X‐ray imaging of virus particles at the European XFEL could eventually allow their complete structures to be solved, potentially approaching the resolution of other structural virology methods. To achieve this ambitious goal with today's technologies, about 1 ml of purified virus suspension containing at least 1012 particles per millilitre is required. Such large amounts of concentrated suspension have never before been obtained for enveloped viruses. Tick‐borne encephalitis virus (TBEV) represents an attractive model system for the development of enveloped virus purification and concentration protocols, given the availability of large amounts of inactivated virus material provided by vaccine‐manufacturing facilities. Here, the development of a TBEV vaccine purification and concentration scheme is presented combined with a quality‐control protocol that allows substantial amounts of highly concentrated non‐aggregated suspension to be obtained. European brown hare syndrome (EBHS) is a highly contagious and fatal viral disease, mainly affecting European brown hares (Lepus europaeus). The etiological agent, EBHS virus (EBHSV), belongs to the Lagovirus genus within the Caliciviridae family. The Italian hare (Lepus corsicanus) is endemic to Central-Southern Italy and Sicily and is classified as a vulnerable species. L. corsicanus is known to be susceptible to EBHS, but virological data available is scarce due to the few cases detected so far. In this study, we describe the occurrence of EBHS in two free-ranging L. corsicanus, found dead in a protected area of Central Italy. The two hares were identified as L. corsicanus using phenotypic criteria and confirmed through mitochondrial DNA analysis. Distinctive EBHS gross lesions were observed at necropsy and confirmed by subsequent histological examination. EBHSV was detected in the livers of the two animals initially using an antigen detection ELISA, followed by an EBHSV-specific reverse transcription-PCR, thus confirming the viral infection as the probable cause of death. The EBHS viruses detected in the two hares were identical, as based on blast analysis performed for the VP60 sequences and showed 98.86% nucleotide identity and 100% amino acid identity with strain EBHSV/GER-BY/EI97.L03477/2019, isolated in Germany in 2019. Phylogenetic analysis places our virus in group B, which includes strains that emerged after the mid-1980s. This study supports previous reports of EBHS in L. corsicanus and further expands the knowledge of the pathological and virological characteristics of the etiological agent. The ability of EBHSV to cause a fatal disease in the Italian hare represents a serious threat to the conservation of this vulnerable species, especially in populations kept in enclosed protected areas. Epizootic haemorrhagic disease (EHD) is a Culicoides-borne viral disease caused by the epizootic haemorrhagic disease virus (EHDV) associated with clinical manifestations in domestic and wild ruminants, primarily white-tailed deer (Odocoileus virginianus) and cattle (Bos taurus). In late September 2021, EHDV was reported in cattle farms in central/western Tunisia. It rapidly spread throughout the country with more than 200 confirmed outbreaks. We applied a combination of classical and molecular techniques to characterize the causative virus as a member of the serotype EHDV-8. This is the first evidence of EHDV- 8 circulation since 1982 when the prototype EHDV-8 strain was isolated in Australia. Largemouth bass is an important commercially farmed fish in China, but the rapid expansion of its breeding has resulted in increased incidence of diseases caused by bacteria, viruses and parasites. In this study, moribund largemouth bass containing ulcer foci on body surfaces indicated the most likely pathogens were iridovirus and rhabdovirus members and this was confirmed using a combination of immunohistochemistry, cell culture, electron microscopy and conserved gene sequence analysis. We identified that these fish had been co-infected with these viruses. We observed bullet-shaped virions (100–140 nm long and 50–100 nm in diameter) along with hexagonal virions with 140 nm diameters in cell culture inoculated with tissue homogenates. The viruses were plaque purified and a comparison of the highly conserved regions of the genome of these viruses indicated that they are most similar to largemouth bass virus (LMBV) and hybrid snakehead rhabdovirus (HSHRV), respectively. Regression infection experiments indicated fish mortalities for LMBV-FS2021 and HSHRV-MS2021 were 86.7 and 11.1%, respectively. While co-infection resulted in 93.3% mortality that was significantly (p < 0.05) higher than the single infections even though the viral loads differed by >100-fold. Overall, we simultaneously isolated and identified LMBV and a HSHRV-like virus from diseased largemouth bass, and our results can provide novel ideas for the prevention and treatment of combined virus infection especially in largemouth bass. Never before have the media focused on a single infectious disease as they have in the case of the coronavirus COVID-19 pandemic that started to spread globally from China at the end of 2019. The consequences of the pandemic on health, economics, and the societal conditions of isolated individuals have been discussed from a range of different perspectives. Virologists are expected to be capable of providing immediate answers to many different kinds of questions—how and under what conditions is an individual infectious, what are the relative roles of the different arms of the immune system, do reinfections occur, when will a vaccine preventing infection with the virus become available, what are the possibilities of developing antiviral drugs capable of interfering with the disease, and so on. In many cases there are no immediate answers, since virologists globally are still in the middle of researching the particular problem in the focus of interest. The only proper answer to demanding questions of this kind should be “Welcome to the workshop of virologists.” However, what needs to be emphasized is that the tools available to understand the details of the interaction of a particular virus and the various organs in an infected human host have changed dramatically during the somewhat more than a hundred years of studies of viruses.

