Buffaloes represent an important economic resource for several regions of the world including Romania. In the present study, we examined 104 faecal samples collected from 38 buffalo calves (2-11 weeks old) from household rearing systems in Romania for gastrointestinal parasites. All samples were tested using the saturated salt flotation, McMaster and modified Ziehl-Nielsen staining methods. PCR coupled with sequencing isolates were used to identify assemblages of Giardia lamblia (Kunstler, 1882) and species of Cryptosporidium Tyzzer, 1907. Overall, 33 out of 38 examined buffalo calves were infected with different gastrointestinal parasites: 16 had single infections and 17 had mixed infections with two or three parasites. Species of Eimeria Schneider, 1875 (32/38; 84%) were the most prevalent parasites; eight species were identified according to the oocyst morphology, including the pathogenic E. bareillyi (Gill, Chhabra et Lall, 1963) which was detected for the first time in buffaloes from Romania. The nematodes Toxocara vitulorum (Goeze, 1782) (11/38; 37%) and Strongyloides papillosus (Wedl, 1856) (6/38; 16%) were also detected. Cryptosporidium spp. were found in four (11%) buffalo calves; two of them were molecularly identified as C. ryanae Fayer, Santin et Trout, 2008, and another one clustered in the same clade with C. ryanae, C. bovis Fayer, Santin et Xiao, 2005, and C. xiaoi Fayer et Santin, 2009. Giardia duodenalis assemblage E was also molecularly detected in a single (2.6%) buffalo calf. The presence of other buffaloes in the same barn was identified as a risk factor for infection with T. vitulorum. Our results indicate extensive parasitic infections in buffalo calves from northwestern Romania and underline the necessity of prophylactic treatments for T. vitulorum and E. bareillyi.

Background During the last decades, filarial infections caused by Dirofilaria spp. have spread rapidly within dog populations of several European countries. Increasing scientific interest in filariasis, and the availability of new diagnostic tools, has led to improved knowledge of the biology, morphology, and epidemiology of different species of filarial worms. However, data are still scarce for a number of countries, including the Republic of Moldova. Thus, we assessed the epidemiological status of canine filariasis in the Republic of Moldova to address part of this knowledge gap. Methods A total of 120 blood samples were collected between June 2018 and July 2019 from dogs originating from the cities of Cahul and Chişinău. The samples were examined microscopically, and multiplex polymerase chain reaction was performed to evaluate filarioid species diversity. Results Microscopic examination revealed that 12 dogs (10.0%) were positive for circulating microfilariae. The molecular test showed that one dog was positive for Acanthocheilonema reconditum (0.8%), one for Dirofilariaimmitis (0.8%), six for Dirofilariarepens (5.0%), and four (3.3%) harboured a co-infection with D. immitis and D. repens. Prevalence was significantly higher in dogs aged ≥ 2 years. Conclusions The epidemiological survey presented here for the Republic of Moldova confirmed the presence D. immitis, D. repens and A. reconditum in dogs that had not received any heartworm preventive. Background Coccidiosis represents a serious threat to the poultry industry, affecting production and causing high morbidity, mortality and significant costs resulting from treatment and prophylaxis. In-feed anticoccidials have been used for decades for managing avian coccidiosis and were very effective until drug resistance emerged. The use of natural remedies has become a promising alternative in combating coccidiosis in chickens. Therefore, the purpose of the present study was to assess the efficiency of a commercial herbal formula (H), as oral liquid preparations, in experimental chicken coccidiosis. Methods Two independent controlled battery experiments (BE1 and BE2) were designed and the product was tested in 3 different formulas (H1, H2 and H3): H1 contained a propylene glycol extract of Allium sativum and Thymus serpyllum; H2 contained Origanum vulgare, Satureja hortensis and Chelidonium majus; and H3 contained Allium sativum, Urtica dioica, Inula helenium, Glycyrrhiza glabra, Rosmarinus officinalis, Chelidonium majus, Thymus serpyllum, Tanacetum vulgare and Coriandrum sativum. Chickens were divided into five groups for each BE as follows: (i) uninfected untreated control (UU1, UU2); (ii) infected untreated control (IU1, IU2); (iii) infected treated with amprolium (ITA1, ITA2); and (iv, v) two experimental groups infected treated with H1 (ITH1) and H2 (ITH2) formulas in the BE1 and with H3 (ITH3-5 and ITH3-10) formula in the BE2. The chickens from infected groups were challenged with 5000 (BE1) and 50,000 (BE2) sporulated oocysts of Eimeria spp. (E. acervulina, E. tenella and E. maxima), respectively. The anticoccidial efficacy was assessed by recording the following: oocysts output (OPG), lesion score (LS), weight gain (WG), feed conversion ratio (FCR) and anticoccidial index (ACI). Additionally, polyphenolics and flavonoids (caffeic-chlorogenic acid, apigenin, kaempferol, luteolin, quercitin, quercitrin) from herb extracts found in H3 formula were determined by the liquid chromatography-tandem mass spectrometry (LC-MS/MS) method. Results H1 and H2 reduced the WG, and increased the FCR and OPG compared with controls. H1 reduced the duodenal lesions, whilst H2 reduced the caecal lesions, compared with control. H3 decreased the OPG of Eimeria spp., reduced the total lesion score and improved the zootechnical performance (weight gain and feed conversion ratio). According to ACI value, H1 and H2 had no efficacy on Eimeria spp. infection, but H3 had good to marked anticoccidial effect, the ACI being slightly greater in the group ITH3-5. According to the results of LC-MS/MS, the concentration of polyphenols in H3 formula was the highest, the sum of chlorogenic acid and caffeic acid being 914.9 µg/ml. Conclusions H3 formula is a promising natural anticoccidial and field trials are recommended in order to validate the obtained data. Neospora caninum is a heteroxenous parasitic Apicomplexan protozoan, able to infect a variety of domestic and wild animals, mainly associated with reproductive disorders and abortions in susceptible species. Considering the importance of Romania in the European buffalo industry and the severe economic losses caused by N. caninum infection in livestock, the aim of the present study was to assess the occurrence of neosporosis in buffaloes raised in household and farming systems in northwestern Romania. Overall, 197 serum samples were tested for the presence of N. caninum-specific antibodies by ELISA. Additionally, from 74 slaughtered buffaloes, diaphragm digest, heart, and lymph node samples were tested for the presence of N. caninum DNA. The overall seroprevalence was 68.5%. The seroprevalence was significantly higher in adult animals compared to calves and young ones and in the household system compared to farm. By nPCR, six samples were found to be positive for N. caninum DNA, of which three were serologically negative. The present study reveals a high prevalence of N. caninum-specific antibodies in buffaloes in northwestern Romania and to our best knowledge, it is the first one performed in Romania.

