Abstract

This study was carried out in the Regional Park of San Rossore (Italy) where we found natural regeneration of Stone pine (Pinus piena L.) in an even-aged stand of pine. The objectives of our study were to characterize the forest structure of the Stone pine stand and to quantify the natural regeneration of pine. Our results show that natural regeneration of P. pinea in the Park of San Rossore is a reachable target, however an adequate management is needed. The results are discussed with the intention of providing knowledge to support management of Stone pine forests along the Tyrrhenian coast.

Abstract

Crop adaptation to climate change is in a part attributed to epigenetic mechanisms which are related to response to abiotic and biotic stresses. Although recent studies increased our knowledge on the nature of these mechanisms, epigenetics remains under-investigated and still poorly understood in many, especially non-model, plants, Epigenetic modifications are traditionally divided into two main groups, DNA methylation and histone modifications that lead to chromatin remodeling and the regulation of genome functioning. In this review, we outline the most recent and interesting findings on crop epigenetic responses to the environmental cues that are most relevant to climate change. In addition, we discuss a speculative point of view, in which we try to decipher the “epigenetic alphabet” that underlies crop adaptation mechanisms to climate change. The understanding of these mechanisms will pave the way to new strategies to design and implement the next generation of cultivars with a broad range of tolerance/resistance to stresses as well as balanced agronomic traits, with a limited loss of (epi)genetic variability.

23. Unexpected presence of Fagus orientalis complex in Italy as inferred from 45,000-year-old DNA pollen samples from Venice lagoon

Abstract Background Phylogeographic analyses on the Western Euroasiatic Fagus taxa (F. orientalis, F. sylvatica, F. taurica and F. moesiaca) is available, however, the subdivision of Fagus spp. is unresolved and there is no consensus on the phylogeny and on the identification (both with morphological than molecular markers) of Fagus Eurasiatic taxa. For the first time molecular analyses of ancient pollen, dated at least 45,000 years ago, were used in combination with the phylogeny analysis on current species, to identify the Fagus spp. present during the Last Interglacial period in Italy. In this work we aim at testing if the trnL-trnF chloroplast DNA (cpDNA) region, that has been previously proved efficient in discriminating different Quercus taxa, can be employed in distinguishing the Fagus species and in identifying the ancient pollen. Results 86 populations from 4 Western Euroasistic taxa were sampled, and sequenced for the trnL-trnF region to verify the efficiency of this cpDNA region in identifying the Fagus spp.. Furthermore, Fagus crenata (2 populations), Fagus grandifolia (2 populations), Fagus japonica, Fagus hayatae, Quercus species and Castanea species were analysed to better resolve the phylogenetic inference. Our results show that this cpDNA region harbour some informative sites that allow to infer relationships among the species within the Fagaceae family. In particular, few specific and fixed mutations were able to discriminate and identify all the different Fagus species. Considering a short fragment of 176 base pairs within the trnL intron, 2 transversions were found able in distinguishing the F. orientalis complex taxa (F. orientalis, F. taurica and F. moesiaca) from the remaining Fagus spp. (F. sylvatica, F. japonica, F. hayataea, F. crenata and F. grandifolia). This permits to analyse this fragment also in ancient samples, where DNA is usually highly degraded. The sequences data indicate that the DNA recovered from ancient pollen belongs to the F. orientalis complex since it displays the informative sites characteristic of this complex. Conclusion The ancient DNA sequences demonstrate for the first time that, in contrast to current knowledge based on palynological and macrofossil data, the F. orientalis complex was already present during the Tyrrhenian period in what is now the Venice lagoon (Italy). This is a new and important insight considering that nowadays West Europe is not the natural area of Fagus orientalis complex, and up to now nobody has hypothesized the presence during the Last Interglacial period of F. orientalis complex in Italy.

Abstract

Information on public attitudes towards the use of transgenic trees in forest plantations is important in the decision-making process and policy implementation for safe tree development, particularly at the EU level. In Europe, the use of transgenic forest trees is very limited and therefore such information is completely lacking. To address this issue within the FP0905 European COST Action on the Biosafety of Transgenic Forest Trees a pioneer cross-country pilot survey on public attitudes towards the use of transgenic forest trees was conducted using young population as a focus group. This was decided mainly because this focus group represents the future consumers, policy makers or developers. Specifically, the survey aimed to: i) assess the level of young people’s knowledge about transgenic forest trees, ii) identify issues of concern to them regarding the cultivation of transgenic forest trees and iii) explore whether they approve or disapprove of the use of transgenic forest trees in plantations. Purposive sampling was performed and university students of different disciplines were included in the research as sampling subjects. In total, 1868 completed questionnaires from 15 European and non-European countries were analyzed. The young educated people that took part in the survey appeared to approve of the use of transgenic forest trees in plantations and would be willing to buy forest transgenic products. The potential loss of biodiversity due to a risk of gene flow between transgenic and wild trees was seen as the safety issue of most concern when considering the commercial release of transgenic forest trees. However, a serious perceived lack of knowledge about potential benefits and risks of the cultivation of transgenic forest trees was recorded in most of the countries. K-means clustering was implemented on respondents’ positive responses to identify potential country patterns. No differences in patterns of public attitude towards the acceptance of the commercial growing of transgenic forest trees were observed between European and non-European countries. Abstract

We present BeechCOSTe52; a database of European beech (Fagus sylvatica) phenotypic measurements for several traits related to fitness measured in genetic trials planted across Europe. The dataset was compiled and harmonized during the COST-Action E52 (2006-2010), and subsequently cross-validated to ensure consistency of measurement data among trials and provenances. Phenotypic traits (height, diameter at breast height, basal diameter, mortality, phenology of spring bud burst and autumn-leaf discoloration) were recorded in 38 trial sites where 217 provenances covering the entire distribution of European beech were established in two consecutive series (1993/95 and 1996/98). The recorded data refer to 862,095 measurements of the same trees aged from 2 to 15 years old over multiple years. This dataset captures the considerable genetic and phenotypic intra-specific variation present in European beech and should be of interest to researchers from several disciplines including quantitative genetics, ecology, biogeography, macroecology, adaptive management of forests and bioeconomy.

FPS COST Action FP0905; International audience; This book provides up-to-date information on the environmental impact of transgenic trees on genetically modified tree (GMT) communication strategy. It is useful to public/private organisations as well as to private and public research bodies and universities worldwide since it reports on the global status of GMT research and policy. A high number of genetically modified trees (GMTs) with altered or novel characteristics have been produced in the last 15 years. However, their very low public acceptance is a basic problem in their commercialization. Breeders anticipate economic and ecological benefits, like reduced product costs and less pressure on native forests, while opponents fear risks, such as unintended spread of GMTs. But what is true? To answer this question, the COST Action FP0905 focused on key aspects related to GMTs: (a) biological characterization; (b) assessment of possible environmental impacts; (c) socio-economic implications and public acceptance/concerns; (d) providing science-based information to communicate with the public.
