Abstract The utility of deep neural nets has been demonstrated for mapping hematoxylin-and-eosin (H&E) stained image features to expression of individual genes. However, these models have not been employed to discover clinically relevant spatial biomarkers. Here we develop MOSBY (Multi-Omic translation of whole slide images for Spatial Biomarker discoverY) that leverages contrastive self-supervised pretraining to extract improved H&E whole slide images features, learns a mapping between image and bulk omic profiles (RNA, DNA, and protein), and utilizes tile-level information to discover spatial biomarkers. We validate MOSBY gene and gene set predictions with spatial transcriptomic and serially-sectioned CD8 IHC image data. We demonstrate that MOSBY-inferred colocalization features have survival-predictive power orthogonal to gene expression, and enable concordance indices highly competitive with survival-trained multimodal networks. We identify and validate (1) an ER stress-associated colocalization feature as a chemotherapy-specific risk factor in lung adenocarcinoma, and (2) the colocalization of T effector cell vs cysteine signatures as a negative prognostic factor in multiple cancer indications. The discovery of clinically relevant biologically interpretable spatial biomarkers showcases the utility of the model in unraveling novel insights in cancer biology as well as informing clinical decision-making.

Abstract Carbon nanotube (CNT) has fostered research as a promising nanomaterial for a variety of applications due to its exceptional mechanical, optical, and electrical characteristics. The present article proposes a novel and comprehensive micromechanical framework to assess the viscoelastic properties of a multiscale CNT-reinforced two-dimensional (2D) woven hybrid composite. It also focuses on demonstrating the utilisation of the proposed micromechanics in the dynamic analysis of shell structure. First, the detailed constructional attributes of the proposed trans-scale composite material system are described in detail. Then, according to the nature of the constructional feature, mathematical modelling of each constituent phase or building block’s material properties is established to evaluate the homogenised viscoelastic properties of the proposed composite material system. To highlight the novelty of this study, the viscoelastic characteristics of the modified matrix are developed using the micromechanics method of Mori–Tanaka (MT) in combination with the weak viscoelastic interphase (WI) theory. In the entire micromechanical framework, the CNTs are considered to be randomly oriented. The strength of the material (SOM) approach is used to establish mathematical frameworks for the viscoelastic characteristics of yarns, whereas the unit cell method (UCM) is used to determine the viscoelastic properties of the representative unit cell (RUC). Different numerical results have been obtained by varying the CNT composition, interface conditions, agglomeration, carbon fibre volume percentage, excitation frequency, and temperature. The influences of geometrical parameters like yarn thickness, width, and the gap length to yarn width ratio on the viscoelasticity of such composite material systems are also explored. The current study also addresses the issue of resultant anisotropic viscoelastic properties due to the use of dissimilar yarn thickness. The results of this micromechanical analysis provide valuable insights into the viscoelastic properties of the proposed composite material system and suggest its potential applications in vibration damping. To demonstrate the application of developed novel micromechanics in vibration analysis, as one of the main contributions, comprehensive numerical experiments are conducted on a shell panel. The results show a significant reduction in vibration amplitudes compared to traditional composite materials in the frequency response and transient response analyses. To focus on the aspect of micromechanical behaviour on dynamic response and for the purpose of brevity, only linear strain displacement relationships are considered for dynamic analysis. These insights could inform future research and development in the field of composite materials.

AbstractExperimental analysis is carried out on the mechanical characteristics of fiber-reinforced that can be produced by adding basalt fibers to strengthened jute fiber-reinforced epoxy composites in both the warp and weft orientations. A three-parameter/three-level Design of Experiment technique called the Box–Behnken design (BBD) is used to ascertain the relationship between the input parameters and the response. Mechanical testing was done on the composite plates after fabrication to estimate the tensile strength of the composites in both the warp and weft directions. The basalt fiber content of the composite was around 0.5 wt.%, 1 wt.% and 1.5 wt.%; the sonication period was 20, 30 and 40 min; and the temperature was approximately 60 °C, 70 °C and 80 °C. In the warp and weft directions, the maximum ultimate tensile stresses are measured to be 34.03 MPa and 36.32 MPa, respectively. Analysis of Variance is used to determine the regression equation and the influence of the input parameters. The optimum ‘ultimate tensile stress’ is 40.162 MPa for warp direction and 35.445 MPa for weft direction with 1.5 wt.% filler weight, 40 min of sonication and 60 °C temperature.

