1. Disease burden, management patterns and multidisciplinary clinical approaches for patients with MPS IVA and VI in selected Latin American Countries

Abstract

Background: There is a paucity of real-world epidemiological data on patients with mucopolysaccharidoses (MPS) in Latin America. This real-world study assessed the disease burden, management patterns and multidisciplinary clinical approaches for MPS-IVA and MPS-VI patients in Latin America (Colombia, Ecuador, Mexico, Peru). Methods: Data were collected from physicians/specialists experienced in treating MPS patients between April–June 2020, via an online patient-diary survey. Results: Overall, 29 physicians/specialists participated in this study. Data from 98 patients were analyzed (MPS-IVA, 71 patients and MPS-VI, 27 patients). Mean age for MPS-IVA patients was 17.5 years and for MPS-VI patients was 11.6 years, and the majority were females (52% and 78%, respectively). MPS-IVA and VI patients presented a high absenteeism from school (55% and 37%, respectively; 18 years age), indicating an impact of the disease on some aspects of the patients' quality of life. The onset of the first symptom occurred at the age of 3.1 years for MPS-IVA patients and at 1 year for MPS-VI, with delay in diagnosis (3.5–3.9 years from symptom onset) and enzyme replacement therapy (ERT) initiation (1.1–3.6 years from diagnosis). ERT interruptions were observed for MPS-IVA (48%) and MPS-VI patients (44%), with non-availability of medication recorded as the main reason for non-adherence (46% and 60% patients, respectively). ERT showed noticeable treatment benefits in MPS-IVA/VI patients, with stabilization/reduction in complications or the number of surgeries. A multidisciplinary clinical team approach was used for patient management. Conclusion: The disease burden for MPS-IVA/VI was high in Latin America, with consistent management, treatment and socio-demographic trends throughout the region.

Novel species of fungi described in this study include those from various countries as follows: Australia, Baobabopsis sabindy in leaves of Eragrostis spartinoides, Cortinarius magentiguttatus among deep leaf litter, Laurobasidium azarandamiae from uredinium of Puccinia alyxiae on Alyxia buxifolia, Marasmius pseudoelegans on well-rotted twigs and litter in mixed wet sclerophyll and subtropical rainforest. Bolivia, Favolaschia luminosa on twigs of Byttneria hirsuta, Lecanora thorstenii on bark, in savannas with shrubs and trees. Brazil, Asterina costamaiae on leaves of Rourea bahiensis, Purimyces orchidacearum (incl. Purimyces gen. nov.) as root endophyte on Cattleya locatellii. Abstract

Latent class analysis (LCA) refers to techniques for identifying groups in data based on a parametric model. Examples include mixture models, LCA with ordinal indicators, and latent class growth analysis. Despite its popularity, there is limited guidance with respect to decisions that must be made when conducting and reporting LCA. Moreover, there is a lack of user-friendly open-source implementations. Based on contemporary academic discourse, this paper introduces recommendations for LCA which are summarized in the SMART-LCA checklist: Standards for More Accuracy in Reporting of different Types of Latent Class Analysis. The free open-source R-package package tidySEM implements the practices recommended here. It is easy for beginners to adopt thanks to user-friendly wrapper functions, and yet remains relevant for expert users as its models are integrated within the OpenMx structural equation modeling framework and remain fully customizable. 16. Body mass index and gut microbiome: A cluster-randomized, controlled, pilot feasibility study

Abstract

Background: Prebiotic fiber has been examined as a way to foster gut bacteria less associated with obesity. Tests of prebiotic fiber in reducing obesity have been conducted mainly in animals, adults, and Caucasians when the highest obesity rates are in African American and Latinx youth. Response to prebiotic fiber is determined by the pre-existing intestinal microbiota. The type of microbiota varies based on diet and physical activity (PA), so it is important to examine acceptability and response to prebiotic fiber in those most at risk for obesity. Methods: This cluster-randomized, controlled feasibility trial included an online program designed to improve diet and PA along with administration of a prebiotic fiber for 12 weeks in 123 students of 4th and 5th grade where 98% were eligible for free or reduced-fee lunch. Of these 56% were male; 71% Latinx; 15% African American; and 14% other. Results: Decrease in body fat (BF) was associated with higher pretest BF. Lower body mass index (BMI) was associated with decrease in fecal Tenericutes and increase in Actinobacteria. Conclusions: Prebiotic fiber can be helpful in supporting healthy weight, so inclusion in culturally congruent foods usually eaten by children from groups at high risk for obesity should be considered following additional studies. Determining those most responsive to prebiotic fibers can also permit individual recommendations for greater inclusion in usual diet choices. Clinical Trial Registration Number NCT05671731.