Full genome sequences are presented for two isolates of Spartina mottle virus (SpMoV) from two accessions of the hybrid turf grass Cynodon dactylon × C. transvaalensis (Bermuda grass, green couch), originally from the USA but detected during post‐entry quarantine in Australia. Both isolates had a genome of 9,346 nucleotides, encoding a single polyprotein of 3,029 amino acids and predicted to be cleaved into 10 mature proteins. Phylogenetic analysis strongly supported placement in a new genus within the family Potyviridae. The isolates possess 715–780 × 11–12 nm flexuous virions and produce cylindrical (pinwheel) inclusions in infected cells. They were mechanically transmitted to several species in the Poaceae including Zea mays (maize, sweet corn), Chloris gayana (Rhodes grass), and Echinochloa colona (awnless barnyard grass). Summary: The Kachia Grazing Reserve (KGR) is located in Kaduna state in north‐western Nigeria and consists of 6 contiguous blocks housing 744 defined households (HH), all engaged in livestock keeping. It is considered as a homogenous epidemiological unit and a defined study area. In 2012, all cattle and sheep of 40 selected HH were sampled to determine sero‐prevalence of antibodies to foot‐and‐mouth disease virus (FMDV) and of FMDV. The overall sero‐prevalence of antibodies to the non‐structural 3ABC protein (NSP‐3ABC ELISA) was 28.9% (380/1,315) (30.6% cattle; 16.3% sheep), and in 4.5% (62/1,380) (5% cattle; 0.6% sheep) of the examined sera FMD viral RNA could be detected by real‐time RT‐PCR (rRT‐PCR). Additionally, in 2012 and 2014 serum, epithelium and probang samples were collected from cattle in reported FMD outbreaks and the causative FMDVs were molecularly characterized. Approximately half (28/59) of the outbreak sera reacted positive in NSP‐3ABC ELISA, and 88% (52/59) of the outbreak sera contained detectable viral RNA. Overall, antibodies against five FMDV serotypes (O, A, SAT1, SAT2 and SAT3) were detected by solid phase competitive ELISA with combinations of two or more serotypes being common. Of the 21 FMDVs that could be isolated 19 were sequenced and 18 were confirmed as SAT2 (lineage VII) while one was characterized as serotype O (EA‐3 topotype). Phylogenetic analysis revealed a close relationship between Nigerian FMDV strains and strains in this region and even with strains in North‐Africa. Our findings indicate that FMD constitutes an endemic health problem to cattle rearing in the agro‐pastoralist community in the KGR and that the KGR is not a closed epidemiological unit. Insight into the local FMDV epidemiology and in the circulating FMDV serotypes/strains is of support to the relevant authorities in Nigeria when considering the need for an FMD control policy to improve animal production in grazing reserves. Apple mosaic disease is one of the most widely distributed and destructive diseases in apple cultivation worldwide, especially in China, whose apple yields account for more than 50% of the global total. Apple necrotic mosaic virus (ApNMV) is a newly identified ilarvirus that is closely associated with apple mosaic disease in China; however, basic viral protein interactions that play key roles in virus replication and the viral life cycle have not been determined in ApNMV. Here, we first identify an ApNMV–Lw isolate that belongs to subgroup 3 in the genus Ilarvirus. ApNMV–Lw was used to investigate interactions among viral components. ApNMV 1a and 2apol, encoded by RNA1 and RNA2, respectively, were co-localized in plant cell cytoplasm. ApNMV 1a interacted with itself at both the inter- and intramolecular levels, and its N-terminal portion played a key role in these interactions. 1a also interacted with 2apol, and 1a’s C-terminal, together with 2apol’s N-terminal, was required for this interaction. Moreover, the first 115 amino acids of 2apol were sufficient for permitting the 1a–2apol interaction. This study provides insight into the protein interactions among viral replication components of ApNMV, facilitating future investigations on its pathogenicity, as well as the development of strategies to control the virus and disease.