ABSTRACTThe objective of this work is to examine the fire resistance of natural composite materials (NCM). The reinforcements used in this work are coir, cotton, Palmyra leaf stream, sisal fiber, and matrix selected for natural rubber resin (latex). The fibers as reinforcement and natural rubber as the matrix phase were considered, and then the composite was manufactured through the hand lay-up open mold method. The chemical compositions of specimens treated with inorganic flame retardant chemicals (Al(OH)3, Mg(OH)2, zinc borate, ZnO, and zirconium oxide). The selection of flame retardant chemicals is based on their high melting temperature, low toxicity, low smoke emission, low burning rate, easy availability, and low cost. The thermal analysis of the horizontal burn test, TGA, DSC, and cone calorimeter analysis was carried out to understand the material’s thermal stability. The important finding of this experimental work is to analyze the micro structural and thermal stability of various compositions of flame-retardant composite materials. In all aspects, the zinc-borate and zirconium oxide-added composites show more advantages in flame retardant properties compared to other composite materials.

Salmonella enterica is a leading cause of foodborne illness in the U.S. In the meat industry, one action taken to address pathogen contamination incidence is an intense sanitization (IS) of the entire processing plant that many large processors perform annually or semiannually. However, this procedure’s immediate and long-term impact on environment microbial community and pathogen colonization are unknown. Here we investigated the impact of IS procedure on environmental biofilms and the subsequent S. enterica colonization and stress tolerance. Environmental samples were collected from floor drains at various areas 1 week before, 1 week, and 4 weeks after the IS procedure at a beef plant with sporadic S. enterica prevalence. Biofilm formation by microorganisms in the drain samples without S. enterica presence was tested under processing temperature. The ability of the biofilms to recruit and/or protect a co-inoculated S. enterica strain from quaternary ammonium compound (QAC) treatment was determined. The community structure of each drain sample was elucidated through 16S rRNA amplicon community sequencing. Post-IS samples collected from 8 drains formed significantly stronger biofilms than the respective pre-IS samples. S. enterica colonization was not different between the pre- and post-IS biofilms at all drain locations. S. enterica survival in QAC-treated pre- and post-IS mixed biofilms varied depending upon the drain location but a higher survival was associated with a stronger biofilm matrix. The 16S rRNA amplicon gene community sequencing results exhibited a decrease in community diversity 1 week after IS treatment but followed by a significant increase 4 weeks after the treatment. The IS procedure also significantly altered the community composition and the higher presence of certain species in the post-IS community may be associated with the stronger mixed biofilm formation and Salmonella tolerance. Our study suggested that the IS procedure might disrupt the existing environmental microbial community and alter the natural population composition, which might lead to unintended consequences as a result of a lack of competition within the multispecies mixture. The survival and recruitment of species with high colonizing capability to the post-IS community may play crucial roles in shaping the ensuing ecological dynamics.

