Abstract

Experiments on rotating annuli that are differentially heated in the radial direction have been largely contributing to a better understanding of baroclinic instabilities. This configuration creates waves at a laboratory scale that are related to atmospheric circulations. Pioneer studies in baroclinic tanks have shown that experiments with low aspect ratios are more suitable to reproduce small-scale inertia gravity waves, but these tanks have a larger free surface, which leads to higher interactions with its surrounding environment. Considering the heat transferred through the free surface, the present work investigates its impacts on the baroclinic instability using direct numerical simulations (DNS).

Abstract

Abstract The RGD (Arg-Gly-Asp)-binding integrins αvβ6 and αvβ8 are clinically validated cancer and fibrosis targets of considerable therapeutic importance. Compounds that can discriminate between homologous αvβ6 and αvβ8 and other RGD integrins, stabilize specific conformational states, and have high thermal stability could have considerable therapeutic utility. Existing small molecule and antibody inhibitors do not have all these properties, and hence new approaches are needed. Here we describe a generalized method for computationally designing RGD-containing miniproteins selective for a single RGD integrin heterodimer and conformational state. We design hyperstable, selective αvβ6 and αvβ8 inhibitors that bind with picomolar affinity. CryoEM structures of the designed inhibitor-integrin complexes are very close to the computational design models, and show that the inhibitors stabilize specific conformational states of the αvβ6 and the αvβ8 integrins. In a lung fibrosis mouse model, the αvβ6 inhibitor potently reduced fibrotic burden and improved overall lung mechanics, demonstrating the therapeutic potential of de novo designed integrin binding proteins with high selectivity.

Abstract

The present paper describes a combined experimental and high performance computing study of new specific behaviors of the Strato-Rotational Instability (SRI). The confrontation of low frequency Particle Image Velocimetry (PIV) experimental data with Direct Numerical Simulations reveal new non-linear phenomena and patterns not yet observed in the SRI, that can contribute for our understanding of astrophysical flows, specially regarding phenomena related to accretion disk theories. The experiment designed to investigate these SRI related phenomena consists of a classical Taylor-Couette (TC) system with two concentric cylinders that can rotate independently under stable density stratification due to temperature gradients imposed in the axial direction. Abstract

Tucked away in the Italian Alps, in the town of Oberbozen-Soprabolzano lies the villa that the family of Bronislaw Malinowski and his first wife, Elsie Masson, called home from 1922 to 1935. Yet Malinowski himself never wrote about South Tyrol or the Alps in general. This volume features a series of essays that explicitly ponder Malinowski’s intriguing influence on Alpine anthropology: Despite not having worked directly in or on the Alps, he nonetheless left anthropological traces through the works of others. The Malinowski Forum for Ethnography (MFEA) aims to uncover the ineffable presence in Alpine anthropology of Malinowski, a founder of modern social anthropology.

Abstract

Comparative studies of mountain areas have long been at the core of the discussion about the relations between nature and culture as well as on environmental and social change. This volume searches for ways to develop further critical comparative perspectives in the study of cultures in mountain areas by drawing inter- and transdisciplinary links amongst anthropology, geography, folklore studies, montology, and global history. Drawing on examples mostly from the Alps and the Andes, but also extending to the global mountains, the authors explore socioecological environments, historical and political processes, borderscapes, demographic dynamics, forms of domestic organization, rituals, religiosities, and human-non-humanrelations., Gli studi comparativi sulle aree montane sono da tempo al centro del dibattito sul rapporto tra natura e cultura e sui cambiamenti ecologici e sociali. Questo volume è alla ricerca di nuovi percorsi per sviluppare ulteriormente prospettive comparative critiche per lo studio delle culture delle aree montane, tracciando collegamenti inter- e transdisciplinari tra antropologia, geografia, studi di folklore, montologia e storia globale. 16. High-order Large Eddy Simulations of Confined Rotor-Stator Flows