ABSTRACT We herein propose the transfer of Metaphareus punctatus Roewer, 1913, to the genus Eutimesius Roewer, 1913 (Heterostygninae) based on the genital morphology of the species and morphology of tarsi III and IV, resulting in the new combination Eutimesius punctatus (Roewer, 1913), comb. nov. New geographical records for E. albicinctus (Roewer, 1915) are offered, recording sympatry between the congeneric species E. albicinctus and E. punctatus and data on the reproductive behavior of E. punctatus are presented, suggesting the existence of parental care in this species.

Abstract

The international RAIA Observatory (www.marnaraia.org) resulted from the effort of 12 research and academic institutions and public agencies (Spanish and Portuguese) working in the field of meteorology and oceanography. The RAIA Observatory serves the main maritime activities of the Galicia-Northern Portugal Euroregion and contributes to collaborative observational networks. Under the framework of coastal risk management, environmental indicators are fundamental tools for the evaluation and mitigation of environmental risks, showing the current state and helping to predict future changes on ecosystem health regarding environmental risks. In last years, the different partners of the RAIA Observatory have identified 38 environmental indicators, in which 12 key risks affecting the ecosystem services of the Euroregion Galicia-Northern Portugal are being evaluated. Data was analyzed and compiled by the various partners of the RAIA Observatory and the development and optimization of the environmental indicators has been done according to the specifications provided by the European Environmental Agency (EEA) and the International Panel for Climate Change (IPCC). The resulting environmental indicators are included and shared on a publicly-accessible Web service, georeferenced and accompanied by graphics (https://marrisk.inesctec.pt/public/#!/indicators). So far, the current status of the indicators has allowed us to establish risk assessment protocols for the Euroregion, and identify critical gaps in temporal and spatial coverage. The inclusion of the environmental indicators in the RAIA Observatory is of great relevance for national and international data exchange and promotes future collaborations. Abstract

Plant synthetic biology is a fast-evolving field that employs engineering principles to empower research and bioproduction in plant systems. Nevertheless, in the whole synthetic biology landscape, plant systems lag compared to microbial and mammalian systems. When it comes to multigene delivery to plants, the predictability of the outcome is decreased since it depends on three different chassis: E. coli, Agrobacterium, and the plant species. Here we aimed to develop standardised and streamlined tools for genetic engineering in plant synthetic biology. We have devised Mobius Assembly for Plant Systems (MAPS), a user-friendly Golden Gate Assembly system for fast and easy generation of complex DNA constructs. MAPS is based on a new group of small plant binary vectors (pMAPs) that contains an origin of replication from a cryptic plasmid of Paracoccus pantotrophus. The functionality of the pMAP vectors was confirmed by transforming the MM1 cell culture, demonstrating for the first time that plant transformation is dependent on the Agrobacterium strains and plasmids; plasmid stability was highly dependent on the plasmid and bacterial strain. We made a library of new short promoters and terminators and characterised them using a high-throughput protoplast expression assay. Our results underscored the strong influence of terminators in gene expression, and they altered the strength of promoters in some combinations and indicated the presence of synergistic interactions between promoters and terminators. Overall this work will further facilitate plant synthetic biology and contribute to improving its predictability, which is challenged by combinatorial interactions among the genetic parts, vectors, and chassis.

Abstract

Adaptation to climate change requires the implementation of services that translate scientific knowledge into practical results, so that policymakers and stakeholders can understand the risks and increase their resilience. In the coast those risks are related to flooding, erosion or physico-chemical changes of seawater. The main aim of MarRisk project was to generate this type of services for the NW of the Iberian Peninsula, relying on the experience of a coastal oceanographic observatory (RAIA). These services have been developed based on indicators and models, through a process of co-creation. Thus, a resilience index for harbours or estimations of physical-chemical changes of seawater that can impact sectors such as fishing or aquaculture have been generated. Moreover we have calculated maps of vulnerable areas to flood and erosion. As a conclusion we highlight that the elaboration of a set of indicators together with the expertise of modelization of climate change is not enough to help coastal communities to adapt to climate change. The interaction with different stakeholders is also a needful step to create climate services.

46. iFADO project: contribution to the determination of the Good Environmental Status in the Atlantic Area through modeling and in situ monitoring approaches

Abstract

The project iFADO ( innovation in the Framework of the Atlantic Deep Ocean ) aims to combine conventional monitoring programs (with special focus on novel semi-automatic techniques that can be applied for in situ biological monitoring) with emerging technologies (such as gliders, satellite imagery, etc.) and modeling approaches to assist the implementation of the Marine Strategy Framework Directive (MSFD), providing tools for the optimization of observing strategies for better evaluation and forecasting of the Good Environmental Status (GES). Within this framework, the multidisciplinary iFADO cruise (within the RADPROF time series program, Instituto Español de Oceanografía) was carried out with transnational collaboration on summer 2020 along a standard coastal-oceanic transect off NW Iberian Peninsula, including physical, biogeochemical and biological measurements. Additionally, an effort is being made in the implementation and validation of existing and new biogeochemical models in the Atlantic margin, focusing on key variables for the implementation of MSFD Descriptors. The combination of iFADO models and in situ monitoring approaches not only might contribute data and potential indicators for MSFD Descriptors 1, 2, 3, 4, 5, 6, 7 and 10, from coastal to open-ocean environments, but they are essential tools to detect, model and predict temporal changes in marine ecosystems in the study area.