ABSTRACT Salmonella enterica is a prominent cause of foodborne disease in the United States. However, the mechanism and route of pathogen transmission that leads to Salmonella infection in commercial processing plants are poorly understood. This study aimed to investigate the effect of mixed-species biofilms on S. enterica survival and persistence under sanitizer stress [Quaternary ammonium compounds (QACs)] by analyzing 78 floor drain samples from a meat processing facility and three S. enterica strains (serovars Cerro, Montevideo, and Typhimurium) isolated from that facility and an unrelated source. The four test groups were as follows: control, QAC treatment, Salmonella addition, and QAC treatment with Salmonella addition. DNAs were extracted, and 16S rRNA gene based on the variable region V4 amplicon sequencing was performed to analyze the relative abundance, core microbiome, and Alpha and Beta diversity using the qiime2 pipeline. At the genus level, the Brochothrix (45.56%), Pseudomonas (38.94%), Carnobacterium (6.18%), Lactococcus (4.68%), Serratia (3.14%), and Staphylococcus (0.82%) were shown to be the most prevalent in all drain samples. The results demonstrate that the relative abundance of different bacterial genera was affected by both QAC treatment and Salmonella addition, with some genera showing increases or decreases in abundance. Notably, the correlation network was constructed to understand the relationships between the different bacteria. Nitrospira had the greatest number of connections in the floor drain environment network, with two negative and eight positive correlations. The results suggest that Nitrospira in the mixed-species biofilm community may play a role in converting ammonium in the QAC sanitizer into nitrites. Thus, Nitrospira could be a potentially important genus in providing sanitizer resistance to pathogen-encompassed mixed-species biofilms.IMPORTANCESalmonella contamination in meat processing facilities can lead to foodborne illness outbreaks. Our study characterized the microbiome dynamics in beef facility drains and their response to Salmonella addition and common sanitizer (QAC). Nitrospira could be an important genus in providing sanitizer resistance to pathogen-encompassed mixed-species biofilms. The results provide insight into the impact of mixed-species biofilms on Salmonella survival and persistence under sanitizer stress in meat processing facilities. The results highlight the need to consider mixed-species biofilm effects when developing targeted interventions to enhance food safety.

Abstract Industries play a very vital role in the developed nation. Proportional to the higher production capacity of these industries, there is a surge in the quantity of waste material being discharged. This waste material can be put to effective use; a considerable way is by creating a green composite that is long-lasting, and concocted by using natural fibers and environment-friendly materials as reinforcements. In the following study, an attempt is made to investigate the buckling characteristics of a thin geometrical plate of epoxy-based composite reinforced with Sugarcane fiber/ Fly-ash/ Carbon Nanotube. The investigative study was conducted numerically on the plate by applying axially compressive load. To procure an optimized result on the weight percentages of the composition of the fiber in the composite material, the DOE/optimization tool i.e. a mathematical and statistical technique known as the Response Surface Methodology (RSM) was used. Essential geometrical modeling and the appropriate boundary conditions for the buckling analysis were carried out using the Static Structural and Eigen Buckling standalone systems in the ANSYS software. The analytical tool, Analysis of Variance (ANOVA) was utilized to investigate the influential degree of reinforcement variables on buckling characteristics present in the composite. The results reveal that the critical buckling loads escalate for higher weight percentages for carbon nanotube and fly-ash reinforcements in the composite composition. The optimized parameters obtained can be incorporated to achieve improved critical buckling load and hence many synthetic composites were replaced thus enhancing the sustainability of the environment.

BackgroundMulti-species biofilms pose a problem in various environments, especially food-processing environments. The diversity of microorganisms in these biofilms plays a critical role in their integrity and protection against external biotic and abiotic factors. Compared to single-species biofilms, mixed-species biofilms are more resistant to various stresses, including antimicrobials like sanitizers. Therefore, understanding the microbiome composition and diversity in biofilms and their metabolic potential is a priority when developing intervention techniques to combat foodborne pathogens in food processing environments. MethodsThis study aimed to describe and compare the microbiome profile of 75 drain biofilm samples obtained from five different locations (Hotscale, Hotbox, Cooler, Processing, & Grind room) of three beef-processing plants (Plant A, B & C) taken over two timepoints 2017-18 (T1) and 2021 (T2) by shotgun sequencingResultsCore microbiome analysis found Pseudomonas, Psychrobacter, and Acinetobacter to be the top three prevalent genera among the plants and locations. Alpha diversity analysis demonstrated a high diversity of microbiome present in all the plants and locations across the time points. Functional analysis showed the high metabolic potential of the microbial community with abundance of genes in metabolism, cell-adhesion, motility, and quorum sensing. Moreover, Quaternary Ammonium Compound (QAC) resistance genes were also observed, this is significant as QAC sanitizers are commonly used in many food processing facilities. Multi-functional genes such as transposases, polymerases, permeases, flagellar proteins, and Mobile Genetic Elements (MGEs) were found suggesting these are dynamic microbial communities that work together to protect themselves against environmental stresses through multiple defense mechanisms.ConclusionThis study provides a framework for understanding the collective microbial network spanning a beef processing system. The results can be used to develop intervention strategies to best control these highly communicative microbial networks.

