Lentinus crinitus (L.) Fr is a wild macrofungus that is popular as antimicrobial and various biological activities. This study aims to determine the capacity growth stimulation of Lactobacillus paracasei and antimicrobial activity of aqueous extracts of L. crinitus obtained from wild basidiomata, mycelial biomass by liquid fermentation and spent mushroom substrate obtained by solid-state fermentation. The antimicrobial activity was investigated against bacterial and fungal pathogens and growth stimulation L. paracasei probiotic bacterium. The total carbohydrate and β-glucan contents of the extracts were determined using colorimetric analysis. The aqueous extracts obtained showed inhibition against Fusarium oxysporum., Penicillium sp., Rhizopus oryzae, Aspergillus niger, Escherichia coli and Salmonella typhimurium. The aqueous extract obtained from wild basidiomata, and mycelial biomass showed the highest percentage of stimulation of L. paracasei growth in 48 h. The extracts obtained from L. crinitus have antimicrobial potential and stimulating capacity of the probiotic Lactobacillus paracasei. Additionally, different biotechnological techniques such as liquid and solid-state fermentation can be used to obtain aqueous extracts.

Xylem density is a physical property of wood that varies between individuals, species and environments. It reflects the physiological strategies of trees that lead to growth, survival and reproduction. Measurements of branch xylem density, ρx, were made for 1653 trees representing 598 species, sampled from 87 sites across the Amazon basin. Measured values ranged from 218 kg m−3for a Cordia sagotii (Boraginaceae) from Mountagne de Tortue, French Guiana to 1130 kg m−3 for an Aiouea sp. (Lauraceae) from Caxiuana, Central Pará, Brazil. Analysis of variance showed significant differences in average ρx across regions and sampled plots as well as significant differences between families, genera and species. A partitioning of the total variance in the dataset showed that species identity (family, genera and species) accounted for 33% with environment (geographic location and plot) accounting for an additional 26%; the remaining "residual" variance accounted for 41% of the total variance. Variations in plot means, were, however, not only accountable by differences in species composition because xylem density of the most widely distributed species in our dataset varied systematically from plot to plot. Thus, as well as having a genetic component, branch xylem density is a plastic trait that, for any given species, varies according to where the tree is growing in a predictable manner. Within the analysed taxa, exceptions to this general rule seem to be pioneer species belonging for example to the Urticaceae whose branch xylem density is more constrained than most species sampled in this study. These patterns of variation of branch xylem density across Amazonia suggest a large functional diversity amongst Amazonian trees which is not well understood.

This study examined how having had a similar experience to a target person's experience affected three facets of empathy: empathic concern, empathic accuracy, and perceived empathy. Women who had never been mothers, who were pregnant with their first child, or who had just given birth to their first child (20 in each group) served as perceivers, watching videotapes of new-mother targets (N = 20) and providing measures of emotional and cognitive empathy. When perceivers had experienced the same life events as the targets, they expressed greater empathic concern and reported greater understanding of targets. However, experience had a much smaller effect on empathic accuracy, limited to comparisons between new-mother and never-pregnant perceivers and only for accuracy at guessing stereotypic attitudes, not individual thoughts. Perceived empathy, in contrast, appeared to be influenced by targets' knowledge of whether perceivers had experienced similar events.