Background: During the last decades, the spread of the infection with filarial nematodes Dirofilaria spp. in dog’s population in various European countries has been dynamic and rapid. The increased interest of the scientific community as well as the availability of new diagnosis tools, allowed a better knowledge on the biology, morphology and epidemiology of different species of filarial worms. However, in some countries, data are still scarce on this topic. To address some knowledge gaps, we assessed the epidemiological status of different canine filariasis in the Republic of Moldova. Methods: A total of 120 blood samples were collected between June 2018 and July 2019 from dogs originating from Cahul and Chișinău cities. The samples were microscopically examinated and multiplex PCR was performed to evaluate filarioid species diversity. Molecular test showed that 1 dog was positive for Acanthocheilonema reconditum (0.83%), 1 for D. immitis (0.83%), 6 for D. repens (5%), and 4 (3.33%) harboured a coinfection with D. immitis and D. repens. Conclusions: This study is the first epidemiological survey of filaroid worms in dogs from the Republic of Moldova and the first to show the presence of D. repens and A. reconditum in this species.

Toxoplasma gondii is a widespread zoonotic protozoan that infects most species of mammals and birds, including poultry. This study aimed to investigate the course of T. gondii infection and the efficacy of diclazuril and Artemisia annua in preventing infection in experimentally infected chickens. Seventy-five 1-month-old chickens, female and male, were randomly divided into five groups (n = 15 each) as follows: (1) uninfected untreated (negative control, NC); (2) infected with T. gondii genotype II/III isolated from a wild cat (group WC); (3) infected with T. gondii genotype II isolated from a domestic cat (group DC); (4) infected with T. gondii domestic cat strain and treated with the anticoccidial diclazuril (group DC-D); and (5) infected with T. gondii domestic cat strain and treated with the medicinal plant Artemisia annua (group DC-A). Clinical signs, body temperature, mortality rate, weight gain, feed conversion ratio, hematological parameters, and the presence of T. gondii–specific IgY antibodies were recorded in all groups. Five chickens per group were euthanized 28 days post-infection (p.i.) and their brains, hearts, and breast muscle tested for T. gondii by mouse bioassay and polymerase chain reaction (PCR). No clinical signs related to the experimental infection were observed throughout the study period. T. gondii–specific antibodies were detected by day 28 p.i., but not in all infected chickens. Overall, T. gondii DNA was detected (bioassay or tissue digests) in all infected and untreated chickens (10/10), while viable parasite (bioassay) was isolated from 7 out of 10 chickens. The parasite was most frequently identified in the brain (7/10). There were no differences in the T. gondii strains regarding clinical infection and the rate of T. gondii detection in tissues. However, higher antibody titers were obtained in chickens infected with T. gondii WC strain (1:192) comparing with T. gondii DC strain (1:48). A. annua reduced replication of the parasite in 3 out of 5 chickens, while diclazuril did not. In conclusion, broiler chickens were resistant to clinical toxoplasmosis, irrespective of the strain (domestic or wild cat strain). The herb A. annua presented prophylactic efficacy by reduced parasite replication. However, further studies are required aiming at the efficacy of diclazuril and A. annua for the prevention of T. gondii infection in chickens using quantitative analysis methods.