Recently, a novel atypical porcine pestivirus (APPV) in pig was reported. In this study, two APPV strains, APPV-China/GZ01/2016 (GZ01) and APPV-China/GD-SD/2016 (GD-SD), were identified in two newborn piglet herds with congenital tremor from China. The open reading frame of the two strains shared an 83.5% nucleotide identity. Phylogenetically, the APPV strains were placed into two groups: GZ01 belonged to group I and GD-SD belonged to group II. A high viral load was detected in the cerebellum (quantification cycles < 26). Further studies should be carried out to thoroughly elucidate the development of congenital tremors caused by APPV. Abstract: This study fully describes a severe disease outbreak occurred in 2016 in black bullhead catfish farmed in Italy. Affected fish showed nervous clinical signs as well as emaciations and haemorrhagic petechiae on the skin at the fin bases, abdomen and gills. Viral isolation in cell culture allowed the subsequent identification of a rhabdovirus, tentatively named ictalurid rhabdovirus (IcRV), through electron microscopy, immunofluorescence and whole genome sequencing (WGS). The newly isolated virus, together with 14 additional viral strains stored in our repository and detected during similar mortality episodes in the period 1993–2016, was phylogenetically analysed on the basis of the nucleoprotein and the glycoprotein nucleotide and amino acid sequences. The genetic distances among Italian IcRV strains were also estimated. Our results show that all the IcRV strains belong to the genus Sprivivirus and are closely related to the tench rhabdovirus (TenRV). Italian catfish production is constantly decreasing, mainly due to viral infections, which include the newly characterized IcRV. Data presented in this work will assist to investigate the molecular epidemiology and the diffusive dynamics of this virus and to develop adequate surveillance activities. [ABSTRACT FROM AUTHOR]

Control measures for foot and mouth disease ( FMD) in Nigeria have not been implemented due to the absence of locally produced vaccines and risk-based analysis resulting from insufficient data on the circulating FMD virus ( FMDV) serotypes/strains. In 2013-2015, blood and epithelial samples were collected from reported FMD outbreaks in four states (Kaduna, Kwara, Plateau and Bauchi) in northern Nigeria. FMDV non-structural protein ( NSP) seroprevalence for the outbreaks was estimated at 80% (72 of 90) and 70% (131 of 188) post-outbreak. Antibodies against FMDV serotypes O, A, SAT1, SAT2 and SAT3 were detected across the states using solid-phase competitive ELISA. FMDV genome was detected in 99% (73 of 74) of the samples from FMD-affected animals using rRT- PCR, and cytopathic effect was found in cell culture by 59% (44 of 74) of these samples. Three FMDV serotypes O, A and SAT2 were isolated and characterized. The phylogenetic assessments of the virus isolates showed that two topotypes of FMDV serotype O, East Africa-3 ( EA-3) and West Africa ( WA) topotypes were circulating, as well as FMDV strains belonging to the Africa genotype (G- IV) of serotype A and FMDV SAT2 topotype VII strains. While the serotype O ( EA-3) strains from Nigeria were most closely related to a 1999 virus strain from Sudan, the WA strain in Nigeria shares genetic relationship with three 1988 viruses in Niger. The FMDV serotype A strains were closely related to a known virus from Cameroon, and the SAT2 strains were most closely related to virus subtypes in Libya. This study provides evidence of co-occurrence of FMDV serotypes and topotypes in West, Central, East and North Africa, and this has implication for control. The findings help filling the knowledge gap of FMDV dynamics in Nigeria and West Africa subregion to support local and regional development of vaccination-based control plans and international risk assessment. Influenza virus infections are responsible for more than 250,000 deaths annually. Influenza virus isolation, propagation, and characterization protocols are critical for completing reproducible basic research studies and for generating vaccine seed stocks. Detailed protocols for the isolation and identification of influenza virus have been recently reported (Eisfeld et al., 2014). However, there are few standardized protocols focused on the propagation and characterization of viral isolates, and as a result, viruses propagated in different conditions in different laboratories often have distinct in vitro and in vivo characteristics. Here, we focus on influenza A virus propagation and characterization in the laboratory taking into consideration the overall quality and composition of the virus stock to achieve consistency in virus yield, virulence, and immunostimulatory activity.