Abstract

In many engineering and industrial applications, the investigation of rotating turbulent flow is of great interest. In rotor-stator cavities, the centrifugal and Coriolis forces have a strong influence on the turbulence by producing a secondary flow in the meridian plane composed of two thin boundary layers along the disks separated by a non-viscous geostrophic core. Most numerical simulations have been performed using RANS and URANS modelling, and very few investigations have been performed using LES. This paper reports on quantitative comparisons of two high-order LES methods to predict a turbulent rotor-stator flow at the rotational Reynolds number Re=400000. The classical dynamic Smagorinsky model for the subgrid-scale stress (Germano et al., Phys Fluids A 3(7):1760-1765, 1991) is compared to a spectral vanishing viscosity technique (S\'everac & Serre, J Comp Phys 226(2):1234-1255, 2007). Numerical results include both instantaneous data and postprocessed statistics. The results show that both LES methods are able to accurately describe the unsteady flow structures and to satisfactorily predict mean velocities as well as Reynolds stress tensor components. A slight advantage is given to the spectral SVV approach in terms of accuracy and CPU cost. The strong improvements obtained in the present results with respect to RANS results confirm that LES is the appropriate level of modelling for flows in which fully turbulent and transition regimes are involved.

Abstract

This article provides an anthropological appraisal of the demographic, social and climatic changes recently experienced by the Italian Alps and concentrates on the complex negotiations over the future of the Alps that involve local inhabitants, old and new, and external actors. It is suggested that the epistemological turn fostered by the notion of Anthropocene has added new dimensions to the ongoing processes of negotiation in Alpine settings and that the Covid-19 pandemic, while sharpening pre-existing gaps, is also bringing attention back to the social and welfare needs of marginal areas.

Abstract

Severely affected during the twentieth century by depopulation and neglect, the Alps also suffered an erosion of their cultural heritage. Since the beginning of the twenty-first century, however, population has begun to grow in many areas, mainly due to the settlement in the uplands of both “highlanders by choice” (largely youth dissatisfied with city life) and “highlanders by necessity” (economic migrants). In recent years, however, the Alps have been increasingly hosting also asylum-seekers and refugees (“highlanders by force”). This article concentrates on the Italian Alps and on the number and distribution of foreign migrants. It provides quantitative and qualitative data, also considering some specific local cases of immigration, and addresses the following questions: What are the effects of Alpine repopulation, and notably the impact of foreign inhabitants, on local cultures? Should repopulation be seen as an opportunity for social innovation and cultural creativity or rather as a threat to “indigenous” cultures? Is demographic discontinuity precluding cultural continuity? Will foreign immigration, especially if directed to demographically depleted areas, favour over time the emergence of new forms of identity and “tradition,” as a result of the re-invention of the territory? We argue that the “new peopling” of the Alps should not be considered a priori as a threat nor as an enrichment, and that the interactions and negotiations between the locals and the migrants should be studied in-depth and with attention to the local contexts.

The RGD (Arg-Gly-Asp)-binding integrins αvβ6 and αvβ8 are clinically validated cancer and fibrosis targets of considerable therapeutic importance. Compounds that can discriminate between the two closely related integrin proteins and other RGD integrins, stabilize specific conformational states, and have sufficient stability enabling tissue restricted administration could have considerable therapeutic utility. Existing small molecules and antibody inhibitors do not have all of these properties, and hence there is a need for new approaches. Here we describe a method for computationally designing hyperstable RGD-containing miniproteins that are highly selective for a single RGD integrin heterodimer and conformational state, and use this strategy to design inhibitors of αvβ6 and αvβ8 with high selectivity. The αvβ6 and αvβ8 inhibitors have picomolar affinities for their targets, and >1000-fold selectivity over other RGD integrins. CryoEM structures are within 0.6-0.7Å root-mean-square deviation (RMSD) to the computational design models; the designed αvβ6 inhibitor and native ligand stabilize the open conformation in contrast to the therapeutic anti-αvβ6 antibody BG00011 that stabilizes the bent-closed conformation and caused on-target toxicity in patients with lung fibrosis, and the αvβ8 inhibitor maintains the constitutively fixed extended-closed αvβ8 conformation. In a mouse model of bleomycin-induced lung fibrosis, the αvβ6 inhibitor potently reduced fibrotic burden and improved overall lung mechanics when delivered via oropharyngeal administration mimicking inhalation, demonstrating the therapeutic potential of de novo designed integrin binding proteins with high selectivity.

Abstract

Contemporary societies are coping in different ways with the declining strength of kinship and other effects of demographic transitions such as a shift from nuclear to “unclear” family forms and population ageing. One strategy consists in widening and reshaping existing ties through modes of relatedness that come under the heading of “fictive kinship”. Drawing on an ethnographic study of the transformation of the compadrazgo system in Quito (Ecuador), this article considers fictive kinship in comparative perspective, paying attention to its novel fashions but also to changes and continuities in its traditional forms.