Giardia duodenalis assemblage E was also molecularly detected in a single (2.6%) buffalo calf. The presence of other buffaloes in the same barn was identified as a risk factor for infection with T. vitulorum. Our results indicate extensive parasitic infections in buffalo calves from Northwestern Romania and underline the necessity of prophylactic treatments for T. vitulorum and E. bareillyi.

Background: Lungworms are recognized as important agents in the pathology of the respiratory system in domestic cats. While Aelurostrongylus abstrusus is worldwide known and studied, Troglostrongylus brevior has gained the attention of the scientific community only in the last decade. The pathogenicity of this species seems to be higher than A. abstrusus, causing more severe clinical presentations and being potentially fatal, especially in young animals. Methods: In this study, 371 DNA isolates of faecal samples were tested by multiplex polymerase chain reaction for the presence of A. abstrusus, T. brevior, and Angiostrongylus chabaudi. Results: The results showed that 30.2% and 6.7% of the investigated domestic cats were positive for A. abstrusus and T. brevior respectively, stressing out the importance of these parasites as agents of respiratory conditions in domestic cats from Romania. None of the samples were positive for A. chabaudi. The age, the outdoor access, and the lack of deworming were identified as significant risk factors for infection with A. abstrusus. Conclusions: This paper represents the first report of T. brevior in domestic cats from Romania. Moreover, it presents the most northern localization in Europe of T. brevior.

Thelazia callipaeda has been regarded in Europe in the last few decades as a "new" agent of an ocular disease in dogs, cats, several wild species, and not least in humans. After the first European description in Italy, in 1989, the parasite has been reported in many other countries, showing a widespread distribution of this parasite. The first case of canine thelaziosis in Romania has been reported in 2015, and it has been followed by other studies confirming the presence of the nematode in dogs or broadening the host spectrum with some wild carnivore species that have been found positive. Despite being considered suitable primary hosts for T. callipaeda, infection with T. callipaeda in domestic cat is rarely reported in Europe, probably due to some behavioral and physical particularities of this host. This study describes the first autochthonous report of T. callipaeda infection in a domestic cat from Romania. Additionally, to these findings, new geographical records of canine thelaziosis are being revealed, confirming the increased risk of this emerging vector-borne zoonosis.

In the present study, we aimed to assess the toxicity of artemisinin on the haematological system and its effect on the productive performance of broiler chickens. Eighteen-day-old chickens were randomly divided into four groups of 30 chickens (three replicates of 10 broilers/group): control group and three experimental groups: ART5 - diet with 5 ppm of artemisinin; ART50 - diet with 50 ppm of artemisinin; and ART500 - diet with 500 ppm of artemisinin. Artemisinin enhanced the productive performances of broiler chickens at the lowest concentration (5 ppm), but at the highest concentration (500 ppm), it negatively affected weight gain and the feed conversion ratio. The performance characteristics of the chickens whose diets were supplemented with 50 ppm artemisinin were similar to those of the control group. Additionally, 5 ppm artemisinin in feed did not significantly affect the haematological parameters of the chickens, but 50 and 500 ppm artemisinin induced a gradual decline of the total leukocytes, lymphopenia, monocytosis, and eosinopenia, and the highest concentration caused anaemia. Artemisinin at a low concentration could be used as a feed additive in the poultry industry to improve organic broiler production performance with no serious side effects.

Toxocara canis (Werner, 1782) is a zoonotic nematode commonly parasitizing dogs worldwide with great public health importance as the aetiological agent of human toxocariasis. In this respect, the aim of this study was to evaluate the effect of six disinfectant products commonly used in kennels, veterinary clinics and as household cleaning products on the embryogenesis and viability of T. canis eggs. The composition of active ingredients in these commercial disinfectants was sodium hypochlorite (A); a mix of N-(3-aminopropyl)-N-dodecylpropan-1.3-diamine and didecyldimethylammonium chloride (B); sodium dichloroisocyanurate dehydrate (C); a mix of glutaraldehyde, quaternary ammonium compounds, benzyl-c12-18-alkyldimethyl and chlorides (D); a mix of 2-propanol, ethanol, benzalkonium chloride and glucoprotamin (E); a mix of pentapotassium bis (peroxymonosulphate) bis (sulphate), sodium C10-13-alkylbenzenesulphonate, malic acid, sulphamidic acid, sodium toluenesulphonate, dipotassium peroxodisulphate and dipentene (F). After dilution, the tested disinfectants had the maximal concentration recommended by the manufacturer in order to achieve a biocidal effect. Each product was tested on approximately 10,000 T. canis eggs, having five different contact times (5, 10, 15, 30, 60 min). Three replicates were tested for each diluted disinfectant and for each contact time. After the treatment, eggs were washed and incubated in distilled water at 27 °C for 2 weeks. None of the tested products had a significant inhibitory effect on the embryogenesis and viability of T. canis eggs, regardless of the contact time. Moreover, after 2 weeks, in all tested samples, eggs containing motile infective larvae were identified, showing that routinely used disinfectants do not eliminate risk of infection by T. canis.