Abstract

The MyCOAST project (http://mycoast-project.org/) is an INTERREG Atlantic Area project designed to demonstrate that marine services for tackling coastal risks can be jointly developed. The main innovation and originality of the project stems from the implementation of transferable tools able to improve the risk management systems operated in the Atlantic Area. A successful outcome was achieved by identifying mature existing tools and selecting those that could be further developed by partners during the project duration. Demonstration of the tools in pilot actions showed that they are effective in supporting end users and relocatable among different regions in the Atlantic Area., Le projet MyCOAST (http://mycoast-project.org/) est un projet INTERREG de l'Espace Atlantique destiné à démontrer que les services maritimes pour faire face aux risques côtiers peuvent être développés conjointement. La principale innovation et originalité du projet réside dans la mise en œuvre d'outils transférables capables d'améliorer les systèmes de gestion des risques exploités dans l'Espace Atlantique. L'identification d'outils matures existants et la sélection de ceux qui pourraient être développés par les partenaires pendant la durée du projet ont permis d'atteindre un résultat positif. La démonstration des outils dans des actions pilotes a montré qu'ils sont efficaces pour soutenir les utilisateurs finaux et qu'ils peuvent être transférés dans différentes régions de l'Espace Atlantique.

49. Complementary and alternative medicine mind-body approaches used among racially and ethnically diverse adolescents

Abstract

The aim of this paper is to examine complementary and alternative medicine (CAM) use among racially and ethnically diverse adolescents. Greater understanding of CAM use among this group is warranted to better inform health care providers in delivering a culturally relevant health promotion approach. Design and methods: A secondary data analysis was conducted using the 2012 Child Complementary and Alternative Medicine Supplement of the National Health Interview Survey (CAM-NHIS) data, which was collected from a national sample of adolescents aged 12–17 years. A logistic regression test was employed to investigate the predictors associated with CAM use among racially and ethnically diverse adolescents. Results: While Black and Hispanic adolescents were the least likely to use CAM compared to their White counterparts, families with higher incomes, higher education attainment, and adolescents who experienced pain were more likely to use CAM. Conclusions: Findings suggest the need for future research to gain a greater understanding of CAM use among racially and ethnically diverse adolescents, and insights into how health disparities impact CAM use. Greater understanding of how CAM use intersects with health beliefs and outcomes is also warranted. Practice implications: Based on the CAM-NHIS survey, few racially and ethnically diverse adolescents have reported use of CAM. Development of culturally appropriate instruments and methods to assess CAM use among racially and ethnically diverse adolescents may yield specific data for this population. Informed health care providers can advocate for improved access to CAM for minority adolescents and alter disparate use.

50. An fMRI study of cognitive reappraisal in major depressive disorder and borderline personality disorder

Abstract

BACKGROUND: One common denominator to the clinical phenotypes of borderline personality disorder (BPD) and major depressive disorder (MDD) is emotion regulation impairment. Although these two conditions have been extensively studied separately, it remains unclear whether their emotion regulation impairments are underpinned by shared or distinct neurobiological alterations. METHODS: We contrasted the neural correlates of negative emotion regulation across an adult sample of BPD patients (n = 19), MDD patients (n = 20), and healthy controls (HCs; n = 19). Emotion regulation was assessed using an established functional magnetic resonance imaging cognitive reappraisal paradigm. We assessed both task-related activations and modulations of interregional connectivity. RESULTS: When compared to HCs, patients with BPD and MDD displayed homologous decreased activation in the right ventrolateral prefrontal cortex (vlPFC) during cognitive reappraisal. In addition, the MDD group presented decreased activations in other prefrontal areas (i.e., left dorsolateral and bilateral orbitofrontal cortices), while the BPD group was characterized by a more extended pattern of alteration in the connectivity between the vlPFC and cortices of the visual ventral stream during reappraisal. CONCLUSIONS: This study identified, for the first time, a shared neurobiological contributor to emotion regulation deficits in MDD and BPD characterized by decreased vlPFC activity, although we also observed disorder-specific alterations. In MDD, results suggest a primary deficit in the strength of prefrontal activations, while BPD is better defined by connectivity disruptions between the vlPFC and temporal emotion processing regions. These findings substantiate, in neurobiological terms, the different profiles of emotion regulation alterations observed in these disorders.