Abstract: The technique of chromatofocusing was applied to the characterization and purification of three bacteriophages that are routinely used for testing virus filters: φX174, PR772, and PP7. Chemically well-defined eluent buffers were used, instead of the more commonly used chromatofocusing polyampholyte buffers. Chromatographic column packings were selected to minimize band broadening by confining bacteriophage adsorption solely to the exterior particle surface. Under the conditions used it was determined that bacteriophages could be made to focus into narrow bands in a retained pH gradient with recoveries of live phage that ranged from 15 to nearly 100% as determined by a plaque-forming assay. Retention times and apparent isoelectric point data were obtained for samples consisting either of purified bacteriophage, or samples consisting of crude preparations of bacteriophages containing host cell impurities. Isoelectric point estimates were obtained using modified, previously described models. The results obtained suggest that chromatofocusing is a simple and rapid method for obtaining approximate isoelectric points for bacteriophages and probably other types of viruses. Starting with the influences of having a father who was an agricultural plant pathologist, I sketch my career through university and research institute from field epidemiology, basic virus characterization to molecular biology. I note what I consider to be the highlights of my scientific career and the events that shaped the development of my thinking. These include secondment to teach in a university in Uganda, a sabbatical year in the University of California, Davis, where I became aware of the emerging DNA technology; studying the molecular biology of Cauliflower mosaic virus, rice tungro viruses, and Banana streak virus with the aim of developing diagnostics and approaches to control of viruses. Bringing these experiences together, I am now involved in facilitating the uptake of the application of biotechnology to crop improvement in developing countries. I conclude with some thoughts on opportunities for young plant pathologists over the next years of rapid change. Abstract: Background: Human rhinoviruses (HRVs) are the most common cause of viral illness worldwide but today, less than half the strains have been sequenced and only a handful examined structurally. This viral super-group, known for decades, has still to face the full force of a molecular biology onslaught. However, newly identified viruses (NIVs) including human metapneumovirus and bocavirus and emergent viruses including SARS-CoV have already been exhaustively scrutinized. The clinical impact of most respiratory NIVs is attributable to one or two major strains but there are 100+ distinct HRVs and, because we have never sought them independently, we must arbitrarily divide the literature''s clinical impact findings among them. Early findings from infection studies and use of inefficient detection methods have shaped the way we think of ‘common cold’ viruses today. Objectives: To review past HRV-related studies in order to put recent HRV discoveries into context. Results: HRV infections result in undue antibiotic prescriptions, sizable healthcare-related expenditure and exacerbation of expiratory wheezing associated with hospital admission. Conclusion: The finding of many divergent and previously unrecognized HRV strains has drawn attention and resources back to the most widespread and frequent infectious agent of humans; providing us the chance to seize the advantage in a decades-long cold war. Abstract: A nucleopolyhedrovirus (SpliMNPV-Az) was isolated from diseased larvae of Spodoptera littoralis, collected at the Island of S. Miguel in Azores. The virulence of this isolate was tested against S. littoralis larvae in laboratory. LD50 against 2nd and 3rd instars were not significantly different, 1.44×104, 3.89×104 OBs per larvae, respectively, but both were significantly different from that against 4th instar, which was 61.3×104 OBs per larvae. The complete codons sequence of SpliMNPV-Az Polh gene obtained was 750bp (NCBI GenBank Accession No. AY600451). This sequence was compared to other 38 polyhedrin genes from NPVs and to 6 granulin genes from GVs and resulted to be identical to the sequence of a SpliMNPV previously published, thus indicating that the natural host of SpliMNPV-Az must be S. littoralis. Genetic distances estimated from restriction enzymes profiles showed SpliMNPV-Az is close to the Egyptian SpliMNPV type B, despite some degree of genetic divergence suggested by slight differences observed on PstI profile. The relationship between inoculum density and mortality or infection was