This paper presents an experimental study on the low-velocity impact behavior of hemp and jute fiber-reinforced epoxy composite. The Compressive After Impact (CAI) properties of composites post-impact are used for analysis. The type of damage and area of damage is used to analyze the effectiveness of composites for variation in impact energy. The impacted composite specimens were subjected to Compression After the Impact (CAI) test. A two-factor, five-level Central Composite Design (CCD) model of response surface methodology (RSM) is used to analyze the effect of input variables on ultimate load and thereby obtain the optimized input conditions for damage. The experimental results were optimized using Derringer’s Desirability function. The results indicate that jute laminates show a better impact energy absorption capability than hemp laminate composites. From the study, it was observed that the CAI strength of jute/epoxy composite was high compared to hemp/epoxy composite and the jute/epoxy composite exhibited enhanced CAI properties of the composite. From the regression equations and the three-dimensional response surface plots obtained from CCD, optimized ramp values were obtained. The optimized ultimate load of hemp and jute fiber laminate was 5.90 MPa and 10.68 MPa, respectively.

Recent investigations involving specimens for material and process qualification related to the Laser Powder Bed Fusion (L-PBF) additive manufacturing (AM) method, point that microstructural quality and surface properties vary significantly based on feedstock material and process parameters. There is currently the need, therefore, to create systematic ways to design specimens for process and material qualification. This manuscript provides an approach to evaluate the effects on the as-built quality based on specific metrics for specimens produced by L-PBF. Surface quality, as well as micro- and sub-microstructures and defects are related to specific aspects of the L-PBF process. To accomplish this goal, a digital engineering approach was first developed to design geometries based on process simulations to account for mechanical and thermal process effects. A parametric AM fabrication plan was then chosen to produce statistically significant populations of AM parts. Moreover, on axis, melt-pool scale, in-situ monitoring data, collected during the AM process, was examined in conjunction with the formation of flaws. X-ray micro-computer tomography analysis was additionally used to characterize porosity levels. Finally, scanning electron microscopy (SEM) and electron back-scatter diffraction (EBSD), as well as surface profilometry were combined to correlate specific process parameters and conditions with microstructural and surface effects.