The association of the prion protein (PrP) gene with susceptibility to scrapie has formed the basis of selection programs aimed at eradicating the disease from sheep populations. Animals are genotyped for the PrP gene and those with the less susceptible genotypes are selected. The objectives of this study were to determine the effectiveness of predicting PrP genotypes by using information from relatives and to investigate the association of the PrP genotype with lamb performance traits in Suffolk sheep. Data were obtained from a scrapie-affected flock maintained in Scotland. A total of 643 were animals genotyped at codon 171 of the PrP gene with 2 alleles, R and Q. The genotypes of these animals were used to predict the genotypes of 5,173 nongenotyped animals in the same flock using segregation analysis. The genotype of nongenotyped animals was predicted from the probabilities for each possible genotype; further, an overall index for each animal was calculated to reflect the accuracy of prediction. Association analyses of the PrP gene (using animals with both known and inferred genotypes) with BW at birth, at weaning (56 d), and at 150 d, and for backfat and muscle depths at 150 d of age were carried out. A linear mixed model with random direct and maternal additive genetic effects, maternal permanent and temporary environmental effects, and year of birth was tested, and the most appropriate model was used for each trait. The expected number of Q alleles carried (from 0 to 2) by each animal was calculated and used in the model as a linear and quadratic covariate to test for associations with possible additive and dominance PrP gene effects, respectively. Results showed that the genotypes of relatively few animals (235) were inferred with certainty (compared with the 5,173 nongenotyped animals). Approximately 25% of the 5,173 predicted genotypes were inferred with a genotype probability index of 50% and greater. There was no significant association of the PrP gene with any of the performance traits studied (there were no significant additive or dominance effects). Such was the case whether data on animals with known or with both known and predicted genotypes were considered. It can be concluded that selection for PrP-resistant alleles in Suffolk sheep is unlikely to affect performance directly. In this study the effect of artificial selection on BLUP EBV for production traits on the allele frequencies of a pleiotropic QTL affecting both production and disease susceptibility was investigated. Stochastic simulations were used to model artificial selection on a production trait that is controlled, in part, by a biallelic QTL that also controls susceptibility to disease. The QTL allele increasing production also increased susceptibility to disease. Different modes of action and proportions of variation accounted for by the QTL were assessed for the production trait. The main results indicated that alleles that confer susceptibility to the disease could be maintained in the population over a long period, depending on the mode of action of the QTL. In addition, the results of the study indicate that, under various conditions, it is possible to find pleiotropic QTL that control 2 traits despite these traits appearing to be uncorrelated. Therefore, in practice, an estimate of the genetic correlation between 2 traits may be misleading when the presence of such a QTL exists. The results of this study have implications for breeding programs. For example, if a pleiotropic QTL exists that favors heterozygotes for a production trait, it would be very difficult to remove disease susceptibility alleles via traditional selection methods. This study investigated associations of prion protein (PrP) genotype with body composition and weight traits of Scottish Blackface ewes. Body composition was predicted using computer tomography (CT) scans to estimate muscle, carcass fat, internal fat, and bone weights. The traits were measured at 4 key seasonal production points (pre-mating, pregnancy, midlactation, and weaning) over 4 production cycles (2 to 5 yr old). There were 2,413 records for each of the CT traits measured on 335 ewes, and 26,649 records for each of the body condition score and BW traits for 2,356 ewes. From 1999 to 2004, animals were genotyped to determine polymorphisms at codons 136, 154, and 171, which are associated with scrapie susceptibility. Four alleles were found in the population (ARR, AHQ, ARQ, and VRQ). The data were analyzed using a linear mixed random regression model assuming that the direct additive genetic effect was a 2nd order Legendre polynomial function of time. The PrP genotype was included in the model as a fixed effect along with other fixed factors with significant effects (P < 0.05). Five separate analyses were carried out for each trait, depending on the method of classifying the PrP geno type. In the first analysis, animals were categorized according to the genotype. Only the 5 most common genotypes (ARR/ARR, ARR/AHQ, ARR/ARQ, AHQ/ARQ, and ARQ/ARQ) were included. In the last 4 analyses, animals were categorized according to the number of each PrP allele carried. For CT traits and body condition score, results showed that the PrP genotype has no association with the overall mean of the traits (averaged over age). For BW, ewes without the ARQ allele were at least 0.5 kg heavier than ARQ homozygous and heterozygous ewes. On the other hand, there was a significant interaction between PrP genotype and age of the ewe (i.e., the effect of PrP genotype was not the same at different ages for 5 out of the 6 traits studied). In general, ARQ carrying ewes mobilized more fat reserves at times of nutrient deficiency, such as during lactation, and gained it back more quickly by the mating season (when nutrients became abundant) than non-ARQ carriers. Therefore, selecting against this allele would have consequences on BW and seasonal mobilization of body reserves. The number of VRQ alleles (the most scrapie susceptible allele) carried was not significantly associated with any of the traits. The objective of this study was to investigate and estimate the associations of the ovine prion protein (PrP) genotypes with a wide range of performance traits in Scottish Blackface lambs. Performance records of up to 7,138 sheep of known PrP genotypes born from 1999 to 2004 in 2 experimental farms were utilized. Performance traits studied were BW at birth, marking (when the sheep were identified with permanent ear tags at an average age of 52 d), and weaning (average age of 107 d); slaughter traits (BW at slaughter, slaughter age, carcass weight, and carcass conformation); ultrasonic muscle and fat depths; and computerized tomography-predicted carcass composition and carcass yield at weaning. Different linear mixed models, including random, direct animal effect, and up to 3 maternal effects (genetic, permanent, and temporary environmental) were used for the different traits. The PrP genotype was included in the model as a fixed effect, along with other fixed factors with significant effects (P < 0.05). Five separate analyses were carried out for each trait, differing in the method of PrP genotypic classification. The first analysis was based on classifying the sheep into categories according to all 9 available PrP genotypes. In the other 4 analyses, sheep were categorized according to the number of each PrP allele carried. Results showed that there were no significant differences between PrP genotypes for any of the performance traits studied when all 9 genotypes were compared (first analysis). Similarly, performance of the lambs did not significantly differ between genotypes with different numbers of ARR copies. However, there were significant variations in a few traits with respect to the number of ARQ, AHQ, and VRQ alleles carried. Heterozygous lambs for the AHQ or the ARQ allele were significantly heavier at some ages than lambs of the other genotypes. Lambs carrying the VRQ allele required approximately 10 d longer finishing time (P = 0.01) and yielded carcasses approximately 0.5 kg heavier (P = 0.03) compared with noncarriers. The few significant associations found do not have a negative influence on performance when selecting against the most susceptible PrP allele (VRQ) or in favor of the most resistant one (ARR). Overall, there were no major associations of PrP genotypes with most lamb performance traits in Scottish Blackface sheep. This study investigated, through stochastic computer simulation, the extra gains expected from marker-assisted selection (MAS) in an infinitesimal model with linkage. The trait under selection was assumed to be controlled by 2,000 loci of additive small effect and evenly distributed in c chromosomes of one Morgan each (and c = 5, 10, 20, or 30). This approach differs from previous studies on the benefits of MAS that have considered mixed inheritance models. Marker information was used together with pedigree information to compute the relationship matrix used in BLUP genetic evaluations. The MAS schemes were compared with schemes where genetic evaluations were performed using standard BLUP (i.e., the relationship matrix is obtained using pedigree information only). When the number of markers was large enough (approximately one marker every 10 cM), there were increases in the accuracy of selection with MAS, and this led to extra gains compared with standard BLUP for all genome sizes considered. The benefit from MAS increased over generations. At the last generation of selection (Generation 10), the response from MAS was 11, 9, 7, and 5% greater than with standard BLUP for genomes with 5, 10, 20, and 30 chromosomes, respectively. Thus, although small, gains from MAS were nonetheless detectable for genome sizes typical of livestock populations. Stochastic computer simulation was used to investigate the potential extra genetic gains obtained from gene-assisted selection (GAS) by combining 1) optimization of genetic contributions for maximizing 2) gain, while restricting the rate of inbreeding with 2) optimization of the relative emphasis given to the QTL over generations. The genetic model assumed implied a mixed inheritance model in which a single quantitative trait locus (i.e., QTL) is segregating together with polygenes. When compared with standard GAS (i.e., fixed contributions and equal emphasis on the QTL and polygenic EBV), combined optimization of contributions of selection candidates and weights on the QTL across generations allowed substantial increases in gain at a fixed rate of inbreeding and avoided the conflict between short- and long-term responses in GAS schemes. Most of the increase of gain was produced by optimization of selection candidates' contributions. Optimization of the relative emphasis given to the QTL over generations had, however, a greater effect on avoiding the long-term loss usually observed in GAS schemes. Optimized contribution schemes led to lower gametic phase disequilibrium between the QTL and polygenes and to higher selection intensities both on the QTL and polygenes than with standard truncation selection with fixed contributions of selection candidates. The expected benefits from optimized selection in real livestock populations were evaluated by applying dynamic selection algorithms to two livestock populations of sheep (Meatlinc) and beef cattle (Aberdeen Angus). In addition, the effects of introducing BLUP evaluations on the population structure, genetic gain, and inbreeding were investigated. The use of BLUP-EBV accelerated the rates of gain in the Meatlinc, but the effects of BLUP evaluations on Aberdeen Angus are not as evident. Although steady increases in the average coefficient of inbreeding (F) were observed, the inbreeding rates ([DELTA]F) before and after the introduction of BLUP evaluations were not significantly different. The observed [DELTA]F in the last generation was 1.0% for Meatlinc and 0.2% for Aberdeen Angus. T he application of the dynamic selection algorithms for maximizing genetic gain at a fixed [DELTA]F led to important expected increases in the rate of genetic gain ([DELTA]G). When [DELTA]F was restricted to the value observed in both populations, increments per year in [DELTA]G of 4.6 (i.e., 17%) index units for Meatlinc and 3.5 (i.e., 30%) index units for Aberdeen Angus were found in comparison to the [DELTA]G expected from conventional truncation BLUP selection. More relaxed constraints on [DELTA]F allowed even higher expected increases in [DELTA]G in both populations. This study demonstrates that the optimization tools constitute a potentially highly effective way of managing gain and inbreeding under a broad range of schemes in terms of scale and inbreeding level. No losses in genetic gain were associated with the use of dynamic optimization selection when schemes were compared at the same [DELTA]F.