Gastrointestinal parasites are involved in the health and wellbeing of cats and some of them, due to their zoonotic character represent a problem for the public health. This study aimed to assess the prevalence of parasitic infections in cats with digestive symptoms. Faecal samples collected from 137 cats were analysed by the flotation method using sodium chloride saturated solution. The overall prevalence of gastrointestinal (GI) parasites was 56.6%. GI parasites were significantly (p=0.005) more common in cats with digestive symptoms (69.8%) than in clinically healthy ones (23.0%). Toxocara cati (40.2%) was the most frequently identified GI parasite, followed by Cystoisospora spp. (10.2%), Ancylostoma tubaeforme (3.7%), Taenia spp. (2.2), Giardia duodenalis (2.2%), and Toxoplasma gondii-like oocysts (0.7%). T. cati (53.3%, pCystoisospora spp. (15.6%, pT. cati infection by logistic regression analysis. Regardless of the age, symptoms like vomiting, diarrhoea, and inappetence were positively associated with T. cati.

International audience; The purpose of our study was to evaluate the prevalence of Toxoplasma gondii infection in autochthonous Carpathian buffaloes from northwestern Romania by serology, PCR techniques, and mouse bioassay. Agreement between MAT and ELISA, correlation between indirect and direct detection methods, and risk factors were evaluated. The apparent overall seroprevalence of T. gondii was 8.1% by MAT and 6.6% by ELISA. The agreement between ELISA and MAT was fair. The apparent seroprevalence was significantly higher in adult buffaloes (12.5%) compared to calves (0.0%) and juveniles (1.9%) by MAT. Most of the positive adult buffaloes detected by MAT had antibodies at a low sera dilution and the highest dilution was 1:768 in a juvenile female (30 months). No viable T. gondii was detected by mouse bioassay, as no T. gondii cyst or DNA was found in the brain of mice and they did not seroconvert. However, T. gondii DNA was detected in two buffaloes: in a 30-month-old male buffalo by qPCR on the diaphragm digest and in a 252-month-old female buffalo by RE nPCR on the mesenteric lymph node. Both animals were negative in MAT and ELISA. The total prevalence of T. gondii by direct detection methods was 2.7%. There was no correlation between indirect and direct detection methods. Since no viable T. gondii was detected in buffaloes, the risk of human infection from buffalo meat is minimal. Buffaloes' biological response to a T. gondii infection appears to be very similar to the response of cattle.

Four experiments were conceived in order to test the efficacy of artemisinin, a sesquiterpene lactone derived from Artemisia annua, in single experimental infection of broiler chickens with Eimeria acervulina (1 × 10(5) oocysts), Eimeria maxima (5 × 10(4) oocysts) or Eimeria tenella (1 × 10(4) oocysts), and mixed infection with all 3 species (3.2 × 10(4) Eimeria spp. oocysts). For each experiment, three different dosages of artemisinin (5, 50 and 500 ppm) were compared with a negative control (uninfected, unmedicated), a positive control (infected, unmedicated) and a classical anticoccidial (monensin). The weight gain (WG), feed conversion ratio (FCR), oocysts shedded per gram of feces (OPG), lesion score, oocysts sporulation rates and mortality rate were recorded in all groups. The dosage of 5 ppm of artemisinin improved the WG and FCR for the chickens infected with E. acervulina. The OPG was significantly decreased in all the groups medicated with artemisinin and challenged with a mixed infection (p ≤ 0.01). The lesion score of the chickens challenged with Eimeria was reduced by different concentrations of artemisinin, depending on the species involved, but this compound did not have a positive effect on the lesions caused by E. acervulina. Histopathological analysis revealed superficial erosions of the intestinal mucosa, mixt. mononuclear and heterophilic inflammatory infiltrate in the lamina propria and intralesional presence of various developmental stages of parasite in groups infected with Eimeria spp.The sporulation rate of E. acervulina and E. maxima oocysts was significantly affected by 500 ppm of artemisinin, whilst the dosage of 5 ppm affected the sporulation of E. tenella oocysts. These data suggest that artemisinin is not effective against single eimerian infections but could be used as an alternative in mixed coccidiosis, especially if its effect on the oocysts sporulation would be fully investigated.

During six years (April 2010–April 2016) we examined individual feces samples collected from 1314 dogs located in Center and Northwest Romania (Transylvania). Stool samples were analyzed by saturated salt flotation, sedimentation technique and modified Ziehl-Nielsen staining method. The overall prevalence of endoparasitic infections was 66.6% (n = 875). Sixteen species/genera of endoparasites were identified. The most prevalent species were Ancylostoma caninum/Uncinaria stenocephala (33.0%) ( p = 0.0001) followed by Trichocephalus vulpis (25.0%). Mixed infections, were significantly more frequent ( p = 0.0001) than single species infections. The age and the living condition/service of dogs were identified as the main risk factors for infection with endoparasites.