Immunotherapy is now established as a potent therapeutic paradigm engendering antitumor immune response against a wide range of malignancies and other diseases by modulating the immune system either through the stimulation or suppression of immune components such as CD4+ T cells, CD8+ T cells, B cells, monocytes, macrophages, dendritic cells, and natural killer cells. By targeting several immune checkpoint inhibitors or blockers (e.g., PD-1, PD-L1, PD-L2, CTLA-4, LAG3, and TIM-3) expressed on the surface of immune cells, several monoclonal antibodies and polyclonal antibodies have been developed and already translated clinically. In addition, natural killer cell-based, dendritic cell-based, and CAR T cell therapies have been also shown to be promising and effective immunotherapeutic approaches. In particular, CAR T cell therapy has benefited from advancements in CRISPR-Cas9 genome editing technology, allowing the generation of several modified CAR T cells with enhanced antitumor immunity. However, the emerging SARS-CoV-2 infection could hijack a patient’s immune system by releasing pro-inflammatory interleukins and cytokines such as IL-1β, IL-2, IL-6, and IL-10, and IFN-γ and TNF-α, respectively, which can further promote neutrophil extravasation and the vasodilation of blood vessels. Despite the significant development of advanced immunotherapeutic technologies, after a certain period of treatment, cancer relapses due to the development of resistance to immunotherapy. Resistance may be primary (where tumor cells do not respond to the treatment), or secondary or acquired immune resistance (where tumor cells develop resistance gradually to ICIs therapy). In this context, this review aims to address the existing immunotherapeutic technologies against cancer and the resistance mechanisms against immunotherapeutic drugs, and explain the impact of COVID-19 on cancer treatment. In addition, we will discuss what will be the future implementation of these strategies against cancer drug resistance. Finally, we will emphasize the practical steps to lay the groundwork for enlightened policy for intervention and resource allocation to care for cancer patients.

Cold spray process (CSP) is a thermal spray technology in which coating (10–40 µm) is formed in the solid state by the impingement of power particles with supersonic velocity (200–1,200 m/s2) on coupon employing compressed gas jet, below the melting point of coating powder. It is commonly referred as cold gas dynamic spray, high velocity powder deposition, kinetic spray and kinetic energy metallisation process. Using CSP, various engineering materials (metals, polymers and ceramics) and its composites can be deposited. It is unique and promising approach for obtaining surface coating and offers various technological benefits over thermal spray as kinetic energy is employed for deposition rather than thermal energy. This offers great benefits in additive manufacturing (AM) to develop a component denser, low oxide coating free of tensile residual stresses, and undesired chemical reactions compared to conventional AM and coating techniques. Cold spray additive manufacturing (CSAM) is the powerful and emerging technique in the field of AM to develop engineering components with improved performance covering broad range of functionalities of surface, subsurface and interfaces. There are few flaws in this technique; however, extensive research work is going in CSAM and repairing of components to meet the real-time applications. The main objective of this review article is to summarise the history, effect of process parameters on surface coating, research and development in CSP along with its implementation in AM, component repairing and biomedical, antimicrobial and electrical applications. A discussion on future trends in CSAM is also provided at the end part of this article.

Fiber-reinforced composite materials are lightweight and can withstand heavy loading conditions. Reinforcement augments the strength of the composite material, which is assessed by its elastic modulus. An attempt is made to reinforce epoxy with Coir Fiber, Carbon Nanotube (CNT) and Fly-ash. Central Composite Design (CCD), a Response Surface Methodology (RSM) tool, which is a Design of Experiment (DOE) technique, is used to fabricate the experimental samples to study their tensile behavior. Analysis of variance (ANOVA) is employed to investigate the effect of reinforcement percentage of CNT, coir-fiber, and fly-ash on tensile behavior of composite. The ANOVA results follow the trend of the experimental values with a deviation of less than 10% in yield strength, tensile strength, and Young’s modulus. Two models (Artificial Neural Network and Multiple Linear Regression Model) originated resting on the regression equation to speculate the elastic modulus for various reinforcement parameters using the experimental data. The main objective is to optimize reinforcement parameters using both the models having a maximum elastic modulus of 2.602 GPa and 2.682 GPa, respectively, which is achieved by the teaching learning-based optimization technique. Furthermore, confirmation experiments validate the optimization process with an error of less than 4%.