studied for various pepper varieties (Capsicum annuum L.) inoculated with zoospores of two P. capsici isolates. The inoculum concentrations required for 50% mortality (LD 50) varied greatly between pepper varieties and P. capsici isolates: with one isolate, LD 50 was 40 zoospores/ml for a susceptible variety and reached 4,380 to 97,300 zoospores/ml for resistant varieties. For another isolate, LD 50 for the same varieties ranged from 26 to 800 zoospores/ml. Comparisons between LD 50 and inoculum doses required for 50% infection (ID 50) also revealed differenced between varieties but not between isolates. After multiple infection correction, regression slopes of infections/inoculum concentration were low for resistant varieties (0.28 to 0.50) but higher for susceptible varieties (0.72 to 0.94), indicating strong competition between spores for infection of resistant plants, but not for infection of susceptible plants. This analysis provided many criteria which can be used to differentiate susceptible from resistant varieties and to evaluate with precision the resistance level of the different resistance genitors used in our breeding program.. Some unreported characteristics of moroccan pepper virus (MPV) and tombusvirus Neckar (TVN) are given in this paper. Virus particles have a sedimentation coefficient of 140 S (MPV) and 133 S (TVN) and a buoyant density of 1.357 (TVN) and 1.355 (MPV) g/cm3 in CsCl. They are constituted by a single protein species with a molecular weight of 43,000 (MPV) and 45,000 (TVN) which encapsidate a single species of nucleic acid with the apparent size of 4,700 nucleotides. The estimated percentage sequence homology of this genomic RNA with that of the type member of the group was 10% (MPV) and 15% (TVN). A rhabdovirus isolated in Tunisia by mechanical inoculation from honeysuckle {Lonkera sp.) plants with vein yellowing, was compared with a Tunisian isolate of eggplant mottled dwarf virus (EMDV). The two viruses had similar host ranges and caused the same symptoms in artificially infected hosts. The honeysuckle virus induced in eggplant a syndrome indistinguishable from that typical of EMDV. The two viruses could not be differentiated serologically, had particles of the same size and elicited identical cytopathological alterations in naturally and artificially infected hosts. Two giant pandas (Ailuropoda melanoleuca) died of unknown causes in a Chinese zoo. The clinical disease profile suggested that the pandas may have suffered a viral infection. Therefore, a series of detection including virus isolation, electron microscopy, cytobiological assay, serum neutralization and RT-PCR were used to identify the virus. It was determined that the isolated virus was a canine coronavirus (CCV), on the basis of coronavirus, neutralization by canine anti-CCV serum, and 84.3% to 100% amino acid sequence similarity with CCV. Well-differentiated primary airway epithelial cell (AEC) cultures have been widely used for the characterization of several human respiratory viruses including coronaviruses. In recent years, there has been an increase in interest toward animal AEC cultures and their application to characterize veterinary viruses with zoonotic potential, as well as studying host-pathogen interactions in animal reservoir host species. In this chapter, we provide a revised and improved protocol for the isolation and establishment of well-differentiated AEC cultures from diverse mammalian species and the use of the cultures for the characterization of veterinary coronavirus. We also describe immunohistochemistry protocols with validated antibodies for the visualization and identification of viral cell tropism in well-differentiated AEC cultures from human, swine, bovine, and feline origin.

A sap-transmissible virus was isolated from chayote (Sechium edule) in Costa Rica. Infected plants showed chlorotic spots and rings, and blotchy mosaics, which often coalesced to give a complete mosaic and leaf deformation. By electron microscopy, spherical virus-like particles of approximately 29 nm in diameter were visible, and cytological changes associated with the chloroplasts were observed. The virus particles sedimented in sucrose density gradients as two components, a top component of empty protein shells and a bottom component of electron-dense particles. Electrophoretic analysis showed a single-stranded RNA of approximately 5.7 kb and capsid protein (CP) subunits of ∼22 kDa. The virus was identified as a member of the tymovirus group on the basis of particle morphology, size, sedimentation in sucrose gradients, cytopathological effects, and capsid protein and genome properties, and it was tentatively named chayote mosaic virus (ChMV).