Canine dirofilarioses are mosquito-borne zoonotic diseases with a continuous expansion of their geographical distribution, as a consequence of different climatic and ecological factors. Dirofilaria immitis, the aetiological agent of heartworm disease, has gained a major veterinary interest, mainly due to its severe clinical implication. In the last decades, D. repens, despite of being regarded as a less pathogenic species, regained attention due to its recognized zoonotic potential. Romania has been traditionally regarded as a non-endemic country, but recent epidemiological surveys are highlighting the presence of both D. repens and D. immitis. The present case report describes the clinical and pathological features of a natural co-infection with D. repens and D. immitis in 5year old male Boxer from north-western Romania. Based on clinical and laboratory investigations, a diagnosis of kidney failure and chronical cystitis was established. The parasites were identified by morphology and confirmed by PCR and sequencing. To the best of our knowledge this is the first worldwide report of the ectopic presence of D. repens adults in the pelvic cavity and mesentery by natural infection and first report of the ocular localization of this parasite for Romania. The clinical and paraclinical findings suggest the implication of D. repens microfilariae in the aetiology of histopathological lesions. Further investigations are needed in order to establish the actual pathogenic potential of D. repens, a frequently neglected parasite.

The paper reports the prevalence and geographical distribution of Borrelia burgdorferi sensu lato (s.l.) and its genospecies in 12,221 questing Ixodes ricinus ticks collected at 183 locations from all the 41 counties of Romania. The unfed ticks were examined for the presence of B. burgdorferi s.l. by PCR targeting the intergenic spacer 5S-23S. Reverse line blot hybridization (RLB) and restriction fragment length polymorphism (RFLP) analysis were performed for identification of B. burgdorferi genospecies. The overall prevalence of infection was 1.4%, with an average local prevalence between 0.75% and 18.8%. B. burgdorferi s.l. was found in ticks of 55 of the 183 localities. The overall prevalence B. burgdorferi s.l. in ticks in the infected localities was 3.8%. The total infection prevalence was higher in female ticks than in other developmental stages. Three Borrelia genospecies were detected. The most widely distributed genospecies was B. afzelii, followed by B. garinii and B. burgdorferi sensu stricto (s.s.). The study is the first countrywide study and the first report of B. burgdorferi s.s. in Romania. The distribution maps show that higher prevalences were recorded in hilly areas, but Lyme borreliosis spirochetes were also present in forested lowlands, albeit with a lower prevalence.

With the exception of Aelurostrongylus abstrusus, feline lungworms have been poorly studied. Information on their distribution is patchy and mostly limited to case reports. In this study, the occurrence of feline lungworms and co-infecting gastrointestinal parasites has been investigated in 12 European countries (i.e. Austria, Belgium, Bulgaria, France, Greece, Hungary, Italy, Portugal, Romania, Spain, Switzerland and the United Kingdom). An average of 10 domestic cats, with regular outdoor access, was sampled each month for 12 months, and freshly passed faeces were collected. Stools were processed using a McMaster assay and a quantitative Baermann-Wetzel method. Animals positive for lungworms and/or gastrointestinal parasites were treated with a formulation containing fipronil, (S)-methoprene, eprinomectin, and praziquantel (Broadline®, Merial), and re-sampled 28 days post-treatment. The association between lungworm infection and risk factors was analysed using statistical medians/means and the efficacy of the treatment against each lungworm species was assessed. Of 1990 cats sampled, 613 (30.8%) were positive for at least one parasite, while 210 (10.6%) were infected by lungworms. The prevalence of lungworm infection varied between the sampled sites, with the highest recorded in Bulgaria (35.8%) and the lowest in Switzerland (0.8%). None of the cats from Austria or the United Kingdom were infected by lungworms. Aelurostrongylus abstrusus was the species most frequently detected (78.1%), followed by Troglostrongylus brevior (19.5%), Eucoleus aerophilus (14.8%) and Oslerus rostratus (3.8%). The overall efficacy of the treatment was 99% for A. abstrusus and 100% for T. brevior, O. rostratus and E. aerophilus. Data presented provide a comprehensive account of the diagnosis, epidemiology and treatment of feline lungworms in Europe, as well as of the occurrence of co-infections by gastrointestinal parasites.

The aim of this study was to evaluate the seroprevalence of Neospora caninum infection in dogs from Romania at a national level and to detect possible correlations between the value of seroprevalence and a series of factors like sex, age, breed category and area. The prevalence of anti-N. caninum antibodies was determined in sera samples from 1114 dogs located in different regions of Romania using the IFAT method. Antibodies to N. caninum were found in 364 dogs (32.7%). The titers of anti-N. caninum antibodies varied from 1:50 to 1:800.

