7. Dose-dependent activation of distinct hypertrophic pathways by serotonin in cardiac cells

Abstract

There is substantial evidence supporting a hypertrophic action of serotonin [5-hydroxytryptamine (5-HT)] in cardiomyocytes. However, little is known about the mechanisms involved. We previously demonstrated that 5-HT-induced hypertrophy depends, in part, on the generation of reactive oxygen species by monoamine oxidase-A (MAO-A) (see Ref. 3). Cardiomyocytes express 5-HT2 receptors, which may also participate in hypertrophy. Here, we analyzed the respective contribution of 5-[HT.sub.2] receptors and MAO-A in H9C2 cardiomyoblast hypertrophy. 5-HT induced a dose-dependent increase in [sup.3]H]leucine incorporation and stimulation of two markers of cardiac hypertrophy, ANF-luc and [alpha]SK-actin-luc reporter genes. Experiments using 1 [micro]M 5-HT showed that hypertrophic response occurred independently from MAO-A. Using pharmacological inhibitors (M 100907 and ketanserin), we identified a novel mechanism of action involving 5-[HT.sub.2A] receptors and requiring [Ca.sup.2+]/calcineurin/nuclear factor of activated T-cell activation. The activation of this hypertrophic pathway was fully prevented by 5-[HT.sub.2A] inhibitors and was unaffected by MAO inhibition. When 10 [micro]M 5-HT was used, an additional hypertrophic response, prevented by the MAO inhibitors pargyline and RO 41-1049, was observed. Unlike the 5-[HT.sub.2A]receptor-mediated H9C2 cell hypertrophy, MAO-A-dependent hypertrophic response required activation of extracellular-regulated kinases. In conclusion, our results show the existence of a dose-dependent shift of activation of distinct intracellular pathways involved in 5-HT-mediated hypertrophy of cardiac cells. 5-hydroxytryptamine; 5-hydroxytryptamine 2A; monoamine oxidase type A; hypertrophy

Abstract

Metastasis formation is a multi-step process starting from the dissemination of transformed carcinomatous cells from the primary tumor and could occur at a very early stage of oncogenesis, before primary tumor detection. The adult mammary gland provides a unique model to investigate epithelial cell dissemination processes. Tissue clearing techniques allow imaging samples of large volume. uDSICO clearing, one of the latest tissue clearing technique developed, provides optical imaging of whole organ due to organ clearing and tissue size reduction. We wanted to take advantage of this technique to study rare events occurring in vivo.Here, we have established a new analytical pipeline exploiting the regenerative properties of the mammary epithelium following orthotropic transplantation of organoids together with the uDISCO organ size reduction and clearing method to study early cell dissemination in the mammary gland. As proof of concept, we analyzed the localization of epithelial cells overexpressing the oncogenic protein atypical protein kinase C iota (aPKCi+) in the normal mammary gland and we were able to visualize epithelial aPKCi+ cells, surrounded by normal epithelial cells, escaping from the normal mammary epithelium and disseminating into the surrounding stroma.

Abstract

This paper reports our work on the development of thin composite films based on aligned multi-walled carbon nanotubes (CNTs) forest embedded in epoxy polymer matrix dedicated for 3D-interconnection in flexible substrate. To reach this goal, information about local CNT properties is strongly needed. In this study, aligned multi-walled carbon nanotube carpets were prepared by aerosol Assisted catalytical chemical vapor deposition (AA-CCVD) process. Such carpets were then embedded in a polymer matrix, subsequently thinned by chemical mechanical polishing (CMP) process and finally treated by plasma process. Then, to determine local conducting properties of CNT, the surface of the composite film is studied by conductive AFM (C-AFM) and scanning spreading resistance microscopy. These methods are discussed and compared. C-AFM appears to be the most efficient method to obtain high resolution current maps. For the first time, a local electrical property mapping and current versus applied voltage characteristics over single carbon nanotube in vertical arrangement are obtained. These results show metallic behavior for nanotubes and emphasize that the electrical conduction is mainly driven by the external walls. [ABSTRACT FROM AUTHOR]

Abstract

The adsorption of DNA on chemically homogeneous, functionalized, oxide-free single-crystal silicon surfaces is studied by x-ray reflectivity. The adsorption of monodisperse, 294 base-pair double-stranded DNA on a positively charged surface is detected through the deformation of the molecular monolayer of aminated alkyl-chain molecules covalently bonded to the surface. The adsorption of single-stranded DNA does not lead to the same deformation. A detailed quantitative characterization of the density profiles yield surface densities of the covalently grafted, molecular monolayers that are in excellent agreement with infrared spectroscopic measurements. The additional mass density that is measured following the adsorption of DNA corresponds either to the partial embedding of a densely-packed adsorbed layer or to a deeper penetration into the soft surface layer at a lower surface density of the adsorbed double-stranded DNA molecules. The adsorption is found to be irreversible under high added salt concentrations, suggesting a partial dehydration of the double-stranded DNA. [ABSTRACT FROM AUTHOR]

Abstract

We use Raman spectrometry to investigate lattice disorder and strain induced by hydrogen or helium implantation in (001) and (011) Si. The phonon peak intensities and the spatial correlation model are used to estimate the amount of damage affecting the phonon coherence length. The redshift due to reduced coherence length is taken into account to fit the model to the experimental spectra. This allows us to correctly estimate a blueshift attributed to a compressive in-plane strain. We observe that the amount of strain increases linearly with the implant dose. For H implants the dependence of strain on crystallographic orientation was discovered. This effect is attributed to the anisotropic morphology of the H-induced extended defects: two-dimensional platelets with preferred orientations versus spherical nanobubbles formed after He implants. Raman results are correlated with the implant damage simulations and compared with the data obtained by other characterization techniques. [ABSTRACT FROM AUTHOR]

Si(111)-alkyl layer interfaces are prepared with a very low density of electronic traps after careful control of the grafting conditions. Water exclusion from the monolayer has promoted long-term passivation of the silicon surface against oxidation in air and water as well as perfect corrosion inhibition in 20% NH4F.

Abstract

This paper presents the results of a study on issues of reliability and accelerated life testing for radio frequency micro-electromechanical system (RF-MEMS) capacitive devices. A human-body-model electrostatic discharge tester has been used to induce charging by operating at stress levels much higher than would be expected in normal use. Temperature ranges from 300 K to 330 K allows the understanding of physical mechanisms that may be responsible for the device's reliability. Crown Copyright © 2009.

31. Molecularly tunable 'organic capacitors' at silicon/aqueous electrolyte interfaces 1

Abstract

Robust and uniform n-alkyl monolayers were formed on silicon from the reaction of Grignard regents (n-CnH2n+1MgBr, n = 2, 6, 10, and 15) with hydrogen-terminated Si(111). The capacitive properties of these organic thin films on silicon in contact with aqueous electrolytes were evaluated by electrochemical impedance measurements. In particular, the reciprocal capacitance of the organic thin film modified silicon/aqueous electrolyte interfaces is proportional to the film thickness, which is tunable by simply varying the alkyl chain length. The derived dielectric constant of these organic thin films from the best fit of the reciprocal capacitance vs ellipsometric film thickness plot is ε = 3.3 ± 0.6.