Abstract Germline ATM gene variations result in phenotypic heterogeneity characterized by a variable degree of disease severity. We retrospectively collected clinical, genetic, and immunological data of 26 cases with A-T. Clinical manifestations included oculocutaneous telangiectasia (100%), ataxia (100%), fever, loose stools or infection (67%), cerebellar atrophy (50%), nystagmus (8%), dysarthria (15.38%), and visual impairment (8%). Genetic analysis confirmed ATM gene variations in 16 unrelated cases. The most common type of variation was stopgain variants (56%). Immunoglobulin profile indicated reduced IgA, IgG, and IgM in 94%, 50%, and 20% cases, respectively. T cell lymphopenia was observed in 80% of cases among those investigated. Unusual presentations included an EBV-associated smooth muscle tumour located in the liver in one case and Hyper IgM syndrome-like presentation in two cases. Increased immunosenescence was observed in T-cell subsets (CD4+CD57+ and CD8+CD57+). T-cell receptor excision circles (TRECs) were reduced in 3/8 (37.50%) cases.

Aim: To assess distress, insomnia, and psychosocial impact of SARS-CoV-2 outbreak on children with SLE and their caregivers. Methods: Patients with pSLE undergoing treatment in the Department of Pediatrics, PGIMER, Chandigarh, and their caregivers were enrolled. Questionnaires were sent to eligible patients and their parents through email or WhatsApp and telephonic interviews were conducted. Self-designed SLE-COVID-19 stress questionnaire; Peritraumatic Distress Inventory; Insomnia Severity Index, Positive and Negative Affect Schedule were used. Ethical approval was sought from Institutes Ethics Committee (IEC/2020/000583). Results: Telephonic connection was possible with 80 families (160 participants). Telephonic contact was possible with 80 families (160 participants); off these 61 children with pSLE (78.2%) and 55 caregivers (70.5%) responded to the questionnaire. Among participants, 23% patients, and 21.8% caregivers were severely stressed about SARS-CoV-2 infection; 78.7% patients and 80% caregivers had heard about hydroxychloroquine (HCQ) being used for the treatment of COVID-19; 52.7% caregivers exhibited moderate concern about shortage of HCQ; and 52.5% patients, and 43.6% caregivers were worried about side effects of HCQ. We found that 20 (32.8%) patients and 18 (32.7%) caregivers experienced significant distress. Majority of participants reported sleep disturbances. High positive affect scores were seen in 40 (65.5%) patients and 43 (78.2%) caregivers, low positive affect scores were noted in 21 (34.5%) patients and 12 (21.8%) caregivers. Conclusion: Patients with pSLE and their caregivers are at risk of psychosocial problems during the COVID-19 pandemic. Psychological interventions can be very helpful.

Abstract Natural fibre reinforced polymer composites are often a better substitute to man-made fibre reinforced polymer composites because of their wide availability, economical, recyclability and biodegradability. In this paper, natural fibre reinforced polymer composites are made using coir/ fly ash as reinforcements. Influences of polymer types, natural fibre (coir) length and wt. % of fly ash on tensile and flexural behaviors are investigated. General purpose resin, Vinyl ester and Isophthalic resin are the different polymers used in making samples. The samples are prepared according to Taguchi Design of Experiments using L9 orthogonal array. To explore the significance of participating factors such as polymer type, natural fibre (coir) length and wt. % of fly ash on the tensile and flexural behaviors, ANOVA is implemented. Regression equations reveal that the type of polymer influences tensile and flexural behaviors more than that of natural fibre (coir) length and wt. % of fly ash.

