii is a widespread protozoan parasite (phylum Apicomplexa), which causes a zoonotic parasitic disease, known as toxoplasmosis. The aim of this study was to evaluate the occurrence and genotypes of T. gondii in wild boars of southern Italy and thus to assess the risk of infection for consumers. The boars were inspected during the hunting season within the regional project ‘Wild Boar Emergency Plan in Campania’, and molecular analyses were performed on 338 boars analysing a total number of 884 matrices (263 brains, 310 hearts and 311 masseter muscles). Toxoplasma gondii was detected in 134 out of 338 boars (39.6%). No significant statistical difference between genders was found (χ2 = 0.15 p = .70). The prevalence was 47.1%, 39.3% and 39.2% in piglets, yearlings and adults, respectively (χ2 = 0.41; p = .81). The highest prevalence of T. gondii was found in masseter muscles (74/311, 23.8%), followed by the heart (70/310, 22.6%) and brain (58/263, 22.0%), respectively. Microsatellite (MS) analysis of 11 samples revealed eleven T. gondii genotypes (nine atypical, one belonging to type II one to type III). Most of the genotypes found were thus atypical and may be virulent in humans. Hierarchical clustering analysis showed the presence of three distinct clusters, with the majority of atypical genotypes in the GII-GIII cluster. The high prevalence of infection in masseters highlights the potential risk for public health, considering that this muscle is commonly used to prepare raw meat products (‘guanciale’ and sausages), which may be a source of T. gondii infection in humans. Wild boars may act as an interface role between wildlife, livestock and humans. Our data highlight the urgent need to minimize the risk of infection for animals and humans by setting up a surveillance programme and preventive strategies in a One Health approach to wildlife species.

Anti-viral activities of long-chain inorganic polyphosphates (PolyPs) against severe acute respiratory syndrome coronavirus (SARS-CoV)-2 infection were investigated. In molecular docking analyses, PolyPs interacted with several conserved angiotensin-converting enzyme (ACE)2 and RNA-dependent RNA polymerase (RdRp) amino acids. We thus tested PolyPs for functional interactionsin vitroin SARS-CoV-2–infected Vero E6, Caco2 and human primary nasal epithelial cells. Immunofluorescence, qPCR, direct RNA sequencing, FISH and Immunoblotting were used to determine virus loads and transcription levels of genomic(g)RNAs and sub-genomic(sg)RNAs. We show that PolyP120 binds to ACE2 and enhances its proteasomal degradation. PolyP120 shows steric hindrance of the genomic Sars-CoV-2-RNA/RdRP complex, to impair synthesis of positive-sense gRNAs, viral subgenomic transcripts and structural proteins needed for viral replication. Thus, PolyP120 impairs infection and replication of Korean and European (containing non-synonymous variants) SARS-CoV-2 strains. As PolyPs have no toxic activities, we envision their use as a nebulised formula for oropharyngeal delivery to prevent infections of SARS-CoV-2 and during early phases of antiviral therapy.

Summary Introduction. Patient safety and quality in healthcare are inseparable. Examining patient safety culture in staff members contributes to further develop quality in healthcare. In Italy there has been some experience in assessing patient safety culture in staff working in hospital. In this pilot study we explored patient safety culture in public health staff working in Italian Local Health Authorities. Methods. We carried out a descriptive cross sectional study in four Italian territorial Prevention facilities in Northern Italy. We administrated an adapted Italian version of the US Hospital Survey of Patient Safety Culture to all the staff within these facilities. The survey consisted of 10 dimensions based on 33 items, according to the results of a previous psychometric validation. Results. Seventy per cent of the staff responded to the survey (N = 479). Overall, six out of the 10 dimensions exhibited composite scores of positive response frequency for patient safety culture below 50%. While "communication openness" (65%) was the most developed factor, "teamwork across Units" (37%) was the least developed. The work areas with the highest composite scores were Management and the Public Health Laboratory, while in terms of professional categories, Physicians had the highest scores. Patient safety culture in the staff participating in this study was lower than in hospital staff. Discussion. Our descriptive cross sectional study is the first to be carried out in Preventive medicine settings in Italy. It has clearly indicated the need of improvement. Consequently, several interventions with this aim have been implemented.

