Abstract

Alzheimer's disease is recognized as a complex condition influenced by multiple factors, necessitating a similarly multifaceted approach to treatment. Ideally, interventions should prioritize averting the progression to dementia. Given the chronic nature of the disease, long-term management strategies are required. Within this framework, lifestyle modifications and dietary supplements emerge as appealing options due to their minimal toxicity, limited side effects, and cost-effectiveness. This study presents findings from a double-blind, placebo-controlled bicentric pilot clinical trial, demonstrating the significant cognitive preservation associated with genistein, a phytoestrogen found in soy and various other dietary sources, among individuals with prodromal Alzheimer's disease. Our prior investigation utilizing APP/PS1 mice elucidated the specific mechanisms through which genistein operates, including anti-amyloid-β, antioxidant, anti-inflammatory, and antiapoptotic effects. These findings underscore the potential of identifying bioactive compounds from dietary sources for the management of Alzheimer's disease. [ABSTRACT FROM AUTHOR]

Abstract

In previous studies, using transcriptomic analysis, we observed higher levels of aryl hydrocarbon receptor (AHR) gene expression in the peripheral blood cells of centenarians compared to octogenarians. This suggests the potential significance of this receptor in maintaining physiological balance and promoting healthy aging, possibly linked to its critical role in detoxifying xenobiotics. In our current study, we confirmed that AHR expression is indeed higher in centenarians. We employed C. elegans as a model known for its suitability in longevity studies to explore whether the AHR pathway has a significant impact on lifespan and healthspan. Our survival assays revealed that two different mutants of AHR-1 exhibited lower longevity. Additionally, we used a mouse model to examine whether supplementation with pomegranate extract modulates the expression of AHR pathway genes in the liver. Furthermore, we studied a nutritional strategy based on pomegranate extract administration to investigate its potential modulation of life- and healthspan in worms. [ABSTRACT FROM AUTHOR]

Abstract

BackgroundConsiderable variability exists in international prevalence and incidence estimates of dementia. The accuracy of estimates of dementia in the oldest-old and the controversial question of whether dementia incidence and prevalence decline at very old age will be crucial for better understanding the dynamics between survival to extreme old age and the occurrence and risk for various types of dementia and comorbidities. International Centenarian Consortium - Dementia (ICC-Dementia) seeks to harmonise centenarian and near-centenarian studies internationally to describe the cognitive and functional profiles of exceptionally old individuals, and ascertain the trajectories of decline and thereby the age-standardised prevalence and incidence of dementia in this population. The primary goal of the ICC-Dementia is to establish a large and thorough heterogeneous sample that has the power to answer epidemiological questions that small, separate studies cannot. A secondary aim is to examine cohort-specific effects and differential survivorship into very old age. We hope to lay the foundation for further investigation into risk and protective factors for dementia and healthy exceptional brain ageing in centenarians across diverse ethnoracial and sociocultural groups.MethodsStudies focusing on individuals aged ≥95 years (approximately the oldest 1 percentile for men, oldest 5th percentile for women), with a minimum sample of 80 individuals, including assessment of cognition and functional status, are invited to participate. There are currently seventeen member or potential member studies from Asia, Europe, the Americas, and Oceania. Initial attempts at harmonising key variables are in progress.DiscussionGeneral challenges facing large, international consortia like ICC-Dementia include timely and effective communication among member studies, ethical and practical issues relating to human subject studies and data sharing, and the challenges related to data harmonisation. A specific challenge for ICC-Dementia relates to the concept and definition of'abnormal' in this exceptional group of individuals who are rarely free of physical, sensory and/or cognitive impairments.

26. Contribution to the study of the electric transport in cuprate superconducting thin films: supercritical currents and paraconductivity

Abstract

In type-II superconductors, in the regime of very high applied current density, an abrupt transition is experimentally observed from the mixed state to a highly dissipative regime, probably the normal state. This provokes a high voltage jump in the Current-Voltage Characteristics (CVC) of the samples, at a current density J* larger than the critical current Jc. In this work we study Y-123 (YBCO) microbridges sputtered over SrTiO3 substrates. We measure their CVCs for different temperatures close to Tc, and with low applied magnetic field, from 0 to 1 T. We compare the dependences of Jc and J*, and find that some of the typical experimental results in this regime could be explained by only considering the effect of the current self-field. This suggests that the depairing of the supercarriers could be involved in the abrupt transition. We also study the influence of thermal effects, by using a simplified model to describe the thermal evolution of our samples. We find that the uniform self-heating can produce an abrupt thermal runaway, at high current density, which could explain the occurrence of J*, as well as its dependence on temperature and applied magnetic field. These results do not totally exclude the implication of other more intrinsic mechanisms (like Cooper pairs depairing, or a change in the regime of the vortex dynamics), but suggest that the uniform self-heating is more relevant than what was considered up to now. Abstract

We first argue that the collective behaviour of the Cooper pairs created by thermal fluctuations well above the superconducting transition temperature, Tc, is dominated by the uncertainty principle which, in particular, leads to a well-defined temperature, T^C, above which the superconducting coherence vanishes. On the grounds of the BCS approach, the corresponding reduced-temperature, ln(T^C/Tc), is estimated to be around 0.55, i.e., above T^C \approx 1.7Tc coherent Cooper pairs cannot exist. The implications of these proposals on the superfluid density are then examined using the Gaussian-Ginzburg-Landau approximation. Then we present new measurements of the thermal fluctuation effects on the electrical conductivity and on the magnetization in different low- and high-Tc superconductors with different dopings which are in excellent agreement with these proposals and that demonstrate the universality of ln(T^C/Tc)., Comment: LaTeX, 10 pages, 3 figures, as published in Europhysics Letters

Abstract

Reversible and sub-lethal stresses to the mitochondria elicit a program of compensatory responses that ultimately improve mitochondrial function, a conserved anti-aging mechanism termed mitohormesis. Here, we show that harmol, a member of the beta-carbolines family with anti-depressant properties, improves mitochondrial function and metabolic parameters, and extends healthspan. Treatment with harmol induces a transient mitochondrial depolarization, a strong mitophagy response, and the AMPK compensatory pathway both in cultured C2C12 myotubes and in male mouse liver, brown adipose tissue and muscle, even though harmol crosses poorly the blood-brain barrier. Mechanistically, simultaneous modulation of the targets of harmol monoamine-oxidase B and GABA-A receptor reproduces harmol-induced mitochondrial improvements. Diet-induced pre-diabetic male mice improve their glucose tolerance, liver steatosis and insulin sensitivity after treatment with harmol. Harmol or a combination of monoamine oxidase B and GABA-A receptor modulators extend the lifespan of hermaphrodite Caenorhabditis elegans or female Drosophila melanogaster. Finally, two-year-old male and female mice treated with harmol exhibit delayed frailty onset with improved glycemia, exercise performance and strength. Our results reveal that peripheral targeting of monoamine oxidase B and GABA-A receptor, common antidepressant targets, extends healthspan through mitohormesis.