Background: Many urologists continue antibiotics after common urologic procedure beyond the timeframes recommended by professional guidelines. In this study, we sought to evaluate the association between postprocedural antibiotic use and patient outcomes. Methods: We identified all patients who underwent 1 of 3 urologic procedures (transurethral resection of bladder tumor [TURBT], transurethral resection of prostate [TURP], and ureteroscopy) within the Veterans’ Health Administration (VHA) between January 1, 2017, and June 30, 2021. A postprocedural antibiotic was any antibiotic potentially used for a urinary tract–related indication that was prescribed for administration after the day of the procedure. Outcomes were captured within 30 days of the procedure and included (1) return visits, defined as any emergency department or urgent care encounter or hospital readmission, and (2) Clostridium difficile infection (CDI), defined as a positive test for C. difficile and the prescription of an anti-CDI antibiotic. We used log-binomial models with risk adjustment to determine the association between postprocedural antibiotic use and outcomes. We constructed hospital-level observed-to-expected ratios for postprocedural antibiotic use, and we used these models to calculate the probability of each patient receiving postprocedural antibiotics. Results: Overall, we identified 74,629 patients; 98% were male; the mean age was 70 years (SD, 10). Among them, 50% underwent TURBT, 28% underwent TURP, and 23% underwent ureteroscopy. A postprocedural antibiotic was prescribed to 25,738 (35%) cases for a median duration of 3 days (IQR, 3–6). Return visits occurred in 13,489 patients (18%), and CDI occurred in 104 patients (0.1%). Patients exposed to postprocedural antibiotics had 16% more return visits (RR, 1.16; 95% CI, 1.13–1.20) and more than twice as much CDI (RR, 2.22; 95% CI, 1.51–3.26) than patients not exposed to postprocedural antibiotics. In log-binomial risk-adjusted analysis, the risk of return visits did not differ between the 2 groups (RR, 1.00; 95% CI, 0.97–1.04) but the risk of CDI was higher in patients who received post-procedural antibiotics (RR, 1.87; 95% CI, 1.00–3.51). Hospitals (n = 105) varied widely in their observed-to-expected ratios for prescribing postprocedural antibiotics, and the frequency of return visits was similar regardless of the frequency at which postprocedural antibiotics were prescribed (Table 1). Conclusions: Postprocedural antibiotics were prescribed beyond recommended intervals after more than one-third of common urologic procedures, with a large degree of variability across hospitals. The use of postprocedural antibiotics was not associated with fewer return visits but was associated with a nonsignificant increase in CDI risk. Efforts to reduce postprocedural antibiotics are needed.

Nickel aluminides have desirable properties for use in high-temperature applications. Nickel aluminides have certain desirable qualities, but for almost a decade in the 1990s, those benefits were overshadowed by the challenges of processing and machining at room temperature. Manufacturing improvements, increased knowledge of aluminide microstructure and deformation processes, and developments in micro-alloying have all contributed to the development of nickel aluminides. Key developments in nickel aluminides, such as their microstructure, alloy addition and alloy development, are given and discussed at length. Methods of production from the past, such as ingot metallurgy and investment casting and melting are addressed, and developments in powder metallurgy-based production methods are introduced. Finally, the difficulties of producing nickel aluminides and possible solutions are examined. Scientists are drawn to the new green composites because they may demonstrate qualities that are comparable to those of composites made of synthetic fibers due to concerns about environmental contamination. In this work, the potential for using the produced green composite in different buckling load-bearing structural applications is explored. The work on composite buckling characteristics is vital because one needs to know the composite’s structural stability since buckling leads to structural instability. The buckling properties of composite specimens with epoxy as the matrix and chemically treated cellulose microfibrils as reinforcements are examined numerically in this study when exposed to axial compressive stress. The numerical model is first created based on the finite element method model. Its validity is checked using ANSYS software by contrasting the critical buckling loads determined through research for three samples. The numerical findings acquired using the finite element method are then contrasted with those produced using the regression equation derived from the ANOVA. The utilization of the created green composite in different buckling load-bearing structural applications is investigated in this study. As a result of the green composite’s unaltered buckling properties compared to synthetic composites, it has the potential to replace numerous synthetic composites, improving environmental sustainability.