The present work focuses its attention about the need of implementation of an energy recovery system for a renewable energy device as photovoltaic cells. In a specific way the present paper is addressed to the evaluation of a potential use of Peltier cells to recover waste heat of PV, using the Seedback effect. An overview of Peltier cells, mainly focused on their functionality, properties and possible applications, is initially presented. The results of an extensive experimental test campaign is then presented; these tests have mainly been dedicated to the evaluation of Peltier cells properties in the electrical conversion of heating energy produced and/or not dissipated during the photovoltaic phenomena in siliceous PV cells. The last part of the work presents the results of a parallel test campaign where Peltier cells have been used as thermoelectric cooling devices, in the aim to improve the PV cell performance. In fact the efficiency and technical lifetime of these latter elements, is strongly affected by temperature of photovoltaic cell. This point and the dissipated heat are fundamental topics for every installation but above all in the cases of regions with a very hot climate or for concentrated photovoltaic plants.

European Legislation has fixed microbiological, chemical and biotoxicological limits for shellfish but no limits for viruses. In the present study we report the results of an investigation on Salmonella spp., Escherichia coli, Hepatitis A virus (HAV) and Norovirus (NoV) contamination in 59 bivalve shellfish collected during the years 2011-2012 in Southern Italy. All the samples of Mytilus galloprovincialis and of Solen marginatus were negative for HAV whereas 6.8% of them were positive for Norovirus GI (NoVGI) and 11.9% positive for Norovirus GII (NoVGII). Samples were also negative for Salmonella spp., while 16 of them (27%) were positive for E. coli. No correlation was found between E. coli and NoV contamination in bivalve molluscs. Moreover, the Competent Authorities are advised to take into serious consideration additional measures for the legislation in force in order to guarantee the consumer's health.

Case Study 2: Palazzo D'Accursio, Bologna, Italy

The use of composite materials has grown significantly in recent years thanks to new manufacturing processes, which have allowed for competing of laminates with metal alloys in in a broad spectrum of technological applications. As known, a composite is a material made by joining two or more constituents, therefore key concept is to combine more performances in a single final product. For their peculiarities, such as the heterogeneity and anisotropy, composites are materials used as a solution to problems of different areas, taking advantage of lightweight, strength, stiffness, good behavior to fatigue and corrosion and reduced manufacturing costs. Precisely because of their complex nature, the execution into a potential industrialization stage pose to the designer new problems but also targets, dictated by the technical need to clearly characterize both the macroscopic that the microscopic properties. The main goal of this activity has been to assess the mechanical properties of two carbon fiber/epoxy samples by means of experimental tests and numerical simulations with a very good correlation level. The outcomes achieved and described in this paper have been performed within an ambitious research project focused on the development and application of self-healing materials: one of the specimens in fact was filled with microcapsules to trigger the self-repair process in case of damage. Among the significant advantages of these smart materials, the improved ability to dissipate vibration energy has been especially appreciated in this framework. The conducted tests have revealed a higher damping coefficient compared to that one of a standard CFRC. Relying upon the validated FEM, sensitivity nonlinear analyses were carried out to evaluate the stiffness trend with respect to first-ply failure.

The widespread of obesity in childhood is a rising cause of the development of metabolic syndrome, the prevalence of which is clearly increasing. The complexity of the frequent concurrence between metabolic syndrome and type 2 diabetes requires the therapeutic approach to be multidisciplinary and based on the stabilization of glycemia, monitoring of arterial pressure and of the lipid profile, and on the development of an appropriate hypocaloric diet and exercise plan. Moreover, the therapeutic objective of stablilizing glycemia may be achieved with the administration of metformin, the only oral hypoglycemic agent approved for type 2 diabetes children. We here describe a clinical case which highlights the complexities of the diagnostic approach.