In 2010 and 2011, questing ticks were collected from 188 forested locations in all the 41 counties of Romania using the dragging method. The total of 13,771 ticks collected belonged to eleven species: Ixodes ricinus (86.9 %), Dermacentor marginatus (9.5 %), Haemaphysalis punctata (2.6 %), H. concinna (0.6 %), H. sulcata (0.3 %), H. parva (0.1 %), Hyalomma marginatum (0.02 %), D. reticulatus (0.02 %), I. crenulatus (0.007 %), I. hexagonus (0.007 %) and I. laguri (0.007 %). Ixodes ricinus was present in 97.7 % (n = 180) of locations, occurring exclusively in 41.7 % of the locations, whereas it was the dominant species in 38.8 % of the other locations, accounting for over 70 % of the total tick community. The following most common questing ticks were D. marginatus, H. punctata and H. concinna. Ixodes ricinus co-occurred with one, two or three sympatric species. The occurrence of D. reticulatus in forested habitats from Romania was found to be accidental.

Little information is available about the seroprevalence of Toxoplasma gondii and Neospora caninum infections in goats in Romania and even in Europe. During 2007-2010, 735 serum samples were collected from dairy goats located in 4 historical regions (Crişana, Maramureş, Transylvania and Muntenia) of Romania. Sera were analyzed for T. gondii and N. caninum antibodies (IgG type) by enzyme-linked immunosorbent assay (ELISA) using two commercial kits (Chekit Toxotest Antibody ELISA and Chekit Neospora caninum Antibody ELISA; Idexx-Bommeli, Switzerland). Three hundred and eighty-eight out of 735 (52.8%) goats presented T. gondii antibodies and 12 out of 512 (2.3%) goats had N. caninum antibodies. The high seroprevalence of T. gondii suggests that infection with this parasite is common in dairy goats in Romania, and less common the infection with N. caninum. This is the first time that infection with N. caninum in goats has been reported in Romania and the first extended study on seroepidemiology of T. gondii.

Felines are the key species in the epidemiology of Toxoplasma gondii infection, as they are the definitive host of the parasite and are the only species that can shed resistant oocysts in the environment. Different assays are in use for the detection of antibodies against T. gondii in cats. However, assay validation studies are limited. For that reason it was our aim to first evaluate 6 serological tests (one commercial and 2 in-house ELISAs, ImmunoComb, IFAT and MAT) for antibodies (IgG) against T. gondii in cats by Bayesian modeling. Factors associated with seropositivity were evaluated by bivariable and multivariable methods. The test evaluation indicated the commercial ELISA had the highest Youden Index. The estimated sensitivity ranged between 95.7% and 97.1% and the specificity between 97.3% and 97.6%. Using this commercial ELISA 111 out of 236 cats (47%) were positive for T. gondii antibodies. Two peaks in the percentage of strong positive samples (S/P≥200) were observed, around 10-months-old and 8-years-old. In bivariable analysis the seroprevalence was significantly higher in adult cats, cats with mixed diet, with outdoor access, in cats from a rural area and in cats from centre and north-western Romania. Adult age (adults: OR 6.98; 95% CI: 2.02-24.14 and geriatrics (cats older than 10-years): OR 12.01; 95% CI: 1.60-90.15) and outdoor access (OR 6.38; 95% CI: 2.32-17.53) remained significant risk factors in the multivariable logistic regression analysis. Our results suggest that T. gondii infection is common in household cats in Romania, and especially in those with outdoor access.

Ticks and tick-borne diseases represent a great concern worldwide. Despite this, in Romania the studies regarding this subject has just started, and the interest of medical personnel, researchers, and citizens is increasing. Because the geographical range of many tick-borne diseases started to extend as consequences of different biological and environmental factors, it is important to study the diversity of ticks species, especially correlated with host associations. A total number of 840 ticks were collected between 1 April and 1 November 2010, from 66 animals, from 17 species in 11 counties, spread all over Romania. Four Ixodidae species were identified: Dermacentor marginatus (49.2%), Ixodes ricinus (48.3%), Hyalomma marginatum (2.4%), and Rhipicephalus sanguineus (0.1%). The obtained results indicate that D. marginatus is the most abundant tick species and I. ricinus is the most prevalent. As both of them are important vectors for human and animal diseases, the present paper discusses the associated risks for tick-borne diseases.

Few studies were conducted to investigate the pathogenesis and to characterize the histopathological alterations in the intestinal mucosa of kids with cryptosporidiosis. In our study, 58 kids were presented for necropsy, and the faeces of another 83 goat kids were studied. Animals were aged between 1 day and 3 weeks. Infection was confirmed by oocyst identification in the faeces (Ziehl-Neelsen stain modified by Henriksen). Gross examination was followed by examination of the tissue using histopathological procedures (Hematoxylin-Eosin stain), scanning electron microscopy (SEM) and immunofluorescence. Pathological changes occurred mainly in the ileum, but also jejunum and caecum of the infected animals, at the apical surface of intestinal epithelial cells. Oocysts appeared as round or oval formations, with a diameter 4.5 m, sometimes covering the atrophic and denudated free border of intestinal villi. Most histological and morphological characteristics of infection with C. parvum are common on the whole to other Cryptosporidium spp. Massive dentification of cryptospodiosis in kids indicates the possible role of these animals as disease reservoirs for other animals and for humans. This is the first study to characterize the Cryptosporidium infection by confocal technique and SEM in Romania.