Background: In higher education, Teacher Empathy is a term that refers to the empathetic skills of teachers. Researchers in other disciplines have shown that Teacher Empathy reduces teacher burnout and improves teacher satisfaction and student performance. There is little research on Teacher Empathy within engineering education. Purpose: In our current study, we explored the potential changes in perception of Teacher Empathy among three engineering faculty members as they utilized empathetic actions while teaching second-year engineering courses. Understanding the experiences of faculty members will help us to implement and further explore Teacher Empathy in engineering education. Method: Teacher Action Research methodology helped us provide agency to our three participants and research with them instead of on them. We used a combination of values coding and simultaneous coding for data analysis. Findings: Improved confidence in their empathetic skills was observed for two participants as they showed positive evolution of their perception about Teacher Empathy. The other participant increased the number of empathetic approaches he used in his classroom. Conclusion: Reduced teacher burnout, improved teacher satisfaction, and better student performance were some of the major benefits of Teacher Empathy that emerged. The difference in the evolution of each participant's perception about Teacher Empathy indicated that the personality of a faculty member has an influence on implementation and success of Teacher Empathy.

Background: Street vendors spend relatively more time near roadways and are vulnerable to air pollution related health disorders. One of the most recently discovered monogenic disorder of immune dysregulation is COPA syndrome. COPA syndrome is an inherited autoimmune disorder caused by mutations in COPA gene. COPA gene encodes for α subunit of the COP1 protein, which is involved in the reverse vesicular protein transport from Golgi apparatus to the endoplasmic reticulum (ER). The inheritance pattern of COPA syndrome is autosomal dominant, and the patients typically present with interstitial lung disease with pulmonary hemorrhage and subsequently develop arthritis. Immunological features involve autoantibody formation, elevated expression of IL-1β and IL-6, and increase in the number of Th17 cells. Molecular pathophysiology of COPA syndrome is not clearly understood. However, it is known that accumulation of unfolded proteins in ER leads to ER stress, which is an indirect result of aberrant vesicular transport of proteins from Golgi apparatus to ER and defective cellular autophagy. ER stress induces inflammation and is responsible for pathogenesis of a large number of chronic inflammatory diseases. Unfolded protein response process responds to improperly folded proteins and defends against stress in ER to ensure the fidelity of the protein folding. It maintains the expression of stress-response genes and causes initiation of inflammatory signaling pathways essential for the innate immunity. Mutation in COPA gene associated with defective protein sorting to ER has unearthed a new primary immunodeficiency disease with a unique clinical phenotype. This review highlights the clinical and molecular aspects of COPA syndrome. ObjectivesVedolizumab (VDZ) and ustekinumab (UST) are second-line treatments in pediatric patients with ulcerative colitis (UC) refractory to antitumor necrosis factor (anti-TNF) therapy. Pediatric studies comparing the effectiveness of these medications are lacking. Using a registry from ImproveCareNow (ICN), a global research network in pediatric inflammatory bowel disease, we compared the effectiveness of UST and VDZ in anti-TNF refractory UC.MethodsWe performed a propensity-score weighted regression analysis to compare corticosteroid-free clinical remission (CFCR) at 6 months from starting second-line therapy. Sensitivity analyses tested the robustness of our findings to different ways of handling missing outcome data. Secondary analyses evaluated alternative proxies of response and infection risk.ResultsOur cohort included 262 patients on VDZ and 74 patients on UST. At baseline, the two groups differed on their mean pediatric UC activity index (PUCAI) (p = 0.03) but were otherwise similar. At Month 6, 28.3% of patients on VDZ and 25.8% of those on UST achieved CFCR (p = 0.76). Our primary model showed no difference in CFCR (odds ratio: 0.81; 95% confidence interval [CI]: 0.41-1.59) (p = 0.54). The time to biologic discontinuation was similar in both groups (hazard ratio: 1.26; 95% CI: 0.76-2.08) (p = 0.36), with the reference group being VDZ, and we found no differences in clinical response, growth parameters, hospitalizations, surgeries, infections, or malignancy risk. Sensitivity analyses supported these findings of similar effectiveness.ConclusionsUST and VDZ are similarly effective for inducing clinical remission in anti-TNF refractory UC in pediatric patients. Providers should consider safety, tolerability, cost, and comorbidities when deciding between these therapies.
