Abstract

The role of the thyroid hormone in calcium metabolism in developing rat myocytes was investigated by inducing either hypo- or hyper-thyroidism in neonatal rats early in the postnatal period and monitoring the expression of mRNAs and proteins responsible for the transport of calcium in myocytes. Results reveal that thyroid hormone is necessary in the inhibition of the expression of the sodium/calcium exchanger and the induction of the expression of the sarcoplasmic reticulum calcium ATPase.

Abstract

Dietary intake of medium-chain fatty acid (MCFA) affects the changes caused by etomoxir in heart muscle. Carnitine palmitoyltransferase-1 (CPT-1), situated on outer mitochondrial membrane, is inhibited by etomoxir. Uptake of long-chain fatty acid (LCFA) into mitochondria and its oxidation are related to CPT-1 activity. Malonyl-coenzyme A controls the enzyme. A study is conducted on rats to know effects of diet on heart.

Abstract

Analysis of the influence of a different substrate utility for cardiac sarcoplasmic reticulum (SR) Ca2+ transport in normotensive rats by enantiomeric etomoxir treatment indicates that etomoxir suppresses fatty acid synthesis and mitochondrial carnitine palmitoyltransferase-1 (CPT-1) in the rat. Differential increases of 13% and 38% were seen for Ca2+ uptake rates by right and left ventricular homogenates. Phosphoenzyme levels of the Ca2+ pump shows changes while tropomin I, and phosphorylated phospholamban remain constant after etomoxir treatment. Thus, the metabolic effects of etomoxir are more profound on relaxation- and contraction-mediating proteins of the left ventricle.

Abstract

Abstract Background Calcium (Ca2+) handling proteins are known to play a pivotal role in the pathophysiology of cardiomyopathy. However little is known about early changes in the diabetic heart and the impact of insulin treatment (Ins). Methods Zucker Diabetic Fatty rats treated with or without insulin (ZDF ± Ins, n = 13) and lean littermates (controls, n = 7) were sacrificed at the age of 19 weeks. ZDF + Ins (n = 6) were treated with insulin for the last 6 weeks of life. Gene expression of Ca2+ ATPase in the cardiac sarcoplasmatic reticulum (SERCA2a, further abbreviated as SERCA) and phospholamban (PLB) were determined by northern blotting. Ca2+ transport of the sarcoplasmatic reticulum (SR) was assessed by oxalate-facilitated 45Ca-uptake in left ventricular homogenates. In addition, isolated neonatal cardiomyocytes were stimulated in cell culture with insulin, glucose or triiodthyronine (T3, positive control). mRNA expression of SERCA and PLB were measured by Taqman PCR. Furthermore, effects of insulin treatment on force of contraction and relaxation were evaluated by cardiomyocytes grown in a three-dimensional collagen matrix (engineered heart tissue, EHT) stimulated for 5 days by insulin. By western blot phosphorylations status of Akt was determed and the influence of wortmannin. Results SERCA levels increased in both ZDF and ZDF + Ins compared to control (control 100 ± 6.2 vs. ZDF 152 ± 26.6* vs. ZDF + Ins 212 ± 18.5*# % of control, *p Conclusion The current data demonstrate that early type 2 diabetes is associated with an increase in the SERCA/PLB ratio and that insulin directly stimulates SERCA expression and relaxation velocity. These results underline the important role of insulin and calcium handling proteins in the cardiac adaptation process of type 2 diabetes mellitus contributing to cardiac remodeling and show the important role of PI3-kinase-Akt-SERCA2a signaling cascade.

International audience; Clinical and experimental studies indicate that the progression of renal disease is faster in males than females. These observations are corroborated by a sexual dimorphism observed in the polygenetic Munich Wistar Frömter (MWF) rat model. The age dependent spontaneous progression of increased urinary albumin excretion (UAE) in male MWF is influenced by multiple quantitative trait loci (QTL). In contrast, female MWF develop only a slight increase in UAE, while the role of genetic factors for this phenotype is unknown. Abstract

Failing cardiac hypertrophy is associated with an inadequate sarcoplasmic reticulum (SR) function. The hypothesis was examined that pressure overloaded hearts fail to increase SR Ca(2+) uptake rate proportionally to the hypertrophy and that carnitine palmitoyltransferase-1 inhibition by etomoxir ((+/-)-ethyl 2[6(4-chlorophenoxy)hexyl] oxirane-2-carboxylate) can counteract this process. Severe left ventricular pressure overload was induced in rats by constricting the ascending aorta for 8, 10, 14 and 28 weeks leading to cardiac hypertrophy (+62 - +103% of sham-operated rats) and pulmonary congestion. Homogenate oxalate-facilitated SR Ca(2+) uptake rate g wet wt(-1) was reduced (P0.05) by 29.9+/-1.8% irrespective of phospholamban phosphorylation (in the presence of catalytic subunit of protein kinase A) and inhibition of SR Ca(2+) release channel by ruthenium red. SERCA2 protein level was reduced (P0.05) by 30.4+/-0.8%. SR Ca(2+) uptake rate was inversely correlated (P0.05) with left ventricular weight but was not affected by the occurrence of pulmonary congestion. Because SR Ca(2+) uptake rate of whole ventricles was not reduced, a hypertrophy proportional dilution of SR Ca(2+) uptake has to be inferred which precedes pulmonary congestion. Treatment with etomoxir (15 mg kg body wt(-1) day(-1) for 10 weeks) did not affect left ventricular weight but decreased (P:0.05) the right ventricular hypertrophy related to pulmonary congestion. In parallel, SR Ca(2+) uptake rate of left ventricle and myosin isozyme V(1) were increased (P0.05). Etomoxir represents a candidate approach for prevention of heart failure by inducing a hypertrophy proportional increase in SR Ca(2+) uptake rate.