There is no current information regarding the prevalence of parasites in Romanian cats. Consequently, we conducted a study to evaluate the prevalence of endoparasites in a sample of household cats in the Transylvania region, to point out the risk factors for parasite infection and to evaluate the knowledge of cat owners about the zoonotic potential of some parasites. Four hundred fourteen faecal samples were collected and examined by sodium chloride flotation followed by microscopy. Also, questionnaires were administered to cat owners (196). The overall prevalence of endoparasites in household cats was 34.3% (142/414; CI 29.8-39.1). Concurrent infections with two or more parasites were recorded in 17.6% cats. The detected parasites were Toxocara cati (20.3%), Ancylostoma spp. (10.1%), Isospora rivolta (8.9%), Isospora felis (5.3%), Aelurostrongylus abstrusus (5.6%), Strongyloides spp. (3.4%), Capillaria aerophila (3.1%), Taenia taeniaeformis (2.7%), T. gondii/H. hammondi (1.2%), Sarcocystis spp. (1%), Giardia duodenalis (0.7%) and Dypillidium caninum (0.2%). Risk factors for infection with parasites in cats were identified to be age and medium (rural or urban area). Thus, I. felis, I. rivolta and T. cati were more common in cats less than/equal to 1-year old, and Ancylostoma spp. and A. abstrusus were more prevalent in cats older than 1-year of age. 72.4% of the owners applied anthelmintic treatments to their cats, more commonly in urban areas (87.3%) (4 treatments/year) than in rural areas (12.7%) (1 treatment/year). 66.3% of the owners knew about the zoonotic potential of some parasites from cats, and the main source of information was veterinarians (65.4%). In conclusion the prevalence of endoparasites in household cats from Transylvania is high. Consequently, consideration should be given to the use of anthelmintics and to pet owner education.

The first autochthonous Dirofilaria repens infection confirmed by molecular techniques in humans from North-Western Romania will be presented. Because subcutaneous human dirofilariasis may go unnoticed due to lack of awareness, medical profession in the region should be informed about the risk of this disease even in patients with no travel history

During the last decades, Dirofilaria spp. infection in European dogs has rapidly spread from historically endemic areas towards eastern and northeastern countries, but little or no information is available from these geographical regions. The present study provides a picture of filarial infections in dogs from Romania and compares two tests for the diagnosis of Dirofilaria immitis. From July 2010 to March 2011, blood samples were collected from 390 dogs from nine counties of Romania and serological SNAP tests were performed for the detection of D. immitis antigen. The remaining blood clots were subsequently used for DNA extraction followed by multiplex PCR for assessing filarioid species diversity (i.e. D. immitis, Dirofilaria repens and Acanthocheilonema reconditum). Based on molecular detection, an overall prevalence of 6.92 % (n = 27; 95 % confidence interval (CI) 4.70-10.03 %) for D. repens, 6.15 % (n = 24; 95 % CI 4.07-9.14 %) for D. immitis and 2.05 % (n = 8; 95 % CI 0.96-4.16 %) for A. reconditum was recorded, with significant variations according to sampling areas. Coinfections of D. immitis and D. repens were recorded in 23.91 % (n = 11) positive dogs. A slightly higher prevalence for D. immitis was detected at the SNAP test (n = 28, 7.17 %; 95 % CI 4.91-10.33 %), but this difference was not statistically significant (p = 0.66). However, only 53.57 % (n = 15) of antigen-positive dogs were confirmed by PCR, while other dogs (n = 9) PCR positive for D. immitis were negative at the serology. The present study shows that Dirofilaria species are endemic in the southern and southeastern areas of Romania, This article also provides, for the first time, an epidemiological picture of the distribution of A. reconditum in Romania.

Canine leishmaniasis is a vector-borne zoonotic disease caused by the protozoan parasite Leishmania infantum. In Romania between 1955 and 2013, no cases of human autochthonous visceral leishmaniasis were reported. Data regarding canine leishmaniasis is similarly scarce. Since the first report of clinical autochthonous canine leishmaniasis in 1935, there were only three sporadic reports of positive dogs all without any clinical signs. Our study reports the first clinical case of autochthonous canine leishmaniasis in the last 80 years, stressing the importance of a targeted surveillance of Leishmania infection, as infected dogs act as the primary reservoir for zoonotic visceral leishmaniasis.