Several studies to increase train speed are underway with the target to design a new generation pantograph in order to reduce aerodynamic noise. The aerodynamic characteristics as shapes of pantograph are the main issue. The present work deals with a high-speed aerodynamic and aeroacoustic analysis of a three-dimensional pantograph model, through the employment of 3D numerical simulations. In particular, a computational fluid dynamics-based analysis is performed using commercial software code FLUENT AnsysTM and an experimental campaign is performed in the high-speed wind tunnel in order to verify the accuracy of numerical approach. The aerodynamic noise is, then, calculated through the Ffowcs Williams--Hawkings (FW-H) equation to evaluate the flow induced sound pressure level in aeroacoustics. The numerical and experimental results are then compared in terms of sound pressure levels and a well agreement has been found. The numerical results in terms of visualization of pressure distribution on the pantograph provides useful information for pantograph design. The results highlight the possibility to develop a new design of the pantograph for high-speed trains, in order to obtain an aerodynamic noise reduction

Present work focuses on the implementation of numerical and experimental analyses aimed to acoustic performances characterization of a composite sandwich panel used for a high-speed train. Firstly, an experimental and a numerical modal analyses are presented. Starting from both FE simulation and impact testing outcomes, it has been possible to carry out a correlation study through the computation of the Modal Assurance Criterion (MAC). Good agreement between numerical and experimental analyses has been found, therefore the definition of a reliable FE model has been obtained without the necessity of implementing a sensitivity and updating procedure. In this paper, to find a convenient and accurate mean for predicting the panel Transmission Loss parameter, the panel is modeled as a composite sandwich panel, and its TL is predicted with the hybrid FE&SEA (Statistical Energy Analysis) method. The TL result is then compared to the experimental one, carried out through the employment of an intensity sound probe. A very good accordance has been found allowing to use such numerical procedure for further acoustic performances improvements. Hence, future developments could regard the possibility to implement a Reverse Engineering procedure, in order to realize an optimization process by considering different materials and stratifications or different panel thicknesses, to improve the acoustic attenuation properties at those frequencies at which a worse acoustic behavior of the panel, is present.

Phage-resistant mutants have been isolated from Streptococcus thermophilus. Selection was carried out using anti-phage antibodies or Hoechst 33258-labelled phages. Two mutants out of eight tested displayed reduced acidifying capacity. Selection of the bacteria that extruded more rapidly the fluorochrome 5-6-carboxyfluorescein diacetate (CFDA) restored the acidifying capacity of these two mutants to the level of the parental strains. Mutants displaying phage resistance and good acidifying capacity were obtained in 4-5 days. New phages that are able to overcome the protection mechanisms of the existing bacteria arise continually in the dairy environment. The procedures described here permit to replace promptly the starter culture susceptible to newly emerged phages with a resistant one.

Background and purpose To carry out an observational epidemiological survey (Studio Morfeo), to determine: (1) the frequency of insomnia in a large Italian population presenting directly to the general physician (GP); (2) the impact of insomnia on the quality of life, on the use of health-care resources and on co-morbidity. Patients and methods The study was accomplished by GPs, trained by sleep specialists accredited by the Italian Association of Sleep Medicine. Only patients spontaneously presenting to their GP for medical problems were surveyed. Each GP was asked to enroll at least five patients across a routine week of medical activity including both morning and afternoon clinics. The first patient of each weekday was recruited after obtaining written consent. According to the responses to the sleep-related questions, patients were classified into three categories: (1) no insomnia, (2) level 1 insomnia with absence of day-time dysfunction and (3) level 2 insomnia with presence of day-time dysfunction. Results A total of 3284 patients were enrolled by 738 GPs in this Italian survey. Insomnia was reported by 64% of all interviewed patients, with 20% classified as level 1 and 44% as level 2. Logistic analysis indicated that depression (odds ratio, 2.70), involvement of >1 organ systems (odds ratio, 1.24), female gender (odds ratio, 1.19), unemployment (odds ratio, 1.18), low education (odds ratio, 1.18) and increasing age (odds ratio, 1.02) were the major risk factors for insomnia. Conclusions Our findings indicate that insomnia is a frequent disturbance in the Italian primary care population, is associated with high risk of co-morbid conditions, and results in increased use of health-care resources.