Borrelia burgdorferi sensu lato and Anaplasma phagocytophilum are two tick-borne pathogens of medical and/or veterinary importance which are distributed worldwide. Erinaceus roumanicus , the Northern white-breasted hedgehog, is a common synanthropic species that is known to carry not only the hedgehog tick, Ixodes hexagonus , but also I. ricinus , the most common European tick species. I. ricinus is the main vector of both mentioned pathogens. Within this framework and because only limited information is available on the role of E. roumanicus in the ecology of B. burgdorferi s.l. and Anaplasma phagocytophilum in Europe, we carried out an epidemiological surveillance on this species in Romania. From the 57 examined hedgehogs collected in 12 different counties, 24 presented tick infestation. Most ticks ( n = 959) were morphologically identified as larvae, nymphs, or adults of I. ricinus . The prevalence of B. burgdorferi s.l. was 0.4%, and that of A. phagocytophilum 12%. In all positive cases for B. burgdorferi s.l., restriction fragment length polymorphism revealed the genospecies B. afzelii . In Romania, only limited information is available on the epidemiology of B. burgdorferi s.l. and A. phagocytophilum . As hedgehogs commonly share the same environment with humans and other potential reservoir hosts for tick-borne pathogens, our study provides new epidemiological data of public health importance.

Tick-borne diseases are of great concern worldwide. Despite this, in Romania there is only limited information regarding the prevalence of vector-borne pathogens in dogs. In all, 1146 serum samples were tested by SNAP(®) 4Dx(®) (IDEXX Laboratories, Inc., Westbrook, ME) for Anaplasma phagocytophilum, Borrelia burgdorferi, and Ehrlichia canis antibodies, and for Dirofilaria immitis antigen. The correlation between positive cases and their geographic distribution, as well as potential risk factors (age, sex, breed, type of dog, habitat, and prophylactic treatments) were evaluated. Overall, 129 dogs (11.3%) were serologically-positive to one or more of the tested pathogens. The seroprevalence for the four infectious agents were: A. phagocytophilum 5.5% (63/1146), D. immitis 3.3% (38/1146), E. canis 2.1% (24/1146), and B. burgdorferi 0.5% (6/1146). Co-infection with E. canis and A. phagocytophilum was registered in 2 dogs (0.2%). The geographical distribution of the seropositive cases suggests clustered foci in southern regions and in the western part of the country for D. immitis, and in the southeastern region (Constanţa County) for E. canis. A. phagocytophilum and B. burgdorferi showed a homogenous distribution, with a tendency for Lyme-positive samples to concentrate in central Romania. For D. immitis, A. phagocytophilum, and E. canis, administering prophylactic treatments was a risk factor associated with infection. Another associated risk factor was the type of dog (stray dogs were at risk being positive for D. immitis, shelter dogs for E. canis, and hunting dogs for B. burgdorferi). The prevalence of D. immitis was significantly higher in males and in dogs older than 2 years. This survey represents the first data detailing A. phagocytophilum and E. canis seroprevalence in Romanian dogs, and the most comprehensive epidemiological study on vector-borne infections in dogs from this country.

A wide range of swine husbandry practices prevail in Romania: pork for human consumption is derived from hunting wild boar, from household rearing of small numbers of backyard pigs, and from indoor, industrial production of swine raised in confinement indoors. Romania thus represents an instructive place for evaluating the influence of animal management on the exposure risk of the zoonotic parasite, Toxoplasma gondii. The fact that many Romanians eat uncooked or undercooked pork, especially when raised for household consumption, elevates the public health imperative to understand these risks. The aim of the study, therefore, was to evaluate the seroprevalence of T. gondii in pigs and wild boars from Romania. During 2008-2010, we collected 3,595 serum samples from pigs (fattening pigs, sows, backyard pigs) and 150 serum samples from wild boars. The sera were assayed by immunofluorescence antibody test (cutoff, 1:32) for antibodies against T. gondii. The overall seroprevalence of T. gondii infection was 23.1 % (829/3,595) in pigs and 16 % (24/150) in wild boars. The seroprevalence was significantly higher (p < 0.001) in backyard pigs (30.5 %; 783/2,564) than in sows (12.4 %; 46/371) or fattening pigs (none of the sera was positive). The management system (indoor pigs versus backyard pigs) represented the most important factor in the epidemiology of T. gondii infection. The proximity of backyard pigs to the definitive host of this parasite (cats), as well as their access to contaminated meat products, elevated their exposure risk well above that of pigs raised in confinement, and even above that of wild boars inhabiting sylvatic environments.

The current paper is a synoptic review of the distribution and host associations of the 25 species of hard tick fauna (family Ixodidae) in Romania. In addition to a full literature survey, original data is presented, based on eight years of occasional or targeted sample collection. The literature data on geographical distribution was transposed digitally to the decimal degree coordinate system. For each species, an updated distribution map is given together with all historical data and new host associations. Overall, our paper records 58 new tick-host associations for Romania: 20 for Ixodes ricinus, 1 for I. apronophorus, 6 for I. arboricola, 2 for I. hexagonus, 9 for I. redikorzevi, 1 for I. trianguliceps, 2 for I. vespertilionis, 2 for Haemaphysalis punctata, 1 for H. sulcata, 2 for H. concinna, 1 for D. marginatus, 4 for Rhipicephalus sanguineus sensu lato, 1 for R. bursa and 6 for Hyalomma marginatum.
